An immunogenic, high molecular weight lipopolysaccharide (LPS)-protein complex isolated from a potassium thioncyanate extract of a Pasteurella multocida (P multocida ; strain P-2383, capsular type A and somatic type 3) was characterized. Chemical analysis of the complex by gas chromatography on a capillary column demonstrated that this complex contained most of the chemical constituents characteristic of LPS extracted by the phenol-water methed from the whole bacterium. However, there was proportionately more carbohydrate than fatty acid in the complex in contrast to LPS in which fatty acid seemed to be in excess. When toxicity of the complex was evaluated in 10-day-old chicken embryos, the complex was less toxic ($LD_{50}=12.72{\mu}g$) than the purified LPS ($LD_{50}=0.44{\mu}g$). The $LD_{50}$, of the LPS moiety extracted from the complex was $5.24{\mu}g$. Composition of the complex was analyzed by SDS-PAGE with silver staining and Western immunoblotting. The complex did not migrate through the polyacrylamide gel unless dissociated with SDS. The complex dissociated with SDS contained at least 32 different protein and polysaccharide components: 18 components reacted with an antiserum against the complex. There was no significant compositional variation between the complexes from different strains, but quantitative differences in individual components were noted. When cross-protectivity of the complex was evaluated in mice, this complex provided substantial protection not only against the homologous bacteriun but also against different P multocida strains of the same serotype. LPS-protein complexes isolated by the same method from other strains also induced protection against an challenge with P-2383.
구름버섯 배양균사체와 야생 구름버섯 자실체로부터 얻은 protein-bound polysaccharide의 당과 단백질 함량을 비교하였을 때 배양균사체인 경우보다 야생 자실체의 경우가 당 함량이 더 높게 나타났으며 단백질 함량은 더 적게 나타났다. 구름버섯에서 부터 얻은 시료를 methanolysis시킨 후 HPLC로 분석하였을 때 배양균사체인 경우 4종류의 단당이 경출되었고 그중 mannose가 가장 함량이 많은 반면 야생 자실체인 경우 3종류의 단당이 경출되었으며 그중 glucose가 가장 많이 존재하였다. 구름버섯에서부터 얻은 시료에 lactose가 혼합되었을 경우 HPLC로 구별할 수 있었다. 구름버섯에서 얻은 시료의 구성 아미노산을 분석하였을 때 시료 모두에서동일한 17종의 아미노산이 검출되었다.
A gene coding for phosphoketolase, a key enzyme of carbohydrate catabolism in heterofermentative lactic acid bacteria(LAB), was cloned from a Lactobacillus paraplantarum C7 and expressed in Escherichia coli. The gene is 2,502 bp long and codes for a 788-amino-acids polypeptide with a molecular mass of 88.7 kDa. A Shine-Dalgarno sequence(aaggag) and an inverted-repeat terminator sequence are located upstream and downstream of the phosphoketolase gene, respectively. The gene exhibits an identity of >52% with phosphoketolases of other LAB. The phosphoketolase of Lb. paraplantarum C7(LBPK) contains several highly conserved phosphoketolase signature regions and typical thiamine pyrophosphate(TPP) binding sites, as reported for other TPP-dependent enzymes. The phosphoketolase gene was fused to a glutathione S-transferase(GST::LBPK) gene for purification. The GST::LBPK fusion protein was detected in the soluble fraction of a recombinant Escherichia coli BL21. The GST::LBPK fusion protein was purified with a yield of 4.32mg/400ml by GSTrap HP affinity column chromatography and analyzed by N-terminal sequencing. LBPK was obtained by factor Xa treatment of fusion protein and the final yield was 3.78mg/400ml. LBPK was examined for its N-terminal sequence and phosphoketolase activity. The $K_M\;and\;V_{max}$ values for fructose-6-phosphate were $5.08{\pm}0.057mM(mean{\pm}SD)$ and $499.21{\pm}4.33{\mu}mol/min/mg$, respectively, and the optimum temperature and pH for the production of acetyl phosphate were $45^{\circ}C$ and 7.0, respectively.
