Cytogenetic Analysis of Induced Hybrid between Common Carp (Cyprinus carpio) and Crucian Carp (Carassius auratus) (잉어(Cyprinus carpio)와 붕어(Carassius auratus)간 잡종의 세포유전학적 분석)

  • Cytogenetic analyses were carried out with induced hybrid between common carp (Cyprinus carpio) female and crucian carp (Carassius auratus) male. The erythrocytic measurement revealed that cellular and nuclear size of induced hybrids were intermediate between those of paremtal species. The modal chromosome numbers of common carp, crucian carp and its hybid were same as 2n=100. The DNA content of induced hybrids determined based on flow cytometry was 3.7pg/cell which corresponding to intermediate value between the carp (3.6ph/cell) and crucian carp (3.8pg/cell)

  • Goldfish hematopoietic necrosis (GHN) affects hematopoietic organs and causes high mortality in goldfish. The causative agent of GHN is cyprinid herpesvirus 2 (CyHV-2). The purpose of this study was to detect CyHV-2 in ornamental cyprinid fish. CyHV-2 monitoring was conducted on a monthly basis for 1 year using goldfish Carassius auratus and pearlscale goldfish Carassius auratus that had been cultured at a domestic aquafarm and imported from Singapore, respectively. The results of polymerase chain reaction (PCR) assays using specific primers for the CyHV-2 helicase gene showed that 27.8% of goldfish and 33.3% of pearlscale goldfish were positive for CyHV-2. No cytophatic effects were detected in koi fin or common carp brain cells inoculated with tissue homogenates from either fish. Our data provides the useful data for establishing future quarantine and disinfection policies.

  • Genomic DNA isolated from two crucian carp species obtained from Yesan (Carassius auratus) and Dangjin (Carassius cuvieri) in Korea were amplified at several times by polymerase chain reaction (PCR). The amplified products were separated by agarose gel electrophoresis (AGE) with oligonucleotides decamer primer and stained with ethidium bromide. The seven arbitrarily selected primers OPC-11, OPC-14, OPC-18, OPD- 02, OPD-11, OPD-15 and OPD-20 generated the shared loci by each species, the polymorphic and specific loci. The seven primers generated the total 458 loci that can be scored from the crucian carp obtained in C. auratus species. 358 fragments were generated from the species obtained in C. cuvieri species. The size of DNA fragments varies from 150 to 1,600bp. The complexity of the banding patterns varies dramatically between the primers and two locations. In this study, 458 loci were identified in the crucian carp species from Yesan and 358 in the crucian carp species from Dangjin: 84 polymorphic loci (18.3%) in the C. auratus species and 48 (13.4%) in the C. cuvieri species. 154 shared loci by each species, the average 22 per primer, were observed in the C. auratus species and 187 loci, the average 26.7 per primer, in the Dangjin species. Based on the average bandsharing (BS) values of all samples, the similarity matrix ranged from 0.434 to 0.868 in the C. auratus species and from 0.449 to 0.924 in the C. cuvieri species. The average BS value was 0.641±0.013 within the C. auratus species and 0.684±0.013 within the C. cuvieri species. The average BS value between two crucian carp species 0.484 ± 0.007, ranged from 0.307 to 0.682. The BS value between the individual No. 09 and No. 16 was 0.682, which was the highest between two crucian carp species. Compared separately, the BS value of individuals within the C. cuvieri species was higher than the C. auratus species. The dendrogram obtained by the seven primers, indicates three genetic clusters: cluster 1 (AURATUS No. 01, 02, 03, 04, 05, 06, 07, 08, 09, 10 and 11), cluster 2 (CUVIERI No. 12, 13, 14, 15, 16, 17, 18, 19, 20 and 21) and cluster 3 (CUVIERI no. 22). The shortest genetic distance displaying significant molecular difference was between the individual AURATUS No. 09 and AURATUS No. 08 from Yesan (genetic distance=0.064). The longest genetic distance displaying significant molecular differences was between the individual CUVIERI No. 17 and AURATUS No. 11 between two crucian carp species (0.477). RAPD-PCR analysis has revealed the significant genetic distance between two crucian carp species pairs.(Key words: Carassius auratus, Carassius cuvieri, Crucian Carp, DNA Polymorphism, Genetic Distance)

