Contamination of heavy metals in water and sediments along Mangyeong Gang(river) has reached up to critical level The object of the study is to elucidate somed molluscs which inhabit along the river on respect to the modulatory role in reducing the pollution. For the purpose, molluscs which are common in numbers and biomass in the area were collected, and Cadmim(Cd) was subjected as a reference metal in the experiment. The corresponding species were; Cipango;aludina chinensis (muddy snail) and Scapharca subcrenata (seashell). As methods, sample species were kept in laboratory under the natural condition as possible. Soil were brought from the site of the collection, dried autoclaved and wetted with ordinary water. It wad utilized as media to maintain the collected species in vitro all the way of the experiment. CdCI$^{2}$ was mixe in the medium according to experimental design. On the result obtained in the study, it is summarized that molluscs which inhabit along Mangyeong Gang(river)consume heavy mital-containing matters, so far Cd is concerned in the study. The amout of Cd concentration in tissues of the benthic natured invertebrates were dose and time related, and MT-Cd was also similar trend. Thus, Cd may eventually combine with low molecular protein forming metalloprotein, then reduce the toxicity of the heavy metal.
This study was conducted to investigate the antitoxic component in ethanol extract of Saururus chinesis (S. chinesis). The results were as follows: Generally, detoxication effects by s. chinesis extract increased in proportion to the extract concentration. non 8 $\mu\textrm{g}$/g dosage of S. chinesis extract was administered, it showed the highest antitoxic effects in metallothionein induction. After the extract treatment, body weights generally increased In proportion to the extract concentrations. from the above results, S. chinesis extract Increased Metallothionein concentration and decreased the toxicity of cadmium In rats. In vitro the antitoxic activity of ethanol extract of S. chinesis on NIH3T3 fibroblasts was evaluated by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide) and SRB (sulforhodamine B protein) assays. The light microscopic study was carried out to observe morphological changes of the treeated cells. $10^{-2}$mg/ml Concentrations of S. chinesis extract was shown significant antitoxic activity. The number of NIH 3T3 fibroblasts were increased and tend to regenerate. These result suggest that S. chinesis extract retains a potential antitoxic activity.
Tannic acid (0.5mg/ml, 1.0mg/ml, 2.0mg/ml)와 카드뮴(20mg/kg)을 마우스에 경구 투여한 결과는 다음과 같다. 1. Tannic acid와 카드뮴을 투여한 마우스의 증체량과 음수소비량에 변화가 있었으나, 카드뮴투여에 의한 변화는 tannic acid투여에 의하여 감소되었다. 2. 카드뮴투여에 의하여 간장의 상대중량과 뇌 상대중량이 대조군에 비하여 유의한 변화가 있었으며, tannic acid 1.0mg/ml 투여군에서는 간장의 상대중량, 폐장의 상대중량, 흉선의 상대중량도 유의하게 변화하였다. (P < 0.05) 3. Hemoglobin contents, packed cell volume, platelet count, neutrophill count 등의 혈액학적인 변화는 대조군에 비하여 카드뮴투여군에서 유의한 변화가 인정되었다. 그러나, 이러한 유의한 변화가 tannic acid를 동시 투여한 군에서는 나타나지 않았다. 4. 카드뮴을 투여한 군에서는 혈청학적 변화(ALT, AST, BUN와 creatinine)가 있었으나 tannic acid 0.5, 1.0, 2.0mg/ml을 동시투여한 군에서는 회복되는 경향이 나타났다. 위의 결과로 미루어 카드뮴 투여에 의한 독성이 tannic acid을 2.0 mg/ml/day 이상 4주간 투여하였을 때 경감효과가 나타날 수 있었다. 그러나, 카드뮴과 같은 중금속의 독성에 tannic acid가 어떻게 경감효과를 나타내는지에 관한 작용기전의 연구가 더 필요할 것으로 사료된다.
