Effect of Ensilage of Rye Treated with Formic Acid and Lactic Acid Bacteria Inoculant on Ruminal Fermentation Characteristics (개미산과 유산균제 첨가 베일 사일리지의 발효 차이가 반추위 발효 특성에 미치는 영향)

  • This study investigated the effects of silage additives on rumen fermentation characteristics of rye silage. Rye was harvested at ripening stage and treated with different additives in quadruplicate following: without additive (control), with either lactic acid bacteria inoculant (LAB), formic acid (FA), or Ca-formate (Ca-FA). Overall, ensiling characteristics of FA and Ca-FA silages contained 4-fold more (P<0.05) butyrate and 2-fold more (P<0.05) NH3-N concentration (% total nitrogen) than those of control and LAB silages. Cows fed LAB silage showed a diurnal trend with the highest values of propionate concentration compared to the control at 1, 2 and 3 hr after feeding. In contrast, FA and Ca-FA silages increased the proportion of butyrate significantly (P<0.05) at all sampling times compared to control and LAB silage. In conclusion, Forage rye treated with FA or Ca-FA showed different fermentation characteristics during ensilage and in the rumen compared to LAB silage. Further studies are needed to evaluate whether different fermentation characteristics in the rumen between LAB and FA silages had effect on partitioning of nutrients between milk production and body tissue.

Groundwater Quality Characteristics of Pollution Concerned Area in Gyeongnam Using Groundwater Quality Monitoring Data (지하수수질측정망 자료를 활용한 경남 오염우려지역의 지하수 수질 특성)

  • This study analyzed the groundwater quality characteristics according to the main source of pollution and quarter (season) by using data from the pollution exclusive monitoring network in the Gyeongsangnam-do area for five years (2013-2017). The main source of pollution was the industrial complex areas, waste mines, and sewage treatment facilities. The analysis items were field measurement items (water temperature, pH, electrical conductivity, dissolved oxygen, oxide reduction potential), positive ions, and negative ions. Water temperature and pH did not vary significantly according to the main source of pollution. In industrial complex areas, the value of electrical conductivity was the highest, and dissolved oxygen value was the lowest. The mean concentration of positive and negative ions was the largest in industrial complex areas, followed by sewage treatment facilities and waste mines. It was shown that the concentration of sodium ion was the highest in industrial complex areas and calcium ion in waste mines and sewage treatment facilities. The concentration of bicarbonate ion was the highest in all main sources of pollution. Water temperature, pH, and concentrations of cations and anions did not vary significantly from quarter to quarter. Of the water quality types, the Na-HCO3 type accounted for the highest proportion, but the Na-Cl type, which has a high possibility of external contamination, accounted for about 20% of the total data in the pollution exclusive monitoring network.

Characterization of ginsenoside compound K loaded ionically cross-linked carboxymethyl chitosan-calcium nanoparticles and its cytotoxic potential against prostate cancer cells

  • Backgroud: Ginsenoside compound K (GK) is a major metabolite of protopanaxadiol-type ginsenosides and has remarkable anticancer activities in vitro and in vivo. This work used an ionic cross-linking method to entrap GK within O-carboxymethyl chitosan (OCMC) nanoparticles (Nps) to form GK-loaded OCMC Nps (GK-OCMC Nps), which enhance the aqueous solubility and stability of GK. Methods: The GK-OCMC Nps were characterized using several physicochemical techniques, including x-ray diffraction, transmission electron microscopy, zeta potential analysis, and particle size analysis via dynamic light scattering. GK was released from GK-OCMC Nps and was conducted using the dialysis bag diffusion method. The effects of GK and GK-OCMC Nps on PC3 cell viability were measured by using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide assay. Fluorescent technology based on Cy5.5-labeled probes was used to explore the cellular uptake of GK-OCMC Nps. Results: The GK-OCMC NPs had a suitable particle size and zeta potential; they were spherical with good dispersion. In vitro drug release from GK-OCMC NPs was pH dependent. Moreover, the in vitro cytotoxicity study and cellular uptake assays indicated that the GK-OCMC Nps significantly enhanced the cytotoxicity and cellular uptake of GK toward the PC3 cells. GK-OCMC Nps also significantly promoted the activities of both caspase-3 and caspase-9. Conclusion: GK-OCMC Nps are potential nanocarriers for delivering hydrophobic drugs, thereby enhancing water solubility and permeability and improving the antiproliferative effects of GK.

