Effects of Bovine Somatotropin (bST) Administration Combined with Controlled Internal Drug Release (CIDR) on Embryo Quality and Pregnancy of Hanwoo (Korean Native Beef Cattle) during Commercial Embryo Transfer Program

  • Effects of recombinant bovine somatotropin (bST) on plasma hormonal concentration, embryo quality, and pregnancy rate were examined during the superovulation and synchronization treatment in donor and recipient cows. Hanwoos (Korean native beef cattle) were treated with controlled internal drug release (CIDR) combined with bST (CIDR+bST) or without bST (CIDR) as donor cows. The embryos recovered from donors were transferred into Holstein recipient heifers treated with bST (CIDR+bST) or without bST (CIDR) for synchronization. The correlation between IGF-I and P4 showed a positive pattern in the CIDR+bST group (r=0.44, p<0.01), but a negative pattern was shown in the CIDR group (r = -0.59, p<0.02) at day 7 of estrous cycles. Although the number of recovered, transferable, and degenerated embryos was not different, quantities of grade 1 (excellent) embryos in CIDR+bST group were significantly higher than those of the CIDR group (p<0.01). The pregnancy rate was higher in the CIDR+bST recipient group compared to CIDR group (p<0.05), when the embryos were recovered from the donors treated with CIDR. However, the pregnancy was maintained highly in both recipient groups, when the embryos were produced by CIDR+bST treated donors. It can be concluded that bST administration combined with CIDR is an effective method for superovulation and synchronization treatment to stabilize plasma hormonal levels, to obtain excellent quality of embryos, and to get higher pregnancy rate.

  • Artificial insemination (AI) technology has widely applied in domestic animals including cattle for production and preservation of valuable animals. However, AI in goat was poorly studied. We here compared two different estrus synchronization methods, CIDR (controlled intravaginal drug release)+$PGF_2a$ and CIDR+PMSG, in Korean native goats. After treatment of CIDR+$PGF_2a$ or CIDR+PMSG for estrus synchronization, AI was applied to 38 recipients. Of 18 recipients treated with CIDR+$PGF_2a$, 4 recipients produced offsprings. However, no pregnancy was obtained in 20 recipients treated with CIDR+PMSG. Especially, after treatment of CIDR+$PGF_2a$, AI with fresh or frozen sperm showed markedly higher parturition rates than that of CIDR+PMSG. From these results, we suggest that combination of CIDR and $PGF_2a$ could be used as good method for estrus synchronization in Korean native goats.

  • Progesterone을 함유하고 있는 CIDR(Controlled Internal Drug Release)의 질내삽 입은 황체기를 인위적으로 연장시킬 수 있다. CIDR의 삽입이,삽입시 존재했던 우세난포 (dominant follicle)의 반응과 난포의 발육반응 그리고 2회 또는 3회의 난포주기를 가지고 있는 처려우에서 CIDR의 삽입기간동안 난포의 성장 및 발육에 어떠한 영향을 미치는가를 비교검 토하기 위하여 배란후 16일째의 처녀우 4마리에 7일동안 CIDR를 삽입하였다. CIDR의 삽입 은 발정의 발현을 억제시켰으며 그리고 발정주기의 길이를 정상 발정주기보다 유의성있게 연 장시켰다($26.3{\pm} 0.5 vs 20.8{\pm}$ 1.5일, p<0.05). CIDR의 삽입시 혈장 progesterone 농도는 $3.6{\pm}$ 2.7 ng/ml 이었으며, 17일과 23일 사이에는 2.1-4.4 ng/ml($3.6{\pm}1.2 ng/ml$) 사이를 유지했다. 혈 장 estradiol-179의 농도는 난포의 발육 및 배란전 배란난포의 성숙을 나타내는 특징적인 변화 양상을 나타래었다. 4마리의 처녀우중 2마리는 CIDR 삽입전 발정주기당 2회의 난포주기를 가진 반면, 나머지 2마리는 주기당 3회의 난포주기를 가졌다. 그렇지만 CIDR의 삽입기간동안 모든 처녀우는 주기당 3회의 발정주기를 가졌다. CIDR의 삽입전 발정주기당 3회의 난포주기 를 갖는 처녀우에서 CRR의 삽입은 세 번째 난포주기에서 배란성 우세난포의 우세기 (dominant phase)를 연장시켰다. 3회의 난포주기를 갖는 2마리에서 CIDR의 삽입후 배란난포 는 존속시간과 우세기가 유의성있게 연장되었다. CIDR의 삽입전 발정주기당 2회의 난포주기 를 갖는 다른 2마리의 처녀우에서 CIDR의 삽입후 우세난포는 곧바로 퇴행되었고 새로운 난 포주기를 형성하였으며, 우세난포의 우세기와 배란난포의 존속기간을 연장시키지 않았다. CRR의 삽입은 CIDR의 삽입후 이어지는 발정주기동안 난포의 발육 및 성장에 영향을 미치 지 않았으며 발정주기의 길이, 난포주기, 혈장 progesterone 및 estradiol-179 농도에 영향을 미치지 않았다. 결과적으로 황체기 후반부에 CIDR의 삽입은 CIDR삽입전 발정주기동안 3회 의 난포주기를 갖는 처녀우에서 배란성 우세난포의 발육과 배란까지의 기간을 연장시켰고 2회 난포주기를 갖는 처녀우에서는 우세난포를 곧바로 퇴행시킨후, 새로운 난포주기를 형성 하였다.

