Variation of Nutritional and Antioxidant Characteristics of Extract of Lycium barbarum produced by using Different Extraction Processes

  • Lycium barbarum extract has a high potential to be developed as a health functional food due to the various health-promoting effects of Lycium barbarum. This study analyzed changes in nutritional and functional components depending on the extraction solvent (purified water and a mixture of purified water and alcohol) and the condition of the sample. The nutritional components (carbohydrates, protein, fat, ash), organic acids, amino acids, total phenolic compounds, and total flavonoids of the extract produced during the extraction process were analyzed. The nutritional composition and functional substances of the extracts showed some differences depending on the type of solvent and the condition of the sample. The amounts of crude protein (7.61%), crude fat (1.63%), carbohydrate (90.22%), and ash (0.54%) of dried Lycium barbarum extract using purified water as a solvent were similar to those of the powder sample extract. The highest content of citric acid was 4.31 mg/mL, similar to the case of acetic acid, when the powder sample used a mixture of purified water and alcohol as a solvent. The highest amino acid content was 357.39 mg/mL when the powder sample was mixed with purified water and alcohol as a solvent. The total amount of phenolic compounds was 686.16 g/L when the powder sample was extracted with a mixture of purified water and alcohol as a solvent. The highest total flavonoid content was 111.32 g/L when the powder sample was extracted with a mixture of purified water and alcohol as a solvent.

한국산(韓國産) Comfrey의 식품학적(食品學的) 가치(價値)에 관(關)한 연구(硏究) (Study on Food Scientific Value of Korean Comfrey)

  • General components of comfrey collected during the period of July-September 1978 in Pusan area were investigated with a view to determine its possible value as a foodstuff by means of titrimetry, chromometric analysis and fluorometric analysis. As a result, the following data were obtained. 1) The contents of moisture, crude lipid, crude protein and carbohydrates of the comfrey were similar to those of other green vegetables such as spinach, crowndaisy, lettuce, leek and mallow. 2) The inorganic Ca, P and Fe of the comfrey were fairly greater in contents than those of other green vegetables. Compared with spinach, for instance, there were 4 times as much Ca and about 2 times as much Fe in it, respectively. 3) The contents of vitamins of the comfrey were also found in an amount much greater than those of other green vegetables. V-A of the comfrey, for instance, were 6 times greater in content than that of lettuce, and about 2 times greater than that of crowndaisy, leek and mallow, respectively. It also showed a greater content of V-A than even that of spinach, known as a vegetable most rich in V-A. V-B of the comfrey showed a little lower level of content than that of spinach but showed about the same level of content as that of leek and lettuce. V-B of the comfrey was 6 to 8 times greater in content than that of lettuce and leek. The content of niacin of the comfrey was also found much greater than those of other foodttuffs. V-C of the comfrey compared 3.5 times greater with that of lettuce, 2 times greater with that of mallow, and generally a higher level with those of other green vegetables. The above findings seem to firmly establish the food scientific value of the comfrey. Furthermore, the vegetable is very prolific and grows easily. If the general public is made aware of these facts, the vegetable is thought to contribute not a little to their dietary life as a nutritious foodstuff.

Ecophysiological characteristics of Rosa rugosa under different environmental factors

  • Background: Ecophysiological characteristics of Rosa rugosa were analyzed under different environmental factors from May to October 2022. Photosynthesis, chlorophyll fluorescence, chlorophyll content, leaf water content (LWC), osmolality, carbohydrate content, and total ion content were measured to compare the physiological characteristics of R. rugosa at two study sites (i.e., in large pots and in the Goraebul coastal sand dune area). Results: When R. rugosa was exposed to high temperatures, photosynthetic parameters including net photosynthetic rate (PN) and stomatal conductance (gs) in both experiment areas declined. In addition, severe photoinhibition occurs when R. rugosa is continuously exposed to high photosynthetically active radiation (PAR), and because of this, relatively low Y(II) (i.e., the quantum yield of photochemical energy conversion in photosystem II [PSII]) and high Y(NO) (i.e., the quantum yield of non-regulated, non-photochemical energy loss in PSII) in the R. rugosa of the pot were observed. As the high Y(NPQ) (i.e., the quantum yield of regulated non-photochemical energy loss in PSII) of R. rugosa in the coastal sand dune, they dissipated the excessed photon energy through the non-photochemical quenching (NPQ) mechanism when they were exposed to relatively low PAR and low temperature. Rosa rugosa in the coastal sand dune has higher chlorophyll a and carotenoid content. The high chlorophyll a + b and low chlorophyll a/b ratios seemed to optimize light absorption in response to low PAR. High carotenoid content played an important role in NPQ. As a part of the osmotic regulation in response to low LWCs, R. rugosa exposed to high temperatures and continuously high PAR used soluble carbohydrates and ions to maintain high osmolality. Conclusions: We found that Fv/Fm was lower in the potted plants than in the coastal sand dune plants, indicating the vulnerability of R. rugosa to high temperatures and PAR levels. We expect that the suitable habitat range for R. rugosa will shrink and move to north under climate change conditions.