Trichoderma species/isolates exhibited varied degree of agglutination on sclerotial (Sc) and hyphal (Hy) surface of Macrophomina phaseolina. The agglutination efficiencies on Sc and Hy ranged from $11\;to\;57\%$. Isolates of T. harzianum (Th) and T. viride (Tv) showed greater agglutination on Sc ($23-57\%$) and Hy ($16-47\%$). Different enzymes (trypsin, pepsin, proteinase k, a-chymotrypsin, lyticase and glucosidase) and inhibitors (tunicamycin, cycloheximide, brefeldin A, sodium azide, dithiothreitol and SDS) reduced the agglutination potential of conidia of Th-23/98 and Tv-25/98; however, the extent of response varied greatly in different treatments. Different fractions of Th-23/98 and Tv-25/98 exhibited haemagglutinating reaction with human blood group A, B, AB and O. Haemagglutinating activity was inhibited by different sugars and glycoproteins tested. Crude haemagglutinating protein from outer cell wall protein fraction of Th-23/98 and Tv-25/98 were eluted on Sephadex G-100 column. Initially Th-23/98 and Tv-25/98 exhibited two peaks showing no agglutination activity; however, lectin activity was detected in the third peak. Similar to crude lectin, the purified lectin also exhibited haemagglutinating activity with different erythrocyte source. SDS-PAGE analysis of partially purified lectin revealed single band with an estimated molecular mass of 55 and 52 kDa in Th-23/98 and Tv-25/98, respectively. Trypsin, chymotrypsin and b-1,3-glucanase totally inhibited lectin activity. Similarly, various pH also affected the haemagglutinating activity of Th-23/98 and Tv-25/98. From the present observations, it can be concluded that the recognition/attachment of mycoparasite (T. harzianum and T. viride) to the host surface (M. phaseolina) may be most likely due to lectin-carbohydrate interaction.
Two mesophilic trickling bed bioreactors filled with two different types of media, hydrophilic- and hydrophobic-cubes, were designed and tested for hydrogen production via anaerobic fermentation of sucrose. Each reactor consisted of a column packed with polymeric cubes and inoculated with heat-treated sludge obtained from anaerobic digestion tank. A defined medium containing sucrose was fed with changing flow rate into the capped reactor, hydraulic retention time and recycle rate. Hydrogen concentrations in gas-phase were constant, averaging 40% for all conditions tested. Hydrogen production rates increased up to $10.5 L{\cdot};h^{-1}{\cdot}L^{-1}$ of reactor when influent sucrose concentrations and recycle rates were varied. Hydrophobic media provided higher value of hydrogen production rate than hydrophilic media at the same operation conditions. No methane was detected when the reactor was under a normal operation. The major fermentation by-products in the liquid effluent of the both trickling biofilters were acetate and butyrate. The reactor filled with hydrophilic media became clogged with biomass and bio gas, requiring manual cleaning of the system, while no clogging occurred in the reactor with hydrophobic media. In order to make long-term operation of the reactor filled with hydrophilic media feasible, biofilm accumulation inside the media in the reactor with hydrophilic media and biogas produced from the reactor will need to be controlled through some process such as periodical backwashing or gas-purging. These tests using trickling bed biofilter with hydrophobic media demonstrate the feasibility of the process to produce hydrogen gas in a trickle-bed type of reactor. A likely application of this reactor technology could be hydrogen gas recovery from pre-treatment of high carbohydrate-containing wastewaters.
항암 효과와 면역 활성 효과가 있는 영지 균사체의 alkali 추출 단백다당류인 GMPG와 이것을 모체로하여 한외여과를 이용한 고분자 단백다당류(GMPG-UH)와 저분자 단백다당류(GMPG-UL) 그리고 column 분획을 이용한 고분자 단백다당류(GMPG-CH)와 저분자 단백다당류(GMPG-CL)를 얻어서, 면역 항암 활성을 연구한 결과 다음과 같은 결과를 얻었다. GMPG, GMPG 고분자 및 저분자 분획들의 총당과 총단백질 함량을 측정한 결과, GMPG는 85.9% 및 12.8%, 한외여과 분획인 GMPG-UH와 GMPG-UL은 75.5% 및 72.7%의 당과 31.3%의 33.6%의 단백질, 컬럼 분획인 GMPG-CH와 GMPG-CL은 당과 단백질이 각각 96.9%와 87.1% 및 1.9%와 8.1%를 함유하고 있었다. 구성 당은 대부분 glucose, mannose, fructose, galactose, xylose와 소량의 ribose 및 arabinose를 함유하고 있었으며, ${\alpha}$와 ${\beta}-glucose$의 결합 비율이 $0.84{\sim}1.14$로 ${\beta}-glucan$이 주를 이루는 단백다당류이고, 저분자보다는 고분자가 더 많은 ${\beta}$-결합을 이루고 있는 것으로 조사되었다. 구성 아미노산의 조성은 공통적으로 산성 아미노산인 Asp와 Glu, 중성 아미노산인 Ala와 Leu이 다량 함유하고 있는 것으로 조사되었다. IR의 경우는 ${\beta}$-당의 결합양식을 결정 짖는 $890cm^1$ 부근에서의 흡수 peak가 관찰되었다. 또한 $^{13}C-NMR$ spectrum에서 GMPG, GMPG 고분자 및 저분자, ${\beta}-1,3$ anomer를 결정 짖는 C-1 peak가 $103{\sim}105ppm$에서 나타나 ${\beta}-(1{\rightarrow}3)-glucan$ 구조를 갖는 단백다당류임을 알 수 있었다. GMPG-CH와 GMPG-CL은 sarcoma 180 고형암에 대해 각각 79.6% 및 63.4%의 암세포 중식 저해율을 나타내었는데, 이것은 ${\beta}$-결합 당의 함량이 높은 고분자 분획이 항암활성도 높게 나타났다.
