Streptozotocin유도 당뇨쥐의 단기간 capsaicin 섭취와 운동이 혈중 지질, 항산화 효소 및 Akt량에 미치는 영향 (The Effects of Short-term Exercise and Capsaicin Supplementation on Serum Lipids, Antioxidants and Akt Contents in STZ-induced Diabetic Rats)

  • 본 연구는 50 mg/kg의 STZ로 유발된 당뇨쥐에서 단기간 운동과 capsaicin 섭취가 혈중지질, 항산화 효소 및 Akt량에 미치는 영향을 알아보고자 31마리의 흰쥐를 대상으로 실험 하였다. 운동 군과 운동과 capsaicin을 병행한 군에서는 당뇨 유발 일주일 후 부터 15 m/min, 0%의 경사도로 25분간 주5회 2주간 트레드밀 운동을 실시하여 다음과 같은 결과를 얻었다. 혈중 지질 농도는 capsaicin 섭취와 운동을 병행한 군에서 TG는 유의하게 감소하였고, HDL-C는 다른 군에 비해 유의하게 높게 나타났고, ROS는 당뇨군을 제외한 모든 군에서 유의하게 감소하였다. CAT는 capsaicin만 섭취한 군에서 유의하게 증가하였고, SOD은 운동만 처치한 군과 capsaicin만 섭취한 군에서 유의하게 높았으며, GSH는 당뇨군을 제외한 모든 군에서 유의하게 높은 것으로 각각 나타났다. Akt효소는 capsaicin만 섭취한 군, p-Akt는 운동군에서 유의하게 증가하였다.

  • 고추의 저장중에 미생물 오염에 의해서 일어나는 신미성분(辛味成分)의 변화에 대하여 검토하여 다음과 같은 결과를 얻었다. 1. capsaicin이 함유된 배지에서 생육이 강하고 분해력(分解力)이 강한 우수균주로서 Aspergillus oryzae, Aspergillus sp. KF-7 를 오염된 고추로 부터 얻었다. 2. 건조균체량(乾燥菌體量)은 capsaicin의 농도가 $2{\sim}3{\mu}g/ml$일 때는 약간 증가하였지만 농도가 높아지면 감소하였다. 3. 배지중에 첨가한 capsaicin은 일주일 후에는 반으로 감소되고 2주일 후에는 거의 소실되었고 $30^{\circ}C$의 온도, 그리고 $pH\;7{\sim}9$일 때에 분해 소실되는 양(量)이 가장 많았다. 4. 고추분말로 된 고체배지에서 Aspergillus sp. KF-7은 거의 생육하지 못하였고 Aspergillus oryzae는 수분함량이 13%일때 가장 잘 생육하였으며 배양 40일 후에는 capsaicin의 잔존률(殘存率)이 45%가 되었다. 5. 고추씨로 된 고체배지에서 두 균주가 생육하여 배양 4주(週) 후에는 각각 55%, 38%의 capsaicin잔존률(殘存率)을 나타내었다.

Orotic Acid-유발 지방간에 미치는 Capsaicin의 개선효과 (Improvement Effect of Capsaicin Against Orotic Acid-Induced Fatty Liver in Rats)

  • Orotic acid(OA)-유발 지방간 흰쥐의 혈청 및 간장의 지질농도, 임상생화학적 및 조직형태학적 변화에 미치는 cap-saicin(CS)성분의 영향을 검토하였다. 기본식이에 OA 1%, CS 0.02% 및 OA와 CS를 각각 1%와 0.02% 수준으로 첨가하여 4주간 섭취 시켰다. 간장 중성지방 농도는 대조군에 비해 약 4.7배 현저히 증가하여 지방간이 유발되었으나, OA十CS 동시투여에 의해서는 간장 중성지방 농도가 OA군에 비해 38% 감소하여 지방간이 개선된 것으로 나타났다. OA 식이에 의해 간장 콜레스테롤 농도가 증가하였으나, capsaicin 동시투여에 의해 감소하였다. Capsaicin 단독 투여는 간장 중성지방 및 콜레스테롤 농도에 영향을 미치지 못하였다. Capsaicin 투여에 의한 혈청 중성지방 및 콜레스테롤 농도의 감소는 OA 첨가 또는 무첨가 식이군에서 모두 관찰되었다. 간기능 지표 효소인 혈청 GOT 및 GPT 활성은 각 실험군간에 차이가 나타나지 않았다. 간조직의 형태학적 관찰 결과, 대조군과 CS군은 정상적인 간소견을 보였으나 OA군의 간세포에는 소포성 지방 변성이 소엽 중심대로 주로 나타나 있고, 크고 많은 지방세포들이 균일하게 배열되어 있었다. 그러나 OA+CS군의 간세포들은 손상된 세포가 재생될 뿐 아니라 소포성 지방세포들의 크기와 수가 줄어들고 있음을 확인하였다. 이상의 실험 결과에서, capsaicin 0.02% 농도는 OA-유발 지방간을 개선시키는 효과가 있는 것으로 판단되어진다.

