GC/MS analysis of hydrocarbons and 2-alkylcyclobutanones from γ-ray irradiated walnut (Juglans nigra) (호두(Juglans nigra)의 감마선 조사에 따른 hydrocarbon류와 2-alkylcyclobutanone류의 GC/MS 분석)

  • This study was conducted to analyze the hydrocarbons and 2-alkylcyclobutanones as marker compounds in walnuts after the walnuts' exposure to ${\gamma}$ irradiation. The samples were irradiated with gamma rays at 0, 1, 3, 5, 7, and 10 kGy doses. The lipids were extracted via soxhlet extraction using hexane, and were separated by florisil column and identified via gas chromatography / mass spectrometry (GC/MS). The hydrocarbons that were detected were 8-heptadecene ($C_{17:1}$) and 1,7-hexadecadiene ($C_{16:2}$) from oleic acid and 8,11-heptadecadiene ($C_{17:2}$) and 1,7,10-hexadecatriene ($C_{16:3}$) from linoleic acid. The 2-alkylcyclobutanones that were detected were 2-dodecylcyclobutanone (DCB) from palmitic acid, 2-tetradecylcyclobutanone (TCB) from stearic acid, 2-(5'-tetradecenyl)cyclobutanone (TECB) from oleic acid, and 2-(5',8'-tetradecadienyl)cyclobutanone (5',8'-TCB) from linoleic acid. The correlation between the irradiation dose and the concentrations of the hydrocarbons and 2-alkylcyclobutanones in the walnuts was found to be linear. The radio-induced hydrocarbons and 2-alkylcyclobutanones were clearly detected in the irradiated walnuts at 1 kGy and above, but not in the non-irradiated ones. The major hydrocarbons obtained after irradiation were 8-heptadecene from oleic acid and 8,11-heptadecadiene and 1,7,10-hexadecatriene from linoleic acid, and the major 2-alkylcyclobutanones were TECB from oleic acid and 5',8'-TCB from linoleic acid. Therefore, these major compounds were concluded to be the marker compounds for determining the irradiated and non-irradiated samples.

Physicochemical Properties of Ripe and Dry Jujube ($Ziziphus$ $jujuba$ Miller) Fruits (성숙과 건조 대추의 이화학적 특성)

  • This study was carried out to investigate the physicochemical characteristics of maturating and dried jujube. The moisture contents of the unripe and ripe jujube were found to be 84.66 and 66.45%, respectively, but that of the dried jujube was 31.48%. The crude-fat and crude-ash contents of the dried jujube were much higher than those of the maturing jujube. The carbohydrate contents of the unripe, ripe, and dried jujube were found to be 13.51, 27.94, and 61.70%, respectively, and the soluble-protein contents were 0.88, 1.73, and 3.71%. The reducing-sugar content of the dried jujube was 18.82%, higher than that of the ripe jujube. The sucrose contents of the unripe and ripe jujube were 10.15 and 16.66 g/100 g, respectively, and that of the dried jujube was 33.46 g/100 g. The major fatty acids that were found to compose jujube were palmitic, oleic, pamitoleic, linoleic, and linolenic acid. The myristoleic and arachidonic acid contents were higher in the dried jujube than in the ripe jujube, but the palmitoleic acid content was lower in the dried jujube than in the ripe jujube. The major minerals of jujube were found to be Ca, K, Mg, and P, and the total mineral contents of the unripe, ripe, and dried jujube in this study were 343.38, 584.94, and 331.56 mg/100 g, respectively. The polyphenol contents of the unripe, ripe, and dried jujube were 309.12, 248.80, and 23.34 mg/100 g, respectively, while the vitamin C contents were 610.04, 310.22, and 423.27 mg/100 g.

