Oocyte freezing has become a prevalent source for related reproductive technologies. This study was carried out to evaluate viability of post-thawed bovine oocyte injected DTT-treated sperm following by two different activation stimuli (Group 1, 5 M ionomycin, 5 min + CR1aa, 3 h . 1.9 mM dimetylaminopurine (DMP), 3 h; group 2, ionomycin + 10 $\mu\textrm{g}$/$m\ell$ cycloheximide(CHX), 5h). The techniques of ultra-rapid freezing used in this study were essentially similar to those of described by Vajta et al (Theriogenology 1999; 52:939-948), Denuded oocytes at 22 h of culture were exposed to cryoprotectant (3.2 M Ethylene glycol, 2.36 M DMSO, 0.6 M sucrose), and followed by freezing in electron microscopic grid. After thawing the oocytes were transferred back into the drop of maturation medium and cultured for additional 2 h before being subjected to ICSI. All eggs were then cultured in CRlaa medium, and transferred into M199+10% FCS on day 4. The culture was maintained until day 9. In Experiment 1, frozen-ICSI eggs were compared on development into blastocyst to those of unfrozen and IVF control. Those eggs were activated with the method of group 2. A higher proportion of unfrozen-ICSI and IVF eggs developed into cleavage and blastocysts than of frozen-ICSI eggs (65% and 13%; 71% and 23% vs. 39% and 8%; P<0.05). In Experiment 2, development and ploidy of embryos made from group 1 were compared to those from group 2. Between groups there did not differ on the rates of development, however, chromosomal abnormality in group 1 was significantly higher than in group 2 (49% vs. 30%; P<0.05). The present result suggests that frozen bovine oocytes can be used for ICSI.

In this study, we developed emission factors from solid-fuel fired combustors. In order to increase the reliability of emission factors, we conducted a joint research with the Institute of Health and Environment. As a result, PM average concentration was $8.19mg/m^3$. $SO_2$ and $NO_x$ were respectively 8.46 ppm, 50.64 ppm. Hazardous air pollutants such as Cr, Pb and Hg were detected in trace amounts continuously for 2 years in some solid-fuel fired combustors. The emission factors for the three kinds of PM, $SO_x$, $NO_x$ were developed based on the measurement data. For the PM emission factors, that of SRF was 15.93 g/kg and that of Bio-SRF was 14.18 g/kg. Compared with those of US. EPA, emission factors of this study showed the results of low values. $SO_x$ emission factors were 4.42 g/kg for SRF and 1.39 g/kg for Bio-SRF. $NO_x$ emission factors were 13.21 g/kg and 4.43 g/kg, respectively. Through the results of this study, we would support atmospheric administration policies such as the emission factor notification revision.
In this study, A bacterium with an ability to resist toxic heavy metals was isolated from reeds in wetland. The isolated strain was identified to Alcaligenes sp. KC-1 by 16S rDNA sequencing. Heavy metals such as Pb, $Cr^{6+}$, Cd, Zn and Cu were supplied to media. The ecotoxic treat of the heavy metals on the growth of strain KC-1 was performed when the isolated strain Alcaligenes sp. KC-1 cultured with Cu ranging from 0 mM to 20 mM. It showed the resistance of $EC_{50}$(7.34 mM) and cell growth ($OD_{600\;nm}$ : 0.83 after 42 hours) when it was cultured in Cu.

