  • Conjugated linoleic acid (CLA) was chemically synthesized using the alkaline isomerization method of com oil. CLA-TG was synthesized by reaction with sodium methoxide. Five different treatments were devised based on differences in the amount of CLA-TG added into the pressed han. for controls, 10% of pork back fat among the total component was only added without any CLA-TG. For the first treatment, 5% of CLA-TG among the lard component added into the press ham was replaced. For the 2nd, 3rd and 4th treatments, 10%, 15% and 20% of CLA-TG was respectively replaced. Pressed ham manufactured using CLA-TG was vacuum packaged and then stored for 1, 7, 14, 21 and 28 days at $4^{\circ}C$. Samples were analyzed for chemical composition, physico-chemical properties (pH, color), and texture characteristics. Typical chemical composition characteristics (moisture, crude protein, crude fat) were not significantly different between controls and CLA-TG treatment groups. Crude fat content of CLA-TG treatment groups was significantly lower than that of controls (p<0.05). pH values of controls was higher than that of CLA-TG treatment groups. The pH of control and CLA-TG treatment groups increased significantly as the storage period increased (p<0.05). Meat color (CIE $L^*,\;b^*$) of CLA-TG treatment groups was higher than that of controls. $a^*$ values were decreased by replacement of CLA-TG, but appeared to be unaffected by storage length. There was no significant difference in texture between controls and CLA-TG treatment groups. Based on these findings, we conclude that the physico-chemical properties and texture characteristics of manufactured pressed ham were not affected by CLA-TG addition. Also, our results indicate that high-quality pressed ham can be manufactured with CLA accumulation.

  • CLA was chemically synthesized by alkaline isomerization method using corn oil. CLA-TG was synthesized by chemical reaction using sodium methoxide. For the control, 10% of back fat among the total component was only added without the annex of CLA-TG. For the first treatment, 5% of CLA-TG among the lard component added into the press ham was replaced. For the 2nd, 3rd and 4rd treatments, 10%, 15% and 20% of CLA-TG was respectively replaced. Manufacture press ham using CLA-TG were vacuum packaged and then stored during 1, 7, 14, 21 and 28 days at 4℃. Samples were analyzed for shear force value, sensory evaluation, TBARS, fatty acid composition and CLA content. Shear force value of control was significantly higher than that of CLA-TG treatment groups(P<0.05). All treatments were increased by the passage of storage time. No remarkable differences were found in sensory properties among control and CLA-TG treatment groups. CLA-TG treatment groups showed significantly(P<0.05) lower TBARS value than the control. TBARS value was increased significantly during storage in all treatment. In the change of fatty acid composition, the contents of C14:0~C20:4 were decreased significantly by CLA-TG additive. Whereas the increase level of CLA-TG additive resulted in the significantly higher unsaturated fatty acid and CLA content. Summing up the a forementioned results, press ham manufacturing with CLA-TG additive was not affected in sensory evaluation. Also, it may be assumed that the high quality press ham can be manufactured with the extent of storage period and CLA accumulation.

  • Most fatty acids in food matrices are triglyceride(TG) forms. Conjugated linoleic acid(CLA) produced from linoleic acid by microorganisms or chemicals is a free form. To apply the CLA to food systems, the TG containing CLA(designate CLA-TG) was synthesized by Lipozyme-catalyzed esterification method. An optimum reaction con-dition for the esterification of free CLA(FCLA) to glycerol by Lipozyme was determined as follows; Lipozyme(50mg) effectively catalyzed the esterification of CLA(500mg) to glycerol(1150mg) dissolved in isooctane (3ml) in a shaking incubator (200 rpm, 5$0^{\circ}C$) for 48 hr. Under the reaction condition, the resultant contained 52.4% CLA-TG as well as 31.1% Di-CLA-glycerol(CLA-DG), 7.6% mono-CLA-glycerol(CLA-MG), and 9.0% other CLA(un-reacted FCLA plus CLA dimer). These results suggest that the Lipozyme could be a useful enzyme for the production of CLA-TG to be employed in foods.

