Sensing NO3-N and K Ions in Hydroponic Solution Using Ion-Selective Membranes (이온선택성 멤브레인을 이용한 양액 내 질산태 질소 및 칼륨 측정)

  • Rapid on-site sensing of nitrate-nitrogen and potassium ions in hydroponic solution would increase the efficiency of nutrient use for greenhouse crops cultivated in closed hydroponic systems while reducing the potential for environmental pollution in water and soil. Ion-selective electrodes (ISEs) are a promising approach because of their small size, rapid response, and the ability to directly measure the analyte. The capabilities of the ISEs for sensing nitrate and potassium in hydroponic solution can be affected by the presence of other ions such as calcium, magnesium, sulfate, sodium, and chloride in the solution itself. This study was conducted to investigate the applicability of two ISEs consisting of TDDA-NPOE and valinomycin-DOS PVC membranes for quantitative determinations of $NO_3$-N and K in hydroponic solution. Nine hydroponic solutions were prepared by diluting highly concentrated paprika hydroponic solution to provide a concentration range of 3 to 400 mg/L for $NO_3$-N and K. Two of the calibration curves relating membrane response and nutrient concentration provided coefficients of determination ($R^2$) > 0.98 and standard errors of calibration (SEC) of < 3.79 mV. The use of the direct potentiometry method, in conjunction with an one-point EMF compensation technique, was feasible for measuring $NO_3$-N and K in paprika hydroponic solution due to almost 1:1 relationships and high coefficients of determination ($R^2$ > 0.97) between the levels of $NO_3$-N and K obtained with the ion-selective electrodes and standard instruments. However, even though there were strong linear relationships ($R^2$ > 0.94) between the $NO_3$-N and K concentrations determined by the Gran's plot-based multiple standard addition method and by standard instruments, hydroponic $NO_3$-N concentrations measured with the ISEs, on average, were about 10% higher than those obtained with the automated analyzer whereas the K ISE predicted about 59% lower K than did the ICP spectrometer, probably due to no compensation for a difference between actual and expected concentrations of standard solutions directly prepared.

  • A gene encoding a putative phospholipase D was isolated from Bacillus licheniformis and cloned into pGEM-T easy vector. The gene was expressed in E. coli BL21 (DE3) using a pET-21(a) vector containing His6 tag. Affinity purification of the recombinant phospholipase D with nickel-nitrilotriacetic acid (Ni-NTA) resin resulted major one-band by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis. The purified enzyme showed a molecular weight of 44 kDa. The optimum activity of enzyme was around pH 7.0 and the enzyme was also the most stable around this condition. The optimum temperature was about $40-45^{\circ}C$ and the enzyme still showed considerable activities at wide range of temperature. Among various detergents, Triton X-100 significantly increased the enzyme activity, resulting in 181% activity of control at 0.6 mM of the detergent. Calcium ion did not significantly affect the enzyme activity, suggesting that the enzyme might be classified into $Ca^{2+}$-independent PLD.

Removal of Aqueous Iron Ion by Micellar Enhanced Ceramic Microfiltration Adding Surfactant (계면활성제를 첨가한 미셀 형성 세라믹 정밀여과에 의한 용존 철 이온 제거)

  • In this study sodium dodecyl sulfate (SDS), which was anionic surfactant, was added for forming micelles to remove iron ion that could be contained with small amount in industrial water. Then aggregates binding between iron ions and micelles were rejected by a ceramic microfiltration membrane. As result of SDS concentration effect on removal rates of iron and SDS in modified iron solution, the removal rate of iron was the highest value of 92.26% and the removal rate of SDS was 61.10% a little higher than the result of calcium ion at 8 mM which was CMC (Critical micelle concentration) of SDS. As final resistance of membrane fouling $R_f$ increased the more at the higher SDS concentration, it showed the highest value at 4 mM and the lowest at 10 mM of SDS. The final permeate flux $J_{180}$ had the highest value and the largest total permeate volume could be finally acquired at SDS 10 mM. In case of CMC 8 mM, low $R_f$ was shown as same as that of 10 mM until 80 minutes of operation, and tended to increase dramatically to 120 minutes and increase slowly again until 180 minutes.

