Effect of pH on the binding of hGM-CSF to ion exchange resin

  • 본 연구에서는 형질전환된 식물세포배양으로 생산한 (hGM-CSF)를 cationic exchange resin인 CM-sepharose와 anionic exchange resin인 DEAE-sepharose에 통과키며 pH의 변화를 주었을 때 흡착 여부를 확인하였다. hGM-CSF는 pH가 5-7사이의 범위에서 가장 안정함을 보였으며 이 결과를 바탕으로 각각의 pH 범위를 결정하였다. Buffer exchange를 했을 때 cationic exchange resin인 CM-sepharose의 경우 pH 4.8에서 상대적으로 가장 높은 흡착율(77%)을 보였으며 anionic exchange resin인 DEAE-sepharose를 이용한 흡착에서는 pH 5.5에서 높은 흡착율(74%)을 보였다. 이러한 결과를 바탕으로 buffer exchange 없이 hGM-CSF가 secretion된 배지를 pH를 맞춘 후 좁은 범위에서의 흡착 실험을 수행하였다. 그 결과 pH 4.6에서 CM-sepharose를 이용했을 때 흡착율이 84%로 가장 좋았다. 이러한 결과를 이용하면 외래 단백질을 생산하는 식물세포 배양시에 가장 문제가 되는 protease에 의한 목적 단백질의 degradation을 해결할 수 있는 in situ adsorption이 가능하리라 사료된다.

횡격흉막에서 발생한 고립성 섬유성 종양 (Solitary Fibrous Tumor of the Diaphragmatic Pleura)

  • 우리는 단순 흉부 방사선 촬영에서 우연히 발견한 47세 여자의 횡격흉막에서 발생한 양성 고립성 섬유성 종양 1례를 보고한다. 단순 흉부 방사선 촬영, 복부 초음파 검사, 전산화 단층 촬영과 자기공명영상으로 주변과 경계가 분명한 다엽성의 흉막 섬유성 종양으로 진단하였다. 우측 개흉 수술시 종괴는 대부분 유착이 없었으나 $5{\times}4$cm 정도의 횡격막 중앙부와 유경으로 붙어 있었다. 붙어 있는 횡격막과 함께 종괴를 완전히 절제하였다. 종양은 $23.5{\times}13.5{\times}8.0$cm 크기였으며, 병리조직학 진단은 양성 섬유성 종양이었고 함께 절제된 횡격막에는 종양세포가 없었다.

백호에서의 전이성 유선암종 (Metastatic Mammary Gland Adenocarcinoma in a Tiger (Panthera tigris altica))

  • A 17-year-old female tiger (Panthera tigris altica) was found dead after suffering from continuously growing mass at the right mammary gland area. At necropsy, a firm tan mass approximately 25 cm in diameter was noted at the ventral abdomen. The mass was not fixed to the adjacent tissue and mottled tan to red on cut sections. Chains of similar nodules ranging from 2 to 5 cm in diameter were also present along the right mammary glands region. Histologically, the neoplastic masses consisted of lobules that were filled with pleomorphic neoplastic cells and separated by fibrious conntective tissue. The neoplastic cells have hyperchromatic nuclei with prominent nucleolus and moderate amount of cytoplasm. The degree of mitosis was high. Multiple areas of necrosis, hemorrhage, mineralization and tumor emboli were also noted. Metastasis to the regional lymph nodes, lung, liver, spleen, kidney, and adrenal gland were observed. Based on the gross and histopathologic examinations, a diagnosis of lobular type metastatic mammary gland carcinoma was made.

구개부에 발생한 다형성선종의 트랩도어 수술법에 대한 증례보고 (Trapdoor Approach for Removal of the Pleomorphic Adenoma in the Palate: Case Report)

  • A 27-year-old female presented to our hospital with a slow growing, hard and soft palate mass on the right that had been present for the several months. Physical examination showed a 2 cm, firm, well-circumscribed, painless mass on the right side of the palate. PNS computer tomographic imaging showed a $1.5{\times}1.3{\times}2$ cm well-defined cystic mass on the right side of both the hard and soft palate without any underlying bone change. The lesion was completely excised under general anesthesia. In order to preserve the palatal mucosa, trapdoor approach for removal of the pleomorphic adenoma was done. This technique provided more comfortable healing of the operative site. Three years after surgery, there was no evidence of recurrence. If pleomorphic adenoma without bony and mucosal destruction exists, we suggest consideration of the trapdoor approach to protect the palatal mucosa. In view of the potential for tumour recurrence, long-term follow-up and careful examination are necessary.

정중선 이소성 갑상선에서 주사상의 변화 추적 - 증례보고 - (A Changing Pattern of Isotope Scan of the Midline Ectopic Thyroid - A Case Report -)

  • A 17 month-old girl presented in the pediatric clinic on November 27th, 1990 with a neck mass. The mass was 2 cm in diameter, firm in consistency and movable on the upper pole of the thyroid cartilage in the midline. The technetium thyroid scan showed a hot reactivity at the compatible site of the mass, but no other radioactivity in either site of the normal thyroid positions. At her second visit on January 23th, 1996, the mass had enlarged up to 3.5cm in diameter in the same location of the neck. The follow up thyroid scan revealed a walnut sized, snowman-like radioactivity. One of the snowman-like double images seemed to be a lingual ectopic thyroid and the other a midline ectopic thyroid remnant in the infrahyoid level. This interpretation was supported by the computed tomography of the neck, which showed a ligual mass in the foramen cecum area and an another mass in the anterior comis-sure of the larynx in the mildline. Thyroid function test was normal except a slightly increased TSH. As a result of this changing pattern of thyroid radioactive images, a case of a lingual thyroid as well as another midline ectopic thyroid tissue at the infrahyoid level is reported.

