Statistical Optimization of Medium Composition for Bacterial Cellulose Production by Gluconacetobacter hansenii UAC09 Using Coffee Cherry Husk Extract - an Agro-Industry Waste

  • During the production of grape wine, the formation of thick leathery pellicle/bacterial cellulose (BC) at the airliquid interface was due to the bacterium, which was isolated and identified as Gluconacetobacter hansenii UAC09. Cultural conditions for bacterial cellulose production from G. hansenii UAC09 were optimized by central composite rotatable experimental design. To economize the BC production, coffee cherry husk (CCH) extract and corn steep liquor (CSL) were used as less expensive sources of carbon and nitrogen, respectively. CCH and CSL are byproducts from the coffee processing and starch processing industry, respectively. The interactions between pH (4.5-8.5), CSL (2-10%), alcohol (0.5-2%), acetic acid (0.5-2%), and water dilution rate to CCH ratio (1:1 to 1:5) were studied using response surface methodology. The optimum conditions for maximum BC production were pH (6.64), CSL (10%), alcohol (0.5%), acetic acid (1.13%), and water to CCH ratio (1:1). After 2 weeks of fermentation, the amount of BC produced was 6.24 g/l. This yield was comparable to the predicted value of 6.09 g/l. This is the first report on the optimization of the fermentation medium by RSM using CCH extract as the carbon source for BC production by G. hansenii UAC09.

Biological conversion of biomass to succinic acid

  • Batch cultivations of Anaerobiospirillum succiniciproducens have been systematically studied for the economical production of succinic acid from wood hydrolysate with corn steep liquor(CSL) as a nitrogen source. CSL was found to be an alternative complex nitrogen source for A. succiniciproducens when glucose and wood hydrolysate were used as carbon sources. Compared with polypeptone and/or yeast extract, CSL had similar effects on fermentation performance such as succinic acid yield and a ratio of succinic acid to acetic acid in the fermentation of wood hydrolysate as well as glucose. This means that succinic acid can be produced more economically from wood hydrolysate and CSL than relatively expensive carbon and nitrogen sources. Besides its low cost, the alternative medium served as a green technology for succinic acid production because it gives a net-zero effect on global warming.

  • 새로운 고지혈증 예방 제품 개발을 위한 기초 자료를 얻고자 우선 전통 된장으로부터 HMG-CoA reductase 저해 물질을 강력하게 세포외로 생산하는 D-3 세균을 최종 분리 하였다. D-3 균의 미생물학적 특성을 조사한 결과 그람양성의 간균으로 운동성이 있었고 통성 혐기성이였다. 이들 특성과 16S rRNA 염기서열의 분석 결과 등을 종합하여 Bergey's manual로 동정한 결과 Bacillus cereus D-3로 동정되었다. HMG-CoA reductase 저해물질 생산 최적 조건을 조사한 결과 Bacillus cereus D-3를 glucose 2%, corn steep liquor $0.6%,\;K_{2}HPO_4\;0.04%,\;KH_{2}PO_4\;0.05%$를 함유한 Glucose-CSL 배지에 접종하여 $30^{\circ}C$에서 36시간 배양하여 얻은 상등액의 HMC-CoA reductase 저해활성이 39.4% 저해물질을 가장 많이 생성 하였다.

Development of a Practical and Cost-Effective Medium for Bioethanol Production from the Seaweed Hydrolysate in Surface-Aerated Fermentor by Repeated-Batch Operation

