Anti-allergic Effect of Seungmagalgeun-tang through Suppression of NF-${\kappa}B$ and p38 Mitogen-Activated Protein Kinase Activation in the RBL-2H3 Cells

  • In previous report, Seungmagalgeun-tang (SGT) could exert its anti-inflammatory actions in the BV-2 microglial cells. However, study on the anti-inflammatory effect of SGT in mast cells has not been identified. Therefore, we examined on the anti-inflammatory effect of SGT on the phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187-induced rat basophilic leukemia (RBL-2H3) cells. SGT inhibited the release of ${\beta}$-hexosaminidase and secretion and expression of pro-inflammatory cytokines such as tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-4 on RBL-2H3 cells, without affecting cell viability. The protein expression level of nuclear factor (NF)-${\kappa}B$ (p65) was decreased in the nucleus by SGT. In addition, SGT suppressed the degradation of inhibitory protein $I{\kappa}B-{\alpha}$ protein, the activation of p38 mitogen-activated protein kinase (MAPK), and the expressions of cyclooxygenase (COX)-2 mRNA and protein level in RBL-2H3 cells. These results suggest that SGT could be involved anti-allergic effect by control of NF-${\kappa}B$ (p65) translocation into the nucleus through inhibition of $I{\kappa}B-{\alpha}$ degradation and suppression of COX-2 expression.

Inhibitory Effect of Chan-Su on the Secretion of PGE2 and NO in LPS-stimulated BV2 Microglial Cells

  • 본 논문은 오랫동안 민간요법으로 염증치료에 사용되어오던 섬수가 lipopolysaccharide(LPS)-자극된 BV2 소교 세포의 nitric oxide(NO) 분비에 미치는 효과에 대해 연구한 내용이다. 실험 결과 섬수는 세포 생존력에 대한 영향 없이 BV2 소교 세포에서 NO 분비를 억제시켰고, nitric oxide synthase (iNOS) 단백질도 감소시켰다. 또한 섬수는 prostaglandin E2 (PGE2) 생산 및 cyclooxygenase (COX)-2 발현을 저지하였고, proinflammatory cytokines과 ${IkB-\alpha}$감소를 억제시켰다. 따라서 섬수가 $I{\kappa}B-{\alpha}$감소를 억제함으로써 NO 합성을 저해하여 항염증작용을 할 수 있다는 내용이다.

감마선을 이용한 육종 차조기의 항염증 효과 (Effects of anti-inflammatory on Perilla frutescens var. crispa Induced by mutants with γ-Ray)

  • 본 연구는 감마선을 이용한 육종 차조기 추출물을 통해 항염증 효능을 평가하고자 하였다. RAW 264.7 세포에서 MTT를 통해 세포 생존율을 평가하였으며, LPS로 유도한 RAW 264.7 세포에서 ROS, NO, 염증성 사이토카인, $NF-{\kappa}B$, COX-2 등을 ELISA, Luminex 및 PCR로 측정하였다. 그 결과, 육종 차조기 추출물은 $25{\mu}g/m{\ell}$ 이하에서 세포독성이 없었으며, LPS로 유도된 RAW 264.7 세포에서 ROS, NO, 사이토카인($IL-1{\beta}$, IL-6, $TNF-{\alpha}$)의 생성을 억제하였다. 또한, $NF-{\kappa}B$, COX-2의 발현을 감소시켜 육종 차조기 추출물은 뛰어난 항염증 효과를 보였다. 이와 같은 결과는 염증 매개체로 인해 발생하는 질환을 개선하기 위한 새로운 건강식품 및 치료제의 원료로 개발될 수 있다.

