Development of Gel Formulation Containing Alkaloid of Cacao Extract (카카오 추출물의 알칼로이드 성분이 함유된 겔제형의 개발)

  • In this study, methanol (CAMe) and ethanol (CAEt) extract of Theobroma cacao L. were tested to see possibility as natural functional cosmetic agent. Cacao extract and cacao extract loaded gel were evaluated for various properties such as antioxidant, antimicrobial, cell viability, anti-inflammatory. Inhibition of nitric oxide production in lipopolysaccaride (LPS) stimulated raw 264.7cells, at concentration of $0.125{\sim}1{\mu}g/ml$ a dose-dependent manner. Gel formulation (3% extract) were stable for 60day in both extract. In conclusion, gel containing extract of Theobroma cacao L. indicated strong possibility for functional cosmetic ingredient.

Cacao bean husk: an applicable bedding material in dairy free-stall barns

  • Objective: The objectives of the study were to assess the effect of cacao bean husk as bedding material in free-stall barn on the behavior, productivity, and udder health of dairy cattle, and on the ammonia concentrations in the barn. Methods: Four different stall surfaces (no bedding, cacao bean husk, sawdust, and chopped wheat straw) were each continuously tested for a period of 1 week to determine their effects on nine lactating Holstein cows housed in the free-stall barn with rubber matting. The lying time and the milk yield were measured between d 4 and d 7. Blood samples for plasma cortisol concentration and teat swabs for bacterial counts were obtained prior to morning milking on d 7. The time-averaged gas-phase ammonia concentrations in the barn were measured between d 2 and d 7. Results: The cows spent approximately 2 h more per day lying in the stalls when bedding was available than without bedding. The milk yield increased in the experimental periods when cows had access to bedding materials as compared to the period without bedding. The lying time was positively correlated with the milk yield. Bacterial counts on the teat ends recorded for cows housed on cacao bean husk were significantly lower than those recorded for cows housed without bedding. Ammonia concentration under cacao bean husk bedding decreased by 6%, 15%, and 21% as compared to no bedding, sawdust, and chopped wheat straw, respectively. The cortisol concentration was lowest in the period when cacao bean husk bedding was used. We observed a positive correlation between the ammonia concentrations in the barn and the plasma cortisol concentrations. Conclusion: Cacao bean husk is a potential alternative of conventional bedding material, such as sawdust or chopped wheat straw, with beneficial effects on udder health and ammonia concentrations in the barns.

Structure Determination of Glucosyltransferase Inhibitors from Cacao Bean Husk (Cacao Bean Husk로부터 Glucosyltransferase 저해물질 구조결정)

  • For glucosyltransferase(GTase) inhibitors, two flavan-3-ols were isolated from Theobroma cacao beam husk. They showed positive reaction with $anisaldehyde-H_2SO_4$ solution, $FeCl_3$ to be confirmed as dimeric flavan-3-ols on TLC and were identified as procyanidin B-1 [(-)-epicatechin-$(4{\beta}{\rightarrow}8)$-catechin] and procyanidin B-3 [(+)-catechin-$(4{\beta}{\rightarrow}8)$-catechin by spectroscopic analysis. Their inhibitory effect on glucosyltranaferase activity was also investigated and procyanidin B-1 showed 50% inhibition at 0.3 mM. They inhibited on the glucosyltransferasa noncompetitively and dimeric flavan-3-ol containing (-)-epicatechin had higer inhibitory activity.

Purification and Structure Determination of the GTase Inhibitor from Cacao Bean Husk Extract (Cacao Bean Husk 추출물로부터 Glucosyltransferase 저해 활성 물질의 분리 및 구조 동정)

  • The isolation of active compounds showing the inhibitory effect on glucosyltransferase(GTase) from cacao bean husk(CBH) extract was carried out for screening of anti-plaque agents. These active compounds were purified by additional column chromatography of MCI-gel CHP-20 and Sephadex LH-20 and their chemical structures were determined by NMR and mass spectroscopy. Two compounds showing the inhibitory effect on GTase from CBH extract were obtained. These compounds showed positive reactions with anisaldehyde-H2SO4 solution and FeCl3, and were identified as dimeric flavan-3-ols on TLC. By NMR and MS data analyses, the structures of two different flavan-3-ols were identified as procyanidin B-1 and procyanidin B-3, respectively.