The electrical properties of the praseodymium-based ZnO varistor ceramics, which are composed of Zn-Pr-Co-Cr-Dy oxides were investigated with $Dy_2O_3$ amount. The average grain size of varistor ceramics was greatly decreased from 18.2 to 4.6 pm with increasing $Dy_2O_3$ amount. The calculated nonlinear exponent$({\alpha})$ in varistor ceramics without $DY_2O_3$ was only 4.9, whereas the a value of the varistors with $DY_2O_3$ was abruptly increased in the range of 48.8 to 58.6. In particular, the maximum value of a was obtained by doping of 1.0 mol% $DY_2O_3$, reaching 58.6. The measured leakage current$(I_l)$ value in varistors without $DY_2O_3$ was $85.45{\mu}A$, whereas the $I_{\ell}$ value of the varistors with $DY_2O_3$ was very abruptly decreased in the range of 1.10 to $0.12{\mu}A$. In particular, the minimum value of $I_{\ell}$ was obtained by doping of 0.5 mol% $DY_2O_3$, reaching $0.12{\mu}A$. The tan $\delta$ varied in V-shape, with minimum 2.28% at 0.5 mol% $DY_2O_3$. The donor concentration and the density of interface states were decreased in the range of $(4.66{\sim}0.25){\times}10^{18}cm^3$ and $(5.70{\sim}1.39){\times}10^{12}/cm^2$, respectively, as $DY_2O_3$ amount is increased.
In this research, a 2-stage intermittently aerated activated sludge system(IA) and intermittently aerated dynamic flow activated sludge system(DF) were investigated for the removal of nutrients in domestic wastewater. Wastewater was characterized by low C/N( organics/nitrogen) ratio. $COD_{cr}$, $BOD_s$, TKN and TP concentrations of domestic wastewater were 235, 47, 32 and 5.4 mg/L, respectively. Three sets of IA and one set of DF were operated. Three of four systems were added with fermented settled sludge taken from primary settling tank as an external electron donor and the other(IA) for control reactor was operated without addition of electron donor. All systems were operated at same sludge retention time of 20 days and hydraulic retention time of 12hrs. The supplemental electron donor was supplied into the anoxic mode. A higher denitrification rate was observed from the reactors with fermented settled sludge as an electron donor for denitrification compared to that of without addition of organic source. The result of this study indicates that the settled primary sludge, if the fermented at the acid stage, was an excellent electron donor for denitrification. 81 % of TN and 80% of TP were removed from the systems with the supplemental organic source added. However, the control reactor without addition of electron donor showed only 39% of TN and 43% of TP.
A silica mine monitoring was conducted from March to December in 2008 to measure rainfall, runoff amounts and pollution loads. A total of 13 rainfall-runoff events were measured and analyzed with respect to runoff ratio, pollutant concentration and load, and initial flush. Over rainfall-runoff events, 95% confidence range of SS concentration was 942.5~2,056.2 mg/L. Other measured water quality indices also showed relatively large variation. This wide concentration variation was thought to be caused by the bare working ground of the mine that was used to store, process and transport the mined silica. Total pollution load of the 13 rainfall-runoff events was SS 17,901 kg/ha, $COD_{Cr}$ 160.9 kg/ha, $COD_{Mn}$ 111.24 kg/ha, BOD 79.6 kg/ha, T-N 13.8 kg/ha, T-P 3.5 kg/ha, and TOC 39.3 kg/ha. Initial flush was not well observed except SS. Very high SS concentration and load was occurred when rainfall was large. Therefore, it was recommended to manage the bare ground not to discharge excessive pollutants during wet days by covering the ground or constructing runoff treatment systems such as a sediment basin.
The purpose of this study was to investigate Rn concentration and annual radiation exposure level in the basement and first floor. The Rn Cup monitors were placed in different environments such as shopping stage, office building, Apartment, Hospital, house in Seoul from Match 1996 to April 1997 and CR-39 films were collected every two months. The mean radon concentration in the basement of house($88.6\;Bq/m^3$) showed the highest level among the areas, while radon concentration on the first floor of house($50.5\;Bq/m^3$) showed the higher than other areas. The annual radiation exposure dose that person on the floor / in the basement of differential place in the seoul can be exposed during living was estimated from 24.11 to 87.64 mRem/yr. This radiation dose is significantly lower than 130mRem maximum radiation dosage from the radon nuclide prescribed by the ICRP, with respect to the overall average exposure of the working adult. this study indicated that possible radon sources on the first floor / in the basement areas are radon intrusion from soil gas, construction materials, or ground water leaking. Further study is needed to quantitatively assess major contributions of radon-222 and health effect to radon exposure.
This study was conducted to evaluate the ELISA kit for measuring the level of cortisol in the urine. The CV of within-run variation and day to day variation were 0.4~2.8 and 1.8~5.7, respectively. The minimum limitation of measurement was 1ng/ml. The cross reaction was high ($CR_{50}(%)=11.4{\sim}43.2$) in prednisolone, 11-deoxycortisol, 21-deoxycortisol and predinosone. There was low and no cross reaction in other steroid. To develop the ELISA kit we measured the cortisol level in diluted urine with PBS (procedure I), extracted urine with methylene chloride (procedure II) and extracted methylene chloride-extracted urine from thin-layer chromatography (procedure III). The CV value of procedure I, II, III was 9.4~28.3%, 7.2~8.9% and 2.5~5.7%, respectively. There was significant difference between procedure I with II, and pro-cedure I with III(p < 0.01), but no difference between procedure II with III significantly(p < 0.01). The mean UCCR of urine collected through am 8 to 10 was $9.5{\pm}7.6$(0.14~28.0) in 12-month-old dog(n = 47). In this study we can measure the cortisol level in extracted urine with methylene chloride and sequential thin-layer chromatography accurately using ELISA kit.

Aerosol slurry samples were collected in 60-min interval using Korean Semi-continuous Elements in Aerosol Sampler (KSEAS) between May 19 and June 6, 2010 at an urban site of Gwangju. The $PM_{2.5}$ samples were collected with a flow rate of 16.7 L/min and particles are grown by condensation of water vapor in a condenser maintained at ${\sim}5^{\circ}C$ after saturation by direct injection of steam. The resulting droplets are collected in a liquid slurry with a airdroplet separator. Concentrations of 16 elements (Al, Fe, Mn, Ca, K, Cu, Zn, Pb, Cd, Cr, Ti, V, Ni, Co, As, Se) in the collected slurry samples were determined off-line by ICP-MS. KSEAS sample analysis encompassed the sampling periods for which 24-hr average elemental species concentrations were calculated for comparison with those derived from 24-hr integrated filter samples. Relationship between elemental species measured by two methods indicated high correlation coefficients (r), mostly greater than r of 0.80. However, we note that concentrations of Al, K, Ca, Mn, and Fe, which are often associated with crustal elemental particles, in the KSEAS samples, were substantially lower (1.4~11 times) than those found in the typical filter-based samples. This discrepancy is probably due to difficulties in transferring insoluble dust particles to the collection vials in the KSEAS. Temporal profiles of elemental concentrations indicate that some transient events in their concentrations are observed over the sampling periods. For the elemental species studied, atmospheric concentrations during the transient events increased by factors of 4 in Mn~80 in Zn, compared to their background levels. Principle component analyses were applied to the hourly KSEAS data sets to identify sources affecting the concentrations of the metal constituents observed. In this study, we conclude that hourly measurements for particle-bound elemental constituents were extremely useful for revealing the short-term variability in their concentrations and developing insights into their sources.
