Control of Metal-Oxide Nanostructures for $H_{2}-Alcohol$ Fuel Cells (수소-알코올연료전지를 위한 금속-산화물 나노구조제어)

  • Due to their excellent catalytic activity with respect to methanol oxidation on platinum at low temperature, platinum nanosized catalysts have been a topic of great interest for use in direct methanol fuel cells (DMFCs). Since pure platinum is readily poisoned by CO, a by-product of methanol electrooxidation, and is extremely expensive, a number of efforts to design and characterize Pt-based alloy nanosized catalysts or Pt nanophase-support composites have been attempted in order to reduce or relieve the CO poisoning effect. In this review paper, we summarize these efforts based upon our recent research results. The Pt-based nanocatalysts were designed by chemical synthesis and thin-film technology, and were characterized by a variety of analyses. According to bifunctional mechanism, it was concluded that good alloy formation with $2^{nd}$ metal (e.g., Ru) as well as the metallic state and optimum portion of Ru element in the anode catalyst contribute to an enhanced catalytic activity for methanol electrooxidation. In addition, we found that the modified electronic properties of platinum in Pt alloy electrodes as well as the surface and bulk structure of Pt alloys with a proper composition could be attributed to a higher catalytic activity for methanol electooxdation. Proton conducting contribution of nanosized electrocatalysts should also be considered to be excellent in methanol electrooxidation (Spillover effect). Finally, we confirmed the ensemble effect, which combined all above effects, in Pt-based nanocatalsyts especially, such as PtRuRhNi and $PtRuWO_{3}$, contribute to an enhanced catalytic activity.

Raoultella ornithinolytica as a Potential Candidate for Bioremediation of Heavy Metal from Contaminated Environments

  • Disposal of waste containing heavy metals into the environment is a major threat to human health and can result in toxic or chronic poisoning in aquatic life. In the current study, metal-resistant Raoultella ornithinolytica was isolated from metal-contaminated samples collected from the Tanjaro River, located southwest of Sulaymaniyah, Iraq. R. ornithinolytica was identified by partial amplification of 16S rRNA. The uptake potency of heavy metals was assessed using inductively coupled plasma-optical emission spectroscopy (ICP-OES) and indicated that R. ornithinolytica removed 67, 89, 63.4, 55.6, 56.5, 65, and 61.9% of Cd, Pb, Cr, Ni, Zn, Co, and Fe, respectively. These removal rates were influenced by temperature, pH, and contact time; at 35℃ and pH 5 with a change in the incubation time, the reduction rate improved from 89 to 95% for Pb, from 36.4 to 45% for Cu, and from 55.6 to 64% for Ni. Gene analysis indicated that R. ornithinolytica contained pbrT, chrB, nccA, iroN, and czcA genes, but the pcoD gene was absent. Energy-dispersive X-ray spectroscopy (EDS) images showed evidence of metal ion binding on the cell wall surface with different rates of binding. Transmission electron microscopy (TEM) detected different mechanisms for metal particle localization; cell surface adsorption was the main mechanism for Pb, Zn, and Co uptake, while Cd, Ni, and Fe were accumulated inside the cell. The current study describes, for the first time, the isolation of R. ornithinolytica from metal-contaminated water, which can be used as an eco-friendly biological expedient for the remediation and detoxification of metals from contaminated environments.

