Enhanced Occlusiveness of Nanostructured Lipid Carrier (NLC)-based Carbogel as a Skin Moisturizing Vehicle

  • In order to develop a topical preparation which has a high occlusive property with skin moisturization, nano-structured lipid carrier (NLC) systems along with solid lipid nanoparticle (SLN) were designed. Various NLC dispersions were successfully formulated with Compritol 888 ATO as a solid lipid, Labrafil M 1944 CS as an oil, and Tween 80 as a surfactant. The increase of oil content (5 to 50%) led to the decrease in the occlusion factor in the order of SLN > NLC-5 > NLC-15 = NLC-30 > NLC-50. Particle size of lipid particulates was in the range of 100 to 160 nm. NLC-based carbogels were prepared by the employment of humectants such as urea, glycerin, and Tinocare GL to carbomer gel. NLC-30 gel formulations containing 4 or 8 % of lipid particles showed improved occlusive effect in vitro, compared to NLC-free gel base. Even though NLC-free gel base revealed comparable occlusion effect by itself, the occlusion factor of 4 % NLC-30 gel was about 2-fold higher than that of NLC-free gel base.

Skin Brightening Effect of Gel Essence Containing High Concentrated-Marine Minerals in Silicon Copolyol Network Film in vivo (고농도의 미네랄을 실리콘 네트워크 필름에 포집한 젤 에센스 제형의 피부 밝기 개선 효과)

  • Since minerals are in ionic forms in aqueous phase (like deep sea water), it is very hard to incorporate concentrated minerals in gel like cosmetic formulations which are generally composed of carbomer or acrylates polymer. In this study, we invented a salt-resistant network formulation made of silicon copolyol. It was found that the network formulation traps minerals (especially from marine) in a mesh-like silicone copolyol structure. Since the mineral-containing formulation makes a very thin film on the skin, minerals could be easily absorbed into stratum corneum. In 6 weeks in vivo test, it was found that the novel mineral formulation increased the amounts of amino acids, major components of natural moisturizing factors (NMFs), as well as levels of minerals. It was also found that the moisture contents and the brightness of the skin increased.

Anti-inflammatory Effects of Hydrogels Containing Piroxicam and Hydrolyzed Products of Scutellariae Radix on Collagen-induced Arthritis (콜라겐으로 유발된 관절염에 대한 피록시캄 및 황금 가수분해물 복합 히드로겔의 항염 효과)

  • In order to access the suppressive effects of piroxicam (PX) and hydrolyzed products of Scutellariae Radix (PSH) on arthritis, we investigated whether PSH gel could suppress the progression of collagen-induced arthritis. PX, one of nonsteroidal anti-inflammatory drugs has been used in the systemic and topical treatment in a variety of inflammatory conditions. Scutellariae Radix, one of the herbal medicines, was used for the purpose of anti-inflammatory and anti-bacterials. For the purpose of transdermal absorption of the hydrogel preparations, two classes of hydrogels (PX, PSH) were formulated with carbomer 940, diethylene glycol monoethyl ether, polyethylene glycol-8-glyceryl caprylate/caprate and triethanolamine. In carrageenan-induced edema in rat hind paws, inhibition of foot swelling was more increased in PSH than PX hydrogel. Rheumatoid factors including serum IgG, IgM and collagen specific antibody were present much lower in PSH gel treated mice than control. Histological examination revealed that PSH hydrogel inhibited infiltration of inflammatory cells into affected paw joint, compared with control. The PSH hydrogel would be a suitable preparation to increase transdermal treatment for anti-inflammatory effects on collagen-induced arthritis.