본 연구는 GC 및 GC/MS에 의한 벌꿀과 시판음료 중에 함유되어 있는 fructose, glucose, sucrose의 TMS 및 TMSO 유도체화를 통한 동시분석방법을 확립하고자 하였다. 3종 당류를 분석하기 위한 동시분석방법으로 당류의 TMSO 유도체화가 TMS 유도체화 방법보다 정성 및 정량성이 우수한 것으로 나타났으며, TMSO화된 당류의 GC 최적분석조건으로 초기 oven온도 $175^{\circ}C$에서 5분간 머무르게 하다가 $10^{\circ}C/min$로 $280^{\circ}C$까지 승온시키는 조건으로 3종의 당류를 19분 이내에 분석할 수 있었다. GC 분석에서 fructose와 glucose 그리고 sucrose의 직선성은 50-5000 ${\mu}g/mL$ 농도 범위 내에서 $r^2$값이 모두 0.9999 이상이었으며, 회수율은 각각 100.5, 101.0, 99.7%이였으며, 검출한계와 정량한계는 0.68, 0.47, 0.53 ${\mu}g/mL$와 2.27, 1.58, 1.77 ${\mu}g/mL$이었다. 일내 정확도와 정밀성은 fructose, glucose, sucrose에서 각각 99.0-100.0%와 0.47-1.06%, 일간 정확도와 정밀성은 100.1-101.2%와 1.09-1.79%로 나타났다. 위와 같은 우수한 분석 값은 GC/MS에 의한 분석에서도 유사하게 나타났다. 확립된 GC 분석방법으로 아카시아 벌꿀 시료 12종의 당 함량을 조사한 결과, 아카시아 벌꿀의 fructose와 glucose 평균함량은 각각 $42.58{\pm}1.97%$, $27.74{\pm}1.16%$이었으며, F/G ratio는 $1.53{\pm}0.07$로 HPLC로 분석한 값과 거의 동일하였다. 그러나 sucrose 함량은 GC분석에서는 평균함량이 $0.79{\pm}0.52%$였으나, HPLC $NH_2$ column을 사용 시 $2.24{\pm}4.62%$를 나타내었고, carbohydrate column으로 분석했을 때는 분리도의 저하로 몇 개의 시료에서는 정확한 함량분석에 어려움이 있었다. 따라서 본 연구에서 실시한 GC, GC/MS에 의한 fructose, glucose, sucrose의 TMSO 유도체 동시분석방법은 감도, 분리도, 회수율, 직선성, 정밀성, 정확도면에서 우수한 정량분석방법이라고 판단된다.
