  • A strontium titanate ($SrTiO_3$)-based material with a perovskite structure is considered to be one of the promising alternatives to $LaCrO_3$-based materials since $SrTiO_3$ perovskite shows a high chemical stability under both oxidizing and reducing atmospheres at high temperatures. $SrTiO_3$ materials exhibit an n-type semiconducting behavior when it is donor-doped and/or exposed to a reducing atmosphere. In this work, $Sr_{1-x}La_xTi_{1-y}M_yO_3$ materials doped with $La^{3+}$ in A-sites and aliovalent transition metal ions ($M^{n+}$) in B-sites were synthesized by the modified Pechini method. The X-ray diffraction analysis indicated that the materials synthesized by the Pechini process exhibited a single curbic perovskite-type structure without any impurity phases, and are tolerant, to some extent, to cation doping. The sintering behaviors of $Sr_{1-x}La_xTi_{1-y}M_yO_3$ in $H_2/N_2$ and air were characterized by dilatometry and microstructural observations. The electrical conduction mechanism and the dopant effect are discussed based on the defect structures and the electrical conductivities measured at various oxygen partial pressures and temperatures.

실혈 후 및 혈압상승 후의 소화기 조직 혈액량 및 산소 섭취량 -제 1 편 정맥혈압과 소화기 조직 혈액량- (Gastrointestinal Tissue Blood Volume Affected by Venous Pressure Change)

  • Changes in gastrointestinal tissue blood volume induced by variations of venous pressure between 6 and 40 mmHg were studied in 32 rabbits. Venous pressure lowering was produced by withdrawal of appropriate volume of blood and venous pressure elevation was obtained by partial occlusion of intra-thoracic vena cava inferior. Estimation of regional tissue blood volume was performed by means of regional distribution of injected $Cr^{51}-labeled$ red blood cells. The following results were obtained. 1. At the normal control venous pressure value of 18 mmHg, spleen showed the highest value of tissue blood volume expressed on weight basis, namely, $111{\mu}l/gm$, Liver tissue blood volume was $95\;{\mu}l/gm$, small intestine 24 and stomach $21\;{\mu}l/gm$, respectively. 2. Linear relationships were observed between venous pressure change and gastrointestinal tissue blood volume. The coefficients of correlation were: in spleen r=0.723; in liver r=0.791; in stomach r=0.704, respectively. In small intestine the relationship was less clear and r=0.358. Tissue blood volume of extrabdominal tissue, such as M. gastrocnemius was not influenced by venous pressure change. 3. The highest change in tissue blood volume expressed on weight basis was observed in spleen. The liver tissue showed the next highest change. Change in total tissue blood volume, however, was greatest in liver and next greatest in small intestine. This was interpreted by the fact that total weight of these two organs was much greater than that of spleen. 4. The mechanism that the change in tissue blood volume lies in the venous system which has a great compliance was discussed.

베로 세포에서 생산된 2세대 일본뇌염 백신의 마우스에서의 면역원성 (The Immune Response of Mice Vaccinated with Japanese Encephalitis Vaccine, CJ50003 Produced in Vero Cells)

  • 베로 세포에 적응시켜 개발된 약독화 일본뇌염 바이러스인 CJ5003 주를 이용해 제조된 새로운 2세대 일본뇌염 백신의 효과를 마우스에서 조사하였다. Adjuvant 로 aluminiu hydroxide gel을 사용한 경우, 그렇지 않은 경우에 비해 높은 일본뇌염 바이러스 특이 항체 유도능과 10배 향상된 중화항체 유도능을 보였으며, 항원의 양을 변화시켜 면역시킨 결과 0.5 ng 의 적은 항원양으로도 항체가 유발되었고 방어 가능한 수준인 1:10 이상의 중화항체가 유지되었다. 500ng의 항원양으로 면역된 마우스를 면역 초기부터 24주간 관찰한 결과 중화항체가가 14주까지 1:200 이상으로 계속 유지되었으며 24주에도 1:160을 유지하여 일본뇌염에 대한 방어효력이 장기간 유지되고 있음을 나타냈다. 또한 면역된 마우스 혈청은 Nakayama-NIH, SA14, P3 등 3종의 신경독성 일본뇌염 바이러스주에 대해서 각각 유사한 수준의 일본뇌염 바이러스 특이 항체가를 보여주어 다양한 일본뇌염 바이러스 주에 대한 방어 가능성을 보여주었다. 마우스를 이용한 뇌내 직접 공격 시험결과, CJ5003 후보 백신은 90% 이상의 높은 생존율을 나타냈다. 이상의 in vitro, in vivo 시험 결과는 베로 세포에서 생산된 CJ5003 후보 백신의 차세대 백신으로 개발 가능성을 시사한다.

