고마심신환거주사방(古魔心賢丸去朱砂方)이 우울증(憂鬱症) 모형(模型) 흰쥐의 우울(憂鬱) 성향 및 PVN 의 c-Fos 발현(發顯)에 미치는 효과(效果) (The Effects of Koamsimsinwhan(without Cinnabarite) on Depression in Chronic Mild Stress(CMS)-treated Rats)

  • This study was performed to evaluate the effects of Koamsimsinwhan (without Cinnabarite) on depression in CMS-treated rats. Rats of trial groups were exposured to chronic mild unpredictable stress for 4 weeks. These CMS-treated rats were stratified into Koamsimsinwhan(without Cinnabarite) group and vehicle(non-drug) group. And there were control rats excluding both CMS and Koamsimsinwhan(without Cinnabarite). Forced swimming test, general activity test and immunohistochemical test(measuring the level of manifestation of c-Fos in PVN) were executed to evaluate the effects of Koamsimsinwhan(without Cinnabarite) on depression after 4 weeks of CMS treatment. The results were as follows 1. In FST, depression was induced from CMS treatment and was supressed with Koamsimsinwhan(without Cinnabarite). 2. The increased activity of nerve cell in PVN, which refers to stress, was induced from CMS treatment and normalized with Koamsimsinwhan (without Cinnabarite).

반추위 미생물의 질소공급원으로서 Condensed Molasses Solubles (CMS)의 사료 가치 평가 (An Evaluation of Condensed Molasses Solubles (CMS) as a Source of Nitrogen for Ruminal Microbes In Vitro)

  • 본 연구는 monosodium glutamate의 부산물인 condensed molasses solubles (CMS)가 반추가축의 질소 공급원으로서 반추위 미생물 단백질 합성에 미치는 영향을 in vitro 실험을 통하여 조사하였다. 실험 1에서는 반추위 미생물에 대한 질소 공급원으로서 CMS의 가치를 요소와 비교하였고, 실험 2는 미생물의 생성양이 최대가 되는 시간대를 조사하기 위하여 수행하였고, 실험 3에서는 실험 1과 2에서 배양 후 실시된 원심분리 (1000g×5분)에 의해 생성된 침전물이 총 질소의 공급양과 측정된 질소양에 차이를 발생시킴에 따라 침전물의 성분을 조사하고자 수행하였고, 실험 4는 배양 2시간대에서 CMS와 요소로부터 발생되는 암모니아의 발생양을 조사하기 위하여 수행하였다. 이상의 결과를 요약하면 다음과 같다. 1. 반추위 미생물태 질소 합성량은 CMS/당밀 처리구 (7.79mg/100ml)와 요소/당밀 (7.61mg/100ml)의 처리구간에 유의적인 차이는 없었지만 CMS/당밀의 처리구가 암모니아태 질소의 함량을 유의적으로 감소시켰다. 2. CMS/당밀 처리구의 암모니아태 질소 함량과 pH는 당밀/요소 처리구보다 유의적으로 감소하였다. 하지만 미생물태 질소의 양은 처리구간에 차이가 없었다. 최대 미생물의 성장률은 배양 후 10시간에서 발생하였다. 3. 당밀 대신 100% 수용성인 설탕을 이용하였을 경우에도 침전물이 발생하여 침전물이 당밀에 의한 것이 아님이 확인되었고, 또한 배양 시간이 짧았을 경우 (2시간) 질소의 손실양도 상대적으로 크게 감소하여, 총 질소의 공급양과 측정된 질소양의 차이는 침전물에 박테리아가 포함되었기 때문인 것으로 조사되었다. 4. 배양 2시간 동안 CMS로부터 발생되는 암모니아의 생성속도는 요소로부터 발생되는 속도보다 유의적으로 빠르게 나타났다. 본 실험의 결과 CMS는 반추위 미생물 단백질을 합성하는 데 있어 최소한 요소의 가치를 지니고 있음이 조사되었다.

