  • This study evaluated the efficacy and safety to determine the therapeutic responses of oclacitinib in canine atopic dermatitis (CAD) and identified factors related to the therapeutic response. Overall, 13 dogs with CAD were treated with oclacitinib for 56 days. Owners and veterinarians assessed visual analog scale (VAS) scores of pruritus and dermatitis. The examined dogs were grouped according to treatment success and failure based on changes in the VAS scores. To identify factors related to the therapeutic responses of oclacitinib, signalments (breed, sex, age, and body weight), mean progression time of CAD, mean Owner and Veterinarian VAS scores at day 0, and preexisting infection rate were compared between the two groups. Among the 13 dogs, 7 (53.8%) met the criteria of treatment success. In the success group, the Owner VAS scores were significantly lower from day 6 to 56 compared with the score at day 0 (P < 0.05). Additionally, the Veterinarian VAS scores were significantly decreased at days 14 and 42 compared with the score at day 0 (P < 0.05). There were no changes in hematological indices after the administration of oclacitinib. The most common abnormality reported was otitis externa (30.8%), followed by pyoderma (23.1%), and vomiting (7.7%). Factors related to responses of oclacitinib were not identified. This study demonstrated that oclacitinib was safe and moderately effective in dogs with CAD. This is the first report of the clinical application of oclacitinib in South Korea.

  • Atopic dermatitis (AD) is a common skin disease in animals and several therapeutic trials with various drugs have been conducted for more effective management of AD. However, these trials have not been able to properly address all the aspects of AD management because of the lack of good efficacy or due to significant side effects of the drugs being tested. Synbio-glucan functional spray is a functional skin spray using Synbio-glucan composed of β-glucan and probiotics. We designed a functional spray composed of Synbio-glucan (patent application number:10-1805863), distilled water, glycerin, solubilizer, and 40% alcohol. We tested the efficacy and safety of the functional spray on six dogs with AD. The trial was conducted with the consent of the caregivers. The spray was applied to the skin lesions, including the trunk, axillae, inguinal region, or periocular areas, thrice a day for 30 days. To evaluate the efficacy of this functional spray, we assessed the pruritus visual analog scale (PVAS) and the canine atopic dermatitis extent and severity index (CADESI)-4. At the end of one month, the results clinical scores after functional spray treatment showed a significant decrease in the PVAS (p = 0.03) and CADESI-4 (p = 0.03) in all the subject dogs with AD. This study thus confirmed that the Synbio-glucan functional spray is efficacious and safe for the treatment of AD in dogs.

  • An 11-year-old, toy poodle dog was presented with dermatologic lesions and mammary gland tumor (MGT) evaluation. A solitary, lobulated MGT (size $2.5{\times}3.5$ cm) was affecting the 5th left mammary gland. Firm, oval plaque skin lesions were present on the left dorsal carpal area. The skin lesions were alopecic and salivary staining. The dog had historical separation anxiety and excessive licking of skin lesions were observed. Based on the clinical and histopathologic examinations, MGT was diagnosed with mammary complex adenoma and the skin lesions were diagnosed with acral lick dermatitis. Behavior modification treatment using oral clomipramine was effective.

  • Probiotics improve the immune system. However, the effects of its lactic acid bacteria on atopic dermatitis relief and inflammation improvement is not fully understood. Recently, one of the probiotics, Lactobacillus helveticus HY7801 (HY7801), was found to have an anti-inflammatory effect. In this study, we investigated the effects of HY7801 on atopic dermatitis-induced animal models. After four weeks of oral administration, the group treated with HY7801 showed amelioration of the atopic dermatitis compared to the group receiving placebo. In the HY7801 treated group, the epidermal hyper-proliferation and collagen deposition were inhibited compared to the placebo group, and the secretion amount of the inflammatory factors, such as TNF-α, IL-4 were reduced. In conclusion, these results suggest that HY7801 acts as a functional probiotic via amelioration of the atopic dermatitis such as a decrease of epidermal hyper-proliferation, and collagen deposition and anti-inflammatory effects.

  • The purpose of this study is to establish experimental canine skin barrier disruption model in dog. The study was designed to investigate the predictive properties of acetone damage using as effect parameters transepidermal water loss (TEWL) and stratum corneum (SC) hydration. To compare the structures of SC intercellular lipids in normal and acetone damaged skin, TEM observations were performed. Six clinically normal, male Beagles without dermatological problems were chosen for this study. Acetone damage was performed at 48hrs after clipping. Efficacy measurements for TEWL and skin hydration were performed before ($t_1$) and after ($t_2$) damage in a temperature- and humidity-controlled room ($2{0\sim}22^{\circ}C$, 50-60%). TEWL and SC hydration values were decreased in the acetone damaged model compared with non damaged skin. In TEM observation of acetone damaged sample, the intercellular lipid lamellae exhibit abnormal and incomplete structure compared with those of normal skin. It seems that acetone damage would be one of canine skin epidermal barrier disruption model for the study of canine atopic dermatitis (AD) as well as dry skin in veterinary dermatology research.

  • We examined the responses of PBMCs to house dust mite (HDM) allergen in atopic and healthy, non-atopic dogs to identify differences in lymphocyte reactivity that might reflect the immunologic status of atopic dermatitis. Thirteen of 20 (65%) atopic dogs showed a positive lymphocyte proliferative response to HDM allergen. The rate of response was significantly higher in the atopic dogs than that in healthy, non-atopic dogs insensitive to the allergen (P = 0.007). The proliferative responses were positively correlated with the level of HDM-specific IgE in serum (P = 0.035), and were thereby confirmed to reflect the activity of lymphocytes competent to promote IgE production. These results suggest that HDM-specific lymphocytes were present in peripheral blood and played a role in the pathogenesis of canine atopic dermatitis.
