  • Garlic occupies a special position among the many foods of vegetable origin because it is the sole food for Koreans during the their lives. And vitamin A has been ingested by forms of food or additives. Cadmium has been described as one of the most dangerous trace elements in the food and environment of man and livestocks. Since the de novo synthesis of stress proteins can be detected early after exposure to some agents, analysis of cadmium-induced changes in gene expression , ie. alterations in patterns of protein synthesis, may be useful to develop as biomarkers of exposure and damage for food hygiene. He acute and chronic combine effects of cadmium (Cd, CdCl2 20mg/kg), garlic oil(Dds: diallyl disulfide 50mg/kg, 3 times a week) and vitamin A(Ra: retinol acetate 50,000 IU/kg, 3 times a week) on Wistar male rats were evaluated concerning cadmium contents, tissues enzyme activity, HSP expression histopathological and electron microscopical examinations. The results of the study are as follows ; 1. Less cadmium was absorbed through the digestive tracts, but the ratio of contents in tissue were not changed by the simultaneous adminstration of diallyl disufide or retinol acetate. 2. ALT(alanine aminotransferase) , AST(aspartate aminotransferase), glucose, BUN (blood urea nitrogen), creatinine, the key indices of the clinical changes in hepatic and renal function were significantly hanged by the cadmium treatment after 1 week in liver, after 4 weeks in kidney. 3. Histopathological changes in cadmium treated rats were appeared at 8 weeks age treatment in kidneys. Homogenous eosinophilic material was accumulated in cortical and collecting tubular lumens at 16 weeks. Degenerated or necrotized tubular cells were observed in cortex and medulla. Degenerated seminiferous tubules and homogeneous eosinophilic material was seen in interstitial tissue of rat treated with cadmium for 16 weeks. Calcium deposits were seen in degenerated seminiferous tubules and the tubules showed severe calcification of rat treated with cadmium for 16 weeks. Electron microscope changes in kidney were observed in rats treated with CdCl2 20 mg/kg. Proximal convoluted tubule cells showed selling of cytoplasm and narrow lumen. Capillary endothelial cells showed cytoplasmic vacuoles and swelling. Degenerated epithelial cells were accumulated in tubular lumen of kidney. 4. Enhanced synthesis of 70 KDa relateve molecular mass proteins were detected in 2 hours after cadmium, exposure, with maximum activity occurring at 8~48 hours. Induction of HSP 70 was evident at proximal tubules and glomeruli in kidney. Testicular cells produced enough HSP to be detected normally. From the above results, it could be concluded that HSP70 induction by the cadmium treatment was a rapid reaction to indicated the exposure of xenobiotics, and retinol acetate reduced the cadmium induced nephrotoxicity.

Expression of Cu/Zn Superoxide Dismutase (Cu/Zn-SOD) mRNA in Shark, Schyliorhinus torazame, Liver during Acute Cadmium Exposure

  • Superoxide dismutase (SOD), an antioxidant enzyme catalyzing the first step for scavenging the reactive oxygen species is important as an early warning indicator to address various biological stresses. For this reason, the monitoring the expressed pattern of SOD gene in fish organs is one of important biomarkers to assess the aquatic pollution caused by many toxic chemicals. Based on the Northern blot hybridization, semi-quantitative and/or realtime RT-PCRs, the alteration of SOD gene transcripts in shark liver was examined during the experimental acute exposures to cadmium. The expression of SOD at mRNA level was up-regulated both by injection (0, 0.5, 1 or 2 mg $CdCl_2/kg$ body weight for 48 hours) and by immersion (0 or $5{\mu}M$ Cd for 0, 1, 4 and 7 days) treatments of cadmium. The transcriptional stimulation of shark SOD gene by cadmium exposure was dependent upon doses and durations: there was a trend toward more increase in higher dose and longer durations of exposure. The hepatic SOD mRNA levels showed also a general agreement with the tissue cadmium concentrations accumulated in immersion exposure. This result may provide useful strategy to develop a fine molecular biomarker at mRNA level for detecting aquatic pollution caused by toxic metals.