Hydrogeochemical study of a watershed in Pocheon area: controls of water chemistry

  • The groundwater in the Pocheon area occurs from both a fractured bedrock aquifer in igneous and metamorphic rocks and an alluvial aquifer with a thickness of <50 m, and forms a major source of domestic and agricultural water supply. In this study, we performed a hydrogeochemical study in order to identify the control of geochemical processes on groundwater quality. For this study, groundwater level and physicochemical parameters (EC, Eh, pH, alkalinity) were monitored once a month from a total of 150 groundwater wells between June 2003 to August 2004. A total of 153 water samples (13 surface water, 66 alluvial groundwater, 74 bedrock groundwater) were also collected and analyzed in February 2004. Groundwater chemistry in the study area is very complex, depending on a number of major factors such as geology, degree of chemical weathering, and quality of recharge water. Hydrochemical reactions such as the leaching of surficial and near-solace soil salts, dissolution of calcite, cation exchange, and weathering of silicate minerals are proposed to explain the chemistry of natural groundwater. Alluvial groundwaters locally have very high TDS concentrations, which are characterized by their chloride(nitrate)-sulfate-bicabonate facies and low Na/Cl ratio. Their grondwater levels are highly fluctuated according to rainfall event. We suggest that high nitrate content and salinity in such alluvial groundwaters originates from the local recharge of sewage effluents and/or fertilizers. Likewise, high concentrations of nitrate were also locally observed in some bedrock groundwaters, suggesting their effect of anthropogenic contamination. This is possibly due to the bypass flow taking place through macropores. Tile degree of the weathering of silicate minerals seems to be a major control of the distribution of major cations (sodium, calcium, magnesium, potassium) in bedrock groundwaters, which show a general increase with increasing depth of wells. Thermodynamic interpretation of groundwater chemistry shows that the groundwater in the study area is in chemical equilibrium with kaolinite and Na-montmorillonite, which indicates that weathering of plagioclase to those minerals is a major control of hydrochemistry of bedrock groundwater. The interpretation of the molar ratios among major ions, as well as the mass balance calculation, also indicates the role of both dissolution/precipitation of calcite and Ca-Na cationic exchange as bedrock groundwaters evolves progressively.

Removal of Ca2+ and Fe3+ in Acid Mine Drainage by Tripolyphosphates (트리폴리인산염을 이용한 산성광산배수 내 칼슘 및 철이온 제거)

  • This study evaluates tripolyphosphate's ability to treat AMD (Acid Mine Drainage). Based on the batch test results for reaction between tripolyphosphate and AMD obtained from Munkyung coal mine, $4.7{\times}10^{-3}$ mole is the optimum dosage of tripolyphosphate for AMD treatment. $Ca^{2+}$ concentration is decreased from $16.4mg/{\ell}$ to $5.6mg/{\ell}$, in other words, the removal rate of $Ca^{2+}$ is 65.9%. $Fe^{3+}$ concentration is decreased from $3.7mg/{\ell}$ to $0.02mg/{\ell}$, that is, the removal rate of $Fe^{3+}$ is 99.5%. $SO{_4}^{2-}$ concentration ranges from $526.8mg/{\ell}$ to $566.5mg/{\ell}$, which shows no obvious decrease. After dosing up tripolyphosphate, $Na^+$ concentration in AMD ranges from $549.8mg/{\ell}$ to $599.3mg/{\ell}$ and orthophosphate concentration in AMD ranges from $6.82mg/{\ell}$ to $7.60mg/{\ell}$. It was found that the precipitate in the order of amount is Apatite${\gg}{\beta}$-tricalcium phosphate > $Fe(OH)_3$ from SEM, XRF, XRD analyses. Consequently, the treatment by tripolyphosphate is effective in pH buffering and in the removal of $Ca^{2+}$ and $Fe^{3+}$.

