Many skin diseases are caused by microbial infections. Representative pathogenic fungus and bacterium that cause skin diseases are Candida albicans and Staphylococcus aureus, respectively. Malassezia pachydermatis is a fungus that causes animal skin diseases. In this study, we propose a method for removing pathogenic microorganisms from the skin using relatively safe edible herbal extracts. Herbal extracts were screened for skin health through the removal of pathogenic microorganisms, and combinations for effective utilization of the screened extracts were identified. In this study, among methanol extracts of 240 edible plants, C. albicans, S. aureus, and M. pachydermatis were killed by extracts of 10 plants: Acori Gramineri Rhizoma, Angelicae Tenuissimae Radix, Cinnamomi Cortex, Cinnamomi Ramulus, Impatientis Semen, Magnoliae Cortex, Moutan Cortex Radicis, Phellodendri Cortex, Scutellariae Radix, and Syzygii Flos. By evaluating the synergistic antifungal activities against C. albicans using all 45 possible combinations of these 10 extracts, five new synergistic antifungal combinations, Acori Gramineri Rhizoma with Magnoliae Cortex extracts, Acori Gramineri Rhizoma with Phellodendri Cortex extracts, Angelicae Tenuissimae Radix with Magnoliae Cortex extracts, Magnoliae Cortex with Phellodendri Cortex extracts, and Phellodendri Cortex with Syzygii Flos extracts, were identified. By utilizing the selected extracts and five combinations with synergistic antifungal effects, this work provides materials and methods to develop new and safe methods for treating candidiasis using natural products.
Four secondary metabolites from an unidentified tunicate were isolated by treatment with trichloroethyl chloroformate(TECF) or acetic anhydride in pyridine. Their structures were determined by an extensive NMR analysis and the configuration of diacetyl derivatives(3a, 4a) was assigned by comparing with NMR data of a similar compound. Three new naturally occurring compounds (1, 3, 4) showed potent brine shrimp lethality and antifungal effect against Candia albicans.
Candida albicans, a polymorphic fungus, causes systemic and local infections. Recent reports show that the fungus is a main etiological agent for the arthritis. For trea tment, antifungal drugs and/or rheumatoid drugs are used, but resistance and side effects limit application of the drugs. In search of new sources for treatment of the fungal arthritis, we choose Egb 761 (extract of Ginkgo leaves 761), one of the most popular over-the-counter herbal medicines. The Egb 761 contains two major ingredients such as terpene and flavonoid. In the present study, we examined if the terpene portion of Egb 761 had anti-inflammatory activity against C.albicans-caused arthritis. The terpene was extracted with combination of methanol and water from the Egb 761, followed by gel-permeation chromatography. Presence of terpene was determined by the Salkowski colorimetric method and HPLC analysis. For an animal model of inflammation induction, mice were given an emulsion form of C.albicans cell wall mixed with Complete Freund's Adjuvant (CFA) by footpad-injection. Results showed that intraperitoneal administration of the water-soluble portion that contained terpene and flavonoid reduced the inflammation. Whereas the terpene had anti-inflammatory activity, flavonoid portion had no such activity, For determination of possible mechanism of the activity, the terpene seemed to be suppression of nitric oxide (NO) production from LPS-treated macrophages. Taken together the Ginkgo terpene may have anti-inflammatory effect against C.albicans-caused arthritis, possibly by blocking NO production.
Candida spp. are yeast form fungi, which cause an opportunistic infections in a immune suppressed patients however it is a normal flora of the respiratory system, the gastrointestinal system, and the urogenital system of healthy person. It is investigated that the distribution of Candida spp. cause an oral disease from oral cavity of healthy people and also identified Candia dubliniensis. Distribution and identification of the yeast form fungi in oral cavities of healthy people was investigated by an automatic identifier, VITEK2 system. We found 21 strains of Candida albicans, 3 strains of Candida famata, one strain of Candida tropicalis, Candida haemulonii, Candida krusei, and Candida dubliniensis. In addition, one strain of Cryptococcus spp., Saccharomyces spp., and two unknown strains were isolated. Candida dubliniensis which forms a mass by more than 2 chlamydospores was isolated from a healthy person for the first time. Candida dubliniensis was not grown at $42^{\circ}C$ whereas Candida albicans was grown well. It is known that Candida dubliniencis was isolated in AIDS while it is found in healthy people from this study, which will be helpful to investigate the distribution of Candida spp.

