Bacterial and fungal community composition across the soil depth profiles in a fallow field

  • Background: Soil microorganisms play key roles in nutrient cycling and are distributed throughout the soil profile. Currently, there is little information about the characteristics of the microbial communities along the soil depth because most studies focus on microorganisms inhabiting the soil surface. To better understand the functions and composition of microbial communities and the biogeochemical factors that shape them at different soil depths, we analyzed microbial activities and bacterial and fungal community composition in soils up to a 120 cm depth at a fallow field located in central Korea. To examine the vertical difference of microbial activities and community composition, ${\beta}$-1,4-glucosidase, cellobiohydrolase, ${\beta}$-1,4-xylosidase, ${\beta}$-1,4-N-acetylglucosaminidase, and acid phosphatase activities were analyzed and barcoded pyrosequencing of 16S rRNA genes (bacteria) and internal transcribed spacer region (fungi) was conducted. Results: The activity of all the soil enzymes analyzed, along with soil C concentration, declined with soil depth. For example, acid phosphatase activity was $125.9({\pm}5.7({\pm}1SE))$, $30.9({\pm}0.9)$, $15.7({\pm}0.6)$, $6.7({\pm}0.9)$, and $3.3({\pm}0.3)nmol\;g^{-1}\;h^{-1}$ at 0-15, 15-30, 30-60, 60-90, and 90-120 cm soil depths, respectively. Among the bacterial groups, the abundance of Proteobacteria (38.5, 23.2, 23.3, 26.1, and 17.5% at 0-15, 15-30, 30-60, 60-90, and 90-120 cm soil depths, respectively) and Firmicutes (12.8, 11.3, 8.6, 4.3, and 0.4% at 0-15, 15-30, 30-60, 60-90, and 90-120 cm soil depths, respectively) decreased with soil depth. On the other hand, the abundance of Ascomycota (51.2, 48.6, 65.7, 46.1, and 45.7% at 15, 30, 60, 90, and 120 cm depths, respectively), a dominant fungal group at this site, showed no clear trend along the soil profile. Conclusions: Our results show that soil C availability can determine soil enzyme activity at different soil depths and that bacterial communities have a clear trend along the soil depth at this study site. These metagenomics studies, along with other studies on microbial functions, are expected to enhance our understanding on the complexity of soil microbial communities and their relationship with biogeochemical factors.

Geological Structure of the Moisan Epithermal Au-Ag Mineralized Zone, Haenam and its Tectonic Environment at the Time of the Mineralization (해남 모이산 천열수 금-은 광호대의 지질구조와 광화작용 당시의 지구조환경)

  • An Epithemal Au-Ag mineralized zone is developed in the Moisan area of Hwangsan-myeon, Haenam-gun, Jeol-lanam-do, Korea, which is located in the southwestern part of the Ogcheon metamorphic zone. It is hosted in the Hwangsan volcaniclastics of the Haenam Formation of the Late Cretaceous Yucheon Group. This research investigated the characteristics of bedding arrangement, fold, fault, fracture system, quartz vein and the time-relationship of the fracture system to understand the geological structure related to the formation of the mineralized zone. On the basis of this result, the tectonic environment at the time of the mineralization was considered. Beds mainly trend east-northeast and gently dip into north-northwest or south-southeast. Their poles have been rearranged by subhorizontal-upright open fold of (east)-northeast trend as well as dip-slip fault. Fracture system was formed through at least 6~7 different deformation events. D1 event; formation phase of the main fracture set of EW (D1-1) and NS (D1-2) trends with a good extensity, D2 event; that of the extension fracture of NW trend, and conjugate shear fracturing of the EW (dextral) and NS (sinistral) trends, D3 event; that of the extension fracture of NE trend, and conjugate shear refracturing of the EW (sinistral) and NS (dextral) trends, D4 event; that of the extension fracture of NS trend showing a poor extensity, D5 event; that of the extension fracture of NW trend, and conjugate shear refracturing of the EW (dextral) and NS (sinistral) trends, D6 event; that of the extension fracture of EW trend showing a poor extensity. Frequency distribution of fracture sets of each deformation event is D1-1 (19.73 %)> D1-2 (16.44 %)> D3=D5 (14.79 %)> D2 (13.70 %)> D4 (12.33 %)> D6 (8.22 %) in descending order. The average number of fracture sets within 1 meter at each deformation event is D6 (5.00)> D5 = D4 (4.67)> D2 (4.60)> D3 (4.13)> D1-1 (3.33)> D1-2 (2.83) in descending order. The average density of all fractures shows 4.20 fractures/1 m, that is, the average spacing of all fractures is more than 23.8 cm. The frequency distribution of quartz veins at each orientation is as follows: EW (52 %)> NW (28 %)> NS (12 %)> NE (8 %) trends in descending order. The average density of all quartz veins shows 4.14 veins/1 m, that is, the average spacing of all quartz veins is more than 24.2 cm. Microstructural data on the quartz veins indicate that the epithermal Au-Ag mineralization (ca. 77.9~73.1 Ma) in the Moisan area seems to occur mainly along the existing D1 fracture sets of EW and NS trends with a good extensity not under tectonic stress but non-deformational environment directly after epithermal rupture fracturing. The D1 fracturing is considered to occur under the unstable tectonic environment which alternates compression and tension of NS trend due to the oblique northward subduction of the Izanagi plate resulting in the igneous activity and deformation of the Yucheon Group and the Bulguksa igneous rocks during Late Cretaceous time.

