Production of Glucosyl-xylitol Using Encapsulated Whole Cell CGTase (캡슐 고정화 전세포 CGTase를 이용한 Glucosyl-xylitol 생산)

  • We tried to prepare encapsulated whole cell cyclodextrin glucanotransferase(CGTase) in order to produce glycosyl-xylitol using xylitol as glucosyl acceptor. The organic nitrogen source was more effective for the production of CGTase from Bacillus macerans IFO 3490 than the inorganic one. Most of the CGTase which had been produced during cultivation was excreted to the growth medium. B. macerans cells inocculated in the capsule failed to grow to the high cell density. Adsorbents such as activated charcoal, Sephadex and Amberite resins could not adsorb efficiently the CGTase from the broth solution. We obtained successfully the encapsulated whole cell CGTase by immobilizing the concentrated broth solution in the calcium alginate capsules. The encapsulated whole cell CGTase carried out the transglycosylation reaction which converts xylitol into glucosyl-xylitol using dextrin as glucosyl donor.

성막직전 기판 열처리가 롤투롤 스퍼터를 이용하여 성장시킨 터치 패널용 ITO 투명 전극의 특성 미치는 효과 연구

  • 본 연구에서는 저가격, 대면적화를 위한 롤투롤 스퍼터를 설계&개발하고, 성막직전 PET 기판의 열처리 유무를 통한 ITO 박막을 성막 시킨 저항막 방식의 터치 패널용 투명 전극에 대하여 전기적, 광학적, 구조적, 표면적 특성을 분석하였다. 롤투롤 스퍼터는 degassing챔버와 스퍼터 챔버가 한 시스템에 구성되었고, Degassing 챔버는 좌우측의 Rewinder/Unwinder 롤러에 의해 감고 풀어지는 PET기판의 수분 및 가스를 중앙부에 위치한 히터를 통해 제거하며, 수분 제거 후 스퍼터 챔버로 옮겨진 1250 mm폭의 PET기판을 Unwinder/Rewinder 롤러에 장착하며, Unwinder 롤러로부터 풀려진 PET 기판은 guide 롤러를 거쳐 cooling drum과의 물리적 접촉에 의해 PET 기판의 냉각이 일어나게 된다. ITO 캐소드 전에 장착된 할로겐 히터 상부로 기판이 지나가면서 열처리가 진행되고 열처리 후 두 개의 ITO 캐소드 상부를 지나면서 연속적으로 ITO 박막이 PET 기판에 성막 되게 된다. ITO 박막의 주요 성막 변수인 DC Power, Ar/$O_2$ 가스 유량비, 기판의 속도는 최적으로 고정하고, 성막 직전 기판의 열처리에 유무에 따른 ITO박막의 필름을 각각 고온 챔버에서 $140^{\circ}C{\times}90min$ 동안 열처리를 통한 내열성 테스트를 진행하여 ITO 필름의 특성 향상을 비교 분석하였다. 분석을 위해 전기적 특성은 four-point probe로 측정했고, 투과도는 Nippon Denshoku사(社)의 COH-300A를 이용해 가시광(550nm)에서 분석했고, FE-SEM으로 ITO박막 의 표면 상태를 분석하였다. 또한 Bending Tester(Z-100)를 이용하여 기계적 안정성을 분석하였다. 성막직전 PET 기판의 열처리를 하지 않은 ITO박막은 고온의 챔버 에서 $140^{\circ}C{\times}90min$ 동안 내열성 테스트 후 면저항이 511($\omega/\Box$)에서 630($\omega/\Box$)으로 높아졌으나, 성막직전 열처리를 통한 ITO 박막인 경우에는 465($\omega/\Box$)에서 448($\omega/\Box$)로 안정화 되었고, 투과율은 성막직전 열처리를 통해 1%향상되어 89%를 보였고, 유연성 또한 보다 우수한 특성을 보였다. 표면 조도는 평균 0.416 nm의 낮은 값을 보였다. 이는 PET 기판의 degassing 공정 중 충분히 제거되지 않은 가스나 불순물을 성막직전 열처리 공정으로 충분히 제거하여 깨끗한 PET 기판 상에 ITO 박막을 성막시키고, 열처리시 기판에 주어진 열에너지에 의해 보다 밀도가 높은 ITO 박막이 성장했기 때문으로 사료 된다.