Acute Toxicity of Carassius auratus and Pungtungia herzi Lavra on Mercury, Lead and Copper Exposure (수은, 납 및 구리에 대한 붕어(Carassius auratus)와 돌고기(Pungtungia herzi) 자어의 급성독성)

  • Larva of Carassius auratus and Pungtungia herzi were exposed to mercury ($HgCl_2$), lead ($PbCl_2$) and copper ($CuSO_4$) to determine acute toxicity. The toxicity tests were conducted triplicate and the $LC_{50}$ values (24, 48, 72 and 96 hours) were determined for two species. Data obtained from the toxicity tests were evaluated using the probit analysis. Although sensitivities of two species to mercury were relatively similar, C. auratus was more susceptible than P. herzi to the exposure of lead but P. herzi was more sensitive than C. auratus on copper. The sensitivity on three metals tested may be ranked in the following order from highest to lowest toxicity on larval stage of these fish: mercury > copper > lead. It is suggested that acute toxic test at the larval stage of C. auratus and P. herzi, indigenous species in Asia area, is an important part of the ecotoxicological assessment of some heavy metals.

A Study on the Toxicity of Pb and Cu Compound in Carassius auratus(goldfish) (납 및 구리화합물이 Carassius auratus(goldfish)에 미치는 독성에 관한 연구)

  • In order to investigate acute toxicity and bioconcentration of heavy metals for a freshwater fish, the fish used in this experiment was goldfish, Carassius auratus. Each ten goldfish was accommonidated in a water and was treated with different concentration of Pb and Cu compound. The 24 $hr-LC_{50}$ was obtained by plotting on the log-normal distribution graph. Furthermore, the combined effect of Pb and Cu was also investigated the fish was treated with Pb or Cu compound only, and Pb and Cu compound together, respectively. These results were summarized as follows: 1. The 24 $hr-LC_{50}'s$ of Pb and Cu were 7.48 mg/l and 0.666 mg/l, respectively. 2. When single or/and combined treatment with Pb(7.0 mg/l) or/and Cu(0.6 mg/l) to Carassius auratus for 24 hours were performed, there was significant difference between the single or/and the combined treatment in their bioaccumulated Cu concentrations. Cu concentrations in goldfish were higher in the combined treatment than in the single treatment. 3. When Carassius auratus was exposed to 0.748 mg/l (1/10 of 24 $hr-LC_{50}$) and 1.496 mg/l of Pb (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 79.14 and 100.11 for Pb, respectively. The BCF of Pb was obtained as a linearity according to the concentration and exposure time as follows log BCF=1.014 log $P\cdot T$+1.011 ($r^2$=0.9041) where, P: pollutant concentration(mg/l) T: exposure time(day) 4. When Carassius auratus was pxposed to 0.0666 mg/l (1/10 of 24 $hr-LC_{50}$) and 0.1332 mg/l of Cu (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 55.42 and 63.24 for Cu respectively.   • Histopathological changes and cadmium accumulations in the gills were investigated in crucian carp (Carassius auratus) exposed to 0, 0.01, 0.1, 0.5 mg/1 cadmium (Cd) concentrations for 25 days. After 25 days of exposure. cadmium accumulations increased in each of the different exposure concentrations. Histopathological changes in the gills of curcian carp exposed to cadmium included the acidification of mucous cell, the terminal clubbing of lamellae, the hyperplasia of epitherial cell and the curved of secondary lamellae. The similar histopathological changes were observed once the accumulations of cadmium reached the similar levels found in the present study with the different exposure schemes. Also the acidification of mucous cell in gills appeared when cadmium accumulations reached 5 $\mu\textrm{g}$/g and more. These results suggested that histopathological changes in the gills, an appearance of mucous cell acidification. be used to estimate the relation between toxic effect and cadmium accumulations in the gills.