임진강변 퇴적층 내 탄소물질들의 분포 및 특성 연구 (A Study on the Distribution and Property of Carbonaceous Materials in the Subsurface Sediments near the Imjin River)

  • The fate of hydrophobic organic contaminants (HOCs) in ground water is highly affected by the distribution and property of the carbonaceous materials (CMs) in subsurface sediments. CMs in soils consist of organic matters (e.g., cellulose, fulvic acid, humic acid, humin, etc.) and black carbon such as char, soot, etc. The distribution and property of CMs are governed by source materials and geological evolution (e.g., diagenesis, catagenesis, etc.) of them. In this study, the distribution and property of CMs in subsurface sediments near the Imjin river in the Republic of Korea and HOC sorption property to the subsurface sediments were investigated. The organic carbon contents of sand and clay/silt layers were about 0.35% and 1.37%, respectively. The carbon contents of condensed form of CMs were about 0.13% and 0.45%, respectively. The existence of black carbon was observed using scanning electron microscopes with energy dispersive spectroscopy. The specific surface areas (SSA) of CMs in heavy fraction(HFrCM) measured with N2 were $35-46m^2/g$. However, SSAs of those HFrCM mineral fraction was only $1.6-4.3m^2/g$. The results of thermogravimetric analysis show that the mass loss of HFrCM was significant at $50-200^{\circ}C$ and $350-600^{\circ}C$ due to the degradation of soft form and condensed form of CMs, respectively. The trichloroethylene (TCE) sorption capacities of sand and clay/silt layers were similar to each other, and these values were also similar to oxidzed layer of glacially deposited subsurface sediments of the Chanute Air Force Base (AFB) in Rantoul, Illinois. However, these were 7-8 times lower than TCE sorption capacity of reduced layer of the Chanute AFB sediments. For accurate prediction of the fate of hydrophobic organic contaminants in subsurface sediments, continuous studies on the development of characterization methods for CMs are required.

Condensed Molasses Solubles(CMS) / 당밀 혼합물이 반추위 미생물 단백질 합성량에 미치는 영향 (The Effects of Condensed Molasses Solubles(CMS) / Molasses Mixtures on Ruminal Microbial Protein Synthesis)

  • 본 연구는 monosodium glutamate의 생산 후 발생되는 부산물인 condensed molasses solubles(CMS)가 반추가축의 질소 공급원으로서 반추위 미생물 단백질 합성에 미치는 효과를 조사하기 위하여 수행하였다. 반추위 canulae가 부착된 4마리의 비착유소를 4 ${\times}$ 4 라틴 방각법에 적용하여 실험을 수행하였으며, 4개의 처리구는 다음과 같다. (1) 기초사료(앞착된 보리 3kg/일과 보리짚의 자유채식), (2) 기초사료에 당밀 200g/일 그리고 물 300g/일 첨가, (3) 기초사료에 당밀 200g/일, CMS 100g/일 그리고 물 200g/일 첨가 (4) 기초 사료에 당밀 200g/일, CMS 200g/일, 그리고 물 100g/일 첨가. CMS의 첨가수준에 따른 반추위내 발효양상은 처리구간에 유의적인 차이가 없었으나, CMS를 200g/일을 첨가한 처리구가 다른 처리구들에 비해 반추위내 미생물 단백질 합성량의 표시로서 이용된 allantoin/creatinine의 비율을 증가시키는 경향이 나타났다(P < 0.10).