등골나물 추출물의 항염증 효과 (Anti-inflammatory Effect of Ethanol Extract from Eupatorium japonicum)

  • 등골나물(Eupatorium japonicum)은 식용으로 이용되어온 식물이지만 생리활성에 대한 연구가 없었다. 따라서 본 연구진은 등골나물 에탄올추출물이 쥐 대식세포인 Raw264.7 세포에 LPS로 유도된 염증 반응에 미치는 영향에 대하여 연구를 수행하였다. 등골나물 꽃 부분에 70% 에탄올을 가하여 얻은 등골나물 에탄올추출물(EJE)을 Raw264.7 세포에 LPS와 함께 0, 1, 2.5, 5, 10 mg/L로 처리하여 세포를 배양하였다. LPS에 의해 생성된 NO 및 $PGE_2$ 분비는 EJE를 처리함에 따라 감소하였고 이 결과는 EJE의 독성에 의한 것이 아님이 증명되었다. Raw264.7 세포에 LPS에 의해 생성된 iNOS, COX-2의 단백질과 mRNA의 발현이 EJE의 농도 의존적으로 억제되었으며, 염증 반응시 생성되는 IL-6, IL-${\beta}$, TNF-${\alpha}$의 mRNA 발현도 등골나물 추출물에 의해 현저히 억제되었다. 더욱이 EJE의 분획물 중 EA와 MC 분획물이 독성이 적으면서 효과적으로 NO의 생성을 억제하는 것으로 나타났다. NO 생성 억제효과가 뛰어난 이들 분획물 내의 생리활성 물질에 대한 연구가 추가적으로 수행되어야 한다고 사료된다. 등골나물 꽃을 추출물로 항염증효과를 연구한 논문이 없고 다른 식용 가능한 천연재료들의 항염증효과와 비교하여(data not shown) EJE 분획물의 항염증효과가 낮은 농도에서 탁월한 점을 미루어 볼 때, 등골나물 추출물은 상당히 낮은 농도에서 뛰어난 항염증 물질을 가진 단일물질을 포함할 가능성이 높다고 사료된다. 따라서 위의 결과는 등골나물 추출물이 독성과 부작용이 적은 염증 치료제로 활용될 수 있는 가능성이 높음을 제시한다.

  • 목 적 : 이 연구는 천식, 기관지염, 폐렴, 결핵, 산후감모 등의 호흡기 질환에 사용되는 청폐화담탕(淸肺化痰湯)의 항염작용(抗炎作用)의 효과에 대해 알아보기 위해 시행되었다. 방 법 : 청폐화담탕(淸肺化痰湯)의 항염작용(抗炎作用)의 효과를 평가하기 위해 세포독성에 미치는 영향, NO, TNF-${\alpha}$, IL-l${\beta}$, IL-6 생성량에 미치는 영항, TNF-${\alpha}$, IL-l${\beta}$, IL-6 유전자 발현에 미치는 영향, iNOS, 염증cytokine 유전자 및 단백질 발현에 미치는 영향, PGE$_2$ 합성에 미치는 영향 및 NF-${\kappa}$B 활성에 미치는 영향에 대한 실험평가를 하였다. 결 과 : 청폐화담탕(淸肺化痰湯)은 MTT 분석을 통한 RAW 264.7 세포주의 생존력 평가에서 세포독성이 없었고, LPS로 유도된 RAW 264.7 세포주에서 NO 생성량을 농도 의존적으로 억제하였다. 청폐화담탕(淸肺化痰湯)은 400 g/ml 농도에서 LPS로 유도된 RAW 264.7 세포주에 대해 TNF-${\alpha}$, IL-1${\beta}$, IL-6 생성량을 각각 41.86${\pm}$2.26 %, 61.11${\pm}$2.54 %, 55.33${\pm}$3.65 % 억제하였으며 TNF-${\alpha}$, IL-1${\beta}$ 및 IL-6 유전자 발현을 농도 의존적으로 억제하였고, LPS로 유도된 RAW 264.7 세포주에서 iNOS와 COX-2 유전자 및 단백질 발현은 농도 의존적으로 억제하였다. 또한 그 농도에 따라 PGE$_2$ 생성량이 현저하게 억제하였고, LPS로 유도된 NF-${\kappa}$B 전사활성을 농도 의존적으로 억제함으로써 iNOS와 염증Cytokine 유전자 발현을 하향조절 하였다. 결 론 : 이상의 실험을 통해 청폐화담탕(淸肺化痰湯)은 LPS로 유도된 macrophage에서 NO와 염증Cytokine 생성량을 억제하였고 murine macrophage에서 NF-${\kappa}$B 활성을 억제함으로써 iNOS와 염증Cytokine 유전자 발현을 하향조절 하였다. 이러한 청폐화담탕(淸肺化痰湯)의 항염작용으로 천식, 기관지염, 폐렴, 결핵, 산후감모 등의 호흡기 질환에 응용할 수 있으리라 사료된다.

