새로운 경구용 세팔로스포린의 약효평가 (Studies of Pharmacological Activity on New Oral Cephalosporins)

  • ${\beta}-Lactamase$ stability, chemotherapeutic activity, and pharmacokinetics of 7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyridyl)piperazinyl]thiocarbonylthiomethyl-3-cephem-4-carboxylic acid(CEN1), 7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyrimidyl)piperazinyl]thiocarbonylthiomethyl-3-cephem-4-carboxylic acid(CEN2), pivaloyloxymethyl-7-[(Z)-2-(2-aminothizaole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyridyl)piperazinyl]thiocarbonyl-thiomethyl-3-cephem-4-carboxylate(CEN1P), and pivaloyloxymethyl-7-{(Z)--2-(2-aminothizaole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyridyl)piperazinyl]thiocarbonyl-thiomethyl-3-cephem-4-carboxylate(CEN2P) were examined. CEN1, CEN2, CEN1P, and CEN2P were very stable to the ${\beta}-lactamase$ obtained from three strains(Enterobacter cloacae P99, Escherichia coli TEM, and Citrobacter freundii). Chemotherapeutic activities$(ED_{50})$ of CEN2 and CEN2P against experimental systemic infections due to Streptococcus pyogenes 77A and Escherichia coli 078 were superior to those of CEN1 and CEN1P, respectively. The $ED_{50}$ values of CEN1, CEN2 were 5.82 mg/kg, 0.89 mg/kg(s.c., S. pyogenes 77A) while those of CEN1P, CEN2P were 14.56mg/kg, 6.40mg/kg(p.o., S. pyogenes 77A), respectively. The pharmacokinetics of CEN1, CEN2, CEN1P, and CEN2P were investigated in mice and rats. In mice, peak blood levels of $1.25\;{\mu}g/ml$ were recorded within 20 min after oral administration of a single dose equivalent to 40 mg/kg CEN1P. Cmax of CEN1P was much higher than that of CEN1 in mice and rats. Oral absorption of CEN2P was much higher than that of CEN2.

  • Physicochemical properties and hydrolysis kinetics of new some oral cephalosporins were examined in buttered solution and human plasma or rat liver homogenate. The test cephalosporins were 7-[(Z)-2-(2-aminothiazole-4-yl)-2- methoxyiminoacetamido]-3-[4-(2-pyridyl)piperazinyl] thiocarbonylthhiomethyl-3-cephem-4-carboxylic acid (CEN1), 7-[(Z)-2-(2-aminoth iazole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyrimidyl)piperazinyl]th iocarbonylthiomethyl-3-cephem-4-carboxylic acid (CEN2), pivaloyloxymethyl-7-[ (Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyridyl)piperazi nyl]thiocarbonylthiomethy1-3-cephem-4-carboxylate (CEN1P), and pivaloyloxymethyl-7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamido]-3-[ 4-(2-pyrimidyl)piperazinyl]thiocarbonyl-thiomethyl-3-cephem-4-carboxylate (CEN2P). The partition coefficient(Ko/w) of CEN1P, CEN2P were higher than those of CEN1, CEN2. The calculated pKa values of CEN1, CEN2, CEN1P, and CEN2P were 7.09, 7.75, 4.92, and 5.39, respectively. The hydrolysis of CEN1P and CEN2P were not depend on the composition of pH of the test medium except weak alkaline buffered solution (pH 8.00). CEN1 and CEN2 were very stable in pH 6.80 and 8.00 buffer solutions. CEN1P and CEN2P were rapidly deesterified to CEN1 and CEN2 in human plasma and in rat liver homogenate. Half-lives$(t_{1/2})$ of CEN1 and CEN2 were 3.49 and 4.93 hr in human plasma, 1.47 and 1.26 hr in rat liver homogenate, respectively.

새로운 경구용 세파로스포린의 합성 및 항균작용 (Synthesis and Antibacterial Activities of New Oral Cephalosporins)

  • In order to develop oral cephalosporin having a new substituent at 3 position, the synthesis of cephalosporins modified at C-3 and the effect of the substituents on the oral absorption is studied. 7-[(Z)-2-(2-Aminothiazole- 4-yl)-2-methoxyiminoacetamidol-3-[4-(2-pyridyl )piperazinyl] thiocarbonylthiomethyl-3-cephem-4-carboxylic acid (CEN1) and 7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyrimid yl)piperazinylthiocarbonylthiomethyl-3-cephem-4-carboxylic acid (CEN2) were synthesized from 4-(2-piridyl)piperazinyl dithiocarbamate potassium salt or 4-(2-pirimidyl)piperazinyl dithiocarbamate potassium salt and cefotaxime. Also pivaloyloxymethyl esters of CEN1 and CEN2, pivaloyloxymethyl 7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyridyl )piperazinyllthiocarbonylthiomethyl-3-cephem-4-carboxylate (CENIP) and pivaloyloxymethyl 7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamidol-3- [4-(2-pyrimid yl)piperazinyllthiocarbonylthiomethyl-3-cephem-4-carboxylate (CEN2P) were synthesized. The in vitro activities of two new oral cephalosporins, CEN1 and CEN2, were compared with the in vitro activities of cefaclor and cefotaxime against a variety of bacterial species. CEN2 has a broad antibacterial spectrum covering Gram-positive and Gram-negative bacteria, similar to that exhibited by CEN1 and cefotaxime. CEN1 and CEN2 were more active in vitro than cefaclor against Streptococcus pyogenes, Klebsiella aerogenes and Enterobacter cloacae.