우울증(憂鬱症) 모델 흰쥐에 대한 조위승청탕(調胃升淸湯)과 St. John's Wort의 항우울효과(抗憂鬱效果)에 관한 비교 연구 (The effects of Jowiseungcheongtang versus St. John's wort in the chronic mild stress model of depression in rats)

  • The aim of this study was to evaluate the effects of Jowiseungcheongtang compared with St. John's wort in the chronic mild stress(CMS) animal model of depression. Wistar rats were used for this study. The subjects were divided into 4 groups (Naive group: without CMS procedure, CMS-vehicle: water was administered during CMS procedure, CMS-Jowiseungcheongtang: Jowiseungcheongtang was administered after 5 weeks of CMS procedure, CMS-St. John's wort: St. John's wort was administered after 5 weeks of CMS procedure) During 9 weeks of CMS procedure, The change of the consumption of sucrose and the changes of weights were measured. After CMS procedure, Morris water maze test, open field test, elevated plus maze test and Western blotting were measured. The results were as follows. 1. The consumption of sucrose solution was significantly reversed in Jowiseungcheongtang group and relatively reversed in St. John's Wort group at 7 week. 2. In open field test, Neither Jowiseungcheongtang nor St. John's wort group showed statistically significant change of exploratory activity. In EPM test, St. John's Wort group showed significant decrease of total arm entry in comparison with Naive group. And Jowiseungcheongtang group was showed no significant change. 3. In Morris water maze test, St. John's Wort group showed significant increase in escape latency of the last Morris water maze trial. And in water maze probe test, there was no significant change. 4. St. John's Wort group showed relative increase in LP1 division of 5HT1A receptor compared with Naive group. Both St. John's Wort and Jowiseungcheongtang group showed relative increase in P2 division of GluRl compared with Naive group. These results suggest that Jowiseungcheongtang is as effective as St. John's Wort in the treatment of depression.

This study consists of (1) defining concept of CMS (2) sellecting ${\ulcorner}Structuring\;method{\lrcorner}$ for designing CMS (3) designing ${\ulcorner}Conceptual\;Framework{\lrcorner}$ for CMS. A conceptual framework of the CMS was designed by system design method based on ${\ulcorner}Normative\;approach{\lrcorner}$, and ${\ulcorner}Explorative\;approach{\lrcorner}$; that is ${\ulcorner}Ddeductive\;method{\lrcorner}$ and this is ${\ulcorner}Inductive\;method{\lrcorner}$.

Effect of condensed molasses fermentation solubles (CMS) to replace molasses on the growth performance, nutrient digestibility, and backfat thickness in growing pigs

  • The purpose of this study was to investigate the effects of condensed molasses fermentation solubles (CMS) on the growth performance, nutrient digestibility, and backfat thickness of growing pigs. In an eight-week trial, a total of 120 cross-bred ([Landrace × Yorkshire]× Duroc) growing pigs with an average initial body weight (BW) of 25.84 ± 1.60 kg were used. The pigs were randomly assigned to one of two dietary treatments based on body weight and sex. The dietary treatments were as follows: CON - corn-soybean meal-based basal diet (CON) + 2% molasses; and TRT1 - CON + 2% CMS. Each treatment had 12 replicates with five pigs (three gilts and two castrated barrows) per pen. Though there were no significant differences observed in terms of the growth performance and nutrient digestibility (p < 0.05), the backfat thickness in pigs fed 2% CMS tended to increase in week 4 (p < 0.1). Thus, 2% CMS could be optimal to replace molasses given that it does not exert any adverse effects on the performance of growing pigs.

당밀농축용액이 토양의 화학 및 생물학적 성질과 질소의 무기화에 미치는 영향 (Effects of Condensed Molasses Soluble on Chemical and Biological Properties of Soil, and Nitrogen Mineralization)