청간해울탕(淸肝解鬱湯)과 십륙미유기음(十六味流氣飮)의 유방암에 대한 항암, 항염 효능 연구 (Research on the Anti-Breast Cancer and Anti-Inflammatory Effects of Chungganhaewool-tang and Shipyeukmiyeugi-eum)

  • Objectives: The purpose of this study is to evaluate anti-breast cancer and anti-inflammatory effects of Chungganhaewool-tang and Shipyeukmiyeugi-eum. Methods: MDA-MB-231 cells were used to measure cytotoxicity, Reactive oxygen species (ROS) production, protein expression amounts of Bcl-2-associated X protein (Bax), B-cell lymphoma 2 (Bcl-2), B-cell lymphoma-extra large (Bcl-xl), Cytochrome C Caspase-3, Caspase-7, Caspase-9, Poly ADP-ribose polymerase (PARP), Nuclear factor erythroid-2-related factor 2 (Nrf2), Heme oxygenase-1 (HO-1) and NAD (P) H Quinone Oxidoreductase 1 (NQO1) to evaluate the anti-breast cancer effects of Chungganhaewool-tang (CHT) and Shipyeukmiyeugi-eum (SYE), and THP-1 cells, differentiated into macrophage and induced inflammation with Lipopolysaccharide (LPS), were used to measure production amounts of ROS, Nitric oxide (NO), and protein expression amounts of Inducible nitric oxide synthase (iNOS), Cyclooxygenase (COX-2), Interleukin-1 beta (IL-1β), Interleukin-6 (IL-6) and Tumor necrosis factor-alpha (TNF-α) to evaluate the anti-inflammatory effects of CHT and SYE. Results: CHT and SYE reduced MDA-MB-231 cell counts, increased protein expression of Bax and Cytochrome C, and decreased protein expression of Bcl-2, Bcl-xl. The protein expression amounts of Caspase-3, 7, and 9 decreased, but amounts of the active form, cleaved Caspase-3, 7, and 9, increased. In addition, PARP protein expression decreased, the amount of PARP protein in the cleaved form increased, and the amount of protein expressions of Nrf2 and HO-1 decreased, but NQO1 showed no significant difference. In THP-1 cells CHT and SYE reduced ROS and NO, and reduced protein expressions of iNOS, COX-2, IL-1, and TNF-α, but only SYE groups reduced IL-6. Conclusions: This study suggests that CHT and SYE have potential to be used as treatments for breast cancer.

아쿠아포닉스 기술을 이용한 친환경 항노화 제품 개발 (Development of Sustainable Anti-aging Products Using Aquaponics Technology)

  • 본 연구에서는 국내 자생식물로부터 지속가능한 천연 항노화 화장품 신소재 개발을 위해 수생식물인 어리연꽃의 친환경 아쿠아포닉스 시스템 적용 가능성 및 항노화 효능을 검증하였다. 어리연꽃은 아쿠아포닉스 시스템내에서 부엽형태의 담액식 방식에서 성장 가능하고, 근경번식을 통해 증식됨이 확인되었다. 또한, 어리연꽃의 재배에 질산염, 칼륨 및 수온이 크게 영향을 미치며, 최적 조건은 각각 $80{\mu}g/mL$, $63.5{\mu}g/mL$$25^{\circ}C$로 확인되었다. 특히 주성분 2종(3,7-di-O-methylquercetin-4'-O-${\beta}$-glucoside 및 sweroside)이 약 $5{\mu}g/mL$ 이상 함께 존재 시 시너지 유효성이 있었다. 어리연꽃 추출물은 환경 유해물질 $benzo[{\alpha}]pyrene$, ammonium nitrate, formaldehyde에 의해 손상된 피부 세포의 회복에 유의미한 효과가 있으며, 염증성 조절 인자인 $PGE_2$, $TNF-{\alpha}$ 및 COX-2를 억제하고, 콜라겐 분해 효소인 MMP-1의 생성을 억제함으로써 항염 및 항노화 효과가 있음이 확인되었다. 따라서 아쿠아포닉스 기술 기반 어리연꽃 추출물의 원료 표준화는 화장품 분야에서 신규 항노화 기능성 소재로서의 활용 가능성이 있을 것으로 사료된다.