  • The effects of CaO, $CaO+MgSO_4$, and $CaO+MgSO_4$+compound fertilizers(NPK) on soil chemical properties of acidified forest soils were studied in Quercus spp. and Pinus rigida stands in Mt. Namsan and Mt. Surak in urban areas, and Kwangnung in a mountain area. The soil samples were collected in November 1995 after every year fertilization from November 1991 through April 1995. The fertilizations affected soil chemical properties. Soil pH increased after fertilizations compared with control. However, the effect was different between the stand types and the areas. Organic matter and total nitrogen content were not changed, while exchangable cations such as calcium and magnesium increased after fertilizations. However, these ration concentrations after fertilizer treatments were lower in P. rigida than in Quercus spp. stands. These rations also showed increased leaching characteristics more in the urban area than in the mountain area.

Anticoagulant Activity of Sulfated Polysaccharides Isolated from Codium fragile (청각 산추출물에서 정제한 함황다당류의 항응고활성)

  • We have isolated two anticoagulant polysaccharides from an acidic extract of Codium fragile. The purification was conducted using three consecutive chromatographies of DEAE-Toyopearl 650C, Sephadex G-100 (G-75), and Sepharose CL-6B by measuring activated partial thromboplastin time (APTT). The two purified anticoagulant polysaccharides, CF-1-VIa-1 and CF-1-VIIa-1, were found to be nearly homogenous on HPLC using a gel permeation column and appeared to have molecular weights of about 80,000 Da and 40,000 Da, respectively. The polysaccharides consisted mainly of arabinose and galactose in a molar ratio of about 2 : 1, and also comprised 12-13% of sulfates at their constituent sugars. CF-1-VIa-1 and CF-1-VIIa-1 inhibited blood coagulation via both the intrinsic and the extrinsic pathways. The polysaccharides unlike heparin showed an inhibitory activity on thrombin when a pure fibrinogen without antithrombin III was used as a substrate. Structural modifications using sulfation and desulfation affected the anticoagulant activities directly, suggesting that the content of sulfate plays an important role in the blood coagulation cascade. The polysaccharides may inhibit some proteases involved in the blood coagulation cascade, judging from the independence of calcium concentrations in their anticoagulant activity.

Effects of Iron Supplementation on Iron Status of Anomic High School Girls (철 보충제 섭취가 빈혈 여고생의 철 영양상태에 미치는 영향)

  • This study was designed to investigate the effect of iron supplementation on the iron nutritional status and anemia of high school girls in Korea. One hundred thirty-five female students residing in Ulian metropolitan city in Korea diagnosed as having anemia or iron deficiency participated in this study. One or two tablets of iron medicine(80-160 mg Fe as ferrous sulfate/day) were administered to all participants for 3 months. Subjects were evaluated with a questionaire, measurement of hematological indices before and after iron supplementation. The average height and weight of respondents were 161.62 $\pm$ 4.68 cm and 53.87 $\pm$ 6.10 kg, respectively. Daily intakes of energy were 1597.8 $\pm$ 302.35 kcal(76.0% RDA). Iron intakes were 13.72 $\pm$ 4.17 mg (76.3% of RDA) and calcium intakes were 580.74 $\pm$ 177.21(72.5% of RDA) before iron supp]ementation. At baseline, 63% of all participants had depleted store(serum ferritin 12 ug/ml and/or transferrin saturation(TS) < 14%). After iron supplementation, this proportion declined to 19.3%. 55.6% of subjects had 12 ug/m1 of basal ferritin concentration before iron supplementation, and this proportion declined to 16.3% after iron supplementation. The basal hemoglobin(Hb) concentrations were 12.13 $\pm$ 1.01 g/dl and they increased to 12.79 $\pm$ 0.81 g/dl, which showed significant difference artier iron supplementation(p < 0.001). The basal ferritin and TS(%) were 13.24 $\pm$ 11.66 ng/ml, 18.42 $\pm$ 10.12% and they significantly increased to 32.95 $\pm$ 21.14 ng/ml, 33.53 $\pm$ 16.64%, respectively(p < 0.001). The basal total iron binding protein(TIBC) were 467.81 $\pm$ 97.24 ug/dl and they significantly decreased to 325.05 $\pm$ 48.89 ug/dl(p < 0.001) after iron supplementation. The number of tablets administered was positively correlated with serum iron(t = 0.553, p < 0.01), serum ferritin(t = 0.557, p < 0.01), TS(%)(t = 0.588, p < 0.01) and negatively correlated with TIBC(t= -0.409, p <0.01). The anemia symptoms such as ‘Shortening of breath when going upstairs(p < 0.01)’, ‘Tired out easily(p < 0.01)’, ‘Feeling blue(p < 0.001)’, ‘Decreased ability to concentrate(p < 0.01)’, and ‘Poor memory(p < 0.001)’improved significantly after iron supplementation. In this study, daily iron supplementations were efficacious in improving the iron status and anemic symptoms of female high school students. Regular check-ups and nutrition education for adolescents are necessary because of their vulnerability to iron deficiency. Further studies are needed to determine the minimum effective dose of iron and to examine the adverse effect of long-term iron supplementation.