Dielectric Characteristics due to the nano-pores of SiOCH Thin Flm (기공형성에 의한 SiOCH 박막의 유전 특성)

  • We have studied dielectric characteristics of low-k interlayer dielectric materials was fabricated by plasma enhanced chemical vapor deposition (PECVD). BTMSM precursor was introduced with the flow rates from 24 sccm to 32 sccm by 2 sccm step in the constant flow rate of 60 sccm $O_2$. Then, SiOCH thin film deposited at room temperature was annealed at temperature of $400^{\circ}C$ and $500^{\circ}C$ for 30 minutes in vacuum. The vibrational groups of SiOCH thin films were analyzed by FT/IR absorption lines, and the dielectric constant of the low-k SiOCH thin films were obtained by measuring C-V characteristic curves. With the result that FTIR analysis, as BTMSM flow rate increase, relative carbon content of SiOCH thin film increased from 29.5% to 32.2%, and increased by 32.8% in 26 sccm specimen after $500^{\circ}C$ annealing. Dielectric constant was lowest by 2.32 in 26 sccm specimen, and decreased more by 2.05 after $500^{\circ}C$ annealing. Also, leakage current is lowest by $8.7{\times}10^{-9}A/cm^2$ in this specimen. In the result, shift phenomenon of chemical bond appeared in SiOCH thin film that BTMSM flow rate is deposited by 26 sccms, and relative carbon content was highest in this specimen and dielectric constant also was lowest value

A Methodology for Finding the Convergence Research Area by Measuring Convergence Index in Government Research Institutes (융합지수 측정을 통한 출연연 융합연구영역 발굴모형 연구)

  • In conjunction with securing R&D competitiveness through technology convergence, there is a growing interest in the strategy to derive innovation based on the measurement of convergence. In this paper, we present a data-based model that suggests convergence strategies for the research projects of government research institutes (GRIs) in Korea. The convergence characteristics of the research projects are represented by inherent attributes of technology and behavioral attributes of actors. The measuring process of the proxy variables (Rao-Stirling and Herfindahl-Hirschman Indices) for each attributes are proposed. These two indices are used to construct the convergence diagram, through which the research projects of GRIs are distributed into four areas: AllianceIntra, Alliance-Inter, Competition, Mission-Oriented. Based on the convergence diagram proposed in this study the convergence index is measured to derive 30 convergence research areas of GRIs. As a result, we expect to provide appropriate guidelines for setting the policy direction of the convergence research projects.

  • Objective: To investigate the association of FSH receptor (FSHR) polymorphism at position 680 with outcomes of controlled ovarian hyper-stimulation for IVF-ET in Korean women. Design: Genetic polymorphism analysis. Materials and Methods: The FSHR polymorphism was analyzed by PCR-RFLP in 172 ovulatory women below the age of 40 year. Patients with polycystic ovary syndrome, endometriosis, or previous history of ovarian surgery were excluded. Results: Genotype distribution was 41.9% for the Asn/Asn, 47.7% for the Asn/Ser, and 10.5% for the Ser/Ser FSHR genotype group. There was no difference in age of subjects and infertility diagnosis between genotype groups. When the patients were grouped according to their FSHR genotype, the basal levels of FSH (day 3) were significantly different among the three groups ($6.0{\pm}0.3\;IU/L$ (mean $\pm$ SEM), $5.8{\pm}0.3\;IU/L$, and $8.6{\pm}1.2\;IU/L$ for the Asn/Asn, Asn/Ser, and Ser/Ser groups, respectively, p=0.002). The Ser/Ser group showed a higher total doses of gonadotropins required to achieve ovulation induction, and a lower serum estradiol levels at the time of hCG administration compared with other two groups, but the differences were of no statistical significance. The numbers of oocytes retrieved were significantly different among the three groups ($8.6{\pm}0.8$, $9.9{\pm}0.6$, and $6.3{\pm}0.9$, for the Asn/Asn, Asn/Ser, and Ser/Ser groups, respectively, p=0.049). Clinical pregnancy rates were 42.4%, 25.9%, and 29.4% for the Asn/Asn, Asn/Ser, and Ser/Ser groups, respectively. Conclusion: Homozygous Ser/Ser genotype of FSHR polymorphism at position 680 was associated with decreased ovarian response to gonadotropin stimulation for IVF-ET.