  • To develop a practical and cost-effective medium for bioethanol production from the hydrolysate of seaweed Sargassum sagamianum, we investigated the feasibility and performance of bioethanol production in CSL (corn-steep liquor)-containing medium, where yeast Pichia stipitis was used and the repeated batch was carried out in a surface-aerated fermentor. The optimal medium replacement time during the repeated operation was determined to be 36 h, and the surface aeration rates were 30 and 100 ml/min. Under these conditions, the repeated-batch operation was successfully carried out for 6 runs (216 h), in which the maximum bioethanol concentrations reached about 11-12 g/l at each batch operation. These results demonstrated that bioethanol production could be carried out repeatedly and steadily for 216 h. In these experiments, the total cumulative bioethanol production was 57.9 g and 58.0 g when the surface aeration rates were 30 ml/min and 100 ml/min, respectively. In addition, the bioethanol yields were 0.43 (about 84% of theoretical value) and 0.44 (about 86% of theoretical value) when the surface aeration rates were 30 ml/min and 100 ml/min, respectively. CSL was successfully used as a medium ingredient for the bioethanol production from the hydrolysate of seaweed Sargassum sagamianum, indicating that this medium may be practical and cost-effective for bioethanol production.

  • Pyrimidine nucleoade N-ribosidase (pyrimidine 5'-nucleotide phosphoribo (deoxyribo) hydrolase/ pyrimidine 5'-nucleotide nucleosidase, EC 3.2.2.10)는 CMP와 UMP를 직접 분해하여 cytosine과 uracil를 생성하는 가수분해효소이다. 본 연구에서는 Pseudomonas oleovorans ATCC 8062의 생육과 pyrimidine nu-cleotide N-ribosidase생성에 미치는 탄소원과 질소원의 영향 및 생육인자 등에 대하여 검토했다. 효소생성의 최적배양조건은 2% fumarate, 1.5% peptone, 5% corn steep liquor (CSL)과 1% ammonium chloride의 배지조성(초기 pH 7.0)으로 $28^{\circ}C$, 48시간 진탕 배양이 양호했다. 효소의 활성은 생육이 최대에 도달하는 정지기 후기에 최대에 도달하며, 그 이후부터 급속히 불활성화 되었다. P. oleovorans의 pyrimidine nucleocde N-ribosidase는 UMP에 의하여 유도 생성되지 않으므로 구성효소이며, 내생효소였다.

5균-GMP 생산 융합균주 RC102의 고정화에 의한 5균-GMP 생산 (Production of 5균-GMP by Immobilized 5균-GMP Producing Fusant RC102)

  • 5'-GMP(5'-Guanylic acid) 생산 융합균조 fusant RC102를 고정화하여 5'-GMP을 연속적으로 생산하고자 하였다. 균체 고정화용 담체로서 k-carrageenan, polyacrylamide, Ca-alginate 및 agar 등 4가지 담체에 고정화한 결과 최적 담체로서 3% k-carrageenan을 선정하였으며, 이담체에 고정화한 균체를 이용하여 5'-GMP 연속적으로 생산하였다. 고정화 균체에 의한 5'-GMP 생산의 최적 온도 및 pH은 $32^{\circ}C$, pH 8.0, 미량온소로서 $Mn^{2+}과\;Zn^{2+}$의 농도는 각각 $30\mu\textrm{g}/L,\;1{\times}10^{-6}%$이었다. $Mn^{2+}$이 풍부하고 값싼 CSL(Corn Steep Liquor)을 배지로서 사용하기 위해서 penicillin G와 D-cycloserine 및 POSEA(polyoxyethylene stearylamine) 농도 최적 농도는 각각 0.8unit/ml, 0.8unit/ml, 그리고 5mg/ml으로 나타났으며, 항생물질 보다 계면활성제가 효과적이었다. 이상의 최적 조건에서 고정화 fusant RC102는 15일간 안정되게 5'-GMP를 생산하였다.

Acetobacter xylinum BRC5에 의한 미생물 셀룰로오스의 발효 특성

  • The characteristics of cell growth and cellulose production by Acetobcter xylinum BRC5 were studied in shaking flasks and jar fermentors. Of the substrates tested, the highest cellulose yield was obtained from the fructose medium. Some inductive cultivation was observed and then cellulose was produced with cell growth. When glucose alone or mixture of glucose and fructose was used as the carbon source, cellulose could be biosynthesized under the glucose limitation. Corn steep liquor (CSL), as a low-cost nitrogen source, was a good substitute for yeast extract. In a jar fermentor experiment, 4.14 g/l of disintegrated cellulose was obtained from 8% CSL- medium containing 0.5% glucose and 1.5% fructose.