Bone mineral density in type 2 diabetic patients aged 50 years or older in men and postmenopausal women in Korea

  • Relationship between bone mineral density (BMD) and type 2 diabetes is still inconsistent. Recently, many epidemiologic data show that fracture risk is increased in type 2 diabetic patients regardless of BMD status. In this study, we used nation-wide data from 2008 to 2011 Korea National Health and Nutrition Examination Survey (KNHANES) to analyze the BMD status in patients with type 2 diabetes compared to non-diabetics. We included subjects aged 50 years or older in men (N=2,959, 2,430 without diabetes, 529 with type 2 diabetes) and postmenopausal women (N=2,902, 2,479 without diabetes, 423 with type 2 diabetes). Subjects with history of medication for osteoporosis or with illness or malignancy affecting bone metabolism were excluded. Data of anthropometric measurements and demographic characteristics were collected by trained examiner. Serum was separated from peripheral venous blood samples obtained after 8 hours of fasting. BMD was measured at lumbar spine and femur using dual-energy X-ray absorptiometry (DXA). There was a significant positive association between lumbar spine BMD and type 2 diabetes after adjusting age, gender, body mass index, monthly house income, education level, physical activity, daily calcium intake and vitamin D concentration by multiple regression analysis in all subjects. In the subgroup analysis by gender, this association was maintained both in male and female after adjusting those confounding factors. However, femur BMD was not different between type 2 diabetic and non-diabetic subjects. In conclusion, lumbar spine BMD was significantly higher in type 2 diabetic patients aged 50 years or more in men and postmenopausal women compared to non-diabetic subjects.

The association between body composition and bone mineral density in subjects aged 50 years or older in men and postmenopausal women in Korea

  • The effect of body composition such as lean mass and fat mass on bone mineral density (BMD) is complex and still controversial. In this study, we investigated the relationship between body composition and bone mineral density using nation-wide data from 2008 to 2011 Korea National Health and Nutrition Examination Survey (KNHANES) in 2,139 men and 2,193 postmenopausal women aged 50 years or older. Subjects with history of medication for osteoporosis or with diseases or malignancy affecting bone metabolism were excluded. Data of anthropometric measurements and demographic characteristics were collected by trained examiner. Fasting blood sample was obtained for blood chemistry analysis. BMD of the lumbar spine, total femur, and femoral neck, and body composition such as total lean mass (TLM), total fat mass (TFM), truncal fat mass (TrFM) were measured using dual-energy X-ray absorptiometry (DXA). There were significant positive correlations between body composition indices such as lean mass and fat mass with BMD. In multiple regression analysis, TLM was positively associated with BMD after adjusting age, body mass index, monthly house income, education level, physical activity, daily calcium intake and vitamin D concentration in both men and postmenopausal women. BMD at lumbar spine and femur in lowest quartile of TLM was significantly lower than other quartiles after adjusting those confounding factors in both gender. TrFM was negatively associated with total femur BMD in male and femur neck BMD in postmenopausal women after adjusting confounding factors. In conclusion, TLM is very important factor in maintaining BMD in subjects aged 50 years or older in men and postmenopausal women.

Comparison of Antifungal Activity, Plant Growth Promoting Activity, and Mineral-Solubilizing Ability of Bacillus sp. Isolated from Rhizosphere Soil and Root (근권 토양과 뿌리로부터 분리된 Bacillus sp.의 항진균 활성, 식물 생장 촉진 활성 및 미네랄 가용화능 비교)

  • The purpose of this study was to evaluate the antifungal activity, plant-growth-promoting activity, and mineral solubilization ability of 10 species of phytopathogenic fungi to select a Bacillus sp. from rhizosphere soils and roots that can be used as a microbial agent. The antifungal activity for phytopathogenic fungi varied based on the Bacillus sp. Among the selected strains, DDP4, DDP16, DDP148, SN56, and SN95 exhibited antifungal activity for nine or more species of phytopathogenic fungi. Regarding nitrogen-fixation ability, all Bacillus sp. showed similar levels of activity, and siderophore production ability was relatively high in ANG42 and DDP427. The indole-3-acetic acid production abilities were in the range of 1.83-67.91 ㎍/ml, with variations in activity based on the Bacillus sp. One strain with a high activity was selected from each species, and their mineral solubilization abilities were examined. Most Bacillus sp. could solubilize phosphoric acid and calcium carbonate, and DDP148 and SN56 could solubilize silicon and zinc, respectively. These results suggested that Bacillus sp. can be considered potential multi-purpose microbial agents for plant growth promotion and disease prevention.

Synthesis parameters of hydroxyapatite preparation by a precipitation process (합성조건이 침전법에 의한 Hydroxyapatite 제조에 미치는 영향)

  • Hydroxyapatite (HAp) was synthesized from calcium hydroxide (Ca(OH)2) reacting with phosphoric acid (H3PO4) in aqueous solution. HAp powders were synthesized from extremely high concentration of precursor solutions over 3 M of Ca(OH)2 aqueous suspension using modified process parameters such as phosphoric acid (H3PO4) pouring rate, aging time and post ball milling process. Regardless of phosphoric acid pouring rate, the DCPD (dicalcium phosphate dihydrate) was formed at room temperature and when heated above 700℃, β-TCP (tricalcium phosphate) was synthesized and the amount reached its maximum at 900℃. When the synthesized powder was sintered at 1150℃, β-TCP, a high temperature impurity phase, remained. The single HAp phase without DCPD was obtained from post ball-milled precipitates followed by 3 day aging. For the ball-milled precipitates even without the aging process, the desired single HAp phase without β-TCP could be obtained by heat treatment above 500℃. The post ball milling process provided a convenient route for HAp synthesis.