  • In the course of our studies, novel flavan-3-ol structure isolated from Theobroma cacao L. husk was established by the thiolysis, desulfurization and spectroscopic method. The structure was identified for cinnamtannin A-2 containing the tetrameric epicatechin and molecular weight was [1153] by FAB-MS negative ion. The inhibitory effect on the glucosyltransferase activity was investigated. Cinnamtannin A-2 showed complete inhibition at 0.03 mM and inhibited on the glucosyltransferase noncompetitively. The hydroxyl group of flavan-3-ol was supposed to be the essential element for inhibition on the glucosyltransferase.

Endophytic Association of Trichoderma asperellum within Theobroma cacao Suppresses Vascular Streak Dieback Incidence and Promotes Side Graft Growth

  • Trichoderma species are able to persist on living sapwood and leaves of cacao (Theobroma cacao) in an endophytic relationship. In this research, we evaluated the ability of Trichodema asperellum introduced at the incision site in the bark for side grafting with the concentration of 4 g/10 mL, 4 g/100 mL, and 4 g/1,000 mL (suspended in water) in suppressing vascular streak dieback (VSD) incidence and promoting growth of side grafts in the field. The incidence of VSD in two local clones of cacao, MCC1 and M04, without application of T. asperellum was 71.2% and 70.1% at 21 wk after grafting, respectively. However, when the two clones were treated with a concentration of 4 g/10 mL T. asperellum, the incidence was 20.6% and 21.7%, respectively, compared to 29.1% and 20.9% at 4 g/100 mL and 18.2% and 15.6% at 4 g/1,000 mL. By comparing to the control, the treatment with the same concentrations of T. asperellum listed above, the total number of stomata in MCC1 decreased by 41.9%, 30.2%, and 14.0% and in M04 by 30.5%, 21.9%, and -2.5% (exception), respectively. Otherwise, the total area of stomata opening increased by 91.4%, 99.7%, and 28.6% in MCC1 and by 203.8%, 253.5%, and 35.9% in M04, respectively. Furthermore, the number of buds and branches treated with a mixture concentration on the the two clones increased by 90.7% and 21.7%, respectively. These data showed that the application of T. asperellum to cacao scions while grafting can decrease VSD incidence in side grafts and increase growth of grafts in addition to decreasing total number of stomata, increasing total area of opened stomata, and increasing number of buds and branches.

Effect of Suppository Bases on Absorption of Lithium Carbonate (좌제기제가 탄산리튬의 흡수에 미치는 영향)

  • This paper was designed to investigate the influence of different suppository bases on both the rectal absorption and dissolution rate of lithium carbonate, and to compare bioavailability from rectal administration with that from oral administration. The dissolution rates were in such order as PEG 4000, surfactant A (Witepsol 15+sodium lauryl sulfate), surfactant B (Witepsol 15+cholic acid), Witepsol 15 and cacao butter. Among various suppository bases, the blood level of lithium carbonate after rectal administration was increased in the following order: surfactant A>surfactant B>PEG 4000>Witepsol 15>cacao butter. When it comes to compare oral with rectal administration in AUC values, surfactants and PEG 4000 showed similar blood levels to oral administration, but lipophilic bases such as Witepsol 15 and cacao butter showed far lower blood level than oral administration. Peak time in oral administration was 2 hrs, but those in rectal administration using various suppository bases were $6{\sim}8$ hrs.

Functional Activities of Cacao Nibs and Couvertures according to Process Conditions (카카오닙과 커버춰의 가공 조건에 따른 기능성 분석)

  • This study was conducted to provide information regarding changes in antioxidant activity in response to conching temperatures, conching times, and cacao mass content (CMC) in dark chocolate. The radical scavenging activities and functional components of cacao nibs were highest for raw cacao nib (R0) under all conditions. Moreover, antioxidant activities and functional compounds increased during roasting for 25 min. As the conching temperature increased, the radical scavenging activities and functional components increased. Quantitative analysis of major catechin-derived compounds by HPLC revealed that R0 had the highest value for other roasted cacao nibs in all aspects (P<0.05). The content of procyanidin B2, catechin, and epicatechin increased during roasting for 25 min. Finally, evaluation of couvertures revealed that procyanidin B1 content increased as conching time increased to 48 h, except for 70% CMC and conched at $60^{\circ}C$ (HH) and 70% CMC and conched at $50^{\circ}C$. Overall, HH48 contained the richest catechin-derived components.