Genetic Diversity of Korean Native Chicken Populations in DAD-IS Database Using 25 Microsatellite Markers (초위성체 마커를 활용한 가축다양성정보시스템(DAD-IS) 등재 재래닭 집단의 유전적 다양성 분석)

  • A number of Korean native chicken(KNC) populations were registered in FAO (Food and Agriculture Organization) DAD-IS (Domestic Animal Diversity Information Systems, http://www.fao.org/dad-is). But there is a lack of scientific basis to prove that they are unique population of Korea. For this reason, this study was conducted to prove KNC's uniqueness using 25 Microsatellite markers. A total of 548 chickens from 11 KNC populations (KNG, KNB, KNR, KNW, KNY, KNO, HIC, HYD, HBC, JJC, LTC) and 7 introduced populations (ARA: Araucana, RRC and RRD: Rhode Island Red C and D, LGF and LGK: White Leghorn F and K, COS and COH: Cornish brown and Cornish black) were used. Allele size per locus was decided using GeneMapper Software (v 5.0). A total of 195 alleles were observed and the range was 3 to 14 per locus. The MNA, $H_{\exp}$, $H_{obs}$, PIC value within population were the highest in KNY (4.60, 0.627, 0.648, 0.563 respectively) and the lowest in HYD (1.84, 0.297, 0.286, 0.236 respectively). The results of genetic uniformity analysis suggested 15 cluster (${\Delta}K=66.22$). Excluding JJC, the others were grouped in certain cluster with high genetic uniformity. JJC was not grouped in certain cluster but grouped in cluster 2 (44.3%), cluster 3 (17.7%) and cluster8 (19.1%). As a results of this study, we can secure a scientific basis about KNC's uniqueness and these results can be use to basic data for the genetic evaluation and management of KNC breeds.

Combination Therapy with Ciclosporine, Ketoconazole and 0.1% Tacrolimus Ointment for Fast Reduction of Perianal Fistulae Lesions in Three Military Working Dogs (싸이클로스포린, 케토코나졸, 0.1% 타크로리무스 연고제제로 병용 치료한 군 사역견의 항문주위 누공 3례)

  • Three military working dogs (MWDs) were diagnosed with perianal fistulae after a complete physical examination and rectal palpation. The initial treatment protocol was used with short-term oral ciclosporine (CyA) and ketoconazole and 0.1% topical tacrolimus ointment. When lesions were nearly healed, treatment was continued with 0.1% tacrolimus ointment only at a lower frequency. The lesions improved significantly within 2-8 weeks with no noticeable side effects. Complete recovery was determined at weeks 13, 6, and 4 for cases 1, 2, and 3, respectively. This combination protocol was effective at resolving or reducing perianal fistulae (PAF) lesions even communicating with anal sacs in three dogs. The major benefit of this protocol was the rapid reduction of fistulae lesions and this could be helpful to extend the pain-free period. Moreover, this protocol appeared cost-saving as well as time-saving option for PAF dogs.