  • This study was aimed to evaluate the stability of conjugated linoleic acids (CLAs) by nano-encapsulation against in vitro ruminal biohydrogenation by microbial enzymatic conversion. CLAs (free fatty acid form of CLA [CLA-FFA], nano-encapsulated CLA-FFA, triglyceride form of CLA [CLA-TG], and nano-encapsulated CLA-TG) were used in the in vitro fermentation experiments. When Butyrivibrio fibrisolvens (B. fibrisolvens) was incubated with CLA-FFAs, the concentrations of cis-9, trans-11 CLA and vaccenic acid (VA) slightly was decreased and increased by nano-encapsulation, respectively. When B. fibrisolvens was incubated with CLA-TG, the concentrations of cis-9, trans-11 CLA and VA decreased, but these were increased when B. fibrisolvens was incubated with nano-encapsulated CLA-TG. The nano-encapsulation was more effective against the in vitro biohydrogenation activity of B.fibrisolvens incubated with CLA-FFA than with CLA-TG. In the in vitro ruminal incubation test, the total gas production and concentration of total volatile fatty acids incubated with nano-encapsulated CLA-FFA and CLA-TG were increased significantly after 24 h incubation (p<0.05). Nano-encapsulated CLA-FFA might, thus, improve the ruminal fermentation characteristics without adverse effects on the incubation process. In addition, nano-encapsulated CLA-FFA increased the population of Fibrobacter succinogenes and decreased the population of B. fibrisolvens population. These results indicate that nano-encapsulation could be applied to enhance CLA levels in ruminants by increasing the stability of CLA without causing adverse effects on ruminal fermentation.

  • Conjugated linoleic acid(CLA) is a naturally occuring group of derivatives of linoleic acid found in beef and dairy products. CLA has reported to reduce body fat. This study was designed to observe the effect of CLA supplementation on fat accumulation and degradation in male Sprague Dawley rats. Seventy two rats, weighing 150-180g, were divided into 2 groups according to the types of dietary fat(beef tallow or fish oil) and then each group was divided into 2 groups depending on CLA supplementation, i.e., BT, BT-CLA, FO, FO-CLA. All rats were fed experimental diet containing total fat at 12%(w/w) including CLA at 1% for 30 weeks. At 30 weeks, rats were sacrificed to measure TG, free fatty acid level in plasma, TG, lipogenic enzymes in lever and fat cell size, LPL and HSL activities in epididymal fat fad. Fish oil supplemented with CLA diet showed significant reduction in the food efficiency and weight in 30 weeks-fed rats. CLA supplement did not effect on plasma TG, hepatic TG levels and lipogenic enzymes activities in rats, but, fish oil significantly reduced, The LPL and HSL activities did not affected by CLA supplement and n-3 fatty acid rich fish oil. In conclusion, the results suggest that CLA supplement was not a proper way to reduce the fat accumulation in Sprague Dawley rats. Fish oil supplemented with CLA might better way to reduce the body fatness than fish oil itself. Therefore, It is recommended that further study be performed related to physiological and biochemical effects of CLA supplementation and n-3 fatty acid in rats for the reduction of body fatness.(Korean J Nutrition 34(4) : 367∼374, 2001)

  • The present study explored the short-term effects of dietary conjugated-linoleic acid (CLA) on liver lipid metabolism in starved/refed Otsuka Long Evans Tokushima Fatty (OLETF) rats. Male OLETF rats (12 weeks old) were starved for 24 hours, then refed for 48 hours with either a CLA diet [7.5% CLA and 7.5% Safflower oil (SAF)] or a SAF control diet (15% SAF). The results demonstrated a 30% reduction of hepatic triglyceride (TG) concentration in the CLA group when compared to the control group. Liver cholesterol concentration was also 26% lower in the CLA fed rats. The activity of mitochondrial carnitine palmitoyltransferase, the rate-limiting enzyme of fatty acid oxidation, was moderately elevated by 1.2-fold in the livers of the CLA group when compared to the control. In contrast, phosphatidate phosphohydrolase, the rate-limiting enzyme for TG synthesis, was found to be 20% lower in the livers of the CLA-fed rats. Therefore, dietary CLA evidently lowers liver lipid concentrations through a reduced TG synthesis and enhanced fatty acid oxidation in starved/refed OLETF rats.