  • The ultimate goal of treatment of carton monoxide poisoning is to promote dissociation of carboxyhemoglobin and to maintain arterial $PO_2$ above 50mmHg throughout the course of treatment to protect vital organs from damage caused by hypoxia. The hyperbaric chamber designed and manufactured for this purpose has obviousely made an enormous contribution and yet has several handicaps to be overcome by any means. These handicaps are: the financial impact to purchase the chamber (especially in a small, remote community), an extra manpower requirement to operate the device, limitation in the capacity of the chamber (one man type), and the possible hazard of oxygen intoxication and dysbarism. The primary objective of this study is to develope a new therapeutic measure as an alternative to the hyperbaric chamber when it is not available or contraindicated. The effect of intestinal perfusion with hydrogen peroxide has been studied by many investigators and was known to be an excellent way of extrapulmonary oxygen supply. the advantage of this method will include; 1) much more amount of oxygen is delivered to the tissue than one would expect from 100% saturation with oxygen at 1 ata, 2) the procedure is simple and most economical, 3) neither sophisticated equipment nor extra manpower is required. As a study preliminary to the clinical application, authors conducted a series of experiment to observe the effect of hydrogen peroxide enema on dissociation of carboxyhemoglobin in intoxicated rabbit blood. Using an animal gas chamber, 20 rabbits were exposed to CO gas of 6,000 ppm for 60 minutes. Ten rabbits of control group were given 10cc of warmed normal saline solution by reactal perfusion and for the other 10 of the experimental group, the same amount of 1% $H_{2}O_{2}$ solution was given by the same way. Two blood specimens were drawn from each rabbit: the first one immediately following the exposure and the second one after rectal perfusion, about 30 minutes after the first sampling. The result was as follows; 1) The decrease in carboxyhemoglobin concentration during the first 30 minutes in the control and experimental group were $18.18{\pm}4.49%\;and\;23.03{\pm}4.13%$ respectively shelving the significant difference (p<0.05) between the two groups. 2) Hemoglobin and hematocrit value showed no significant difference between two groups and not altered significantly by intestinal perfusion with $H_{2}O_{2}$.

  • Hyperbaric oxygen therapy(HBO) is defined as breathing 100% oxygen while in an enclosed system pressurized to greater than on atmosphere(sea level). This increased oxygen delivery furthers your body's ability to kill germs and to increase healing. HBO is a supplemental therapy to be used in addition to the current medical and surgical therapy you are receiving. HBO typically is used to complement treatments of medical problems such as bone infections, complication of radiotherapy, and certain chronic, non-healing wounds. On an emergency basis, the chamber also is used to treat problems such as carbon monoxide poisoning and the decompression sickness. We analysed stastically cases which are treated by hyperbaric oxygen therapy in point of oral and maxillofacial region on the Kangnam General Hospital for aid in comprehension and application of this therapy. Total 760 patients were treated at Gangnam General Hospital from July 1996 to September 1999. They were classified by region to Decompression sickness(DCS), Carbon monoxide poisoning(CO), General surgery(GS), Orthopedics(OS), Oral and Maxillofacial surgery(OMFS), others. Patients of Oral and Maxillofacial surgery were divided by diseses to Osteomyelitis, Osteoradionecrosis, Reconstruction, Bone graft, Difficult wounds, others. The results were as follows. 1. This institute conducts HBO therapy for DCS which takes up 62% and 10.5% for OMF region. 2. In OMF region, Osteomyelitis is 40%, Osteoradionecrosis is next, Bone graft, and Reconstruction is a row. 3. According to our precious study, HBO has been frequently conducted in OMF region compared to past, however, it is less actively conducted in this area for research than other conturies. Therefore, We need further application to the clinical use.

Antibacterial and Antioxidant Activity of Chamaecyparis obtusa Extracts (편백나무 추출액의 항균 및 항산화 활성)

  • In this study, we investigated the biological antioxidant and antibacterial activity of Chamaecyparis obtusa (C. obtuse) extracts by measuring DPPH radical scavenging and ABTS radical scavenging, and SOD-like activities. The DPPH and ABTS radical scavenging activities were increased in a dose-dependent manner, with maximum activities of 78% and 62% at an extract concentration of $50{\mu}l/ml$. The C. obtusa extracts also showed high SOD-like activity, with a maximum activity of 92.85% at a concentration of $50{\mu}l/ml$. The antibacterial activities of C. obtusa extracts were measured against six types of bacteria known to cause food poisoning and disease. Antibacterial activity was investigated against three gram-positive and three gram-negative bacteria using the paper disc agar diffusion method. The C. obtusa extracts showed antibacterial activities against B. cereus, E. coli, L. monocytogenes, S. aureus, S. typhi and V. parahaemolyticus, among which the activity against B. cereus was greatest. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of C. obtusa extracts were $30-40{\mu}l/ml$ for the 6 strains that showed an antimicrobial response by the paper disc agar diffusion method. These results suggest that C. obtusa extracts could serve as potential antibacterial agents to inhibit the growth of pathogens responsible for food poisoning and disease.

