Comparison of in vitro propagation and occurrence of morphological and genetic variation in strawberry tissue culture with various plant hormone treatments (딸기 조직배양 시 여러가지 식물호르몬 처리에 따른 기내 증식 및 형태적, 유전적 변이 발생 비교)

  • The objective of this study was to carry out treatment of various plant hormones in order to determine morphological and genetic variation degree of tissue-cultured strawberry. The cultivar used in this experiment was 'Goha' and 'Seolhyang', the plant hormones used for experiment were benzyladenine (BA), N-(2-Chloro-4 pyridyl)-N'-phenylurea (CPPU) and thidiazuron (TDZ), and the concentrations were 0.5, 1.0, 2.0, $4.0mg{\cdot}L^{-1}$ with each hormone. The BA treatment of the proliferation efficiency of tissue-cultured strawberry 'Goha' and 'Seolhyang' was the highest. When processing BA, CPPU and TDZ, morphological variation and genetic variation happened in strawberry 'Goha' and 'Seolhyang', especially, the variations appeared highly in CPPU treatment. The genetic variation in 'Goha' appeared at the concentration more than BA $0.5mg{\cdot}L^{-1}$ as 1.1%, appeared at the concentration of CPPU $0.5mg{\cdot}L^{-1}$ as 15.3%, and at the concentration of TDZ $2.0mg{\cdot}L^{-1}$ as 1.2%. The genetic variation in 'Seolhyang' appeared at the concentration of BA $4.0mg{\cdot}L^{-1}$ as 2.3%, and at the concentration of CPPU $0.5mg{\cdot}L^{-1}$ as 14.3%. Therefore, CPPU should not be treated during strawberry tissue culture, and BA and TDZ should be treated at low concentration.

Multiple Shoot Induction on the New Cltivar, Saxifraga fortunei 'Greenstar' by Different Media and Plant Growth Rregulators (배지 및 생장조절제 종류를 이용한 바위떡풀 신품종 '그린스타' 의 다신초 유도)

  • This study was conducted to develop in vitro propagation techniques of new cultivar, 'Greenstar', bred by Highland Agriculture Research Center. The multiple shoot induction and plant growth of in vitro plant were analyzed by MS media concentration (1/2 MS, 1 MS and 2 MS medium), plant growth regulators and its proper concentration; CPPU [Forchlorfenuron(N-(2-chloro-4-pridyl)-3-phenylurea) (0, 0.5, 1.0 and $2.0mg{\cdot}L^{-1}$), thidiazuron [(TDZ), (0.01, 0.1, 0.5 and $1.0mg{\cdot}L^{-1}$), zeatin (0, 0.5, 1.0 and $2.0mg{\cdot}L^{-1}$), and BA [6-benzylaminiopurine(BA), (0, 0.5, 1.0 and $2.0mg{\cdot}L^{-1}$)] in MS (3% sugar and 0.8% agar with pH 5.7) media. The highest number of induced shoots, leaves and roots were shown in 1/2 MS medium concentration. On the 1/2 MS medium, shoot numbers, shoot length, leaf numbers and root numbers were 11.0, 1.9 cm, 24.7, and 8.0, respectively. On the absence of CPPU in the 1/2 MS medium, shoot length and root numbers was greater than CPPU treatment, but the highest number of shoots was induced by the $2.0mg{\cdot}L^{-1}$ of CPPU concentration in 1/2MS medium. TDZ, zeatin, and BA treatments were not effective on the induction of multiple shoot in vitro culture. As a result, in vitro culture of new Saxifraga fortunei, 'Greenstar' with $2.0mg{\cdot}L^{-1}$ of CPPU in 1/2 MS medium was most effective for the rapid multiplication.

Effect of PGRs and various co-packing materials on storage quality in 'Shine Muscat' grapes

  • This investigation assessed the berry quality after two months of low-temperature storage (3 ± 1℃) of 'Shine Muscat' grapes, which were treated with CPPU (N-[2-chloro-4-pyridyl]-N'-phenylurea) or TDZ (1-phenyl-3-[1,2,3-thiadiazol-5-yl] urea) in combination with gibbrellic acid (GA3). The berry shatter rate was the lowest (1%) in 4 cm + CPPU treatment, while it was the highest (2.4%) in 3 cm floral length treated with CPPU. On the other hand, the 4 cm + TDZ treatment resulted in a shatter rate of 2.0%, which was twice as high as that observed after 4 cm + CPPU treatment. The 4 cm + TDZ treatment resulted in a 5.5% berry decay rate, which was the highest among all treatments. Alternatives to using a sulfur dioxide (SD) pad to maintain the quality of 'Shine Muscat' grapes, namely, using ethylene scrubbers (ESs, 3 g × 2 sachet) and alcohol releasers (ARs, 2 g × 2 sachet) in a 2 kg carton package for export, were explored in this study. The berry shatter rate with ES treatment (1.0%) was found to be comparable to that with SD treatment (0.6%) during three months of cold storage. Regarding the berry decay rate, that of the untreated control surged to 36.0% in the three months of storage, followed by 19.9% and 15.5% in samples subjected to AR and ES treatments respectively. Compared with the untreated control, the samples subjected to SD treatment showed a decay rate of 2.2%, which was the most effective in reducing berry decay by 95%. These results demonstrated that SD pad treatment of 'Shine Muscat' grapes was the most effective method of maintaining berry quality, and ES treatment partially reduced the berry shatter and berry decay rates.

Effects of Synthetic Cytokinin, Thidiazuron, on Berry Size and Quality of 'Kyoho'(Vitis labruscana) Grapes (시토키닌 활성물질 Thidiazuron 처리가 포도 '거봉'(Vitis labruscana)의 과립비대(果粒肥大) 및 품질(品質)에 미치는 영향(影響))

  • This experiment was conducted to find the effect of a non-purine cytokinin, thidiazuron(TDZ), on the increase of berry size and fruit quality in 'Kyoho' grapes. For the comparison, the effect of forchlorfenuron(CPPU) was also examined The number of shot berries per cluster was significantly increased by the application of TDZ at 5 days after calyptra fall, resulting in the increase of cluster weight but no increase of each berry weight. The increase of berry weight was found when TDZ was applied at 10 to 15 days after calyptra fall. Gibberellin(GA $25mg{\cdot}L^{-1}$) was also effective on the increase of berry weight and the combined application of GA and TDZ further increased the berry weight. TDZ showed stronger effect on the increase of berry weight than CPPU at the same concentration. Berry ripening was more delayed at higher concentration of TDZ but no chemical injury was observed even at $20mg{\cdot}L^{-1}$. Results indicated that TDZ was effective on the quality improvement through berry weight increase and the recommended concentration TDZ for 'Kyoho' grapes was 5 to $10mg{\cdot}L^{-1}$.