Autosomal-dominant Polycystic Kidney Disease in a Family of Scottish Fold Cats (스코티쉬 폴드 고양이 가족에 발생한 상염색체 우성 다낭성 신병)

  • Autosomal-dominant polycystic kidney disease (AD-PKD) is common in Persian and Persian-related breeds, and is sporadically reported in Scottish Fold cats. A 5-year-old male Scottish Fold cat was diagnosed with polycystic kidney disease based on screening tests and abdominal ultrasonography and died 3.5 months after diagnosis. The cat had 14 kittens with three queens, including his female sibling, with an age range of 3 months to 8 years. Genetic testing to confirm the genetic transmission of AD-PKD which detects the mutated PKD1 gene was performed. Abdominal ultrasonography confirmed the presence of renal cysts. Nineteen cats were screened in the present study (13 males and 6 females), with an age range of 3 months to 8 years. The results of renal ultrasonography agreed with the genetic test results in the 19 cats in which both tests were performed and 8 cats were diagnosed as ADPKD based on these tests. AD-PKD has not been investigated in cats in South Korea. Moreover, this is the first report of AD-PKD in a family unit of Scottish Fold cats.

Site-Specific Mutagenesis on the 32-T and 39-T of E. coli $tRNA^{phe}$ Gene (E. coli $tRNA^{phe}$ 유전자의 32-T와 39-T 염기의 부의 특이적 돌연변이)

  • There are three pseudourdine ($Psi$)bases in the E. coli $tRNA^{phe}$ In order to study the function of the pseudouridine bases in the $tRNA^{phe}$, changes of bases $tRNA^{phe}$ gene to other bases were undertaken by the site-specific mutagenesis. Site-specific mutagenesis of T in the pheW gene, a $tRNA^{phe}$ gene of E. coli, corresponding to the baseat the No.32 position to C and also T corresponding to the base at the No.39 position to C were performed using Kunkel's uracil-containing template method. Identification of mutants were undertaken by the KNA sequencing techniques of the mutated pheW genes and activities of the mutated pheW genes complementing to E. coli NP37 mutant($pheS^{-ts}$) using the recombinant plasmid containing the mutated genes. Neither NP37 harboring pheW gene mutated at No.32 position nor NP37 harboring pheW gene mutated at No.39 position can be grown at non-permissive temperature. The result means that both mutated pheW genes can not complement to E. coli NP37, and that the pseudouridine bases are essential to the activity of the E. coli $tRNA^{phe}$ in vivo.

Effects of age on in vitro maturation and fertilization of immature oocytes from stimulated cycles in human IVF-ET program (체외수정시술 시 획득한 미성숙난자의 환자 연령에 따른 체외성숙률 및 수정률 비교)

  • Objective: To investigate the effects of female age on in vitro maturation and fertilization of immature oocytes from controlled ovarian hyperstimulation (COH) in human IVF-ET program. Method: A total of 96 immature oocytes (GV & metaphase I) obtained from 40 cycles of IVF-ET (29 patients). The mean age of female patients was $31.8{\pm}3.1years$. Ovulation was triggered by urinary or recombinant hCG. Immature oocytes were cultured with YS medium containing 30% of patients' human follicular fluids, LH (1 IU/mL), FSH (1 IU/mL) and EGF (10 ng/mL), and then matured oocytes were fertilized by ICSI. In vitro maturation and fertilization of immature oocytes were analyzed according to age of female (< 34 or ${\geq}34years$). Results: The maturation rate was similar between two groups (68% vs 64%). The fertilization rate of in?vitro-matured oocytes was higher in patients < 34 years old, but there was no statistical significance (64% vs 50%, p=0.347). The fertilization rate of in-vitro-matured oocytes was significantly lower compared with those of in-vivo-matured oocytes in both age groups (64% vs 79%, p=0.035, 50% vs 86%, p=0.007). Conclusion: In older female group, fertilization rate of in-vitro-matured oocytes seems to be decreased. Further investigations should be warranted to increase fertilization potential of in-vitro-matured oocytes.
