Petrogeochemistry of Granitic Rocks Distributed in the Geumsan District, Korea (금산지역에 분포하는 화강암류의 암석지구화학)

  • The Mesozoic Geumsan granitic rocks of various composition are distributed in the Geumsan district, the central part of the Ogcheon Fold Belt. About 40 ore deposits of $CaF_2{\pm}Au{\pm}Ag{\pm}Cu{\pm}Pb{\pm}Zn$ are widely distributed in this district and are believed to be genetically related to the granitic rocks. Based on their petrography and geochemistry, the granitic rocks in this district can be classified into two groups ; the Group I( equigranular leucocratic granite, porphyritic biotite granite, porphyritic pink-feldspar granite, seriate leucocratic granite) and the Group II(seriate pinkfeldspar granite, equigranular alkali-feldspar granite, equigranular pink-feldspar granite, miarolitic pink-feldspar granite, equigranular biotite granite). Interpreted from their isotopic dating data and geochemical characteristics, the Group I and the Group II are inferred to be emplaced during the Jurassic(~184Ma), and the Cretaceous to the early Tertiary period(~59Ma), respectively. Both Group I and Group II generally belong to magnetite-series granitoids. The Cretaceous granitic rocks of Group II are more highly evolved than those of the Jurassic Group I. The Rb-Sr variation diagram suggests that the granitic rocks of the Jurassic Group I and of the Cretaceous Group II be evolved mainly during the processes of fractional crystallization and partial melting, respectively.

Purification of Chitinase from an Antagonistic Bacterium Bacillus sp.7079 and Pro-Inflammatory Cytokine Gene Expression by PCTC

  • Chitinase was purified from an antagonistic bacterium Bacillus sp. 7079 by ammonium sulfate precipitation, QAE-Sephadex anion exchange chromatography, Sephadex G-100 gel filtration, and SP-Sephadex cation exchange chromatography. The molecula. weight of purified chitinase (PC-1) was approximately 66.5 kDa on SDS-PACE. PC-1 exhibited optimum pH and temperature of pH 7.5 and $45^{\circ}C$, respectively. More than $80\%$ of PC-1 was stable at pH 5.0 to 9.0, and more than $90\%$ at $40^{\circ}C$. $Fe^2+\;and\;Ca^2+$ inhibited the chitinase activity about $20\%$, and EDTA and p-CMB by about $30\%$, whereas $Ag^+$ inhibited the activity up to $65\%$. The $K_m$ value of PC-1 was 1.215 mg/ml with colloidal chitin as a substrate. We also investigated the effect of PC-1 treated chitin (PCTC) on the pro-inflammatory cytokine gene expression in macrophage RAW 264.7 cells. The expression of IL-$1{\alpha}$ and IL-$1{\beta}$ mRNA gene was investigated using reverse transcriptase polymerase chain reaction (RT-PCR). IL-$1{\alpha}$ and IL-$1{\beta}$ mRNA were induced by the treatment of PCTC and chitin only in RAW 264.7 cells. These expressions were induced as early as 2 h and sustained up to 24 h in RAW 264.7 cells. IL-$1{\alpha}$ and IL-$1{\beta}$ mRNA were more strongly expressed by the treatment of PCTC than chitin treatment alone in RAW 264.7 cells.

Association of Genetic Polymorphisms at 1q22 but not 10q23 with Gastric Cancer in a Southern Chinese Population