This study was condo더ed to investigate the antitoxic component in aqueous extract of Houttuynia cordata $T_{HUNB}$. The results were as follows: Generally, detoxification effects by Houttuynia cordata $T_{HUNB}$ extract increased in proportion to the extract concentrations in rats. When 40 mBtg dosage of Houttuynia cordata $T_{HUNB}$ extract was administrated, Houttynia cordata $T_{HUNB}$ extract showed the highest antitoxic effects in metallothionein induction. After the extract treatment, body weights increased in proportion to the extract concentrations. However, after 3 weeks, the body weight decreased insignificantly. From the above results, Houttuynia cordate $T_{HUNB}$ extract increased metallothionein concentration and decreased the toxicity of cadmium in rats. In vitro the antitoxic activity of aqueous extract of Houttuynia cordata $T_{HUNB}$ on NIH 373 fibroblasts was evaluated by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetra-zoliumbromide) and SRB (sulforhodamine B protein) assays. The waste acceptance criteria for heavy metals in mixed waste should be developed by reflecting the leaching behaviors that could highly depend on the repository design and environment surrounding the waste. The current standards widely used to evaluate the leaching characteristics of heavy metals would not be appropriate for the silo-type repository since they are developed for landfills, which are more common than a silo-type repository. This research aimed to explore the leaching behaviors of cementitious waste with Pb, Cd, and Sb metallic and oxide powders in an environment simulating a silo-type radioactive waste repository. The Toxicity Characteristic Leaching Procedure (TCLP) and the ANS 16.1 standard were employed with standard and two modified solutions: concrete-saturated deionized and underground water. The compositions and elemental distribution of leachates and specimens were analyzed using an inductively coupled plasma optical emission spectrometer (ICP-OES) and energy-dispersive X-ray spectroscopy combined with scanning electron microscopy (SEM-EDS). Lead and antimony demonstrated high leaching levels in the modified leaching solutions, while cadmium exhibited minimal leaching behavior and remained mainly within the cement matrix. The results emphasize the significance of understanding heavy metals' leaching behavior in the repository's geochemical environment, which could accelerate or mitigate the reaction.

Cadmium (Cd) is of particular concern because of its widespread occurrence and high toxicity and may cause serious morpho-physiological and molecular abnormalities in in plants. The present study was performed to explore Cd-induced morpho-physiological alterations and their potentiality associated mechanisms in Sorghum bicolor leaves at the protein level. Ten-day-old sorghum seedlings were exposed to different concentrations (0, 100, and $150{\mu}M$) of $CdCl_2$, and different morpho-physiological responses were recorded. The effects of Cd exposure on protein expression patterns in S. bicolor were investigated using two-dimensional gel electrophoresis (2-DE) in samples derived from the leaves of both control and Cd-treated seedlings. The observed morphological changes revealed that the plants treated with Cd displayed dramatically altered shoot lengths, fresh weights, and relative water content. In addition, the concentration of Cd was markedly increased by treatment with Cd, and the amount of Cd taken up by the shoots was significantly and directly correlated with the applied level of Cd. Using the 2-DE method, a total of 33 differentially expressed protein spots were analyzed using MALDI-TOF/TOF MS. Of these, treatment with Cd resulted in significant increases in 15 proteins and decreases in 18 proteins. Significant changes were absorbed in the levels of proteins known to be involved in carbohydrate metabolism, transcriptional regulation, translation and stress responses. Proteomic results revealed that Cd stress had an inhibitory effect on carbon fixation, ATP production and the regulation of protein synthesis. In addition, the up-regulation of glutathione S-transferase and cytochrome P450 may play a significant role in Cd-related toxicity and stress responses. Our study provides insights into the integrated molecular mechanisms involved in response to Cd and the effects of Cd on the growth and physiological characteristics of sorghum seedlings. The upregulation of these stress-related genes may be candidates for further research and use in genetic manipulation of sorghum tolerance to Cd stress.
카드뮴중독으로 혈소판응집이 항진되어 고혈압 또는 동맥경화증이 발생한다는 연구보고를 감안하여 가토와 흰쥐의 생체내 실험으로서 미세혈전의 형성, Malondialdehyde(MDA) 및 thromboxane $B_2(TXB_2)$치에 미치는 카드뮴의 효과를 검토하고, in vitro실험으로 카드뮴을 처치한 가토 대동맥절편이 혈소판응집에 미치는 영향을 관찰하였으며, prostacyclin의 합성능력을 측정하여 그 결과를 다음과 같이 요약하였다. 1)Cadmium chloride 2mg/kg을 매주 동물의 복강내에 주입하였을 때 미세혈전형성이 증가되었다. 2) 카드뮴 중독동물의 platelet-rich plasma (PRP)는 MDA와 $TXB_2$형성이 정상동물에서 보다 현저히 증가되었다. 3) 카드뮴을 중독시킨 가토의 platelet-poor plasma (PPP)에서의 lipid peroxide치는 대조군과 차이가 없었다. 4) In vitro 실험으로, 가토대동맥 절편에서의 6-keto-$PGF_{1{\alpha}}$의 생성은 카드뮴 농도에 비례하여 억제되었고 이때 혈소판 응집률의 증가와 평행하였다. 5) 이상의 결과로서 카드뮴은 동맥내피세포에서 prostacyclin 합성을 억제할 뿐만 아니라 혈소판에서 $TXA_2$합성을 촉진시켜 그 결과로 혈소판 응집률을 증가시켰음을 알 수 있었다.