Bio-monitoring System for Early Detection of Toxic Dinoflagellate Alexandrium pacificum Using the Shell Valve Movements of Bivalves (유독 와편모조류 Alexandrium pacificum의 조기 탐지를 위한 이매패류 패각운동 생물모니터링 시스템 연구)

  • We examined changes in the shell valve movements (SVMs) of Mytilus edulis and Crassostrea gigas using a Hall element sensor to investigate the early detection of the toxic dinoflagellate Alexandrium pacificum. No increase or decrease was observed in SVMs caused by the non-toxic algae Isochrysis galbana in both M. edulis and C. gigas. However, when M. edulis and C. gigas were exposed to A. pacificum, which causes paralytic shellfish poisoning, the average SVMs for 12 hours before and after exposure increased from 1.25 times/hr to 2.13 times/hr and 2.23 times/hr to 8.91 times/hr, respectively. After exposure to A. pacificum, the SVMs of M. edulis increased rapidly within 1 hour and then decreased gradually. However, C. gigas showed high SVMs until 4 hours after exposure. SVMs of C. gigas appeared to be more sensitive to toxic dinoflagellate than those of M. edulis. Therefore, these results are expected to be used as basic data for the establishment of a biological monitoring system for early detection of the toxic dinoflagellate A. pacificum.

Analysis of Emission Characteristics and Emission Factors of Carbon Monoxide and Nitrogen Oxide Emitted from Wood Pellet Combustion in Industrial Wood Pellet Boilers Supplied According to the Subsidy Program of Korea Forest Service (산림청 지원사업에 따라 보급된 산업용 목재펠릿보일러에서 목재펠릿 연소 시 배출되는 일산화탄소와 질소산화물의 배출 특성 및 배출계수 분석)

  • Korea Forest Service has supplied 76 industrial wood pellet boilers from 2011 to 2015 through subsidy programs. Since carbon monoxide (CO) and nitrogen oxides ($NO_x$) generated during boiler combustion are substances that lead to death in the case of acute poisoning, it is very important to reduce emissions. Therefore, the CO and $NO_x$ emission values of 63 boilers excluding the hot air blower and some boilers initially supplied were analyzed. The emission factor was also calculated from the measured exhaust gas concentration (based on exhaust gas $O_2$ concentration of 12%). The average value of CO emitted from industrial wood pellet boilers was 49 ppm and it was confirmed that the CO concentration was decreasing as the years passed. The emission factor of CO was 0.73 g/kg. The average value of $NO_x$ emitted from industrial wood pellet boilers was 67 ppm and the emission factor of $NO_x$ was 1.63 g/kg. Unlike CO, there was no tendency to decrease according to the installation year. Both CO and $NO_x$ measurements met the limits of the Ministry of Environment. These $NO_x$ emission factors were compared with the $NO_x$ emission factors produced by certified low $NO_x$ burners. The $NO_x$ emission factor of industrial wood pellet boilers was about 1.9 times that of certified low $NO_x$ LNG combustors and about 0.92 times that of coal combustion.