일부 자원자를 통한 납, 카드뮴의 다경로 인체 노출평가 (Human Multi-route Exposure Assessment of Lead and Cadmium for Korean Volunteers)

  • Objectives: We wanted to evaluate the main route of exposure to lead and cadmium for the general population in Korea by using multi-route and multi-media exposure assessment Methods: Samples of air at the homes, samples of the food and water and peripheral blood samples were collected from thirty volunteers living in Seoul (the metropolitan area), Yong-in (the suburban area) and Ansan (the industrial area) in 2001. Graphite furnace atomic absorption spectrometric methods were used for the determination of the Pb and Cd levels in the air, food, water and blood samples. Results: The average intake of lead through the air, drinking water and food were $5.06{\mu}g/day\;(26.3%)\;0.002{\mu}g/day\;(0.1%),\;and\;16.4{\mu}g/day\;(73.6%)$, respectively. The average intake of cadmium through the air, drinking water and food were $0.082{\mu}g/day\;(0.9%),\;0.001{\mu}g/day\;(0.007%),\;and\;12.61 {\mu}g/day\;(99.0%)$, respectively. The blood lead level was statistically higher in the male subjects than in the female subjects (3.39 and $2.22{\mu}g/dl$, respectively), and only gender was a significant variable on the multiple regression analysis for blood lead. Conclusions: This study showed that the intake of lead and cadmium through food was the major route of exposure. A policy to reduce the pollutants according to the exposure routes should be established. However, more studies will be needed to support these data for the general population.

Cadmium Exposure and Potential Health Risk from Foods in Contaminated Area, Thailand

  • Man-made cadmium (Cd) emissions can be transported between environmental matrices and the food chain. Food is the primary source of Cd exposure among general population as a consequence of the bioconcentration of Cd from soil. Chronic Cd exposure has been reported to be associated with chronic kidney disease, osteoporosis, diabetes, cardiovascular disease and cancer. The Joint FAO/WHO Expert Committee on Food Additives (JECFA) established the safe level of Cd intake as provisional tolerable monthly intake (PTMI) of $25{\mu}g/kg\;bw$ in 2010. The major food groups that contribute to the most Cd exposure are rice and grains, shellfish and sea food, meat including edible offal, and vegetables. A number of studies reported the high Cd contaminated levels in foods from polluted areas in Thailand. The results are of high concern since the contaminations occur in foods that are major Cd contributors. Thus, in this review, the current situations of Cd contaminated foods in polluted areas of Thailand are summarized. In addition, the Cd intakes from selected scenarios are estimated to assess the potential health risk to consumers and the suggestions are also included.

Cadmium removal by Anabaena doliolum Ind1 isolated from a coal mining area in Meghalaya, India: associated structural and physiological alterations

  • The cyanobacterium Anabaena doliolum Ind1 isolated from a coal mining site was tested for removal of cadmium at optimum pH 7.0 and temperature $25^{\circ}C$. The organism recorded high percentage of metal removal (92-69%) within seven days of exposure to 0.5-2.0 ppm cadmium. Biosorption onto the cell surface was the primary mode of metal removal. Fourier transform infrared spectroscopy (FTIR) established hydroxyl, amides, carboxyl, sulphate and carbonyl groups to be the major functional groups on the cell surface involved in cadmium binding. Cellular ultrastructure and a range of vital physiological processes (i.e., photosynthetic pigments, respiration, photosynthesis, heterocyst frequency and nitrogenase activity) remained unaffected upon 0.5 ppm treatment; higher concentrations of cadmium exerted visible adverse effects. Amongst the five photosynthetic pigments tested, phycocyanin was the most targeted pigment (inhibition was 15-89%). Both respiration and photosynthetic activities were inhibited by cadmium with more severe effect seen on respiration. 2.0 ppm cadmium exposure also had drastic negative effect on nitrogenase activity (87% decreased).