Growth of Maize and Sorghum-Sudangrass Hybrid Affected by Soil Texture and Ground Water Levels (토성 및 지하수위에 따른 사료용 옥수수와 수수-수단그라스 잡종의 생육)

  • This study was conducted to know optimum ground water level for maize(Suwon 19) and sorghum-sudangrass hybrid(Pioneer855F) and investigated effect of different soil types and ground water levels on growth of above two crops at containers were controlled so as to be 25, 35, 50, 70 and 100cm from the soil surface. Both crops showed better growth in sandy loam soil. Optimum ground water level for maize was l00cm and for sorghum-sudangrass hybrid was 70cm in sandy loam soil. Clay loam was unfavorable for both crops even though low ground water level maintained. Sorghum-sudangrass hybrid was adapted better to clay loam soil and high ground water levels. In microscopic study on root anatomy of both crops, destructed cortical tissues were observed. Frequency of the tissues was higher in maize than in sorghum-sudangrass hybrid and higher at mid or tip parts than basal parts of roots in both crops. Arrangement of cortical cells of roots in sorghum-sudangrass hybrid were more regular than those of maize. In root anatomy, no consistent trend with soil types or ground water levels was detcted.

The faunal composition of meiobenthic community in the deep-sea sediment, northeastern Pacific Ocean (북동 태평양 심해저퇴적물에 서식하는 중형저서생물의 군집조성)

  • This study was carried out to investigate the faunal composition and distribution pattern of meiobenthic community in the deep sea sediments of northeastern Pacific Ocean. Faunal samples were collected at 10 stations using a multiple corer and a spade type box corer during May 1996. A total of 18 meiofaunal groups were found, and Sarcomastigophora was the most dominant group which accounted for 35.8% of total meiofaunal abundance. Nematoda (30.8%), Nauplius larvae of Crustacea (11.9%), Ciliophora (6.8%) and Harpacticoida (3.0%) which were also important components of the meiofaunal community. All of these five faunal groups comprised more than 90% of total meiofauna. The highest density of 195 ind./10 $cm^2$ was found at St. 4 (4960 m water depth), but the lowest one, 85 ind./10 $cm^2$ at St. 1 (4969 m water depth). The overall mean density of meiofauna was estimated as $1.34{\times}10^5\;ind./m^2$. The abundance of meiofauna showed a slow decreasing trend along the water depth. The highest density was observed in the upper 1 cm and density decreased drastically with increasing depth from the surface.

Characteristics of Field Uplift Tests of Continuous Greenhouse using the Load Control Method (하중 제어법을 이용한 파이프 줄기초의 현장 인발저항 특성)

  • Institutional inertia anti-disaster standard was presented mainly on the upper surface, it is necessary to improve to the soil type standard and uplift the resistance standard greenhouse that are vulnerable to strong winds. In this study, we carried out a field test using the load control method in order to evaluate the uplift resistance of continuous foundation of greenhouse with different depths of the rafters. Institutional inertia anti-disaster standard of greenhouse foundation did not protect the greenhouse structure from the damages caused by strong winds and heavy snow. Therefore, field tests for behavior characteristics of continuous greenhouse foundation were carried out to ensure stable facility cultivation. The field test condition was evaluated using different embedded depth as follows: 30cm, 40cm, 50cm and spacing 50cm, 60cm, 70 cm. As a result of the uplift resistance field tests using the load control method, the minimum uplift resistance was found to be over 90kg and uplift resistance displacement was 9.4mm. Uplift resistance of the continuous greenhouse foundation was in the range of 90-180 kg according to embedded depth and spacing. Using the test condition, there was no constant trend in the uplift resistance.

Satisfaction Level and Performance Evaluation for CM Service in Korea (국내 건설사업관리 업무만족도 및 성과평가)

  • The domestic CM business market has continued to grow, but its size is relatively small yet in comparison with the total size of the domestic construction industry. Evaluation of CM projects was conducted through questionnaire surveys. Both clients and CM firms showed positive satisfaction levels. Nevertheless, the clients' satisfaction levels were relatively low in safety management, cost management, and document and information management. Superior areas of CM tasks were time management and recovery scheduling, quality control and technical instruction, and design value engineering. On the other hand, inferior areas of CM tasks were claim analysis and dispute resolution, cost estimation, and life cycle costing. Both entities have agreed with the positive effects of CM involvement in terms of cost saving, time reduction, quality improvement, and safety incidents prevention to at least 0~5% extent.