The Effects of Different Moisture Content and Ensiling Time on Silo Degradation of Structural Carbohydrate of Orchardgrass

  • This study determined the influence of moisture, ensiling time and their interactions on the losses of hemicellulose and cellulose during ensiling of orchardgrass. Orchardgrass containing 80 (HM), 70 (MM) and 55% (LM) moisture was ensiled in 3 laboratory silos of 500 ml capacity for 3, 7, 21 and 91 days. The dry matter (DM), water-soluble carbohydrates (WSC), hemicellulose and cellulose contents of the ensiled orchardgrass was lowered than that of the untreated grass regardless of moisture content. Ensiling orchardgrass for 91 days (d) decreased (p<0.01) hemicellulose contents from 19 to 15%, 20 to 15% and 18 to 12% and cellulose from 31 to 29%, 29 to 26% and 27 to 26% for LM, MM and HM silage, respectively. Results from fermentation of LM and MM silages were within acceptable guidelines except for butyric acid and ammonia after 3 weeks of ensiling of MM which appeared to be lower than ideal. The results of the fermentation of HM silages were poor showing higher concentration of acetic, propionic and butyric acids and traces of isovaleric, valeric and caproic acids with ammonia at all stage of time. While the DM losses from LM and MM silages over the ensiling period were acceptable, that for HM silage increased to 13% after 91 d ensiling, confirming a poor fermentation process occurred. The greatest WSC losses occurred within 7 d of ensiling and the lowest losses occurred after 3 weeks of ensiling. Except in HM silage, the hemicellulose and cellulose losses were highest (p<0.01) in the first 3 weeks of ensiling. Hemicellulose losses were between 19 and 22% and 4.2 and 5.9% up to 3 weeks and after 3 weeks of ensiling LM and MM silages, respectively. Cellulose losses were small. In contrast, hemicellulose losses after 3 weeks of ensiling of HM silage was about 50% higher than over the first 3 weeks possibly due to clostridial type fermentation. The results showed that increasing ensiling time of high moisture orchardgrass would result in the excessive losses of DM, WSC, hemicellulose and cellulose in the silage.

Bacillus subtilis를 이용한 국내산 신이대 잎 발효에 따른 영양성분, 폴리페놀, 항산화능 변화 (Changes of Nutritional Components, Polyphenols, and Antioxidant Activities of Domestic Bamboo Tree (Sasa coreana Nakai) Leaves Fermented with Bacillus subtilis)

  • Bamboo leaf has many nutritional and bioactive compounds such as polyphenols, and it can be used for various foods application. Fermentation is one of appropriate processes that can affect the nutrition, taste and flavor, and antioxidant activities of foods. In this study, a representative domestic bamboo tree (Sasa coreana Nakai) leaves were fermented by Bacillus subtilis KCCM 11315 and the changes of carbohydrates, minerals, amino acids, and phenolic compounds and antioxidant activities were investigated before and after fermentation. During the fermentation period, firstly, the leaf was softened and turned to brown. The constituent carbohydrates were slightly increased from $432.09{\pm}5.38mg/g$ to $458.42{\pm}7.39mg/g$, and free sugars decreased by 95% from $28.12{\pm}2.03mg/g$ to $1.4{\pm}0.14mg/g$. Mineral was $20987.5{\pm}345.1{\mu}g/mL$, which was slightly increased after the fermentation compared to $20804.1{\pm}364.6{\mu}g/mL$ before that. The total amino acids were increased to $73881.94{\pm}137.59mg/100g$ compared to $58464.51{\pm}109.12mg/100g$ before fermentation, and free amino acids decreased by more than 85% from $32782.67{\pm}92.49mg/100g$ to $4827.83{\pm}19.76mg/100g$. Total polyphenols content (TPC) increased from $25.51{\pm}1.04GAE^*mg/g$ to $35.34{\pm}0.91GAE^*mg/g$, and the total flavonoid content (TFC) increased to $80.57{\pm}0.22QE^*mg/g$ compared to $69.64{\pm}0.26QE^*mg/g$. The total catechin content (TCC) of TFC was decreased from $69.64{\pm}0.94mg/L$ to $58.23{\pm}0.76mg/L$. The DPPH radical $IC_{50}$ of bamboo leaves decreased to $2624.85{\pm}37.03{\mu}g/mL$ and the ABTS radical $IC_{50}$ of $187.26{\pm}4.78{\mu}g/mL$ was increased after the fermentation. These results could be used as essential nutritional data before developing processed food products using the bamboo leaf.