Yeosin-san Increases Female Fertility through Inducing Uterine Receptivity and Ovarian Function

  • Despite the development of assisted reproduction technologies (ART) including in vitro fertilization (IVF), the poor ovarian response and endometrial receptivity remains clinically a major unmet need. Although these problems are difficulties to solve in infertility treatment, there are no good therapeutic option yet. Traditional herbal remedies and acupuncture, therefore are being proposed as alternative treatment. Our group found that traditional herbal medicines such as Paeonia lactiflora L.(PL, 芍藥), Cyperus rotundus L.(CR, 香附子), and Perilla frutescens (PF, 紫蘇葉) could improve endometrial receptivity. In this study, we found out Yeosin-san (如神散) as an optimal herbal formula via combination of the previously established herbal medicines. Yeosin-san is a traditional Korean medical formula which was established by Ziming Jin (陳自明) and recorded in Furendaiquanliangfang (婦人大全良方) at first. The formula traditionally used for treating abnormal uterine bleeding and leukorrhea. It showed a highest effect on leukemia inhibitory factor (LIF) expression and on the adhesion between trophoblastic cells and endometrial cells. In addition, it has been shown that the Yeosin-san not only increases the endometrial receptivity to improve the embryo implantation but also enhances the ovary function by expressing the angiogenesis-related genes. Here we suggest that Yeosin-san could be a novel and effective candidate for treating female infertility.

Effects of Changes in Glycosylation Sites on Secretion of Recombinant Human Erythropoietin in Cultured CHO Cells

  • The effects of additions/deletions in glycosylated residues of recombinant human EPO (rhEPO) produced in CHO-K1 on their secretion were examined. hEPO cDNA was amplified from human liver mRNA and cloned into the pCR2.1 TOPO. Using overlapping-extension site-directed mutagenesis method, glycosylation sites at 24th, 38th, 83rd, and 126th were respectively or accumulatively removed by substituting its asparagine (or serine) with glutamine. To add novel glycosylation sites, 69 and 105th leucine was mutated to asparagine. Mutant and wild type rhEPO constructs were cloned into the pcDNA3 expression vector with CMV promoter and transfected into CHO cell line, CHO-K1, to produce mutant rhEPO mutant rhEPO proteins. Enzyme-linked immunosorbant assay (ELISA) and Western analysis with monoclonal anti-EPO antibody were performed using supernatants of the cultures showing transient and stable expressions respectively. Addition of novel glycosylation reduced rhEPO secretion dramatically while deletion mutants had little effect except some double deletion mutants ($\Delta$24/83 and $\Delta$38/83) and triple mutant ($\Delta$24/38/83). This fact suggests that not single but combination of changes in glycosyl groups affect secretion of rhEPO in cell culture, possibly via changes in their conformations.

생물환원전극 미생물연료전지에서 외부저항 및 유입부하에 따른 유기물 및 질소 제거와 전기생산에 미치는 영향 (Effect of the Organic and Nitrogen Removal and Electricity Production on Changing the External Resistor and the Inflow Loading in the Biocathode Microbial Fuel Cell)

  • In order to remove the organic substances and the nitrate-nitrogen contained in wastewater, some researchers have studied the simultaneous removal of organics and nitrogen by using different biocathode microbial fuel cells (MFCs). The operating conditions for removing the contaminants in the MFCs are the external resistances, HRTs, the concentration of the influent wastewater, and other factors. This study aimed to determine the effect of the external resistors and organic loading rates, from the changing HRT, on the removal of the organics and nitrogen and on the production of electric power using the Denitrification Biocathode - Microbial Fuel Cell (DNB-MFC). As regards the results of the study, the removal efficiencies of $SCOD_{Cr}$ did not show any difference, but the nitrate-nitrogen removal efficiencies were increased by decreasing the external resistance. The maximum denitrification rate achieved was $129.2{\pm}13.54g\;NO_3{^-}-N/m^3/d$ in the external resistance $1{\Omega}$, and the maximum power density was $3,279mW/m^3$ in $10{\Omega}$. When the DNB-MFC was operated with increasing influent organic and nitrate loading by reducing the HRTs, the $NO_3{^-}-N$ removal efficiencies were increased linearly, and the maximum nitrate removal rate was $1,586g\;NO^3{^-}-N/m^3/d$ at HRT 0.6 h.