  • Lysine을 추출한 후 발생되는 농축당밀용액 (CMS)을 포장용수량의 60-70%인 수분불포화토양과 시용량을 달리하여 혼화처리한 후 $25^{\circ}C$의 항온조건에서 7주 동안 항온배양하면서 토양의 이화학성, 질소의 무기화 및 미생물상의 변화를 조사하였다. CMS를 경토중량의 0.3%에서 1.5%까지 혼화처리했을 때 T-N 함량은 7주 동안 일정한 양을 유지하였으며, CMS 시용량과 비례하여 증가하였다. $NH_4-N$는 시간경과에 따라 전체적으로 감소하였으며, 시용량이 적을수록 감소 폭은 커졌다. 반면, $NO_3-N$ 함량은 시간이 경과함에 따라 지속적으로 증가하여 수분불포화 조건에서 CMS를 처리하면 초기부터 질산화가 촉진되었으며 시용량이 적은 0.3%와 0.6% 처리구에서 $NO_3-N$의 증가폭이 컸다. CMS를 처리한 토양의 pH는 낮아지고 염농도 및 유기물 함량은 증가되었으나 유효인산의 함량에는 영향을 미치지 않았다. CMS 처리로 토양 미생물의 밀도는 무처리에 비하여 현저하게 증가하였으며, 특히 방선균은 배양 7주 동안 지속적인 밀도 증가를 나타내었고, 질소순환과 관계되는 미생물상의 밀도 변화도 무처리에 비하여 증가하는 경향이었다.

Impact of Hospital Specialization on Hospital Charge, Length of Stay and Mortality for Lumbar Spine Disease Inpatients

  • Background: This study investigates association modified category medical specialization (CMS) and hospital charge, length of stay (LOS), and mortality among lumbar spine disease inpatients. Methods: This study used National Health Insurance Service-cohort sample database from 2002 to 2013, using stratified representative sampling released by the National Health Insurance Service. A total of 56,622 samples were analyzed. The primary analysis was based on generalized estimating equation model accounting for correlation among individuals within each hospital. Results: Inpatients admitted with lumbar spine disease at hospitals with higher modified CMS had a shorter LOS (estimate, -1.700; 95% confidence interval [CI], -1.886 to -1.514; p<0.0001). Inpatients admitted with lumbar spine disease at hospitals with higher modified CMS had a lower mortality rate (odds ratio, 0.635; 95% CI, 0.521 to 0.775; p<0.0001). Inpatients admitted with lumbar spine disease at hospitals with higher modified CMS had higher hospital cost per case (estimate, 192,658 Korean won; 95% CI, 125,701 to 259,614; p<0.0001). However, inpatients admitted with lumbar spine surgery patients at hospitals with higher modified CMS had lower hospital cost per case (estimate, -152,060 Korean won; 95% CI, -287,236 to -16,884; p=0.028). Inpatients admitted with lumbar spine disease at hospitals with higher modified CMS had higher hospital cost per diem (estimate, 55,694 Korean won; 95% CI, 46,205 to 65,183; p<0.0001). Conclusion: Our results showed that increase in hospital specialization had a substantial effect on decrease in hospital cost per case, LOS, and mortality, and on increase in hospital cost per diem among lumbar spine disease surgery patients.

From Bench to Market: Preparing Human Pluripotent Stem Cells Derived Cardiomyocytes for Various Applications

  • Human cardiomyocytes (CMs) cease to proliferate and remain terminally differentiated thereafter, when humans reach the mid-20s. Thus, any damages sustained by myocardium tissue are irreversible, and they require medical interventions to regain functionality. To date, new surgical procedures and drugs have been developed, albeit with limited success, to treat various heart diseases including myocardial infarction. Hence, there is a pressing need to develop more effective treatment methods to address the increasing mortality rate of the heart diseases. Functional CMs are not only an important in vitro cellular tool to model various types of heart diseases for drug development, but they are also a promising therapeutic agent for cell therapy. However, the limited proliferative capacity entails difficulties in acquiring functional CMs in the scale that is required for pathological studies and cell therapy development. Stem cells, human pluripotent stem cells (hPSCs) in particular, have been considered as an unlimited cellular source for providing functional CMs for various applications. Notable progress has already been made: the first clinical trials of hPSCs derived CMs (hPSC-CMs) for treating myocardial infarction was approved in 2015, and their potential use in disease modeling and drug discovery is being fully explored. This concise review gives an account of current development of differentiation, purification and maturation techniques for hPSC-CMs, and their application in cell therapy development and pharmaceutical industries will be discussed with the latest experimental evidence.
