High Calcium in the Diet and Vitamin D Inhibit a Bone Matrix Degrading Enzyme $\beta$-Glucuronidase Activity

  • A lysosomal and matrix degrading enzyme $\beta$-glucuronidase activity was measured in BALS/c mice fed high and low Ca in combination with the i.p. adminstration of calcium-regulating hormones including parathyoid hormone(PTH), calcitonin(CT) and cholecalciferol(Vit D). After feeding experimental diets for five weeks, mice were sacrificed by cervical dislocation and the enzyme was fluometrically measured at 440nm. $\beta$-Glucuronidase activity was inhibited by high calcium in the diet. in addition, vitamin D also inhibited the enzyme activity in the serum regardless of the level of dietary calcium. In contrast, PTH has shown to stimulate the enzyme at all the levels of dietary calcium. Calcitonin, and inhibitor of PTH action for bone resorption, revealed to curb PTH effect in this enzyme, whereas CT stimulated the action of vitamin D in the serum. The above results led us to conclude that osteoclastic bone resorption and senile osteoporosis may be reduced by adequate dietary calcium and vitamin D.

  • The present communication deals with the Pharmacophore modeling, 3D QSAR and docking analysis on series of Pyrimidine derivatives as potential calcium channel blockers. The computational studies showed hydrogen bond donor, hydrogen bond acceptor, and hydrophobic group are important features for calcium channel blocking activity. These studies showed that Pyrimidine scaffold can be utilized for designing of novel calcium channels blockers for CVS disorders.

Activation of Phospholipase D in Rat Thymocytes by Sphingosine

  • Sphingosine is known to regulate a wide range of cell physiology including growth, differentiation, and apoptosis. In this study, we examined the effect of sphingosine on the phospholipase D (PLD) activity in rat thymocytes. Sphingosine potently stimulated PLD in the absence of extracellular calcium, while depletion of intracellular calcium by BAPTA/AM treatment completely blocked activation of PLD by sphingosine. Sphingosine-induced increase of the intracellular calcium concentration was confirmed using a fluorescent calcium indicator Fluo-3/AM. A phosphoinositide-specific phospholipase C inhibitor U73122 partially inhibited the stimulation of PLD by sphingosine. When mouse PLD2 gene was transfected into mouse thymoma EL4 cells, which lack intrinsic PLD activity, sphingosine could stimulate PLD2 significantly while overexpression of human PLD1 had no effect. Taken together, the sphingosine-stimulated PLD activity in rat thymocytes is dependent on the mobilization of intracellular calcium and appears to be due to the PLD2 isoform.

Functional Properties of Calcium Powder of Cuttle Bone Treated with Acetic Acid (아세트산 처리 갑오징어갑 칼슘제의 기능적 특성)

  • The functional properties such as a firming maintenance capacity for fermented vegetables, protein coagulation capacity antibacterial activity, calcium and peroxide absorption capacity of calcium powder of cuttle bone treated with acetic acid (ATC) were examined. In the Korean pickled cucumbers (20 days), the calcium content of ethanol insoluble solids was 5,264.9 mg/100 g and this value was higher than that of control (2,036.4 mg/100 g). Protein coagulation capacity of ATC was $0.9\%$ and its calcium absorption capacity was $49.3\%.$ The peroxide absorption capacity of ATC was shown $0-1.1\%$ range. Antibacterial activity was detected above $0.2\%.$ The ATC was effective in a firming maintenance of fermented vegetables (cucumber), coagulation capacities for soybean milk and antibacterial activity. The ATC was not effective in clarifying wastewater. Peroxide absorption capacity toward linoleic acid could not be checked by ATC, but its calcium absorption ratio was high compared to that of commercial calcium powder.

  • Calcium promoted ethylene production from mungbean hypocotyl segments incubated in the presence of either auxin or cytokinin (kinetin). Time course studies indicated that the calcium effect on ethylene production had a longer latent period (about 6 h) in combination with kinetin than with auxin. Studies on the effects of agents that are known to interfere with either action or transport (uptake) of calcium on ethylene biosynthesis indicated different patterns between auxin- and kinetin-treated tissues. Auxin-induced ethylene production was inhibited by the calmodulin inhibitor, trifluoperazine (TFP), and this inhibition was overcome by high concentrations of calcium applied, but TFP had no significant effect on kinetin-induced ethylene production regardless of calcium in the medium. The calcium channel blocker, verapamil, inhibited auxin-induced, but had little effect on kinetin-induced, ethylene producton. In vivo activity of "ethylene forming enzyme (EFE)" was found to be substantially promoted by calcium treatment. The enzyme activity was further increased by kinetin when segments were simultaneously treated with calcium, but auxin did not have such an effect.an effect.