Stellar populations of Galactic Globular Cluster $\omega$ Cen using HST/WFC3 near-UV observations

  • The most massive galactic globular cluster, $\omega$ Cen has been observed using recently mounted WFC3/UVIS cameras of Hubble Space Telescope at both near-UV and optical bands. We present the photometry of stars in the central region of $\omega$ Cen in F225W, F275W and F336W bands. The near-UV color-magnitude diagrams and color-color diagrams of stars in $\omega$ Cen show multiple red giant branches and main sequences as already discovered in both ground-based and HST/ACS observations in the optical bands. We modelled the stellar populations of $\omega$ Cen using Yonsei Isochrone, where $\alpha$-particles and helium enhancement processes have been included properly. We compare the best-fit stellar populations obtained from the current near-UV observations to the suggested stellar population models based on optical band data. We also discuss the methodological issues arising when dealing with the near-UV observations; red-leak in near-UV filters and the huge interstellar extinction in near-UV bands compared to the optical bands and its non-linear effect in color-magnitude diagrams.

살균소독제의 정량적 표면시험방법별 유효성 비교 (Comparison Study on Efficacies of Disinfectants and Sanitizers Among Methods for Quantitative Surface Test)

  • 현재 살균소독제의 국가 공인시험법은 시험관 내에서 행하는 현탁액시험법으로 실제 현장의 표면 살균을 정확히 평가하기에는 무리가 있다. 본 연구는 국제적으로 표준화되어 있는 3가지 정량적 표면시험법간의 유효성 평가력 비교를 목적으로 E. coli와 S. aureus를 대상으로 시중 유통되는 대표적 5개 계열 살균소독제의 유효성을 평가하였다. 그 결과 E. coli와 S. aureus에 대한 각 방법별 살균소독력은 ASTM(1)법에서 5.18 $\pm$ 0.03, 5.27 $\pm$ 0.04 log cfu/carrier, ASTM (2)법에서 4.63 $\pm$ 0.04, 3.97 $\pm$ 0.03 log cfu/carrier, CEN EN 13697법의 청정조건(CEN(1))에서 6.14 $\pm$ 0.05, 5.31 $\pm$ 0.10 log cfu/carrier, 오염조건(CEN(2))에서 4.37 $\pm$ 0.02, 4.06 $\pm$ 0.01 log cfu/carrier로 나타났다. CEN(1)시험법이 ASTM(2)와 CEN(2) 시험법에 비해 살균소독력이 유의적으로 높게 평가되었다. 적용된 3개의 시험법 중 유리를 담체로 사용한 ASTM(1)법에 비해 스테인리스 스틸을 사용한 ASTM(2)와 CEN법 적용 시 살균소독효과가 낮게 평가되었다. 결론적으로 3 가지 국가 공인시험법간 유효성 평가결과가 달랐으며, 실제 식품제조 현장에서 사용되는 표면에서의 살균소독력을 정확히 평가하기 어렵다는 것을 확인할 수 있었다.

The conversion of ammonium uranate prepared via sol-gel synthesis into uranium oxides

  • A combination of simultaneous thermal analysis, evolved gas analysis and non-ambient XRD techniques was used to characterise and investigate the conversion reactions of ammonium uranates into uranium oxides. Two solid phases of the ternary system NH3 - UO3 - H2O were synthesised under specified conditions. Microspheres prepared by the sol-gel method via internal gelation were identified as 3UO3·2NH3·4H2O, whereas the product of a typical ammonium diuranate precipitation reaction was associated to the composition 3UO3·NH3·5H2O. The thermal decomposition profile of both compounds in air feature distinct reaction steps towards the conversion to U3O8, owing to the successive release of water and ammonia molecules. Both compounds are converted into α-U3O8 above 550 ℃, but the crystallographic transition occurs differently. In compound 3UO3·NH3·5H2O (ADU) the transformation occurs via the crystalline β-UO3 phase, whereas in compound 3UO3·2NH3·4H2O (microspheres) an amorphous UO3 intermediate was observed. The new insights obtained on these uranate systems improve the information base for designing and synthesising minor actinide-containing target materials in future applications.