Changes of Fatty Acids and Amino Acids Contents of Korean Native Commercial Ducks Meats with Different Raising Periods (사육 기간에 따른 토종 실용오리의 도체 내 지방산과 아미노산 함량 변화)

  • This work was carried to evaluate effect of different raising periods on fatty acids and amino acids properties of Korean native commercial ducks. Korean native ducks (n = 90) from National Institute of Animal Science (RDA, Korea) were divided into 6 groups (15 birds/group) and were fed with meat-type duck diets for 8 wk old. When ducks grew at specific wk (6, 7 and 8 weeks), 2 ducks per group were slaughtered at 6, 7 and 8 wk old. Myristic acid (C14:0) and palmitic acid (C16:0) of saturated fatty acid (SFA) were the highest and arachidonic acid of unsaturated fatty acid (USFA) was the lowest at the age of 6 weeks old (P<0.05) at the age of 6 weeks old among treatments. Stearic acid of SFA was the highest at the age of 7 weeks old among treatments. There was no significant difference on other fatty acids among treatments. SFA and USFA have significant difference at the age of 8 weeks (P<0.05). Histidine, leucine, lysine, methionine, phenylalanine and threonine of essential amino acid increased with passing weeks (P<0.05). There was no significant difference on isoleucine and valine (P>0.05). Every non-essential amino acids were the highest at the age of 8 weeks among treatments (P<0.05). In conclusion, fatty acid contents and amino acid contents increased until the age of 8 weeks old, but further research was needed because of deficiency of data to suggest the correct shipping time.

Dietary effect of Lactobacillus plantarum CJLP55 isolated from kimchi on skin pH and its related biomarker levels in adult subjects (김치유산균 Lactobacillus plantarum CJLP55 섭취가 성인 남녀의 피부 산도 및 관련 구성 인자의 변화에 미치는 영향)

  • Purpose: The skin pH is maintained by epidermal lactate, free fatty acids (FFAs), and free amino acids (FAAs). As a significant determinant of skin health, the skin pH is increased (less acidic) under abnormal and aged skin conditions. In a search for dietary alternatives that would promote an acidic skin pH, this study investigated the dietary effects of Lactobacillus plantarum CJLP55 isolated from Korean kimchi on the skin pH, and epidermal levels of lactate, FFAs, and FAAs in adult subjects. Methods: Seventy eight subjects (mean age $24.9{\pm}0.5years$, range 19 ~ 37 years) were assigned randomly to ingest CJLP55, Lactobacillus strain from kimchi, (n = 39, CJLP group) or placebo supplements (n = 39, placebo group) for 12 weeks in a double-blind, placebo-controlled trial. Skin pH and epidermal levels of lactate, FFAs and FFAs were assessed at 0, 6 and 12 weeks. Results: Although significant decreases in skin pH were observed in both the CJLP and placebo groups at 6 weeks, the skin pH was decreased significantly only in the CJLP group at 12 weeks. In parallel, the epidermal level of lactate in the CJLP group was also increased by 25.6% at 12 weeks. On the other hand, the epidermal level of FAAs were not altered in the CJLP and placebo groups, but the epidermal level of total FFAs, including palmitic acid and stearic acid, was lower in the CJLP group than in the placebo group over 12 weeks. The changes in the other FFAs, such as palmitoleic acid and oleic acid, were similar in the CJLP and placebo groups over 12 weeks. Conclusion: Overall, a dietary supplement of CJLP55 promotes acidic skin pH with a selective increase in epidermal lactate in adult subjects.

An Innate Bactericidal Oleic Acid Effective Against Skin Infection of Methicillin-Resistant Staphylococcus aureus: A Therapy Concordant with Evolutionary Medicine

  • Free fatty acids (FFAs) are known to have bacteriocidal activity and are important components of the innate immune system. Many FFAs are naturally present in human and animal skin, breast milk, and in the bloodstream. Here, the therapeutic potential of FFAs against methicillin-resistant Staphylococcus aureus (MRSA) is demonstrated in cultures and in mice. Among a series of FFAs, only oleic acid (OA) (C18:1, cis-9) can effectively eliminate Staphylococcus aureus (S. aureus) through cell wall disruption. Lauric acid (LA, C12:0) and palmitic acid (PA, C16:0) do not have this ability. OA can inhibit growth of a number of Gram-positive bacteria, including hospital and community-associated MRSA at a dose that did not show any toxicity to human sebocytes. The bacteriocidal activities of FFAs were also demonstrated in vivo through injection of OA into mouse skin lesions previously infected with a strain of MRSA. In conclusion, our results suggest a promising therapeutic approach against MRSA through boosting the bacteriocidal activities of native FFAs, which may have been co-evolved during the interactions between microbes and their hosts.