황약자(黃藥子) 메탄올 추출물의 염증억제 효과 (Inhibitory Effect of Dioscorea Bulbifera MeOH Extract on Pro-inflammatory Mediator In Vitro and In Vivo)

  • Dioscorea bulbifera is one of the traditional medicinal herb. It commonly used in the treatment of hematemesis, epistaxis, tuberculous cervical lymphadenitis, laryngitis, acute infectious disease in East Asia. In the present study, we have demonstrated the anti-inflammatory effects of Dioscorea bulbifera MeOH extract (DBME) in macrophage cell line. To investigate mechanism of the anti-inflammatory activity, we examined the effects of the lipopolysaccaride (LPS)-induced production of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), pro-inflammatory cytokines and expression of inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), p-inhibitory ${\kappa}B{\alpha}$ (p-$I{\kappa}B{\alpha}$), and nuclear factor-${\kappa}B$ (NF-${\kappa}B$) in a murine macrophage cell line RAW 264.7. The RAW 264.7 cells were cultured in DMEM + serum medium for 24 hrs. After serum starvation for 24 hrs, the cells were treated with DBME 0.03, 0.10, 0.30 mg/$m{\ell}$ for 1 h, followed by stimulation with LPS (1 ${\mu}g/m{\ell}$) for activation of immune response. After treatment, cell viability was measured by MTT assay, and NO production was monitored by measuring the nitrite content in culture medium. The protein band of iNOS, COX-2, p-$I{\kappa}B{\alpha}$, and NF-${\kappa}B$ was determined by immunoblot analysis and levels of cytokine were analyzed by sandwich immunoassays. There were three experimental groups: carrageenan, DBME 0.3, 1.0 g/kg. Rats were administrated either carrageenan (40% PEG) or carrageenan + DBME (0.3, 1.0 g/kg body weight) for 4 days (p.o.). To induce acute paw edema, rats were injected 1% carrageenan (100 ${\mu}{\ell}$/rat, dissolved in sterilized saline). The effect of DBME in the carrageenan-induced rat paw edema. As results, DBME has an inhibitory effect on the production of NO, PGE2, TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 and on the expression of iNOS, COX-2, p-$I{\kappa}B{\alpha}$ and translocation of NF-${\kappa}B$ to nuclear from cytosol. In addition, DBME effectively inhibited the increases of paw edema induced by carrageenan treatment in vivo. These results suggest that DBME can inhibit production of pro-inflammatory mediators and might be a useful source for treatment of acute inflammatory disease.

탈명산(奪命散)의 항산화 및 항염증효과에 관한 연구 (Anti-oxidative and anti-inflammatory experiments of Talmyung-san in RAW264.7 cells)

  • Objectives : The aim of this study was identification of the anti-oxidative and anti-inflammatory effects of Talmyung-san (TMS) in mouse macrophage, RAW264.7 cells. Methods : To identify the anti-oxidative effect of TMS, scavenging activities of DPPH radical, nitric oxide and peroxynitrite were measured in vitro. In RAW264.7 cells, DCFH-DA assay was conducted to examine the inhibitory effect of TMS on ROS production in response to lipopolysaccharide. And the productions of nitric oxide (NO), $PGE_2$ and pro-inflammatory cytokines were measured. The levels of COX-2, iNOS, nuclear NF-${\kappa}B$ p65 expression and phosphorylation of $I{\kappa}B-{\alpha}$ in cytosol were detected by western blotting analyses. Results : TMS couldn't scavenged DPPH radical, but nitric oxide and peroxynitrite were decreased. TMS decreased intracellular ROS, NO, and IL-$1{\beta}$ production effectively. However, TMS inhibited $PGE_2$ levels only in high concentration ($300{\mu}g/m{\ell}$) and TMS failed to suppress the production of IL-6 and TNF-${\alpha}$. Results from immunoblot analyses revealed that TMS decreased activation of COX-2, iNOS, phosphorylation of $I{\kappa}B-{\alpha}$ and nuclear translocation of p65. Conclusions : TMS has anti-RNS and anti-inflammatory effects via NF-${\kappa}B$ pathway and more intensive studies will be required to evaluate therapeutic potential of TMS.