고효율 효모 유전자 운반체의 개발 (Construction of the Stable and High Copy Number Yeast Vectors)

  • 효모 유전자 운반체의 유형을 특징지우는 ARS1 (autonomous replicating sequence), CEN3 (centromere), 2 $\mu$m OR (yeast plasmid의 origin of replication)의 DNA 절편을 재조합하여 만든 유전자 운반체들의 형질전환력, 안정도 및 운반체 수를 효모균주 SHY4(cir$^+$)와 NNY1(cir$^{\circ}$)에서 비교 조사하였다. CEN3를 갖는 유전자 운반체는 매우 안정하나 세포당 수가 하나로 매우 낮으며 2 $\mu$m OR과 ARS1 을 동시에 갖는 유전자 운반체는 안정할 뿐만 아니라 세포당 수도 높으며 균주내에 존재하는 2 $\mu$m 플라스미드는 2 $\mu$m OR을 가지는 유전자 운반체의 복제에 영향을 준다는 사실을 알 수 있었다.

카사바 당화액을 이용한 실험실용 및 산업용 효모의 에탄올 발효성능 비교 (Comparison of Ethanol Fermentation Properties between Laboratorial and Industrial Yeast Strains using Cassava Hydrolysate)

  • 실험실용 효모 CEN.PK2-1D와 산업용 효모 JHS100와 JHS200의 ethanol 발효성능을 순수배지와 카사바 당화액에서 비교하고 발효특성을 규명하기 위해 세포성장속도와 ethanol 내성, 부산물 glycerol과 acetate의 생성에 대해 비교하였다. JHS100과 JHS200은 CEN.PK2-1D과 비교하여 세포성장이 빨랐으며 ethanol에 대한 내성이 높았다. 순수YP 배지에 300 g/L glucose를 탄소원으로 이용한 회분식 배양에서 세 효모 모두 0.46 g/g의 ethanol 생산수율을 나타냈으며 생산성은 세포성장속도가 가장 빨랐던 JHS100이 3.05 g/L-hr로 가장 높아서 최종적으로 JHS100 균주는 136.6 g/L의 ethanol을 생산하였다. 카사바 당화액에 질소원을 추가하지 않은 배지를 이용한 혐기성 회분식 배양에서 산업용 효모 JHS100은 106.1 g/L ethanol을 최종적으로 생산하였고, 0.42 g/g 생산수율과 3.15 g/L-hr 생산성을 보였다. 특히, 카사바 당화액과 순수 YP배지를 이용한 발효의 ethanol 생산변수를 비교할 경우, 실험실용 효모 CEN.PK2-1D는 생산수율과 생산성이 각각 19%와 17% 감소한 반면, 산업용 효모 JHS100과 JHS200은 생산수율이 8% 감소하였고 유사한 생산성을 보였다. 또한, ethanol 생산과정의 최대 부산물인 glycerol과 acetate의 생산에 대해서 JHS100과 JHS200이 CEN.PK2-1D에 비하여 크게 낮았다. 따라서 산업용 효모인 JHS100과 JHS200의 뛰어난 ethanol 발효성능은 빠른 세포성장과 높은 ethanol 내성, 낮은 질소원 요구성, 부산물인 glycerol과 acetate의 낮은 생산성 등에 기인하는 것으로 예상한다.

근접 식쌍성의 분광관측 (I) - V593 Cen, TY Men, 그리고 BX And의 자료처리와 시선속도 곡선

  • 2003년 2월 11일부터 13일까지 3일간 아르헨티나 CASLEO 천문대의 2.15m 망원경을 이용하여 근접쌍성 TY Men과 V593 Cen을, 2003년 2월 26일부터 27일까지 2일간 보현산 천문대의 1.8m 망원경을 이용하여 근접쌍성 BX And를 에셀분광기로 분광관측하였다. IRAF의 분광처리 패키지를 사용하여 관측자료를 전처리 하였고, IRAF의 XCSAO의 CCF(Cross Correlation Function)와 Rucinski의 BF(Brodening Function)를 이용하여 세별의 단일 시선속도곡선을 각각 구하였다. (중략)

응급전문간호사 인력 수요추계에 관한 조사연구 (Projected Workforce of Certified Emergency Nurses in Korea)

  • Purpose: The purpose of this study was to suggest the projected workforce of certified emergency nurses in Korea. Method: The methods used in this study are 1) demand & ratio model was used for the projected workforce of CEN, 2) Index functional formula was used for the suggestion of the number of general hospitals and hospitals, 3) Experts in Emergency care were contacted to get an opinion and information about the criteria of distribution and scope of CEN, 4) National and international internet data were collected. Result: The demand of CEN were analyzed by two ways; demand of Emergency centers only and demand of emergency centers including community centers. The number of CENs needed to meet the demands in 2002, 2005 and 2010 was estimated at minimum of 1,512 to maximum1,576, 1,640 to 1,704 and 1,892 to 1,956 respectively. The projected number of CENs for the 2002 was 1% total available nurses in Korea, and 3.2 CENs per 100,000 population. Conclusion: It is really desirable that CENs not only work for hospital emergency centers but also for emergency related centers in community.