A Study on the Antioxidative Effect and Quality Characteristics of Cookies Made with Lycii fructus Powder (구기자를 첨가한 쿠키의 품질특성과 항산화효과)

  • The purpose of this study was to investigate the antioxidative effect and quality characteristics of cookies made with Lycii fructus powder(LFP). The cookies were stored at $50^{\circ}C$ for 40 days. The LFP additive increased the moisture, ash, crude protein and volume of the cookies. When LFP was added at $5\%$ into the cookies, the spread ratio was higher than for the control, the $10\%$ and the $20\%$ LFP cookies. As more LFP was added, the L-value decreased, and the a--values and b-values increased for the color values. For the textural characteristics, springiness and cohesiveness of the control cookies were the highest among the samples, the other side, brittleness and gumminess were higher in cookies made with LFP than in the control cookies. Overall, the cookies made with $5\%$ LFP were preferred more than the other cookies, as tested forby sensory evaluation. The cookies made with LFP contained mostly palmitic acid (C16:0) followed by oleic acid (C18: 1) and stearic acid (C18:0). The acid value, peroxide value, and thiobarbituric acid value were lower in cookies made with $5\%$ and $10\%$ LFP than in those cookies made with $20\%$ LFP and the control cookies.

Comparative Studies on the Fatty Acid Compositions of the Korean Salt Marsh Plants in the West Sea (한국서해에 서식하는 염생식물의 지방산 조성 비교)

  • Fatty acid is an important component of many biological processes. However, an imbalance in diet-especially, a n-3 polyunsaturated fatty acids (PUFAs) deficiency-causes several diseases such as diabetes, cardiovascular disease and cancer. In this study, we analyzed the fatty acid contents and compositions of 26 species of Korean salt marsh plants and found high fatty acid contents from S. herbacea ($148.75\;{\mu}g$/mg-dry wt.), S. komarvii ($119.05\;{\mu}g$/mg-dry wt.), C. heterocarpa ($79.23\;{\mu}g$/mg-dry wt.), A. capillaris ($71.65\;{\mu}g$/mg-dry wt.), and L. tetragonum ($67.02\;{\mu}g$/mg-dry wt.). In the case of saturated fatty acids (SFAs) composition, palmitic acid is richest in most salt marsh plants. On the other hand, oleic acid and linoleic acid are major components of monounsaturated fatty acid and n-6 PUFA, respectively. In addition, n-3 PUFAs such as LNA (linolenic acid), EPA (eicosapentaenoic acid), and DHA (docosahexaenoic acid) known as the main fatty acid components of fish oils and seaweeds, were also found in S. herbacea, S. komarvii, T. tetragonoides, A. capillaris and G. littoralis.

Comparisons of Fatty Acid intake of Hypercholesterolemia in Women (고콜레스테롤혈증 여성 환자의 지방산 섭취량 비교)

  • The purpose of this study was to compare with in fatty acid intakes between hypercholesterolemia group and normocholesterolemia group, aged between 30 to 65 years old in women. The subjects were classified as hypercholesterolemia group and normocholesterolemia group based on The Guideline for Korean Hyperlipidemia. Dietary intakes of fatty acids were by means of a 24-hr recall method with food models and measuring tools. We analyzed both data sets together using analysis of variance the chi-square test and student's t-test(SPSS for WINDOWS, version 7.5). Significance was defied as a P value <0.05. The results obtained are summarized as follows. Mean age, BMI and percentage of postmenopausal women in hypercholesterolemia group were significantly higher than those of normocholesterolemia group. Percent of total energy from fat in hypercholesterolemia group tended to be higher than that in normocholesterolemia group. C16:0(Palmitic acid) and saturated fatty acids(SFA) intake of hypercholesterolemia group was significantly higher than that of normocholesterolemia group. Therefore, women who suffered from hypercholesterolemia were recommended to control body weight and consume foods containing low SFA and cholesterol.