손 반사요법이 말기 신부전증 환자의 생기와 면역 반응에 미치는 영향 (The Effects of Hand Reflexology on Saeng-Chi and Immunity in ESRD Patients)

  • The purpose of this study was to identify the Effect of Hand Reflexology on Saeng-chi of physiologic, emotional & motivational responses and Immunity in ESRD patients who received hemodialysis in two general hospital from June to September. 2001 A two group quasi-experimental research with pre and post test design was used. The number of participants in the experimental group was 23, and in the control group, 20 The Hand Reflexology Intervention was developed by the researcher based on hand reflexology by Carter & Weber and Chi-massage by Chia. The Hand Reflexology was applied to both hands for 10 minutes per day. and 5 days by 5 times. To evaluate the effects of the program, Pre and Post evaluations were done. In the physiologic response, the PR was decreased at the 1st times post treatment and at the 5th. In addition BP was decreased at the 1st time, but not the 5th. After 5 treatments, there were significant increase in Hb and significant decreases in the BUN and Cr. levels in the experimental group. In the emotional and motivational responses, there were significant increases in vigor, mood, uplifts and self care agency scores in the experimental group, but there was no significant difference in the Rosenberg's self esteem score. In the experimental group, significantly increased CD4, and h/s ratios were found, also NK cells were significantly decreased, and there was a decrease in the CD8. However, no significant differences between groups were observed. There were significant increases in CD32, CD33, CD34 in the experimental group. The self care agency score correlated negatively with the CD8. From the above results, Hand Reflexology is shown to be an effective mind-body nursing intervention for enhancing Saeng-chi responses and affecting some of the immune responses. However, Immune cell activation and differentiation with hand reflexology will be achieved with future study.

Staphylococcus epidermidis 를 이용한 납 이온의 축적에 관한 연구 (The Uptake of Lead Ion with Staphylococcus epidermidis)

  • 수용액으로부터 $Pb^{2+}$ 의 흡수와 분리를 Staphylococcus epidermidis 를 이용하여 조사하였다. 흡수된 $Pb^{2+}$의 양을 초기 $Pb^{2+}$ 의 양에 대한 비율로 나타낸 것을 흡수율이라고 정의할 경우, 세균체의 농도가 높을수록 흡수율은 증가하였으며 세균체 단위 중량당 흡수율은 감소하였다. $Pb^{2+}$ 의 흡수율은 수용액의 pH 가 상승함에 따라 점차 증가하다가 최고치를 나타낸 후 감소하는 경향을 보였으며 $Pb^{2+}$ 의 농도가 증가할수록 흡수율의 최고치는 더 낮은 pH 에서 나타났다. HCI 과 EDTA 는 효과적인 탈락체로 작용하였으며 흡수와 탈착 cycle 의 횟수에 따른 세균체의 $Pb^{2+}$에 대한 흡수 능력에는 거의 변화가 없었다.$Pb^{2+}$ 이 다른 중금속 이온과 함께 혼재할 경우 세균내의 결합위치에 대한 경쟁적인 흡수로 인하여 $Pb^{2+}$ 의 흡수율이 변화하였으며, 공존하는 중금속 이온의 이온 반경이 작을수록$Pb^{2+}$ 의 흡수율이 더 크게 감소하였다.

성장중인 양서류 여포의 스테로이드 생성능력 획득에 관한 연구 (Development of Steroidogenic Capacity during Follicle Growth in Amphibian Ovarian Follicles)