The deactivation behavior of SCR catalyst by alkali and alkali earth metal (알칼리 및 알칼리 토금속에 의한 SCR 촉매 비활성 거동)

  • The effect of the alkali, alkali earth metal elements on selective catalytic reduction(SCR) catalyst deactivation behavior were investigated in terms of microstructure, surface area, pore volume and De-NOx test. Poisoned SCR catalyst were manufactured by injection of $K_2CO_3$, $Na_2CO_3$, $Ca(CH_3COO)_2{\cdot}H_2O$, $C_4H_6MgO_4{\cdot}4H_2O$, $H_3PO_4$ solutions in the new SCR catalyst at $350^{\circ}C$ for 6 hours. New and poisoned catalysts surface were similar. But specific surface area, pore volume decrease from Na, Mg, K, Ca, P compared to new SCR catalyst. Especially, Na poisoned catalyst surface area and pore size extremely decreased by $10.20m^2/g$, $0.061cm^2/g$. De-NOx test results of new and poisoned catalysts at $150{\sim}450^{\circ}C$ indicated that alkali metal (K, Na) poisoned SCR catalysts have the lowest De-NOx efficiency, alkali earth metal poisoned SCR catalysts (Ca, Mg) De-NOx efficiency are higher than alkali metal poisoned SCR catalysts. P poisoned SCR catalyst De-NOx efficiency is similar new SCR catalyst. It were considered that physical deactivation of SCR catalyst was affected by SCR catalyst surface area and pore volume change.

Antimicrobial Activity of Extracted by Supercritical Fluid from Origanum vulgare, Cinnamomum cassia, Chamaecyparis obtusa and Scutellariae baicalensis (오레가노, 육계, 편백 및 황금의 초임계 유체 추출물의 항균 활성)

  • The variety of functional plants has an attention for new natural food preservation and natural antiseptic development. The extracts from functional plants with various methods (ethanol extraction, hot water extraction and supercritical fluid extraction) tested antimicrobial activity against 10 strains including the pathogenic and food poisoning bacteria, the yeast and fungi. The antimicrobial activities of supercritical fluid extracts were shown higher than ethanol extract and hot water extract when tested with disc-diffusion method and minimum inhibitory concentration (MIC). Antimicrobial activity of supercritical fluid extract was two times higher than ethanol extract in Cinnaonomum cassia. In addition, the supercritical fluid extractions of Chamaecyparis obtuas and the C. cassia showed the higher yield than Origanum vulgare and Scutellariae baicalensis. The supercritical fluid extract of C. cassia showed an antimicrobial activity against all strains tested. The supercritical fluid extract of S. baicalensis showed strong antimicrobial activity on Listeria monocytogenes. Supercritical fluid extraction of O. vulgare and C. obtuas showed strong antimicrobial activity on Salmonella typhimuriium. In MIC test, C. obtuas was shown the best natural material for the preparation of natural antimicrobial agent by supercritical fluid extraction. In conclusion, these results suggest that supercritical fluid extraction technique was effective to obtain functional ingredient with higher antimicrobial activity in the development of new antimicrobial reagent from natural materials.

Okadaic Acid Group Toxins: Toxicity, Exposure Routes, and Global Safety Management (오카다익산 군 독소: 독성, 분석법 및 관리 동향)

  • Okadaic acid (OA) group toxins, including OA and its analogs, such as dinophysis toxins (DTXs), have been reported to cause diarrheal shellfish poisoning (DSP). These toxins are primarily produced by dinoflagellates and are accumulated in bivalves. Recently, the presence of Dinophysis sp., a causative alga of DSP, has been reported along the coasts of Korea, posing a potential risk of contamination to domestic seafood and exerting an impact on both the production and consumption of marine products. Accordingly, the European Food Safety Authority (EFSA) and the World Health Organization (WHO) have established standards for the permissible levels of OA group toxins in marine products for safety management. Additionally, in line with international initiatives, the domestic inclusion and regulation of DTX2 among the substances falling under the purview of management outlined by the 2022 diarrheal shellfish toxin standard have been implemented. In this study, we reviewed the physicochemical properties of OA group toxins, their various exposure routes (such as acute toxicity, genotoxicity, reproductive and developmental toxicity), and the relative toxicity factors associated with these toxins. We also performed a comparative assessment of the methods employed for toxin analysis across different countries. Furthermore, we aimed to conduct a broad review of human exposure cases and assess the international guideline for risk management of OA group toxins.