  • Objective: Data from a recent genome-wide association studiesy of gastric cancer (GC) and oesophageal squamous cell carcinoma in Chinese living in the Taihang Mountains of north-central China suggest that 1q22 and 10q23 are susceptibility-associated regions for GC. However, this has not been confirmed in southern Chinese populations. The aim of this study was to investigate whether these polymorphisms at 1q22 and 10q23 are associated with the risk of GC in a southern Chinese population. Methods: We selected seven top significant associated single nucleotide polymorphisms (SNPs) at 1q22 and 10q23 and conducted a population-based case-control study in a southern Chinese population. Genotypes were determined using MassARRAYTM system (Sequenome, San Diego, CA). Results: Two SNPs at 1q22, rs4072037 and rs4460629, were significantly associated with a reduced risk of GC, best fitting the dominant genetic model. Logistic regression models adjusted for age and sex showed that rs4072037 AG and GG (OR=0.64, P=0.017, compared with AA) and rs4460629 CT and TT (OR=0.54, P=0.0016, compared with TT) significantly reduced the risk of GC. However, no significant results for the five SNPs at 10q23 were obtained in this study. Conclusion: These outcomes indicate that 1q22 is associated with GC susceptibility in this southern Chinese population, while an association for the locus at 10q23 was not confirmed.

The Proteinase Distributed in the Intestinal Organs of Fish 3. Purification and Some Enzymatic Properties of the Alkaline Proteinases from the Pyloric Caeca of Skipjack, Katsuwonus vagans

  • Purification and some properties of alkaline proteinases in the pyloric caeca of skipjack, Katsuwonus vagans, were investigated. Four alkaline proteinases, temporarily designated proteinases I, II, III and IV, were identified from the tissue extract of the pyloric caeca by ammonium sulfate fractionation, DEAE-Sephadex A-50 chromatography, and Sephadex G-100 and G-200 gel filtration. Result of disc-polyacrylamide gel electrophoretic analysis showed that the purified proteinases II and III were homogenous with the yields of $1.5\%\;and\;1.2\%$, and those specific activities were increased to 33 to 37 fold over that of the crude enzyme solution, respectively. Molecular weight of the proteinases II and III determined by sephadex G-100 gel filtration were 28,500 and 24,200, respectively. The optimum conditions for the caseinolytic activity of the two enzymes were pH 9.6 and $48^{\circ}C$. The reaction rates of the two alkaline proteinases were constant to the reaction time to 80 min in the reaction mixture of $3.4{\mu}g/ml$ of enzyme concentration and $2\%$ casein solution. The Km values against casein substrate determined by the method of Lineweaver-Burk were $0.56\%$ for proteinase II and $0.30\%$ for proteinase II. The proteinases II and III were inactivated under the presence of $Ag^+,\;Hg^{2+},\;Ni{2+},\;Fe^{2+},\;and\;Cu^{2+}$, and but activated by $Mn^{2+}\;and\;Ca^{2+}$ and markedly inhibited by the soybean trypsin inhibitor and N-p-toluenesulfonyl-L-lysine chloromethyl ketone. Therefore, the proteinases II and III were found to be a group of serine proteases and assured to be trypsin-like proteinases.

Purification and Characterization of Extracellular Aspartic Proteinase of Candida albicans

  • An extracellular proteinase of Candida albicans was purified by a combination of 0~75% ammonium sulfate precipitation, DEAE Sepharose Fast Flow ion exchange chromatography, and Sephacryl S-200 HR molecular sieve chromatography. Its mlecular weight was approximately 41 kDa on SDS-PAGE and isoelectric point was 4.4. The enzyme was inhibited by pepstain A. Optimum enzyme activity ranged from pH 2.0 to 3.5 with its maximum at pH 2.5 and a temperature of 45$^{\circ}C$. The addition of divalent cations, $Ca^{2+}$, Zn$^{2+}$ and $Mg^{2+}$, resulted in no significant inhibition of enzymatic activity. However, some inhibitory effects were observed by Fe$^{2+}$, Ag$^{2+}$ and Cu$^{2+}$. With BSA as substrate, an apparent $K_m$ was determined to be 7$\times$10$^{-7}$ M and $K_i$, using pepstatin A as an inhibitor, was 8.05$\times$10$^{-8}$ M. N-terminal amino acid sequence was QAVPVTLXNEQ. Degradation of BSA and fibronectin was shown but not collagen, hemoglobin, immunoglobulin G, or lysozyme. The enzyme preferred peptides with Glu and Leu at the P$_1$ position, but the enzyme activity was highly reduced when the P$_2$ position was phe or pro. This enzyme showed antigenicity against sera of patients with candidiasis.