  • 참개구리를 사용하여 난소내 여포들이 성장시기에 획즉하게 되는 스테로이드들의 생성능력을 조사하였다. 개구리로부터 중간(0.7-1.11mm) 크기 이하의 여포들을 분리한 다음 전구스테로이들이나 뇌하수체추출물(FPH)을 포함한 배양액에서 6시간 배양 후 이들에 의해 산물스테로이드로 전환된 양을 방사면역 측정법으로 조사하였다. FPH의 처리는 중간 크기의 여포만 스테로이드들의 생성을 현저히 촉진하고 작은 여포들(0.35-0.7mm)에서는 촉진효과가 매우 미약하였다. Progenenolone의 첨가는 가장 작은 여포 (0.22-0.4mm)들에 의해서도 progesterone(P$_4$)으로 전환되었다. 그러나 가장 작은 여포에서는 P$_4$를 첨가했을 때에도 역시 같은 현상이 나타났다. 작은 여포와 중간 여포들에 의한 전환된 스테로이드들의 양을 단위표면적의 전환량으로 계산 비교해 본 결과 전환된 양은 거의 같았다. 이는 작은 여포의 여포세포들 (granulosa cells)이 이미 E$_2$에 이르는 모든 중간 산물들을 생성할 능력을 가졌다는 것을 의미한다. 그러나 이들이 FPH에 의해서는 스테로이드를 조금밖에 생성하지 못하는 것으로 보아 아직 부족한 또다른 요인들이 있는것으로 추정된다.

홍화약침액(紅花藥鍼液)이 RAW Cell 유전자발현(遺傳子發顯)에 미치는 영향(影響) (Microarray Analysis of Gene Expression in RAW Cells Treated with Carthami Flos Herbal Acupuncture Solution)

  • 목적 : 홍화(紅花)는 활혈거어(活血祛瘀), 해독지통(解毒止痛)의 효능이 있어 관절염, 동맥경화(動脈硬化), 종양(腫瘍), 월경부조(月經不調), 뇌혈전(腦血栓)에 사용되어 왔다. 이에 홍화약침액(紅花藥鍼液)의 분자생물학적 효능 분석을 하고자 Lipopolysaccharide(LPS)로 염증을 유발한 RAW 264.7 cell의 유전자(遺傳子) 발현(發顯)에 미치는 영향을 Microarray를 통하여 관찰하였다. 방법 : RAW cell을 배양하고 홍화약침액(紅花藥鍼液)의 세포 독성을 확인한 후 (1) LPS, (2) 홍화약침액(紅花藥鍼液), (3) 홍화약침액(紅花藥鍼液)과 LPS를 처치했을 때의 유전자 발현양상을 microarray를 이용하여 관찰하였다. 대조군에 비해 2배 이상 발현의 차이가 있는 경우를 유의한 것으로 보았다. 결과 : 8,170개의 유전자 중 (1) LPS를 처치하였을 경우 35개의 유전자에서 발현이 상승되었고, (2) 홍화약침액(紅花藥鍼液)을 처치하였을 경우 11개의 유전자에서 발현이 상승되고 53개의 유전자에서 발현이 억제되었으며, (3) 홍화약침액(紅花藥鍼液)과 LPS를 동시에 처치하였을 경우에는 47개의 유전자에서 발현이 상승되었고 11개의 유전자에서 발현이 억제되었다. LPS 자극으로 발현이 상승되었지만 홍화약침액(紅花藥鍼液)을 처치할 때 발현이 억제되는 유전자는 SUMO1/sentrin specific protease 7(SENP7), Serine(or cysteine) proteinase inhibitor, clade B(ovalbumin), member 7(SERPINB7), M-phase phosphoprotein, mpp8(HSMPP8), Glycogenin 2(GYG2), InaD-like(Drosophila)(INADL), Copine III(CPNE3), Loss of heterozygosity, 11, chromosomal region 2, gene A(LOH11CR2A), Chromosome 9 open reading frame 33(SHC3), NADH dehydrogenase(ubiquinone) 1 beta subcomplex, 2, 8kDa(NDUFB2)로 9개가 있었다. 요약 : 홍화약침액(紅花藥鍼液)이 LPS로 염증을 유발시킨 RAW 264.7 cell의 유전자 발현에 미치는 영향을 Microarray를 통해 분석하였다. 홍화약침액(紅花藥鍼液)이 LPS로 발현을 항진시킨 35개의 유전자 중 9개를 효과적으로 억제하는 것을 확인하여 염증 치료 기전을 시사하는 유용한 자료를 얻을 수 있었으며 홍화약침액(紅花藥鍼液)이 발현을 항진시킨 유전자들을 통해 혈관생성과 종양억제 등 보다 넓은 범위에 대한 연구가 가능할 것으로 사료된다.