Characteristics of White Pigments Used in Jiho Oh and Bonung Gu's Paintings Produced in Modern and Contemporary Period (근·현대 시대 오지호와 구본웅 유화작품에 사용된 백색계 안료의 특성 연구)

  • To investigate the pigments used in modern and contemporary oil paintings, thirty-two paintings by Jiho Oh and Bonung Gu were selected. The white pigment found in the ground and painting layers was identified as lead white (hydrocerussite), zinc white (zinc oxide), titanium white (titanium dioxide in anatase or rutile forms), calcite (calcium carbonate), and barite (barium sulfate). Further, this indicated that pigments differ according to the artist and date of the painting's creation. However, both Oh and Gu used zinc white during the modern and contemporary period, while lead white was replaced by titanium white, barite and calcite. Compared with the overseas studies on pigments and oil paints, the change patterns of pigments were the same with them but the periods of the use were partially different. It seems to be due to the fact that South Korea is linked to the historical background of the art material which was imported from Japan instead of Western countries. Therefore, it is inevitable that any change in the white pigments used for domestic oil paintings occurred at a different time from global transitions. If the results of this study are used in the analysis of art works it is suggested that a database recording such aspects as material properties of oil paints, artistic techniques, and chronology would become important for future conservation science and the study of art history.

Thermostable $\alpha$-Amylase Production by Thermophilic Bacillus sp. TR-25 lsolated from Extreme Enviroment (극한환경에서 분리한 고온성 Bacillus sp. TR-25에 위한 내열성 $\alpha$-amylase의 생산)

  • For screening thermostable $\alpha$-amylase from thermophiles, various samples from extreme environments such as hot spring and sewage near them, and compoat, wereexamined microbial growth in enrichment culture medium at 55$\circ$C on the assumption that enzymes from thermophiles are inevitable thermostable. One strain showing higher $\alpha$-amylase activity was pure cultured and designated as Bacillus sp. TR-25 from the results of morphological, cultural and physiological characteristics. The most important carbon sourses for the enzyme production were soluble starch, dextrin, potato starch and corn starch. Glucose and fructose had a catabolite repression on the enzyme production. The good nitrogen sources for the enzyme production were yeat extract, nutrient broth, tryptone, corn steep liquor and ammonium sulfate. The enzyme production was accelerated by addition of CaCl$_{2}$. $\cdot $ H$_{2}$O. The optimal medium composition for the enzyme production was soluble starch 2.0%, yeast extract 0.55, CaCl$_{2}$ $\cdot $ 2H$_{2}$O 0.015, Tween 80 0.001%, pH8.0, respectively. In jar fermenter culture, this strain shows a rapid growth and required cheaper carbon and nitrogen source. These properties are very useful to fermentation industry. The $\alpha$-amylase of this strain demonstrated a maximum activity at 80$\circ$C, pH 5.0, respectively. And calcium ion did not improve thermostability of the enzyme. At 10$0^{\circ}C$, this enzyme has 235 of relative activity. Transformation was carried out by thermophilic Bacillus sp. TR-25 genomic DNA. As a result, the transformant has increased thermostable $\alpha$-amylase activity.