The antimicrobial activity of methanol extracts from 17 seaweeds was screened using a paper disc method and using three human skin pathogens: Staphylococcus aureus, S. epidermidis and Candia albicans. The serial extraction of Neorhodomela aculeata was also conducted using four different solvents (n-hexane, chloroform, ethyl acetate, and methanol) and the minimal inhibitory concentration (MIC) of each extract was examined for the three pathogens. Of the 17 seaweeds, the MeOH extracts of Ulva conglobata, N. aculeata and Symphyocladia latiuscula showed antimicrobial activities. For the extracts from N. aculeata and S. latiuscula, the inhibition zones were more than 10 mm in diameter against S. aureus and S. epidermidis, and >7mm for C. albicans. The inhibition zone of U. conglobata treatment was about 8 mm for S. aureus only. The MIC of each N. aculeata extract ranged from 8 to 32 mg/mL against the three human skin pathogens, and the lowest value (8 mg/mL was with the methanol extract. These results suggest that the MeOH extract of N. aculeata might be useful for developing new antibiotics against human skin pathogens.

Candida albicans is one of the most frequently isolated fungal pathogens in human. Recently, the prevalence of candida infection has markedly increased, partially due to the increase of immunocompromised hosts. Proposed virulence factors of the pathogenic Candida are the ability to form hyphae to adhere to epithelial cell surfaces, and to secrete acid proteinases and phospholipases. We measured the relative cell surface hydrophobicity (CSH) and the ability of proteinase production (PROT), phospholipase production (PLase), adherence to host epithelium (ADH), and hyphal transition (Germ). The relative risk of virulence factors was analyzed by lethality test in murine model of hematogeneously disseminated candidal infection. According to Cox's proportional hazard analysis, the statistically significant virulence factors were PROT, ADH, and CSH. PROT was the highest risk factor of them. To evaluate the applicability for the diagnosis and treatment of Candidiasis, we examined the protective effect of the active and passive immunizations with the materials purified from virulence factors and antibodies to them in Candia-infected mice model. The mean survival times of active and passive immunized groups were slightly longer than those of non-immunized groups.
Objectives : The present study was designed to investigate effects of mixed extracts (9:1, v/v) of Hwangryunhaedoktang, Chamaecyparis obtusa essential oil. We evaluated the antimicrobial and antioxidant activity by manufacturing mixed extracts as the materials for functional medicinal herb cosmetics. Methods : We performed antimicrobial were tested microbes (Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Propionibacterium acnes) by disc diffusion method and measure clear zone. Antioxidant activities were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. Results : Antimicrobial activities of mixed extracts against S. epidermideis, P. acnes, S. aureus, E. coli, and Candida. A were $10.9{\pm}3.0mm$, $16.9{\pm}2.0mm$, $9.2{\pm}2.0mm$, $10.3{\pm}1.0mm$, $11.8{\pm}3.0mm$ respectively. The Chamaecyparis obtusa essential oil had the highest antimicrobial activities against S. epidermideis, P. acnes, S. aureus, E. coli, and Candia. A and clear zone of microbes ware $16.7{\pm}3.0mm$, $28.8{\pm}0.2mm$, $15.9{\pm}2.0mm$, $11.5{\pm}1.0mm$, $16.3{\pm}3.0mm$. Hwangryunhaedoktang extract showed antimicrobial activity but only P. acnes and S. aureus The antioxidant activities of the mixed extracts were tested through the evaluation of DPPH radical scavenging activity. The 100 % mixed extracts were found to have 90 % DPPH radical scavenging activity. The mixed extracts was presented similar antioxidant activities compared with that of ascorbic acid. As a result, A mixture extract is expected to have antimicrobial effects and free radical scavenging activity was found. Conclusions : Accordingly, It can be concluded that mixed extracts has the potential to cosmetic material.
