Cytotoxicity of Cytosine Deaminase (CD) Adenoviral Vectors(AV) with a Promoter (L-plastin) for Epithelial Cancer Cells.

  • The object of this study was to develop a gene therapy strategy for ovarian cancer. We have previously shown that AV with a L-plastin (LP) promoter infects breast and ovarian cancer cells and expressed ${\beta}$-galactosidase cDNA in preference to normal fibroblast cells and hematopoietic cells. We now report on the cytotoxicity of Ad.LP.CD, an AV carrying a CD cDNA which converts the pro-drug, 5-Fluorocytosine (5-FC) into the toxic drug 5-Fluorouracil (5-FU). Infection of Ad.LP.CD into either 293 cells or ovarian cancer cells generated the functional CD as measured by HPLC analysis. Using a ratio of AV to OVCAR3 cell of 100 and a 5-FC concentration of 100 ${\mu}$M, we achieve an over 95 % of cell growth inhibition. We are using flow cytometry analysis for ${\beta}$ -galactosidase and ovarian cancer associated folate receptor to screen primary ascites samples for infectivity after infection with an adenoviral vector, i.e., Ad.LP.LacZ. This vector system may be of value in the treatment of microscopic disease of ovarian cancer in the peritoneal cavity.

Preparation and Characterization of CdSe nanoparticle for Solar Cell application (태양전지용 CdSe 나노입자의 합성)

  • CdSe nanoparticles were prepared by chemical solution methods using $CdCl_2{\cdot}4H_2O$ (or $Cd(NO_3)_ 2{\cdot}4H_2O$) and $Na_2SeSO_3$. The characteristics of CdSe nanoparticles were controlled by the react ion time, reaction temperature and reaction method as well as the surfactants. Cetyltrimethyl ammonium bromide(CTAB) was used as a capping agent to control the chemical reactions in aqueous solution. Polyvinylalcohol(PVA) was used as a templet in sono-chemical method. CdSe nanoparticles synthesized in aqueous solution showed homogeneous size distribution with relatively stable surface. CdSe nanoparticles synthesized in non-aqueous solution containing diethanolamine(DEA) showed the structure transformation from cubic to hexagonal as the reduction temperature increased from 80 to $160^{\circ}C$. Core shell CdSe was synthesized by sono-chemical method. Characteristics of CdSe nanoparticles were analyzed using transmission electron microscopy(TEM), x-ray photoelectron spectroscopy(XPS), x-ray diffraction(XRD), UV-Vis absorption spectra, fourier transform infrared spectroscopy(FT-IR) and photoluminescence spectra spectroscopy(PL). This paper presents simple routes to prepare CdSe nanoparticles for solar cell applications.

Differential Signaling via Tumor Necrosis Factor-Associated Factors (TRAFs) by CD27 and CD40 in Mouse B Cells

  • Background: CD27 is recently known as a memory B cell marker and is mainly expressed in activated T cells, some B cell population and NK cells. CD27 is a member of tumor necrosis factor receptor family. Like CD40 molecule, CD27 has (P/S/T/A) X(Q/E)E motif for interacting with TNF receptor-associated factors (TRAFs), and TRAF2 and TRAF5 bindings to CD27 in 293T cells were reported. Methods: To investigate the CD27 signaling effect in B cells, human CD40 extracellular domain containing mouse CD27 cytoplamic domain construct (hCD40-mCD27) was transfected into mouse B cell line CH12.LX and M12.4.1. Results: Through the stimulation of hCD40-mCD27 molecule via anti-human CD40 antibody or CD154 ligation, expression of CD11a, CD23, CD54, CD70 and CD80 were increased and secretion of IgM was induced, which were comparable to the effect of CD40 stimulation. TRAF2 and TRAF3 were recruited into lipid-enriched membrane raft and were bound to CD27 in M12.4.1 cells. CD27 stimulation, however, did not increase TRAF2 or TRAF3 degradation. Conclusion: In contrast to CD40 signaling pathway, TRAF2 and TRAF3 degradation was not observed after CD27 stimulation and it might contribute to prolonged B cell activation through CD27 signaling.

Acute Toxicity of Heavy Metal (Cd, Cu, Zn) on the Hatching Rates of Fertilized Eggs in the Olive Flounder (Paralichthys olivaceus) (넙치(Paralichthys olivaceus) 수정란 부화율에 대한 중금속(Cd, Cu, Zn)의 급성독성)

  • Acute toxicity test of heavy metal (Cd, Cu, Zn) were examined using the hatching rates of fertilized eggs in the oliver flounder, Paralichthys olivaceus. Eggs were exposed to Cd, Cu, Zn (0, 10, 100, 500, 1000, 2500, 5000 ppb) and then normal hatching rates were investigated after 48 h. The normal hatching rates in the control condition (not including Cd, Cu and Zn) were greater than 80%, but suddenly decreased with increasing of heavy metal concentrations. Cd, Cu and Zn reduced the normal hatching rates in concentration-dependent way and a significant reduction occurred at concentration grater than 1000, 100, 100 ppb, respectively. The ranking of heavy metal toxicity was Zn>Cu>Cd, with $EC_{50}$ values of 584, 1015 and 1282 ppb, respectively. The no-observed-effect-concentration (NOEC) and the lowest-observed-effect-concentration (LOEC) showed each 100 and 500 ppb of normal hatching rates in exposed to Cu and Zn. The NOEC and LOEC of normal hatching rates in Cd were 500 ppb and 1000 ppb, respectively. From these results, the normal hatching rates of P. olivaceus have toxic effect at greater than the 100 ppb concentrations in Cu, Zn and the 500 ppb concentrations in Cd in natural ecosystems. These results suggest that biological assay using the normal hatching rates of P. olivaceus are very useful test method for the acute toxicity assessment of a toxic substance as heavy metal in marine ecosystems.