A primo vessel-like structure in a dog with inflammatory pseudotumor

  • Inflammatory pseudotumor (IPT) is a term defining a mass characterized microscopically by a proliferation of bland mesenchymal spindle cells infiltrated by diffuse mixed inflammatory cells with a predominance of plasma cells and lymphocytes. Here, we show the primo vessel-like structure of the primo-vascular system (PVS) in a dog with IPT. A 6-years old male Mongrel dog was diagnosed with an abnormal mass (diameter 5.5 cm, weight 22 g) near left preputial area. The dog was submitted to the surgical detectomy of the mass. During the surgical operation, we observed primo vessel-like material. After fixations, the masses appeared macroscopically as lipoid-like, firm, white to grey masses, measuring $5{\times}8cm$. Histologically, cellular infiltration into the muscular layers was frequently seen. The mesenchymal proliferation remained the main component of the mass and was composed of myofibroblastic-like spindle cells characterized by globular, irregular nuclei containing open chromatin and a prominent nucleolus. On the basis of the histopathologic lesions, the subcutaneous mass was diagnosed as IPT. Also, we detected a primo vessel-like structures in some areas of the IPT tissues. These were observed as novel thread-like structures and bundle of tubular structures. To our knowledge, this report is the first case of primo vessel-like structure in a dog with IPT.

개의 발가락에 발생한 전이성 악성흑색종 (Metastatic malignant melanoma in digit of the dog)

  • A 8-year-old, intact male Yorkshire Terrier Dog was presented with dark-black mass on the third digits of the left forelimb. Three months earlier, the dog had experienced an episode of digit amputation because of growing mass with ulcerated nodule of the same area. According to the owner, the recurrence of the mass appeared suddenly and was growing rapidly from the amputation site. The mass was more infiltrative than the first one and measured 1.5 cm in diameter. The clinical signs were anorexia, coughing, respiratory distress, exercise intolerance, cardiac murmurs, and cyanosis on the oral mucous membrane. Plain radiographic findings revealed multiple, various-sized(0.5 to 7 cm in diameter), slightly firm-nodules on the thoracic region but digital bone lysis was not seen. These lesions on the thoracic cavity were considered likely to be metastatic from the digit and the dog was naturally died after 3 weeks from the time. Histologically, the digital mass confirmed the diagnosis of metastatic malignant melanoma that was composed of round melanocytic neoplastic, anaplastic, and melanin-containing cells. This report records clinical information and gross and light microscopic features of metastatic malignant melanoma in a dog.

도축돈에서 발생한 피부 멜라닌 세포성 종양 (Cutaneous melanocytic tumor in a slaughtered pig)

  • A 7-month-old brown pig with a discoid and pedunculate mass measuring $14.0{\times}12.5{\times}2.5cm$ on the skin of the right shoulder was noted at the slaughter house in Jeju. The surface of mass approximately $7{\times}4cm$ was interfaced with skin. The color of mass with firm consistency was mainly black and partially white on cut surface. Histopathologically, numerous unencapsulated endocrine-like cellular nodules of epithelioid cell type with abundant intracytoplasmic black pigment, melanin, were occupied in dermis and subcutis. Most of nodules in deep dermis were surrounded by lightly pigmented spindle cells and loose fibrous tissues. Mitotic figures were infrequently observed. The overlying epidermis was hyperplastic due to the down-growth of rete peg. Based on the gross and histopathologic findings, this case was diagnosed as cutaneous melanoma. In our best knowledge, this is the first case of swine cutaneous melanoma in Korea.

Modified temporalis tendon transfer extended with periosteum for facial paralysis patients

  • Background: We have devised a novel surgical method, termed as temporalis muscle tendonperiosteum (T-P) compound surgical method, by modifying pre-existing techniques. Our method is characterized by elevation of temporalis muscle tendon and the periosteum of the mandibular ramus as a single compound. Here, we describe the concept and clinical outcomes of our method. Methods: We conducted both a cadaveric study and a clinical study. First, we used four human cadavers (two males and two females) to confirm the anatomy of the temporalis muscle tendon and availability of sufficient length extension through the elevation of the T-P compound. Moreover, we obtained measurements of the mouth angle and the philtrum angle from a total of six patients (two males and four females) and compared them between preoperatively and postoperatively. Results: The mean length of the periosteal portion was measured as 2.43± 0.15 cm (range, 2.2-2.6 cm). There was an improvement in the mouth angle postoperatively as compared with preoperatively (7.2°± 3.0° vs. 14.5°± 4.7°, respectively). Moreover, there was also an improvement in the philtrum angle postoperatively as compared with preoperatively (7.2°± 3.4° vs. 17.2°± 6.5°, respectively). Conclusion: Our method is a simple, minimally-invasive modality that is effective in achieving good clinical outcomes. Its advantages include an ability to achieve a firm extension of the temporalis muscle tendon as well as a lack of requirement for a donor site that may cause complications.

원발성 개 늑연골육종 1예 (Primary Rib Chondrosarcoma in a Dog)

  • Primary rib tumors occur in dogs at a much lower frequency than appendicular bone neoplasms. A 3-year-old 3.5kg female maltese was referred to the Veterinary Medical Teaching Hospital because of a 2-month history of severe coughing and a 1-month history of dyspnea, gaging, and anorexia. The plain thoracic radiography revealed a huge well-defined mass in the left thoracic cavity, and a diagnosis of primary rib chondrosarcoma was established by histopathologic evaluation of the surgical biopsy specimen. At operation, a 8-x12-x4.5-cm relatively large firm mass was found filling the lumen of the thoracic, which was diffusely attached to the left 2-9th rib. This mass oppressed the heart, left lung and diaphragm.