봉약침액(蜂藥鍼液)과 Melittin 약침액(藥鍼液)이 RAW 264.7 세포의 PGE2, COX-2 및 NF-kB에 미치는 영향(影響) (The Effects of Bee Venom and Melittin Solution on PGE2, COX-2, and NF-kB Dependent Luciferase Activity in RAW 264.7 Cells)

  • Objective : The purpose of this study was to investigate the effect of Bee Venom and Melittin Solution on the lipopolysaccharide(LPS) and sodium nitroprusside(SNP)-induced expression of prostaglandin $E_2(PGE_2)$, cyclooxygenase-2(COX-2), nuclear factor kappa B($NF-{\kappa}B$) and nuclear factor kappa B($NF-{\kappa}B$) dependent luciferase activity in RAW 264.7 cells, a murine macrophage cell line. Methods : The expression of PGE2 was determined by determination of $PEG_2$, COX-2 was by western blotting with corresponding antibodies, $NF-{\kappa}B$ was by gel mobility shift assay method and $NF-{\kappa}B$ dependent luciferase activity was investigated by luciferase assay in RAW 264.7 cells. Results : 1. LPS and SNP-induced expression of $PEG_2$ was significant after 24hour. 2. The 0.5, 1 and $5{\mu}g/mL$ of bee venom and the 5 and $10{\mu}g/mL$ of melittin solution inhibited significantly LPS-induced expression of $PEG_2$ and, the $5{\mu}g/mL$ of bee venom and the 5 and $10{\mu}g/mL$ of melittin solution inhibited significantly SNP-induced expression of $PEG_2$ compared with control, respectively. The 0.5 and $1{\mu}g/mL$ of bee venom could not significantly inhibit SNP-induced expression of $PEG_2$ compared with control. 3. The $5{\mu}g/mL$ of bee venom and the 5 and $10{\mu}g/mL$ of melittin solution inhibited significantly LPS and SNP-induced expression of COX-2 compared with control, respectively. The 0.5 and $1{\mu}g/mL$ of bee venom inclined to decrease LPS and SNP-induced expression of COX-2 compared with control. 4. The 0.5, 1 and $5{\mu}g/mL$ of bee venom and the 5 and $10{\mu}g/mL$ of melittin solution inhibited significantly LPS and SNP-induced expression of $NF-{\kappa}B$ compared with control, respectively. 5. The 0.5, 1 and $5{\mu}g/mL$ of bee venom and the 5 and $10{\mu}g/mL$ of melittin solution inhibited significantly LPS-induced expression of $NF-{\kappa}B$ dependent luciferase activity and the 1 and $5{\mu}g/mL$ of bee venom and the 5 and $10{\mu}g/mL$ of melittin solution inhibited significantly SNP-induced expression of $NF-{\kappa}B$ dependent luciferase activity compared with control, respectively. The $NF-{\kappa}B$ inhibitor also inhibited significantly LPS and SNP-induced expression of $NF-{\kappa}B$ dependent luciferase activity compared with control. 6. The 0.5, 1 and $5{\mu}g/mL$ of bee venom and the 5 and $10{\mu}g/mL$ of melittin solution inhibited significantly LPS + IFN-${\gamma}$, TNF-${\alpha}$ and LPS + TNF-${\alpha}$-induced expression of $NF-{\kappa}B$ dependent luciferase activity compared with control, respectively. The $NF-{\kappa}B$ inhibitor also inhibited significantly LPS and SNP-induced expression of $NF-{\kappa}B$ dependent luciferase activity compared with control. Conclusions : These results suggest the inhibitory action of bee venom and melittin solution on the inflammatory mediators such as $PEG_2$, COX-2 and $NF-{\kappa}B$.