Effects of clove oil on haematology and blood chemistry in crusian carp (Carassius auratus) (Clove Oil의 붕어(carassius auratus) 혈액 및 혈액화학치에 대한 영향)

  • To determine the effects of anesthesia to clove oil on the secondary stress indices, changes in haematological and blood chemistry were monitored in healthy crucian carps (Carassius auratus). 24 fishes were divided into three groups, and blood was collected before anaesthesia (control group), immediately after anaesthesia at a concentration of 30 mg/l clove oil (anesthetized group), and 24 h after anaesthesia (recovered group). The anesthesia to clove oil significantly increased glucose in anaesthetized group, and constantly decreased lactate dehydrogenase in anaesthetized group and recovered group. However, clove oil had not effect on other biochemical indices. These results suggests that the anesthetic use of clove oil at a concentration of 30 mg/l does not cause irreversible damage in carp.

  • Morphological variation of the crusian carp, Carassius auratus, from Yongnam area in Korea was examined. In morphometric and meristic characters, mean values were different slightly among the polulations, whereas the range of variation was overlapped largely. In the present examination the number of spines in dorsal fin were 4-6. The majority of individuals appeared to be similar to Carassius auratus langsdorfii from Japan, but some individuals were quite different from them in having 66-83 gill rakers.

Phylogenetic Analysis of Carassius auratus and C. cuvieri in Lake Yedang Based on Variations of Mitochondrial CYTB Gene Sequences (예당호 붕어와 떡붕어의 CYTB 유전자를 이용한 유연관계 분석)

  • Two crucian carp species (Carassius auratus and C. cuvieri) inhabit Lake Yedang in South Korea, and C. auratus is known to be native to Korea. Classification of these two freshwater fish species is often confused because of their morphological similarity. To distinguish the two species, we conducted phylogenetic and population genetic analyses of C. auratus and C. cuvieri based on their mitochondrial DNA sequences of the cytochrome b gene (CYTB). We also compared our partial CYTB sequence (<1,056 bp) with 10 Chinese, nine Japanese, and two Russian crucian carp fishes. The results of our phylogenetic analysis showed that C. auratus and C. cuvieri were clearly divided into two phylogroups. The nucleotide diversity (π) of C. auratus from Korea, China, and Japan showed a range of 0.146%~0.421%, while the range of π of C. cuvieri from Korea and Japan was lower than those of C. auratus (0.0%~0.054%). Moreover, the comparison of CYTB divergence among crucian carp fishes in China, Japan, and Korea indicated that Korean Carassius fishes were distantly related to those from China and Japan, with two exceptions: the pairwise Fst value between Korean C. auratus and northern Chinese C. auratus was not significantly different. In addition, no significant genetic divergence between Korean and Japanese C. cuvieri was detected. We conclude that, despite the morphological similarities, C. auratus and C. cuvieri should be considered as separate freshwater fish resources in conservation efforts for genetic diversity.

Monitoring of carp edemavirus disease and herpesviral hematopoietic necrosis in Cyprinus carpio and Carassius auratus imported from China (중국에서 수입된 잉어 및 붕어의 잉어부종바이러스병 및 허피스바이러스성 조혈기괴사증 모니터링)

  • In this study, carp edemavirus disease and herpesviral hematopoietic necrosis were examined in carp (Cyprinus carpio) and crucian carp (Carassius auratus) imported from China. From August 2016 to May 2017, 100 carp and 300 crucian carp were examined. There were no clinical symptoms in all the tested fish, and no internal or external parasitic infections were identified except for the detection of Argulus sp. in one leather carp. Carp edema virus disease was examined by PCR in C. carpio, but all were negative. In C. auratus, herpesviral hematopoietic necrosis was tested by PCR and 4 out of 300 were positive. The results of this study are expected to be useful as basic data for conducting risk analysis on the relevant organisms and counterpart countries or for designating new pathogens for quarantine.