Development of Superovulation Method in Korean Native Goats (한국 재래산양의 과배란 처치 방법 개선)

  • 한국 재래산양 체내수정란 생산에 대한 발정동기화 및 과배란 유도방법과 회수된 수정란의 동결 융해 후 생존율을 조사하였다. 발정동기화를 위해 CIDR+FSH 및 CIDR+PMSG의 방법을 이용한 결과, 배란점 및 회수된 수정란의 수는 CIDR+FSH 처리구에서 16.3개 및 9.4개, CIDR+PMSG 처리구에서 16.4개 및 8.7개를 나타내어 두 처리구간에 유의적 차이는 인정되지 않았다. 회수된 수정란을 형태학적으로 평가한 결과 CIDR+FSH 처리구에서 Gade A, B, C 및 D는 75.8%, 15.2%, 4.5% 및 4.5%를 나타낸 반면 CIDR+PMSG 처리구에서는 52.5%, 16.4%, 16.4% 및 14.8%였으며, 이식 가능한 수정란 (Grade A, B) 수는 CIDR+FSH 처리구가 유의적(P<0.05)으로 높게 나타났다. 회수된 수정란의 완만 동결 융해 후 생존성은 CIDR+FSH 처리구에서 73.3%, CIDR+PMSG 처리구에서 63.3%이었으며, 두 군간의 유의적차이는 인정되지 않았다. 따라서 본 결과는 한국 재래산양 체내수정란의 생산과 회수된 수정란의 보존을 위해서 CIDR+FSH로 발정동기화 시키는 것이 효과적이었다.

Relationship between the Conception Rate after Estrus Induction using CIDR and Other Parameters in Dairy Cows (젖소에서 CIDR 투여에 의한 발정 유도 후 수태율과 다른 인자와의 관계)

  • The purpose of this study was to determine the relationship between conception rate and other parameters (body condition score; BCS, progesterone concentrations and follicle size) before estrus induction with CIDR(intravaginal progesterone-releasing controlled internal drug release). The conception rate in cows with < 2.75, 2.75 to 3.25, and 3.25 <, BCS regardless of AI (artificial insemination) time was 46.6%, 63.3%, and 46.6% at CIDR insertion, respectively. The conception rate regardless of BCS was 54.9% in cows inseminated based on detected estrus, and 48.7% in cows inseminated at 72 to 80 hours (timed artificial insemination, TAI) after removal of CIDR. The conception rate regardless of AI time was 40.0% in cows with low progesterone concentrations (less than 1.0 ng/ml), and 56.6% in cows with high progesterone concentrations (more than 1.0 ng/ml) at CIDR injection. The conception rate regardless of progesterone concentrations was 53.8% in cows inseminated based on detected estrus, and 38.0% in cows of TAI after removal of CIDR. The conception rate regardless of AI time was 43.3% in cows with small follicle (less than 5 mm), 53.3% in cows between 5 mm to 10 mm of follicle, and 63.3% in cows with large folliclc (more than 10 mm) at CIDR injection, respectively. The conception rate regardless of follicle size was 58.4% in cows inseminated based on detected estrus, and 45.9% in cows of TAI after removal of CIDR. These results indicated that if the cows with BCS 2.75 to 3.25, active corpus luteum, and/or large dominant follicle (more than 10 mm) are used for estrus induction, the conception rate will be greater.

