  • Abnormal regulation of the inducible form of cyclooxygenase (COX-2) has been often observed in various types of cancerous and transformed cells. Recently, targeted inhibition of COX-2 is recognized as one of the promising strategies for the prevention or treatment of cancer as well as inflammation, As part of a program to evaluate the cancer chemopreventive potential of anti-inflammatory phytochemicals, we initially determined the COX-2 inhibitory activity of some naturally occurring diarylheptanoids structurally related to curcumin.(omitted)

Raw 264.7 세포에서 섬바디나물 추출물의 iNOS, COX-2 단백질 및 mRNA 발현 억제 효과 (Inhibitory Efficacy of Dystaenia takeshimana Extract on iNOS, COX-2 Protein and mRNA Expression in Raw 264.7 Cell)

  • 본 연구에서는 섬바디 나물의 항염증 효과를 알아보기 위하여 LPS로 염증을 유도한 Raw 264.7 세포에 대한 섬바디나물 80% 에탄올 추출물의 효과를 살펴보았다. 섬바디 나물 추출물을 LPS로 유도된 Raw 264.7 대식세포에서 전염증성인자(iNOS, COX-2)들을 생성하여 측정하였다. 섬바디 나물 추출물의 대식세포에서의 세포 독성 측정을 MTT를 수행하였다. 섬바디 나물 추출물의 세포 독성을 측정한 결과, $1,000{\mu}g/ml$의 농도에서 100% 이상의 세포 생존율을 보였다. 섬바디 나물 추출물의 50, 100, $500{\mu}g/ml$ 농도에서 iNOS와 COX-2의 단백질 발현 억제 효과를 측정하기 위해 western blot을 통해 측정하였고, 양성대조군으로는 ${\beta}-actin$을 사용하였다. 그 결과, 섬바디 나물 추출물을 western blot을 통해 측정한 iNOS, COX-2의 단백질 발현 억제 효과는 $500{\mu}g/ml$ 농도에서 각각 56%, 61.6%로 감소하였다. 섬바디 나물 추출물의 50, 100, $500{\mu}g/ml$ 농도에서 iNOS, COX-2의 mRNA의 발현억제 효과를 측정하기 위해 RT-PCR을 통해 측정하였으며, 양성 대조군으로 GAPDH를 사용하였다. 그 결과, 섬바디 나물 추출물을 RT-PCR을 통해 iNOS, COX-2의 mRNA 발현 억제 효과를 측정한 결과는 $500{\mu}g/ml$에서 각각 77.9%, 83.3%로 감소하였다. 이를 통해, 섬바디 나물 추출물은 염증을 억제시켜 주는 가능성이 있는 항염증 물질로써의 효과가 있을 것으로 보여진다.

An Effective Algorithm of Power Transformation: Box-Cox Transformation

  • When teaching the linear regression analysis in the class, the power transformation must be introduced to fit the linear regression model for nonlinear data. Box and Cox (1964) proposed the attractive power transformation technique which is so called Box-Cox transformation. In this paper, an effective algorithm selecting an appropriate value for Box-Cox transformation is developed which is considered to find a value minimizing error sum of squares. When the proposed algorithm is used to find a value for transformation, the number of iterations needs to be considered. Thus, the number of iterations is examined through simulation study. Since SAS is one of most widely used packages and does not provide the procedure that performs iterative Box-Cox transformation, a SAS program automatically choosing the value for transformation is developed. Hence, the students could learn how the Box-Cox transformation works, moreover, researchers can use this for analysis of data.

The COX-2 -765 G>C Polymorphism is Associated with Increased Risk of Gastric Carcinogenesis in the Chinese Hui Ethnic Population

  • Background: The Chinese Hui ethnic group has diverse origins, including Arab, Persian, Central Asian, and Mongol. The standardized mortality rate of gastric cancer in the Hui population is higher than the overall Chinese population. In this study, we investigated whether COX-2-765G>C polymorphism, an extensively studied polymorphism, contributes to gastric cancer and its precursor lesions (GPL) in the Chinese Hui ethnic group. Materials and Methods: COX-2-765G>C polymorphism was determined by pyrosequencing in 100 gastric cancer cases, 102 gastric cancerand its precursor lesions cases and 105 controls. Data were statistically analyzed using Chi-square tests and logistic regression models. Results: Among the Chinese Hui ethnic group COX-2-765 C allele carriers were at increased risk for gastric cancer (OR=1.977, 95%CI=1.104-3.541). We also found an interaction between COX-2 -765 C carriers and Helicobacter pylori infection and eating pickled vegetables. Conclusions: Our findings suggest a multi-step process of gene-environment interaction contributes to gastric carcinogenesis.