Capsaicin이 백서 간의 Cytochrome $P_{450}$에 미치는 영향 (Effects of Capsaicin on Liver Cytochrome $P_{450}$ in the Rat)

  • It was previously reported that cytochrome P$_{450}$ content in liver was increased when Capsicum acetone extract was given chronically to rats. The present study is aimed to investigate the effect of capsaicin, a principal component of red pepper, on the drug metabolizing enzymes in rat liver. Capsaicin (5mg/kg) was given intraperitoneally once a day for seven days and zoxazolamine paralysis time and hexobarbital sleeping time were determined 24 hrs after the last dose of capsaicin. Plasma hexobarbital concentration was also determined five and 15 min after hexobarbital administration to rats. Zoxazolamine paralysis time and hexobarbital sleeping time were shortened by 31.6% and 37.1%, respectively, compared with control group. Plasma hexobarbital concentration was lowered by 26.2% after five min and by 35.2% after 15 min, respectively, compared with control group. However, administration of single dose of capsaicin did not affect the zoxazolamine paralysis time and hexobarbital sleeping time. Microsomal cytochrome P$_{450}$ content and NADPH-cytochrome C reductase activity were increased by 14.6% and 11.6%, respectively in the rats pretreated with capsaicin for seven days, while cytochrome b$_{5}$ content was not changed. These results suggest that treatment with capsaicin for seven days may induce the drug metabolizing enzyme in rat liver.

Effects of oral caffeine and capsaicin administration on energy expenditure and energy substrates utilization in resting rats

  • Caffeine and capsaicin increase resting energy metabolism. However, most measurements have been conducted in short-term studies. Therefore, we investigated the effects of caffeine and capsaicin on energy expenditure and energy substrate utilization in resting rats for 6 h. The caffeine (Experiment 1) experiment included four male rats aged 5 weeks and measured the effects of oral administration of caffeine (10 or 50 mg/kg) on respiratory gas, energy expenditure, and energy substrate oxidation for 6 h. Experiment 2 included four male rats aged 6 weeks to measure the effects of capsaicin (10 mg/kg) using the same method as in Experiment 1. The results of Experiment 1 indicated that O2 uptake and carbohydrate oxidation after caffeine administration for 2 h was higher in the 10 mg trial than that in the 50 mg or placebo trials (P < 0.05). However fat oxidation was not significantly different. In contrast, capsaicin (Experiment 2) observed no differences between the placebo and the capsaicin trials. In conclusion, caffeine initially increased the resting energy consumption for 2 h, and this energy expenditure was due to carbohydrate oxidation. Capsaicin did not change oxygen uptake, respiratory exchange ratio, fat oxidation, or carbohydrate oxidation.

Effect of Capsaicin on the Body fat and Adipocyte in the Diet induced-obese Mice

  • As the obesity has been known to be related with the hyperlipidemia, cardiovascular disease, cerebral apoplexy, fatty liver, and other chronic diseases, recent researches have focused on the functional food materials and their anti-obesity activities. This study was performed to study the effects of vanilloid family capsaicin, major pungent ingredient of hot chillies and peppers, on anti-obesity activities. ICR male mice were fed one of the pellet diet, basal diet, and high fat diet with capsaicin (45 $\mu\textrm{g}$/day) solution for 5 days. Mice in the corresponding control groups were given water for 5 days. In results, capsaicin reduced body weights in any diet groups. Percent weight and cell size of the abdominal white adipose tissue in mice on the high fat diet with capcaicin were significantly lower compared with those in mice on the high fat diet with water. However, percent brown adipose tissue weight per body weight in mice on the high fat diet was not affected by capsaicin. Capsaicin reduced the levels of s-triglyceride and s-total cholesterol in the pellet diet or high fat diet groups. There was no difference in the s-protein levels between the capsaicin group and the control water group. These data indicate that 1) orally administered capsaicin has a reducing effect on the blood triglyceride and total cholesterol levels, and 2) capsaicin has lowering effects on the body weight, percent weight and cell size of the abdominal white adipose tissue.