본 연구에서는 다양한 낙농 발효유제품에 있어서 다양한 유기산과 탄수화물의 분석이 HPLC 방법으로 동시에 잘 진행되었다. 이 방법의 주요 장점으로는 (1) 간단한 시료 준비와 단일 추출이 가능, (2) 유동상(mobile phase)과 추출 용매에 있어서 편리성과 경제성, (3) 추출제와 유동상에 사용되는 황산의 농도가 매우 낮기에 친환경적인 실험, (4) 하나의 샘플당 30분 안에 HPLC의 분석이 완료 가능, (5) 다양한 농도범위에서도 분석 및 정량 가능, (6) 몇 가지 물질을 제외하고 모든 화합물의 변동계수가 낮음, 그리고 (7) 회수율이 대체적으로 높다는 것이다. 또한, $60{\sim}65^{\circ}C$에서 분석 column을 사용하였기에 column 효율면에서도 손실이 극히 적은 것으로 관찰되었다. 따라서 본 실험에서 분석된 다양한 낙농 발효유제품에서의 acid, 탄수화물, diacetyl, acetoin 등이 HPLC 방법은 매우 신속하고 정확하게 검출되었다. 하지만 다양한 향미의 변화를 최소화하면서 다양한 향미를 효과적으로 추출할 수 있는 HPLC 방법들이 지속적으로 개발되어야 다양한 낙농 발효유제품의 품질개선에 기여할 것으로 사료된다. 왜냐하면 향미 성분의 분석은 성분을 정성하는 것뿐만 아니라, 정성된 화합물이 그 식품의 향에 어느 정도 기여하는지를 중요하게 결정하기 때문이다. 또한 본 실험에 사용된 HPLC 방법으로 다양한 유산균주의 일정한 배양시간동안 유당분해효소(lactase) 능력이 조사 되었는데, 각 Probiotic lactic acid bacteria마다 다양한 유당분해효소(lactase)의 능력을 보였으며, 같은 균종에서도 차이가 있는 것으로 나타났다. 본 실험에서는 Bifidobacterium spp.가 Lactobacillus spp.와 Streptococcus spp.보다 높은 유당분해효소(lactase)의 능력을 보였다. 하지만 유산균주의 유당분해효소 능력을 정확하게 파악하기 위해서는 각 Probiotic lactic acid bacteria의 exo- 및 endo-lactase에 대한 연구가 추가적으로 필요하다. 결론적으로 다양한 낙농발효식품 및 기타 발효식품 등에서 HPLC 방법으로 동시에 유기산과 탄수화물 분석이 공인방법으로써 인정되기 위해서는 더 많은 기초자료 확립과 재현성에 관한 지속적인 연구가 진행되어야 할 것으로 사료된다.

Hydrogen production rate was increased till $10.5\;L{\cdot}h^{-1}{\cdot}L^{-1}$ of bioreactor when influent sucrose concentrations and recycle rates were varied. At the same time, the hydrogen production rate with hydrophobic media application was higher than its hydrophilic media application. No methane was detected when the reactor was under a normal operation. The major fermentation by-products in the liquid effluent of the both trickling biofilters were acetate, butyrate and lactate. In order to run in the long term operation of both reactor filled with hydrophilic and hydrophobic media, biofilm accumulation on hydrophilic media and biogas produced should be controlled through some process such as periodical backwashing or gas-purging. Four sample were collected from each reactor on the opposite hydrogen production rate, and their bacterial communities were compared by terminal restriction fragment length polymorphism (T-RFLP) analysis of PCR products generated using bacterial 16s rRNA gene primers (8f and 926r). It was expressed a marked difference in bacterial communities of both reactors. The trickling bed bioreactor with hydrophobic media demonstrates the feasibility of the process to produce hydrogen gas. A likely application of this reactor technology can be hydrogen gas recovery from pre-treatment of high carbohydrate-containing wastewaters.
본 연구에서는 1-kestose, raffinose, nystose 및 $1^{F}-{\beta}-fructofura-nosylnystose$ 등 4종 올리고당 분석을 위하여 기울기용매가 가능 한 증기화광산란검출기를 사용하여 이동상 조성, 유속 등 최적 조건을 설정하였다. 설정한 분석조건을 이용하여 표준액을 농도별로 희석(50, 100, 200, 400, 800${\mu}g/mL$)하여 검량선을 quadratic curve로 작성한 결과 상관계수는 1-kestose(1.0000), raffinose(0.9999), nystose(1.0000), $1^{F}-{\beta}-fructofuranosylnystose$(1.0000)으로 나타났다. 또한 분석조건의 재현성을 측정하기 위하여 혼합표준액 200${\mu}g/mL$을 5회 주입한 면적 값의 RSD는 1-kestose(1.47), raffinose(8.98), nystose(0.77), $1^{F}-{\beta}-fructofuranosylnystose$(2.02)으로 나타났다. 또한 사용된 시료의 프락토올리고당의 측정된 함량 결정계수에 대한 t-검정결과 모든 표준당에 대해 상관이 유의(p<0.05)함을 알 수 있어 본 HPLC-ELSD방법에 의한 정량이 타당한 것으로 나타났다. 본 방법을 통하여 fructooligosacchrides 및 raffinose분석에 유용한 것으로 사료된다.

제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