Application of Analysis Models on Soil Water Retention Characteristics in Anthropogenic Soil (인위적으로 변경된 토양에서의 수분보유특성 해석 모형의 적용)

  • This study was conducted to assess the propriety of models for soil water characteristics estimation in anthropogenic soil through the measurement of soil water content and soil water matric potential. Soil profile was characterized with four different soil layers. Soil texture was loamy sand for the first soil layer (from soil surface to 30 cm soil depth), sand for the second (30~70 cm soil depth) and the third soil layers (70~120 cm soil depth), and sandy loam for the fourth soil layer (120 cm < soil depth). Soil water retention curve (SWRC), the relation between soil water content and soil water matric potential, took a similar trend between different layers except the layer of below 120 cm soil depth. The estimation of SWRC and air entry value was better in van Genuchten model by analytical method than in Brooks-Corey model with power function. Therefore, it could be concluded that van Genuchten model is more desirable than Brook-Corey model for estimating soil water characteristics of anthropogenic soil accumulated with saprolite.

  • This paper presents conducted emission noise measurement from electronic equipment in frequency range of 1 MHz up to 30 MHz by small loop antenna. Small loop antenna measurement method can measure common-mode (CM) and differential-mode (DM) component of the noise on a pair of power line at the same time. The CM and DM can be measured separately. The theory of this measurement method is introduced and analyzed. The measured results were compared with the conventional measurement by Line Impedance Stabilization Network (LISN) and result a good trend between those methods.

A Study on Ways to Optimize the Period of Hospitalization During High-Dose $^{131}I$ Treatment of the Thyroid Cancer (갑상선암의 고용량 $^{131}I$ 치료 시 입원기간의 최적화방안 연구)

  • This study attempts to propose ways to optimize and improve the period of hospitalization for the radioiodine treatment by calculating the dose rate of patients treated with high-dose radioiodine by distance and time slot. As a result, 24 hours after the patient was treated with 100 mCi high-dose iodine, the dose rate was attenuated to $1,035{\mu}Sv/hr$ at the navel, to $109{\mu}Sv/hr$ at 50 cm, and to $33{\mu}Sv/hr$ at 100 cm. 24 hours after the patient was treated with 150 mCi high-dose iodine, the dose rate was attenuated to $637{\mu}Sv/hr$ at the navel, to $100{\mu}Sv/hr$ at 50 cm, and to $40{\mu}Sv/hr$ at 100 cm. 24 hours after the patient was treated with 180 mCi high-dose iodine, the dose rate was attenuated to $1,251{\mu}Sv/hr$ at the navel, $140{\mu}Sv/hr$ at 50 cm, and to $56{\mu}Sv/hr$ at 100 cm. In light of the current criterion for discharge recommended by the US Nuclear Regulatory Commission being $70.4{\mu}Sv/hr$, the present study indicates earlier discharge is feasible by applying a new scheme. This suggests that the proposed scheme in this study will help to solve the problem of shortage of treatment beds with the increasing trend of patients with thyroid cancer taken into consideration.

Effects of Aspergillus oryzae Inclusion on Corn Silage Fermentation

  • This study is aimed at evaluating the effect of Aspergillus oryzae fermentation extract (AFE) on corn silage fermentation characteristics. Trial included two groups of treatments, with or without AFE inclusion in corn ensilage. Sixty corn silage containers, including two treatments with thirty replicates each, were processed in a laboratory scale mini-silo of 21 cm radius by 45 cm height. Three replicate containers were opened and sampled for analysis at 0, 0.5, 1, 2, 3, 4, 6, 10, 18 and 34 days after being ensiled. One silage container from each treatment was installed with a remote controlled electronic thermometer to record the temperature changes. Analysis included silage temperature, pH, fermentation acids, the water-soluble carbohydrates and chemical compositions and the silage protein fractions. Results showed that on the first day, the temperature of the ensiled corn was slightly higher than room temperature, but returned to room temperature on the second day. The pH and concentrations of WSC, ADF, lignin and acetic acid in the AFE treated silage were significantly lower than the control groups (p<0.05). The lactic acid and crude protein on the other hand were significantly higher in the AFE treated silage as compared to the control (p<0.05) at the end of the ensilage period. The DM content was significantly higher (p<0.05) whereas the butyric acid content of the AFE treated silage was significantly lower (p<0.05) than the control at the end of the 34 day ensilage period. Titratable acid and buffering capacity in the corn silage were not significantly different between treatment groups (p>0.05). Ammonia N concentration in the AFE treated silage showed a trend of decrease (p>0.05). NPN and the protein fraction A in both groups increased during the conservation period, but fraction A in the AFE treated corn silage was significantly higher than the control silage (p<0.05). During the conservation period, the AFE treated corn silage showed a trend toward a decrease in fractions $B_1$, $B_3$ and C (p<0.05). The protein fraction B2 showed a trend toward increase in the control group and an inconsistent trend in the AFE treated silage during the ensiling period. The AFE treated silage showed a better Flieg score over the control silage (97 vs. 75) as calculated from the concentrations of lactic acid, acetic acid and butyric acid.