The effects of regular exercise on capsaicin-induced pulpal pain and pain-induced changes in passive avoidance learning and memory in rats

  • Background: Pulpal pain is one of the most common and severe orofacial pain conditions with considerable adverse effects on physiological processes including learning and memory. Regular exercise is known to be effective on cognitive function as well as pain processing in the central nervous system. Here, the possible effects of regular exercise on pulpal pain response as well as pain-induced changes in learning and memory efficiency in rats were investigated. Methods: Twenty-four male Wistar rats were randomly assigned to the control, capsaicin, exercise, and exercise plus capsaicin groups. Rats in exercise groups were forced to run on a treadmill with a moderate exercise protocol for 4 weeks. Capsaicin was used to induce dental pulp pain. Passive avoidance learning and memory performance was assessed by using a shuttle box apparatus. Results: According to the results, regular exercise could decrease the time course of capsaicin-induced pulpal pain (P < 0.001). Moreover, in capsaicin-treated rats, passive avoidance acquisition was impaired as compared to the control (P < 0.05) and exercise (P < 0.001) groups. Additionally, regular exercise before capsaicin injection could attenuate capsaicin-induced memory impairments (P < 0.05). Conclusions: Taken together, the present data showed that regular exercise has inhibitory effects on capsaicin-induced pulpal pain as well as pain-induced cognitive dysfunction in rats.

Characterization of Putative Capsaicin Synthase Promoter Activity

  • Capsaicin is a very important secondary metabolite that is unique to Capsicum. Capsaicin biosynthesis is regulated developmentally and environmentally in the placenta of hot pepper. To investigate regulation of capsaicin biosynthesis, the promoter (1,537 bp) of pepper capsaicin synthase (CS) was fused to GUS and introduced into Arabidopsis thaliana (Col-0) via Agrobacterium tumefaciens to produce CSPRO::GUS transgenic plants. The CS was specifically expressed in the placenta tissue of immature green fruit. However, the transgenic Arabidopsis showed ectopic GUS expressions in the leaves, flowers and roots, but not in the stems. The CSPRO activity was relatively high under light conditions and was induced by both heat shock and wounding, as CS transcripts were increased by wounding. Exogenous capsaicin caused strong suppression of the CSPRO activity in transgenic Arabidopsis, as demonstrated by suppression of CS expression in the placenta after capsaicin treatment. Furthermore, the differential expression levels of Kas, Pal and pAmt, which are associated with the capsaicinoid biosynthetic pathway, were also suppressed in the placenta by capsaicin treatment. These results support that capsaicin, a feedback inhibitor, plays a pivotal role in regulating gene expression which is involved in the biosynthesis of capsaicinoids.

고추 Oleoresin의 추출에 영향을 미치는 몇가지 인자 (Effect of Some Factors on Oleoresin Extraction from Red Pepper)

  • 고추 oleoresin의 적정 추출조건을 설정하기 위하여 추출에 영향을 미치는 몇 가지 인자들을 대상으로 oleoresin의 수율, capsanthin 및 capsaicin 함량의 변화를 조사하였다. Ethyl alcohol 용매로 추출한 oleoresin은 수율 및 capsanthin 함량이 높고, Ethylene dichloride 용매의 경우는 capsaicin 함량이 높게 나타났다. 혼합용매의 경우는 단일용매의 경우보다 수율은 증가하나 capsanthin 및 capsaicin 함량은 거의 변화가 없었다. 추출에 사용한 3가지 품종 중에서 진솔에서 추출한 oleoresin의 품질이 가장 우수하였다. 적정 추출온도 및 추출시간은 $20^{\circ}C$$3{\sim}5$시간이었다. 장기 저장한 고추일수록 고추분말의 입도가 클수록 추출되는 oleoresin의 품질은 저하되고, 함량도 적었다 고추의 과피에는 capsanthin이 씨에는 capsaicin이 가장 많았다. 고추분말과 추출용매의 적정혼합비는 $1\;:\;3{\sim}1\;:\;4(w/v)$였다.