Deinococcus radiodurans is extremely resistant to various genotoxic conditions and chemicals. In this study, we characterized the effect of a sublethal concentration (100 ${\mu}M$) of cadmium (Cd) on D. radiodurans using a whole-genome DNA microarray. Time-course global gene expression profiling showed that 1,505 genes out of 3,116 total ORFs were differentially expressed more than 2-fold in response to Cd treatment for at least one timepoint. The majority of the upregulated genes are related to iron uptake, cysteine biosynthesis, protein disulfide stress, and various types of DNA repair systems. The enhanced upregulation of genes involved in cysteine biosynthesis and disulfide stress indicate that Cd has a high affinity for sulfur compounds. Provocation of iron deficiency and growth resumption of Cd-treated cells by iron supplementation also indicates that CdS forms in iron-sulfur-containing proteins such as the [Fe-S] cluster. Induction of base excision, mismatch, and recombinational repair systems indicates that various types of DNA damage, especially base excision, were enhanced by Cd. Exposure to sublethal Cd stress reduces the growth rate, and many of the downregulated genes are related to cell growth, including biosynthesis of cell membrane, translation, and transcription. The differential expression of 52 regulatory genes suggests a dynamic operation of complex regulatory networks by Cd-induced stress. These results demonstrate the effect of Cd exposure on D. radiodurans and how the related genes are expressed by this stress.
Zinc (Zn) is an essential element in biological process, however inadequate Zn status in general population have been recognized. To update the knowledge for Zn-cadmium (Cd) interaction, we studied the intestinal uptake and transport, and the expression of metal transporter proteins (divalent metal transporter 1, DMT1 ; metal transporter protein 1, MTP1 ; zinc transporter 1, ZnTl ; metallothionein 1 , MT1) in duodenum after Cd exposure using Zn deficient animal model. Rats were led Zn deficient (ZnD, 0.5-1.0 mgZn/kg) or Zn supplemented (ZnS, 50mg Zn/kg) diet for 4 weeks, and followed single administration of $^{109}$ CdCl$_2$orally. The body Zn flatus and tissue Cd concentration were determined at 24 hrs after Cd administration. Total body burden of Cd and Cd absorption index (AI, %) were estimated based on the tissue Cd analyzed. DMT1, MTP1, ZnTl and MT1 mRNA were analyzed by using RT-PCR method. Feeding of Zn deficient diet for 4 weeks produced a reduced body weight gain and a depletion of body Zn. Tissue Cd concentration, body burden of Cd and Cd absorption index were higher in the ZnD diet fed rats than the ZnS diet red rats. Especially, Cd concentration in the small intestine (duodenum, jejunum and ileum) and the colon of FeD diet fed rats were higher markedly than in the FeS diet group. The expression levels of DMT1, MTP1 and ZnT1 mRNA in FeD diet fed rats were similar to the FeS diet. The level of MT1 mRNA expression was significantly lower in the FeD than the FeS diet fed rats. Taken together, theses results indicate that Zn deficiency in diet induce an increased intestinal absorption and tissue retention of Cd, and down -regulate the MT1 expression in the intestine which might be play a part of role in Cd absorption and transport in mammalian. These findings suggest that deficiency of essential metal could be enhanced the toxicity of toxic, non-esstial metals through the metal-metal interaction.

For the purpose, Scapharca subcrenata(sea shell) which is common in numbers and biomass in the area was collected, and cadmium (Cd) was subjected as a reference metal in the experiment. The seashell was dried and autoclaved, followed by pellet preparation under various concentration of Cd: 50,100,250,500 and 1,500 ug /pellet. Mice were fed for 3 months, althrough last group(5 mice), 1,500 ug/pellet, died within 4 weeksof the administration.MT-Cd in liver and kidney increased in correlation with the amont of Cd concentration in pellet, especially in kidney than in liver. Liver and kidney of the mice showed pathological changes such as cellular infiltration and focal necrosis in liver, and peri-tubular cell infiltration or tubular necrosis etc. The changes were more intensive by amount of the Cd. Overall results in the present study suggest that Cd may eventually combine with soluble-protein forming metalloprotein, then reduce the toxicity of heavy metal, How the formed Mt-Cd is discharged from the kidney and the mechanism shall be the further subject to be clarified.