Imaging and analysis of genetically encoded calcium indicators linking neural circuits and behaviors

  • Confirming the direct link between neural circuit activity and animal behavior has been a principal aim of neuroscience. The genetically encoded calcium indicator (GECI), which binds to calcium ions and emits fluorescence visualizing intracellular calcium concentration, enables detection of in vivo neuronal firing activity. Various GECIs have been developed and can be chosen for diverse purposes. These GECI-based signals can be acquired by several tools including two-photon microscopy and microendoscopy for precise or wide imaging at cellular to synaptic levels. In addition, the images from GECI signals can be analyzed with open source codes including constrained non-negative matrix factorization for endoscopy data (CNMF_E) and miniscope 1-photon-based calcium imaging signal extraction pipeline (MIN1PIPE), and considering parameters of the imaged brain regions (e.g., diameter or shape of soma or the resolution of recorded images), the real-time activity of each cell can be acquired and linked with animal behaviors. As a result, GECI signal analysis can be a powerful tool for revealing the functions of neuronal circuits related to specific behaviors.

Effects of Calcium and Galactose on the Ethylene Production of Persimmon Fruits (감과실의 에틸렌 생성에 미치는 칼슘과 Galactose의 영향)

  • This study was carried out to investigate the effects of calcium and galactose treatments on ethylene productions in persimmon fruits for the study on the study of persimmon fruits. Ethylene was producted in green mature persimmon fruits treated with water, calcium and galactose after 24hrs of treatment. Ethylene productions of persimmon fiuits treated with galactose was very higher than those of persimmon fruits treated with water and calcium after 72hrs of treatment. Ethylene productions of persimmon fruits teated with water and calcium were similarly to that of persimmon fruit tested with calcium. The treatment of glucose was not effected on ethylene production of persiommn fruits. The ACC contents and ACC synthase activity in persimmon fruit treated with galactose were higher than those of other groups after 72hrs of storage, but the ACC contents and ACC synthase activity of persimmon fruits treated with calcium were lower than those of control and persimmon fruits treated with water.

Effects of Amydae Carapax on Bone Metabolism in Ovariectomized Rats (난소 절제 흰쥐의 골대사에 미치는 별갑의 영향)

  • Objectives : This study was undertaken to investigate the effects of Amydae Carapax (AC) on parameter related to bone metabolism in ovariectomized rats. Methods : We measured alkaline phosphatase activity and contents of estrogen, calcium, hydroxyproline, osteocalcin, calcitonin and parathyroid hormone after the ovariectomized rats were treated with AC for 30 days. Results : Serum estrogen, calcium and calcitonin contents in ovariectomized rats significantly decreased, but increased after AC treatment. [Significant increase of serum alkaline phosphatase activity, parathyroid hormone activity and osteocalcin content in ovariectomized rats was remarkably decreased by AC treatment. Increase of urinary calcium and hydroxyproline content in ovariectomized rats was decreased by AC treatment.] Conclusions : These results shows that AC has the ability to counteract abnormal calcium metabolic processes due to sex hormone inequality, promoting bone absorption and inhibiting bone formation.

  • The purpose of this study is to enhance the photocatalytic properties of a mixture of low-cost titanium dioxide and calcium carbonate through a simple mixing process. To increase the photocatalytic activity, the weight ratio of titanium dioxide to calcium carbonate was selected as a variable. Photocatalytic activity was evaluated by measuring the degradation of methylene blue and NOx under ultraviolet light. The results showed that a mixture containing 60% titanium dioxide and 40% calcium carbonate exhibited the highest photocatalytic activity.

Isolation of a Calcium-Binding Fraction from a Hot-Water Extract of Smilax rhizoma (청미래덩굴 뿌리 열수 추출물로부터 칼슘 결합 물질의 분리)

  • We isolated a calcium-binding substance from Smilacis rhizoma hot-water extract using ion exchange, normal phase HPLC, and gel filtration chromatography; fractions were analyzed for calcium-binding activity. Fractions (F6) with the highest calcium-binding activity from the resource Q coulmn were pooled and further purified on an $NH_2$ column. Two major peaks were separated and the fraction (F61) with the higher calcium-binding activity was then loaded onto a $Superdex^{TM}$ column. A single peak (F611) with calcium-binding activity was finally obtained. These results suggest that the isolated calcium-binding fraction could be used as a functional food additive, similar to a calcium supplement, in the food industry.