탄수화물이 Aspergillus, Rhizopus, Candida 세포의 인지질 합성과 지방산 조성에 미치는 영향 (Effects of Carbohydrates on the Biosynthesis of Phospholipids and Fatty Acid Composition in Aspergillus, Rhizopus and Candida Cells)

  • 탄수화물(포도당, 서당, raffinose)을 각각 처리한 배지에서 배양한 진균류(Aspergillus phoenicis, Rhizopus acidus, Candida albicans)의 세포에서 일어나는 지질대사 특히 인지질 및 지방산 조성에 미치는 영향을 분석하였다. 서당은 Aspergillus와 Candida의 생장에, 포도당은 Rhizopus의 생장에 최적의 당이었다. 포도당은 Aspergillus와 Rhizopus total lipid 합성에, Raffinose는 Candida total lipid 합성에 가장 유용한 것으로 조사되었다. 포도당 처리는 Aspergillus의 Pl를 73.7%, Rhizopus의 PC를 292% 증가시켰다. 서당 처리는 Aspergillus의 Pl를 112%, Rhizopus와 Candida의 PC의 함량을 77.7%와 71.8%의 촉진 현상을 보여 주었다. Raffinose 처리는 Aspergillus에서 PC의 함량을 29.5% 증가시컸고, PE의 함량은 Rhizopus에서 50.9%, Candida에서 49.1% 촉진하였다. Aspergillus와 Candida의 당 처리구에서 PC와 PE를 구성하는 주요 지방산은 주로 oleic acid와 linoleic acid이었다. 인지질을 구성하는 주요 지방산으로 Aspergillus에서는 palmitic acid와 linoleic acid, Rhizopus에서는 palmitic acid, Candida에서는 palmitic acid, oleic acid, linoleic acid로 조사되었다. 이들 3진균류가 당 처리시 인지질 합성에 공통으로 이용하는 주요 지방산은 palmitic acid로 분석되었다.

  • This research is to establish the basic data of the nutritive value and improve our diet. In the part of the leaf and stem of the wild and cultivated horned rampion (Phyteuma japonicum Mig), the components such as chemical compositon, vitamin C, free sugar, mineral, nucleotide and its related compounds, composition and free amino acid were analyzed one after another. Content of the crude lipids and proteins was determined much higher in its wildness than in its cultivated horned rampion; while, that of carbohydrates was higher in the former than in the latter. The content of vitamin C was retained higher in the leaf than in the stem horned rampion. And the content of calcium among the detected minerals was outstanding in all of the samples collected, and potassium and magnesium was the next ones in its order. The main components of free sugars in both the wild and cultivated horned rampion were glucose and fructose, and their content was higher in the stem than in the leaf. Nucleotide and its related compounds were identified with 5 kinds of nucieotides such as CMP, UMP, IMP, AMP and hypoxanthine (Hx), and the content of Hx and AMP was the highest I the wild and cultivated samples, respectively. In the composition amino acid of the wild horned rampion, glutamic acid, aspartic acid and phenylalanine was outstandingly abundant; while, such amino acid as methinone and proline was small and besides cysteine couldn't be detected in the stem. Total amounts of composition amino acid in the leaf was 2118.0 and 1120.1mg% in the wild and cultivated sample, respectively. In the free amino acid of horned rampion, the total amount ranged from 8.5 to 50.1mg.%, which were lower level than that of composition amino acid. But the number of free amino acid was 29 kinds, which was bigger in its number than that of composition amino acid detected 17 kinds.