Reproductive Performance of Holstein Cattle Treated with Progesterone and Combination of Progesterone and Estradiol (인공수정 후 외인성 Progesterone과 Estradiol 병용 투여 및 Progesterone 단독 투여가 젖소의 번식 성적에 미치는 영향)

  • This study was carried out to investigate the effects of hormones (progesterone, and combination of progesterone and estradiol) on the reproductive performance of dairy cattle. The intravaginal CIDR was inserted in the vagina of cow on day 14 post-insemination to stimulate progesterone secretion with and without estradiol. The CIDR was removed on the day of next expected estrus. The cows for control group were not treated with CIDR or hormones. Conception rate, estrus interval, estrus intensity and serum progesterone were measured. Conception rates in control, CIDR without estradiol and CIDR treated cows were 15.4, 38.9 and 60%, respectively. Estrus interval in cows treated with CIDR was 26.5 days compared with 37.1 and 48.5 days in control and CIDR without estradiol treated cows. Estrus signs were more intense in cows treated with CIDR compared with control and CIDR without estradiol treated cows. Pregnant cows treated with CIDR showed higher serum progesterone concentration than pregnant and non-pregnant cows in control group. Particularly, serum progesterone was significantly higher in CIDR treated pregnant cows compared to non-pregnant cows at days 1, 2 and 6 of gestation. It may be concluded from present results that CIDR treatment is better than CIDR without estradiol to improve conception rate in dairy cattle.

Effects of Progestagens on Estrous Synchronization and Superovulation in Korean Native Goats (프로게스타겐이 한국 재래산양의 발정동기화 및 과배란유기에 미치는 영향)

  • This study was conducted to determine the effects of progestagen (Synchromate-B, Veramix, CIDR) and hormone (PMSG, FSH, hCG) treatments on superovulation and estrous synchronization in Korean native goats. Goats were initially superovulated by using progestagens (Synchromate-B, Veramix, CIDR) and hormones (FSH, PMSG, hCG). The progestagens were removed after 15 days of insertion and 0.88 mg of FSH was injected intramuscularly twice a day from day 12 to day 15. In addition, 150 IU of PMSG and 200 IU hCG were injected intramuscularly in the morning of day 12 and in the afternoon of day 15, respectively. Estrous synchronization was induced by the progestagen releasing devices for 13 days and intramuscular injection of 400 IU PMSG in the morning of day 11. The results were summarized as follows: 1. The responses of superovulation treated with three types (Synchromate-B, Veramix, CIDR) of progestagen were 98.6, 99.4 and 98.8%, respectively. The average ovulation rates with Synchromate-B, Veramix and CIDR were 12.58 $\pm$ 6.52, 12.91 $\pm$ 7.27 and 11.28 $\pm$ 6.33, respectively. The average ovulation rates of Synchromate-B and Veramix treatments were significantly higher than that of the CIDR treatment (P<0.05). 2. The responses of estrus synchronization treated with Synchromate-B, Veramix and CIDR were 52.9, 72.9 and 75.7%, respectively. Veramix and CIDR treatments on estrous synchronization were significantly higher than the Synchromate-B treatment (P<0.05). Among the estrous synchronized goats, the estrous ovulation rates with Synchromate-B, Veramix and CIDR were 2.11 $\pm$ 1.89, 1.35 $\pm$ 0.87 and 1.43 $\pm$ 0.96, respectively. The estrous ovulation rates of the Synchromate-B treatment were significantly higher than those of the other treatments (P<0.05). 3. The average superovulation rates were 11.76 $\pm$ 6.00 and 11.07 $\pm$ 6.46 for the PMSG treatment and control groups, showing that there was no PMSG effects for the superovulation treated with CIDR.