  • Variations in conjugated linoleic acid (CLA) concentrations in Holstein dairy cows milk, depending on feeding systems in different seasons was investigated. Milk samples were collected from Holstein dairy cows, which either grazed for whole days (WG), only daylight hours (TG), or were offered a total mixed ration (TMR) and experienced no grazing (NG), from April to December of 2005. In April, November and December, the cows in TG and WG treatments received grass silage and some concentrate, while from May to October, the cows grazed on temperate pasture. The cows in NG treatment received the TMR throughout the season. The major fatty acid obtained in the pastures was linolenic acid. There was no significant difference in the pasture's linolenic acid concentrations from May to September, but there was a significant decrease in October. However, the linolenic acid concentrations obtained in the pasture were always much higher than those obtained from the TMR. Linoleic acid was also the major fatty acid in the TMR, but these concentrations were higher in the TMR than in the pasture. There was no significant difference in milk cis9trans11CLA (c9t11CLA) concentrations between the three feeding systems while the cows were fed on conserved pasture in April, November and December. Although c9t11CLA concentrations were lower in the TMR, it was found that the cows which grazed in fresh pasture experienced significantly higher concentrations of c9t11CLA in their milk than those which received only TMR. It was also found that cows in the WG treatment experienced higher c9t11CLA concentrations than those in the TG treatment. In the WG and TG treatments, c9t11CLA concentrations were highest in June, after which, they gradually decreased (p<0.01) until October. For the NG treatment, there was no significant change in the concentrations of c9t11CLA (p>0.05) with season. Overall, trans11C18:1 and c9t11CLA were greatly influenced by season, with higher variation in the WG treatment than in the TG treatment and no variation in the NG treatment.

  • In mice, supplementation of t10,c12 conjugated linoleic acid (CLA) increases liver mass and hepatic steatosis via increasing uptake of fatty acids released from adipose tissues. However, the effects of t10,c12 CLA on hepatic lipid synthesis and the associated mechanisms are largely unknown. Thus, we tested the hypothesis that gut microbiota-producing t10,c12 CLA would induce de novo lipogenesis and triglyceride (TG) synthesis in HepG2 cells, promoting lipid accumulation. It was found that treatment with t10,c12 CLA ($100{\mu}M$) for 72 h increased neutral lipid accumulation via enhanced incorporation of acetate, palmitate, oleate, and 2-deoxyglucose into TG. Furthermore, treatment with t10,c12 CLA led to increased mRNA expression and protein levels of lipogenic genes including SREBP1, ACC1, FASN, ELOVL6, GPAT1, and DGAT1, presenting potential mechanisms by which CLA may increase lipid deposition. Most strikingly, t10,c12 CLA treatment for 3 h increased phosphorylation of mTOR, S6K, and S6. Taken together, gut microbiota-producing t10,c12 CLA activates hepatic de novo lipogenesis and TG synthesis through activation of the mTOR/SREBP1 pathway, with consequent lipid accumulation in HepG2 cells.

  • Conjugated linoleic acid (CLA) has been recently reported to have an anti-obesity effect in animals and humans. The objective of this study was to investigate effects of diglyceride (DG)-CLA on proliferation and differentiation of 3T3-L1 preadipocytes. Cell proliferation was determined using WST-8 analysis and cell differentiation was determined by glycerol-3-phosphate dehydrogenase (GPDH) activity. Lipid accumulation in differentiating 3T3-L1 cells was determined by Oil red O staining. There were four experimental groups including vehicle control (DMSO), CLA, triglyceride (TG)-CLA, and DG-CLA. Treatments of CLA, TG-CLA, and DG-CLA at the concentrations of $10{\sim}1000{\mu}g/ml$ reduced proliferation of preconfluent 3T3-L1 cells in a dose-dependent manner. Among them CLA was the most effective in the proliferation inhibition of preconfluent 3T3-L1 cells with increasing concentrations. Treatments of CLA and DG-CLA at the concentration of $100{\mu}g/ml$ significantly inhibited differentiation of postconfluent 3T3-L1 cells as measured by GPOH activity (p<0.05). In addition, treatments of CLA, TG-CLA, and DG-CLA effectively inhibited lipid accumulation during differentiation of 3T3-L 1 cells. OG-CLA had the most inhibitory effect on the differentiation and lipid accumulation. These results suggest that the compounds including CLA have a respectable anti-obesity effect and that consumption of DG-CLA as a dietary oil may give a benefit for controlling overweight in humans.