일부 사무직 여성근로자들의 카드뮴에 대한 노출정도 (Background Exposure of Healthy Office Workers to Cadmium in Selected Korean Women)

  • Objectives : To estimate the normal range of blood and urine cadmium levels using data from occupationally non-exposed office workers to cadmium in selected Korean women. Methods : A total of 437 women from healthy office workers were analyzed. To analyze blood and urine cadmium levels, blood and urine samples of study subjects were collected carefully and analyzed. Using a structured questionnaire, characteristics of the study subjects were investigated by well trained interviewers. Results : Mean blood cadmium concentration (CdB) was $0.46\;{\mu}g/{\ell}$ (GM; 0.16), whereas mean urine cadmium concentration (CdU) was $0.94\;{\mu}g/g$ creatinine (GM; 0.37). A significant increase of cadmium levels in blood and urine was observed by the rise of age (p=0.007; p=0.002, respectively). In the analysis of smoking state, blood and urine cadmium levels were higher in current smokers than in nonsmokers. In the multiple regression analysis, the rise of age and currently smoking were observed as a significant factor associated with cadmium level. Conclusion : These findings suggest that mean cadmium level in blood and urine indicated a relatively low level, although the rise of age and currently smoking were observed as a significant factor related to cadmium level. It can be used for the basic data to prevent harmful effects of cadmium exposure among female workers occupationally exposed to cadmium.

Effects of Acute Metal Exposures on the Viability and mRNA Expression of Metallothionein in Hemibarbus mylodon Fry

  • Transcriptional modulation of metallothionein (MT) during acute metal exposures (cadmium, copper or zinc) was examined in fry of Hemibarbus mylodon, a threatened fish species in Korean peninsula. Viability of H. mylodon fry was most affected by copper exposure (up to 79% of mortality at 1 ppm for 48 hours) and considerably by cadmium exposure (21 to 54% of mortality). On the other hand, Zn showed the least adverse effect on the viability (0 to 13% of mortality) of this species. Based on the semi-quantitative RT-PCR analysis, the stimulation of MT mRNA in response to metal exposures followed generally in a dose-dependent fashion where cadmium was the most potent inducer for the induction of MT transcripts in fry (up to more than 5-fold) while the lowest response was observed in zinc-exposed group (2-fold at maximum). From the exposure using environmentally realistic doses of cadmium (0 to 0.05 ppm for 24 hours), MT expression at mRNA level was also sensitively modulated toward upregulation up to more than 3-fold as relative to non-exposed control. Results from the present study would be a good basis for understanding the adaptive capacity and stress physiology of this endangered fish species during metal pollution.

다기능 선량계로서의 Cadmium sulfide의 X-선에 대한 특성 평가 (The Measurement and Evaluation of X-ray Characteristics of Cadmium Sulfide as a Multi-function Dosimeter)

  • 다기능 X-선 선량계로서의 cadmium sulfide의 X-선에 대한 특성을 평가하기 위해 cadmium sulfide 기반의 X-선 선량계를 진공 증착법을 사용하여 제작하였으며 X-선 노출에 따른 cadmium sulfide의 반응을 측정하였다. Cadmium sulfide의 X-선 관전압, 관전류, 조사시간에 따른 전압의 변화를 측정하여 이를 분석하였다. X-선의 관전압에 따른 cadmium sulfide의 반응을 회귀분석을 통해 통계처리 한 결과 y=0.0995x-0.1146 ($R^2$=0.9595, $\sigma$=0.08, standard error=2%) 이고 X-선 관전류에 따른 cadmium sulfide의 출력신호를 분석한 결과 y=0.0439x+1.1891 ($R^2$=0.9021, $\sigma$=0.04, standard error=1.8%)였다. 또한 X-선 조사시간에 따른 cadmium sulfide의 반응을 같은 통계 방법으로 통계처리한 결과 y=8.2853x+5.5878 ($R^2$=0.7287, $\sigma$=0.06, standard error=1.9%) 였다. 결론적으로 cadmium sulfide의 X-선에 대한 반응은 X-선의 에너지가 증가됨에 따라 선형적으로 반응함을 알 수 있었으며 제작된 cadmium sulfide의 X-선의 다기능 선량계로서의 그 사용 가능성을 제시하고자 한다.

  • Histopathological changes and cadmium accumulations in the gills were investigated in crucian carp(Carassius auratus) exposed to 0, 0.01, 0.1, 0.5mg/$\ell$ cadmium(Cd) concentrations for 25 days.