Emission Detection of Mercuric Ions in Aqueous Media Based-on Dehybridization of DNA Duplexes

  • To quantify the presence of mercuric ions in aqueous solution, double-stranded DNA (dsDNA) of poly(dT) was employed using a light switch compound, $Ru(phen)_2(dppz)^{2+}$ (1) which is reported to intercalate into dsDNA of a right-handed B-form. Addition of mercuric ions induced the dehybridization of poly(dT)${\cdot}$poly(dA) duplexes to form a hairpin structure of poly(dT) at room temperature and the metal-to-ligand charge transfer emission derived from the intercalation of 1 was reduced due to the dehybridization of dsDNA. As the concentration of $Hg^{2+}$ was increased, the emission of 1 progressively decreased. This label-free emission method had a detection limit of 0.2 nM. Other metal ions, such as $K^+$, $Ag^+$, $Ca^{2+}$, $Mg^{2+}$, $Zn^{2+}$, $Mn^{2+}$, $Co^{2+}$, $Ni^{2+}$, $Cu^{2+}$, $Cd^{2+}$, $Cr^{3+}$, $Fe^{3+}$, had no significant effect on reducing emission. This emission method can differentiate matched and mismatched poly(dT) sequences based on the emission intensity of dsDNA.

Purification and Characterization of Chitinolytic Enzymes Produced by Aeromonas sp. J-5003

  • Chitinase and chitobiase produced by Aeromonas sp. J-5003 were purified and characterized. The chitinase was purified to 19.4 folds by gel chromatography and ion-exchange chromatography with the overall yield of $2.2\%$ and the specific activity of 93.1 unit/mg. The purified enzyme showed a single band on SDS-PAGE with MW 54kDa. The optimum pH and temperature of the purified chitinase were 7.0 and $37^{\circ}C$, respectively, and this enzyme stable in the range of pH 6.0 to 10.0 below $37^{\circ}C$. $Mg^{2+},\;Ca^{2+}\;and\;Na^+$ slightly stimulated the chitinase activity. However, $Hg^{2+}\;and\;Fe^{3+}$ inhibited chitinase activity. The chitobiase was purified by Sephacryl HR-l00 gel chromatography and DEAE-Sephadex A-50 ion-exchange chromatography with 33.5 purification folds and $4.3\%$ yield. The purified enzyme showed a single band with MW 63 kDa. The optimum pH and temperature of the purified chitobiase were 7.0 and $37^{\circ}C$, respectively. And this enzyme was stable in the range of pH 6.0 to 9.0 and at the temperature below $37^{\circ}C$. The enzyme activity was increased by $Mn^{2+}$, but it was inhibited by $Ag^+$.

${\alpha}$-Galactosidase from Bacillus megaterium VHM1 and Its Application in Removal of Flatulence-Causing Factors from Soymilk

  • A bacterial strain capable of producing extracellular ${\alpha}$-galactosidase was isolated from a sample of sugarcane industrial waste. Microbiological, physiological, and biochemical studies revealed that the isolate belonged to Bacillus sp. Furthermore, based on a 16S rDNA sequence analysis, the new isolate was identified as Bacillus megaterium VHM1. The production of ${\alpha}$-galactosidase was optimized based on various physical culture conditions. Guar gum and yeast extract acted as the best carbon and nitrogen sources, respectively. The optimum pH was 7.5 and the enzyme remained stable over a pH range of 5-9. The enzyme was optimally active at $55^{\circ}C$ and thermostable with a half-life of 120 min, yet lost 90% of its residual activity within 120 min at $60^{\circ}C$. One mM concentrations of $Ag^2$, $Cu^2$, and $Hg^{2+}$ strongly inhibited the ${\alpha}$-galactosidase, whereas the metal ions $Fe^2$, $Mn^{2+}$, and $Mg^{2+}$ had no effect on the ${\alpha}$-galactosidase activity, and $Zn^{2+}$, $Ni^{2+}$, and $Ca^{2+}$ reduced the enzyme activity slightly. When treated with the B. megaterium VHM1 enzyme, the flatulence-causing sugars in soymilk were completely hydrolyzed within 1.5 h.