  • 팽화 처리온도를 각각 $140{\sim}220^{\circ}C$로 한 한방차 제품의 성분변화를 분석한 결과는 다음과 같다. 처리온도의 상승에 따라 일부 탄화가 발생하며 조회분 함량이 상대적으로 상승하는 소폭의 변화가 있었고, 조단백질 및 조지방 함량은 거의 변화가 없는 것으로 나타났으며 수분함량은 감소하였다. 한방차의 고형분 용출률은 0.18~0.27%(w/w)로 나타내었는데, 팽화온도가 상승할수록 증가하였다. 고형분의 용출은 온도가 화학적 변화보다 물리적 변화에 의해 식품의 원재료 성분인 탄수화물, 단백질, 지방 등이 천연 상태에서 상호가교 결합이 물리적인 힘으로 어느 정도 파괴되어 성분의 용출이 용이해지기 때문인 것으로 생각된다. 벤조피렌 함량은 0.18~0.24ppb로 처리온도, 원재료에 따라 B(${\alpha}$)P 함량에 차이가 발생한 것으로 나타났다.

추파유채 ( Brassica napus L. ) 생육시기에 따른 비구조성 탄수화물 함량의 변화 ( Changes in the Non-Structural Carbohybrate Content during Growth Period in Forage Raps ( Brassica napus L. ) )

  • The objective of this study is to obtain the basic data for investigating the potentiality of continuous utilization (first cutting in the late fall and regrowth yield in next spring) of forage rape seeded in fall. Non-structural carbohydrates(NSC) in leaves and roots of forage rape seeded on Oct. 3 were analyzed during a growth period. The greatest change in NSC content was observed between the wintering and the early spring period. The total content of soluble sugar in leaves and roots highly increased from the late fall(Nov. 7) to the wintering period (Feb. 4), and then rapidly decreased on the early regrow^ period(Mar. 31). The contents of fructose and glucose were relatively lower, and their quantitative change also was smaller than those of other sugars through entire growth period. The highest accumulation of sucrose occured hum the late fall to wintering period, and then greatly decreased in the early regrowth period. Sucrose content in roots was 2.3 times higher than that of leaves in wintering period. Starch was the largest pool of NSC and its content in leaves and roots showed a similar pattern with that of sucrose through entire growth period. Starch contents in leaves and roots were 38mg and 189mgl gDM in the late fall(Nov. 7), 187 and 497mg/gDM(Feb. 4) in the wintering period and 69 and 79mglgDM(Mar. 28) in the early regrowth period, respectively. The results clearly showed that the main reserve forms of NSC are starch and sucrose, and that they are hlghly stored in roots in overwintering forage rape.

Biochemical Characterization and Genetic Diversity of Pongamia pinnata (L.) Pierre in Eastern India

  • Biochemical characteristics of 24 Pongamia pinnata genotypes (candidate plus trees) from three agroclimatic zones were estimated and molecular characterization through RAPD markers was done. Various biochemical characters viz. seed oil, total carbohydrates, protein, acid value and Iodine number recorded significant variation among different genotypes. The highest seed oil content was 41.87% while seeds of 14 genotypes recorded above average (32.11%) for the trait. Seed oil and protein content exhibited a significant positive correlation and moderate heritability. Out of the initially selected twenty-five random primers, twenty-two RAPD primers were found to be highly reproducible and produced a total of 183 loci of which 147 (80.32%) loci were polymorphic. Percentage of polymorphism varied from 44% to 100% with an average of 80.62%. High level of genetic variation was found among different genotypes of P. pinnata. Both molecular and oil content (biochemical) markers appeared useful in analyzing the extent of genetic diversity in Pongamia and the result of these analyses will help to better understand the genetic diversity and relationship among populations. Overall, the Pongamia genotypes included in the study showed a correlation with their geographical origins such that genotypes from the same region tend to have higher genetic similarity as compared to those from different regions. However, in UPGMA based Nei's analysis, some genotypes were found not to be grouped based on geographical origins possibly due to the exchange of germplasm over time between farmers across the regions. The results from oil content analyses showed that several genotypes in 'Central and Western Plateau' agroclimatic zone of Jharkhand displayed a good potential for high oil content. The study provides insight about P. pinnata populations in Jharkhand (India) and constitutes a set of useful background information that can be used as a basis for future breeding strategy and improvement of the species.