Effect of Vacuum Packaging and Aerobic Packaging on the Physico-Chemical Characteristics of Venison (진공포장과 함기포장이 사슴육이 이화학적 특성에 미치는 영향)

  • 사슴(Elk deer ♀, 월령 28~30개월, 체중 170$\pm$10kg)의 등심부위와 대퇴부위를 도축 직후 함기포장과 진공포장하여 4$^{\circ}C$에서 10일간 냉장하면서 경시적으로 이화학적 특성의 변화를 실험한 결과 지방산은 등심부위와 대퇴부의 모두 palmitic acid가 각각 35.16%와 31.75%가 많고 그 다음으로 oleic acid, stearic acid의 순으로 많아\ulcornerT다. 냉장기간이 경과하면서 포화지방산은 증가하였고 불포화지방산은 감소하는 경향이었는데 함기포장육의 변화 폭이 컸다. 냉장기간 중 TBA가는 등심부위 가 0.0598에서 0.5616 mgMA/kg, 대퇴부위가 0.0650에서 0.3770 mgMA/kg,이었으며, 냉장기간이 결과함에 따라 점차 상승하고 상승폭은 진공포장육보다 함기포장육이 대퇴부위보다 등심부위가 크게 나타났다. 냉장기간중 VBN가는 등심부위가 3.93에서 7.31 mg%, 대퇴부위가 3.98에서 6.35 mg%이 었으며 냉장기간이 경과함에 EK라 진공포장육보다 함기포장육이 대퇴부위보다 등심부위가 더 높게 나타났다. 이상의 결과를 종합하여 볼 때 등심부위와 대퇴부 모두 냉장 10일까지 신선한 상태이었고 진공포장육이 함기포장육보다 선도 유지가 잘 되었다.

Effects of Emulsified Sausage Supplemented with Ginseng Saponin on Lipid Metabolism in Rat

  • The current study was designed to assess the effects of emulsified sausage supplemented with ginseng saponin on lipid metabolism by applying a rat model. Four groups of 8 rats (5 wk old) were each allocated one of 4 treatments: basal feed (C), and basal feed with 20% sausage powder containing 0% (S0), 2% (S2) and 4% (S4) ginseng saponin. The experiment was conducted for 4 wk. The results did not differ among the treatments with different amounts of sausage (ST), but daily feed intake (p<0.01) and feed conversion (p<0.001) were significantly increased in STs compared to C. Both total serum cholesterol and triglyceride concentrations were significantly (p<0.001) reduced, by 45 and 46%, and 48 and 46%, in S2 and S4, respectively, compared to S0. In the liver, the total cholesterol level was dramatically (p<0.05) decreased according to increasing sausage powder levels. In particular, S4 showed approximately 14% reduction compared to S0 (p<0.05). Liver triglyceride content also showed a similar tendency, where S2 and S4 resulted in 7% and 31% reduction. With regard to fatty acid composition in the liver tissues, palmitic acid (16:0), oleic acid (18:1), eicosanoic acid (20:1), and eicosatrienoic acid (20:3) did not differ among the STs, whereas both linoleic acid (18:2) (p<0.01) and linolenic acid (18:3) (p<0.001) showed significant increases in S2 compared to S0. The current data demonstrated that emulsified sausages supplemented with ginseng saponin effectively reduce total cholesterol and triglyceride concentrations in the serum and liver, and increase unsaturated and essential fatty acid in the liver. These data collectively imply that the sausage improved the overall lipid profile in a rat model, and can be further generalized to the result that emulsified sausage can improve lipid metabolism depending on the products' formula.