Studies on proteolytic enzyme from A New Strain in Serratia sp. (Serratia속의 신균주가 생산하는 단백질분해효소)

  • Serratia sp. Y-4 was isolated from soil. Many characteristics of the strain and optimum cultivation condition for protease production were investigated.,The protease from Serratia sp. Y-4 was purified and studied for the properties of the enzyme. The isolated strain was identified to the genus Serratia. The strain was cultivated in 1%-casein, 0.5%-Na$_{3}$PO$_{4}$.7H$_{2}$O, 0.1%-NaCl, 0.05%-KCI, 0.02%-MgSO$_{4}$.7H$_{2}$O, 0.02%-CaCl$_{2}$.2H$_{2}$O, 0.02%-ZnSO$_{4}$.7H$_{2}$O, 0.02%-MnCl$_{2}$.4H$_{2}$O and 0.5%-soy bean oil at pH 7.0 for 35 hrs. The enzyme was purified about 5.89 fold from the culture broth with 31.1% recovery and 19,613 u/mg through ultrafiltration, ammonium sulfate, DEAE-sephacel and Superose-12 chromatography. The purified enzyme was identified as one band by isoelectric focusing, SDS- and native-PAGE. It has maxium activity at $37^{\circ}C$ and pH 9.0. Molecular weight of it is approx. 50 kD and pl is about 6.70. Its Km value for casein was 20 mg/ml. 5 mM-EDTA, 5mM-SDS, Ag$^{+1}$, Cu$^{+2}$, Hg$^{+2}$ and Pb$^{+2}$ inhibited the enzyme.

  • 현재 압전재료로써 사용되고 있는 PZT계 세라믹스는 우수한 압전 및 유전특성으로 초음파 센서, 압전변압기, 액츄에이터, 필터, 레조네이터와 같은 여러 응용분야에 널리 사용되어지고 있다. 그러나 납을 60%이상 포함하므로 환경오염에 의하여 그 사용에 대한 규제가 강화되고 있어 최근에는 납을 함유하지 않은 무연 환경친화형 압전세라믹스가 주목받고 있다. 현재 무연 조성 세라믹스에는 텅스텐-브론즈 형, 비스머스 레이어형, 페로브스카이트 형 등이 이으며, 그 중 페로보스카이트 혈을 제외하고는 보통 소성법 조제시 낮은 압전특성을 갖는다. 그러므로 압전특성을 증가시키기 위하여 Hot pressing, Hot forging, RTGG(Reactive Template Grain Growth), SPS(Spark Plasma Sintering), 그러나 이는 무연 세라믹스의 대량생산 과정에서 어려운 문제를 가지고 있고, 저가격 관점에서 볼 때 보통소성법이 보다 바람직하다. 그래서 보통소성법으로 NKN세라믹스의 소결성을 향상 시키고 비교적 높은 압전특성을 얻기 위해서 $LiSbO_3$, $LiNbO_3$, $LiTaO_3$, alkaline-earth(Mg, Ca, Sr, Ba) 등을 첨가한 논문들이 보고되고 있으며, 이러한 재료들의 kp는 대략 0.3-0.4를 나타내고 있다. 따라서, 본 연구에서는 소결온도를 낮추고자 $Ag_2O$를 소결조제로 사용하였고 유전 및 압전특성을 높이고자 소결시간 변화시켜 시편을 제작하여 NKN 세라믹스의 유전 및 압전특성을 조사하였다.