Electrochemical Deposition Characteristics of Ca2+ on Cu Wire Electrode in CaCl2 Molten Salt (CaCl2 용융염에서 Ca2+의 Cu 전극에 대한 전기화학적 증착 특성평가)

  • With the expansion of the automobile market, the demand for Nd as an essential rare earth material for automobile motors is rapidly increasing. Research on the calcio-thermic reduction process between Nd2O3 and calcium-based alloys has been extensively studied in order to manufacture Nd. In this study, Ca-Cu, as a reducing for Nd2O3, was prepared by electrolysis in CaCl2 molten salt. Cu wire and graphite were employed as a working electrode and a counter electrode for electrolysis reaction, respectively. The reference electrode was manufactured by putting Ag wire in a mixture of AgCl and CaCl2 at a ratio of 1:99 mol%. The cyclic voltammetry results showed that the deposition of Ca2+ on the surface of working electrode was observed from a potential of -1.8 V, and the reduction potential of Ca2+ decreased as the reaction temperature increased. The diffusion coefficient of Ca2+ calculated by the chronoamperometry experiment was found to be 5.4(±6.8)×10-6 cm2/s. In addition, Ca-Cu liquid alloy was prepared by applying a constant potential to Cu electrodes. The element ratio of Ca-Cu alloy formed by applying a potential of -2.0 V was found to Ca:Cu=1:4.

Development and Quality Characteristics of Superheated Steam Roasted Small Yellow Croaker Larimichthys polyactis, Sweet Potato Salad and Tomato Mousse for Senior-friendly Seafood (참조기(Larimichthys polyactis)를 활용한 고령친화식품용 과열증기구이, 고구마 샐러드 및 토마토무스의 개발 및 품질특성)

  • This study was conducted to prepare superheated steam roasted (SSR) small yellow croaker Larimichthys polyactis sweet potato salad (SPS) and tomato mousse (TM), as senior-friendly seafoods (SFS) and to examine their quality characteristics. The hardness values of SSR, SPS and TM were 110.5×1.03, 22.4×1.03 and 14.5×1.03 N/m2, respectively. The nutritional properties of SSR, SPS and TM were 22.4, 6.7 and 10.2 g/100 g, respectively, for protein, 97.01, 3.40 and 17.00 ㎍/100 g, respectively, for vitamin A, 10.26, 2.79 ㎍/100 g and not detected (ND), respectively, for vitamin D, 60.68 mg/100 g, ND and 4.17 mg/100 g, respectively, for vitamin C, 0.18, 0.10 and 0.33 mg/100 g, respectively, for riboflavin 24.28, 10.91 and 13.77 mg/100 g, respectively, for niacin, 76.3, 53.0 and 48.5 mg/100 g, respectively, for calcium, 258.2, 231.3 and 145.2 mg/100 g, respectively, for potassium, and ND, 0.70 mg/100 g and ND, respectively, for dietary fiber. Escherichia coli was not detected in any of the products. These results suggested that the products should be classified as step 1 for SSR, step 2 for SPS and step 3 for TM based on the SFS standard of the Korean industrial standards. Moreover, the nutritional and physical properties of the foods were improved.

Group 1 metabotropic glutamate receptor 5 is involved in synaptically-induced Ca2+-spikes and cell death in cultured rat hippocampal neurons

  • Group 1 metabotropic glutamate receptors (mGluRs) can positively affect postsynaptic neuronal excitability and epileptogenesis. The objective of the present study was to determine whether group 1 mGluRs might be involved in synaptically-induced intracellular free Ca2+ concentration ([Ca2+]i) spikes and neuronal cell death induced by 0.1 mM Mg2+ and 10 µM glycine in cultured rat hippocampal neurons from embryonic day 17 fetal Sprague-Dawley rats using imaging methods for Ca2+ and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assays for cell survival. Reduction of extracellular Mg2+ concentration ([Mg2+]o) to 0.1 mM induced repetitive [Ca2+]i spikes within 30 sec at day 11.5. The mGluR5 antagonist 6-Methyl2-(phenylethynyl) pyridine (MPEP) almost completely inhibited the [Ca2+]i spikes, but the mGluR1 antagonist LY367385 did not. The group 1 mGluRs agonist, 3,5-dihydroxyphenylglycine (DHPG), significantly increased the [Ca2+]i spikes. The phospholipase C inhibitor U73122 significantly inhibited the [Ca2+]i spikes in the absence or presence of DHPG. The IP3 receptor antagonist 2-aminoethoxydiphenyl borate or the ryanodine receptor antagonist 8-(diethylamino)octyl 3,4,5-trimethoxybenzoate also significantly inhibited the [Ca2+]i spikes in the absence or presence of DHPG. The TRPC channel inhibitors SKF96365 and flufenamic acid significantly inhibited the [Ca2+]i spikes in the absence or presence of DHPG. The mGluR5 antagonist MPEP significantly increased the neuronal cell survival, but mGluR1 antagonist LY367385 did not. These results suggest a possibility that mGluR5 is involved in synaptically-induced [Ca2+]i spikes and neuronal cell death in cultured rat hippocampal neurons by releasing Ca2+ from IP3 and ryanodine-sensitive intracellular stores and activating TRPC channels.