에스트로겐에 의한 암쥐의 심장조직의 COX-2, Mapkinases 및 관련된 Apoptotic Markers의 발현의 변화에 관한 연구 (Changes in the Expression Pattern of Cyclooxygenase-2, Mapkinases and Related Apoptotic Markers by Different Levels of Estrogen Supplementation in Mature or Ovariectomized Female Rat Heart)

  • The effects of different concentrations of estrogen supplementation to mature female rats or estrogen supplementation to ovariectomized rats on cyclooxygenase-2 (COX-2) expression, PGE$_2$ production and mapkinases expression were investigated in experimentally induced atherogenic rats with feeding a high fat. high cholesterol diet. In the first experiment using 48-week old mature rats, the supplementation of three different levels of estrogen was compared to the basal diet. The high concentration of estrogen supplementation induced the marked up-regulation of COX-2 protein and the increase in plasma PGE$_2$ production and this seems to be followed by the up-regulation of p38 among mapkinases. The regulation of bax showed in a reverse trend of COX-2 in heart tissues of mature female rats. In the second ex-perimental system, female Sprague-Dawley rats were bilaterally ovariectomized; sham-operated animals were used as controls. Three weeks later, the animals were supplied with basal diet to sham-operated control group and ovariectomized control group, and estrogen supplemented diet to ovariectomized group for an eight-week experimental period. In a group supplemented with a medium dose of estrogen, COX-2 expression was up-regulated. This up-regulation was accompanied by the elevated expression of pERK1/2. Bax was increased in estrogen-fed animals indicating bax might be involved in estrogen feeding state in ovariectomized rats. Further investigations on the relationship between COX-2 and biological activities such as vasodilation by estrogen are required in in vivo system of female rats at the various physiological states.

The Prostaglandin Synthase 2/cyclooxygenase 2 (PTGS2/COX2) rs5277 Polymorphism Does not Influence Risk of Colorectal Cancer in an Iranian Population

  • Background: The prostaglandin-endoperoxide synthase 2 [PTGS2, commonly known as cyclooxygenase-2 (COX-2)] is an enzyme induced by proinflammatory stimuli that is often overexpressed in malignant tissue and involved in the synthesis of prostaglandins and thromboxanes, regulators of processes such as inflammation, cell proliferation, and angiogenesis, all relevant for cancer development. We investigated whether a functional genetic polymorphism, rs5277, in COX-2 may have a risk-modifying effect on sporadic colorectal cancer in an Iranian population. Materials and Methods: We conducted a case-control study on 167 patients with colorectal cancer and 197 cancer-free controls in Taleghani Hospital in Tehran, Iran, between 2007 and 2011. Peripheral blood samples of both groups were processed for DNA extraction and genotyping of the COX-2 gene polymorphism (rs5277) using PCR-RFLP. RFLP results were confirmed by direct sequencing. Logistic regression analysis was performed to calculate the adjusted odds ratio (OR) and 95% confidence interval (95% CI). Results: There was no significant difference in the distribution of COX-2 gene rs5277 polymorphism genotype and the allelic form, among CRC patients compared with the healthy control group (p: 0.867). Conclusions: Our results suggest that rs5277 polymorphism in COX2 could not be a good prognostic indicator for patients with CRC.

  • Box-Cox power transformation is employed for analyzing volatilities in Korean financial time series such as KOSPI, KOSDAQ index and interest rates. Statistical procedures for Box-Cox transformed ARCH models are presented. For illustration, diverse financial time series data are analyzed and appropriate power transformations are suggested for each data.

발효 콩의 NF-κB 활성 억제를 통한 cyclooxgenase-2 활성과 prostaglandin E2 생성 억제 (Inhibition of Cyclooxygenase-2 Activity and Prostaglandin E2 Production through Down-regulation of NF-κB Activity by the Extracts of Fermented Beans)

  • 본 연구에서는 콩 발효 산물 추출물의 항염증 효능에 관한 기초 자료를 제시하기 위하여 PMA에 의해 유도되는 COX-2의 발현 및 $PGE_2$의 생성 증가에 미치는 이들 추출물의 영향을 조사하였다. 본 연구에서 조사된 3가지 콩 발효 산물은 PMA에 의한 COX-2의 발현 증가를 매우 유의적으로 차단하였으며, 이는 $PGE_2$의 생성 억제와 연관성이 있었다. 아울러 PMA에 의한 NF-${\kappa}B$ 활성 증가 또한 콩 발효 산물들에 의하여 유의적으로 억제되었다. 이러한 결과들은 콩 발효 산물에 의한 NF-${\kappa}B$ 활성의 억제가 COX-2의 발현을 저하시켰으며, 이로 인한 $PGE_2$의 생성이 억제된 것으로 추정되어진다. 이러한 콩 발효 산물의 항염증 효과는 대두 추출물보다 아가콩 추출물에서 더욱 효과가 높게 나타났으며, 이는 향후 아가콩 추출물은 염증성 질환 예방/치료를 위한 적용 가능성이 매우 우수함을 제시하여 주는 결과이다.

  • A procedure for investigating the influence of observations in choosing Box-Cox transformation for multivariate data is suggested. It is effective in spotting influential observations. A numerical example is provided for illustration.