회분식 및 연속배양에 있어서 고농도 젖산의 생산을 위한 공업용 배지연구 (A Study on Industrial Media for Production of Lactic acid in Batch and Continuous Fermentations)

  • 젖산 생산에 큰 비중을 차지하는 원료비용 절감을 위한 다양한 공업적 질소원의 이용 가능성을 조사하였다. CSL를 사용하여 회분 발효한 결과, CSL 5%배지 발효시 높은 생산성을 보였으며, 정제 CSL을 사용한 배지에서 젖산 생산성은 CSL보다 약 1.21 배 증가율을 보였다. Yeast extract와 CSL 혼합질소원의 배지조성을 변화시켜 LBM5의 젖산 생성정도를 알아본 결과, 조성 비율 1:8일 때 우수한 발효율을 관찰 할 수 있었다. 발효 폐기물의 대체 질소원 사용가능성을 조사하기 위하여 맥주공장에서 부산물로 배출되는 폐효모를 이용하여 발효시에도 CSL 5%와 비슷한 발효율을 얻었다 연속발효 수행결과, 최적의 희석율은 0.05-0.065-1h였으며 젖산 생산성에 있어서 회분발효보다 1.93.0배증가하였다. 한편 초기 순수배지에 들어가는 yeast extract양의 1/3-2/3를 발효폐액에 첨가하여 회분발효 및 연속발효 실험을 수행하여 높은 젖산 생산성을 얻으므로써 공정상 원료비 절감효과를 기대할 수 있어으며 잔류 영양분을 재사용함으로써 폐액량 감소에 따른 처리비용 절감이라는 이중적인 효과를 얻게 되었다.

식품부산물로부터 유기산의 대량생산공정에 관한 연구 - 세포재순환식 연속발효를 이용한 유기산의 대량 생산 - (Production of Organic Acids from Food By-Products - Mass Production of Organic Acids by Continuous Flow Ceil Recycling Fermentation -)

  • 본 연구에서는 식품부산물을 원료로 사용하여 회분식 발효를 통한 Propionibacterium acidipropionici의 유기산 생산성을 검토하고 세포재순환식 연속발효를 통해 유기산의 대량생산을 위한 기초자료를 마련하고자 하였다. CSL의 함유량에 따른 회분식 발효를 실시하여 균주의 생산성이 최대가 되는 발효조건을 구하여 연속 발효공정에 적용하였다. 희석률을 $0.1{\sim}0.2\;hr^{-1}$로 변화시키며 세포재순환식 연속발효를 실시한 결과 $0.2\;hr^{-1}$에서 최대 세포량은 150 g drycell/L으로 회분식 발효에 비해 21배 증가하였으며, 유기산의 최대 생산성은 3.32 g total acids/L/h로서 13배가 향상된 것으로 나타났다. 따라서 이 발효공정을 scale-up한다면 유기산뿐만 아니라 바이오매스 자원으로서 세포량의 대량생산도 가능할 것으로 판단된다.

The Improvement of Cephalosporin C Production by Fed-batch Culture of Cephalosporium acremonium M25 Using Rice Oil

  • The objective of this study is to improve cephalosporin C (CPC) production byoptimization of medium and culture conditions. A statistical method was introduced to optimize the main culture medium. The main medium for CPC production was optimized using a statistical method. Glucose and corn steep liquor (CSL) were found to be the most effective factors for CPC production. Glucose and CSL were optimized to 2.84 and $6.68\%$, respectively. CPC produc­tion was improved $50\%$ by feeding of $5\%$ rice oil at day 3rd and 5th day during the shake flask culture of C acremonium M25. The effect of agitation speeds on CPC production in a 2.5-L bio­reactor was also investigated with fed-batch mode. The maximum cell mass (54.5 g/L) was obtained at 600 rpm. However, the maximum CPC production (0.98 g/L) was obtained at 500 rpm. At this condition, the maximum CPC production was improved about $132\%$ compared to the re­sult with batch flask culture.