Transcervical or Laparoscopic Insemination of Frozen-thawed Semen in Estrus-synchronized Himalayan Tahrs (Hemitragus jemlahicus)

  • Four estrus-induced Himalayan tahrs (Hemitragus jemlahicus) were inseminated with frozen-thawed semen by laparoscopic or transcervical insemination techniques with no regard to the site of ovulation in non-breeding season. In June and July, 2009, estrus was synchronized by Eazi-Breed $CIDR^{(R)}$ (Controlled internal drug release; Pfizer Animal Health, New Zealand) insertion for 16 days and PG 600 (PMSG 400IU, hCG 200 IU; Intervet, Netherlands) injection (IM) a day before removing $CIDR^{(R)}$. Forty eight hours later, laparoscopic or transcervical insemination was done to each of two tahrs under anesthetic condition inducted by ketamine (1.5 mg/kg) and medetomidine (0.09 mg/kg). For examination of estradiol and progesterone, blood was collected right before $CIDR^{(R)}$ insertion, PG 600 injection, $CIDR^{(R)}$ removal and insemination. Estradiol levels of four tahrs (No. 1, 2, 3, 4) before $CIDR^{(R)}$ insertion and insemination were 13.3, 8.8, 14.3, 12 pg/ml and 23.5, 25.5, 21.1, 11.5 pg/ml, respectively. Progesterone levels of four tahrs (No. 1, 2, 3, 4) before $CIDR^{(R)}$ insertion and insemination were 1.8, 0.05, 0.63, 0.61 ng/ml and 1.03, 0.37, 1.48, 2.12 ng/ml. Except for No. 4 tahr, cervices showed cervical mucus and opened enough to penetrate with embryo transfer gun sheet usually used for cows. Therefore, No.4 was laparoscopically inseminated together with No. 1. In conclusion, none of four Himalayan tahrs was pregnant. However, we proved that estrus could be induced by CIDR and PG 600 injection in non-breeding season, and laparoscopic or transcervical insemination with frozen-thawed semen could be one of assisted reproductive techniques in Himalayan Tahr.

Estrus Synchronization and Artificial Insemination in Dairy Goats (유산양에서 발정동기화 유도와 인공수정 효율)

  • This study was conducted to evaluate the response of CIDR-S on estrus synchronization of dairy goats and the effect of artificial insemination on conception rate. Estrus synchronization of sexually matured Saanen goats was induced with insertion of CIDR-S into vagina for 15 days and injection of 500 IU PMSG before removal of CIDR-S. Artificial insemination was conducted using liquid or frozen semen by intra-cervical insemination with synchronized does. Estrus synchronization was 90% using CIDR-S insertion for 15 days. Conception rate of inseminated does was 20% - 25%, while that of natural mated does was 100%. These results suggested that synchronization of dairy goats can be successfully induced and artificial insemination method has to be improved for the practical use in dairy goats.

Effect of Duration Treated on Reproductive Performance in Holstein Cows Treated with Progesterone with and without Estradiol (인공수정 후 외인성 Progesterone과 Estradiol 병용 및 Progesterone 단독 투여 기간이 젖소의 번식 성적에 미치는 영향)

  • This study was carried out to investigate the effects of hormones treated (progesterone, and combination of progesterone and estradiol) on the reproductive performance of dairy cattle. The intravaginal CIDR was inserted in the vagina of cow on day 14 (P-7, PE-7), 16 (P-5, PE-5) and 18 (P-3, PE-3) post-insemination to stimulate progesterone secretion with (PE) and without estradiol(P). The CIDR was removed on 7 (P-7, PE-7), 5 (P-5, PE-5) and 3 (P-3, PE-3) days after. The cows for control group were not treated with CIDR or hormones. Conception rate, the duration needed for conception after AI and the number of AI services needed for conception were measured. Conception rate in control, CIDR without estradiol (P-7, P-5 and P-3) and CIDR treated (PE-7, PE-5 and PE-3) cows were 15.8%, 40.0%, 31.5%, 28.6%, 62.5%, 50.0% and 0%, respectively. The days needed for conception after AI in control, P-7, P-5, p-3, PE-7, PE-5 and PE-3 were 64.9, 63.0, 59.1, 8.0, 0.0, 18.9 and 83.3 days, respectively. The days needed for conception in cows treated with CIDR (PE-7) has shown longer than control and PE-3 (p<0.05). The number of AI services needed for conception in control, CIDR (PE-7, PE-5 and PE-3) and CIDR without estradiol treated cows (P-7, P-5 and P-3) were 27, 2.3, 1.9, 1.3, 1.0, 1.7 and 2.8 times, respectively. The number of AI services needed for conception have shown significantly lower in PE-7 compared to control and PE-3.