Survey on the Use of Hand Sanitizer and Component Analysis (손소독제 사용 실태 조사 및 성분 분석)

  • Objectives: Hand sanitizer is made with ethyl alcohol as the main ingredient. Problems related to the use of hand sanitizers and cases of harm caused by the use of hand sanitizers are occurring. This study investigated the usage behavior and recognition level of people using hand sanitizer and identified the chemical components listed in the component label of hand sanitizer. In addition, the methanol and isopropanol contained in hand sanitizer were quantified using HS-GC-MSD. Methods: The investigation of the behavior and recognition of hand sanitizer usage was conducted through a survey of 143 college students and adults. The components marked on 34 types of hand sanitizers were investigated, and methanol and isopropanol concentrations were analyzed using the HS-GC-MSD method. Results: According to the survey, 57% of respondents use hand sanitizers two to three times per day, 92.3% of them do so when in public places and 41.3% of them do so at home. Ethanol, purified water, carbomer, glycerin, and triethanolamine were the ingredients listed in the hand sanitizer. Among the 34 samples, methanol and isopropyl alcohol were detected in 33 samples, the concentration range for methanol was ND-567 ppm, and the concentration range of isopropyl alcohol was ND-2121 ppm. Conclusion: The results of this study have shown that hand sanitizers are being used constantly every day, and methanol, which is not included in the marked content, was detected in a significant concentration compared to wet tissue. It has been found that maintenance of hand sanitizer manufacturing standards and training on how to use them are needed.

A Study on Preparation of Water in Oil in Water (W1/O/W2) Emulsion Containing Titrated Extract of Centella asiatica (센텔라 아시아티카 정량추출물을 함유한 Water in Oil in Water (W1/O/W2) 에멀젼 제조에 관한 연구)

  • Titrated extract of Centella asiatica (TECA), which is poorly soluble in water is well known for wound healing and anti wrinkle agent. This study was conducted to find the optimum condition for the preparation of water in oil in water ($W_1/O/W_2$) emulsion containing TECA. Solubility of TECA were measured by UV spectrophotometer. 2.55 g of TECA was dissolved in solution composed of dipropylene glycol (40.0 g), ethanol (20.0 g), and water (10.0 g). Factors affecting stability of the emulsions ($W_1/O$, $W_1/O/W_2$) was investigated. The optimum conditions for the preparation of $W_1/O$ emulsion was composed of dipropylene glycol : ethanol : water : TECA in a weight ratio of 40.0 : 20.0 : 10.0 : 2.5 for water phase and squalane : cetyl PEG/PPG-10/1 dimethicone : cetearyl alcohol in a weight ratio of 22.5 : 4.0 : 2.5 for oil phase. The optimum conditions for the preparation of $W_1/O/W_2$ multiple emulsion was composed of water : $W_1/O$ emulsion : polysorbate 80 : carbomer : triethanolamine in a weight ratio of 55.8 : 40.0 : 4.0 : 0.1 : 0.1.

A Study of the Transdermal Permeation of Lotion Formulations Containing Angelica gigas Nakai Extracts in Franz Diffusion Cells (Franz diffusion cell을 이용한 참당귀 추출물 함유한 로션제형의 피부 투과 연구)

  • Angelica gigas Nakai (AGN) has been used in Korean herbal medicine for various pharmacological activities, such as to create antioxidant and skin whitening effects. Decursin and decursinol angelate of AGN extracts can be used as potential active drugs and cosmetic ingredients. This study investigated the possibility of topical delivery of AGN extracts using a manufactured emulsion system. Lotion was formulated by using Tefose® and paraffin for the oil phase, Kolliphor RH 40 for the surfactant and solubilizing agent-which showed high solubility in water (0.82 mg/ml)-and a water phase with a carbomer. In vitro skin permeation of decursin and decursinol angelate was determined using a Strat-M® membrane in Franz diffusion cells. Lotion samples as the experimental group (248.08±19.72 ug/cm2) significantly increased the permeation of decursin and decursinol angelate for up to 24 hr compared to the control group (119.18±19.23 ug/cm2). The permeability was also characterized by the flux (penetration rates) and Kp (permeability coefficient) values. The experimental group (17.20±1.23 ug/h/cm2 and 5.73±1.39 cm/h*10-3) had higher flux and Kp than the control group (8.22±1.24 ug/h/cm2 and 2.74±0.51 cm/h*10-3). Lotion with decursin and decursinol angelate of AGN extracts could be used for the topical application of drug and cosmetic products.