Preparation and Anti-fouling Properties of PVDF Mixed Matrix Asymmetric Membranes Impregnated with 𝛽-cyclodextrin (𝛽-사이클로덱스트린을 함침시킨 PVDF 혼합기질 비대칭막의 제조와 내오염성 평가)

  • Poly(vinylidene fluoride) (PVDF) membrane has a good membrane durability because of its high mechanical resistance, thermal and chemical stability. However, the strong hydrophobic property of PVDF membrane can induce a low water permeability and easy fouling by proteins and organic matters. In order to improve the anti-fouling properties of PVDF membrane, the PVDF mixed matrix asymmetric membranes impregnated with biofunctional material 𝛽-cyclodextrin (𝛽-CD) in the membrane structure were prepared by phase inversion method. The membrane filtration experiments of pure water and BSA solution were performed using the PVDF/𝛽-CD mixed matrix asymmetric membranes prepared according to the 𝛽-CD contents. The experiments showed that the introduction of 𝛽-CD into the PVDF polymer matrix contributed to increase in the hydrophilic property of the PVDF membranes, and this led to the reduction of contact angles and improvement of anti-fouling properties. The PVDF/𝛽-CD membrane which was prepared using the dope solution with a 2 wt% 𝛽-CD content represented 64 L/m2·h of pure water flux, 95% of BSA rejection and maximum 80% of flux enhancements compared to flux results of the pristine PVDF membrane.

  • Simultaneous liquefaction and cyclodextrin (CD) production were conducted on potato starch using cyclomaltodextrin glucanotransferase (CGTase) from a mutant strain MNNG 8 of Bacillus stearothermophilus No. 239. A high concentration (30%) of potato starch was converted to cyc1o-dextrins (CDs) with 29% yield in the conditions of pH 6.0, temperature $80^{\circ}C$, 4.3 mM $CaCl_2$, CGTase addition of 3.0 dextrinizing activity unit (DAU) at $40^{\circ}C$/g starch.

Facile Synthesis of CdTe Nanorods from the Growth of Te Nanorods

  • One-dimensional CdTe nanorods (NRs) are obtained by the reaction of various Cd precursors with single crystalline Te nanorod templates, which are pre-synthesized from Te precursors by a simple and reproducible solvothermal method. Throughout the process, the diffraction intensity of different crystal facets of single crystalline Te NRs varied with reaction times. Finally, by alloying Cd ions along the axial direction of Te NRs, polycrystalline cubic phase CdTe NRs with diameters of 80-150 nm and length up to $1.2-2.4{\mu}m$ are obtained. The nucleation and growth processes of Te and CdTe NRs are discussed in details, and their properties are characterized by XRD, SEM, TEM, Raman scattering, and UV-vis absorption spectra. It was found that the key elements of synthesizing CdTe NRs such as reaction temperatures and Cd sources will strongly influence the final shape of CdTe NRs.

CD83 expression induced by CpG-DNA stimulation in a macrophage cell line RAW 264.7

  • CpG-DNA has various immunomodulatory effects in dendritic cells, B cells, and macrophages. While induction of cytokines by CpG-DNA has been well documented in macrophages, the expression of costimulatory molecules in CpG-DNA treated macrophages has not yet been defined. Therefore, we investigated the effects of CpG-DNA on the expression of costimulatory molecules in RAW 264.7 cells. The surface expression of CD80 was slightly increased and CD83 expression was significantly increased in response to CpG-DNA. However, the expression of CD86 and MHC class II was not changed. As expression of CD83 mRNA was also increased by CpG-DNA, CD83 expression is regulated at a transcriptional level. To understand the contribution of signaling pathways to CD83 induction, we used pathway specific inhibitors. The NF-${\kappa}B$ inhibitor significantly reduced surface expression of CD83 as well as phagocytic activity of RAW 264.7 cells. Therefore, CD83 expression may contribute to the immunostimulatory effects of CpG-DNA in macrophage cells.

Influence of Feeding β-Cyclodextrin on Reducing the Content of Cholesterol in Pork (돼지고기의 콜레스테롤 함량 감소에 관한 β-Cyclodextrin의 급여효과)

  • The main objective of the present study was to determine the effect of dietary ${\beta}-cyclodextrin\;({\beta}CD)$ on reducing the cholesterol content of pork. Twelve swine of 50 kg body weight were randomly distributed into four groups of three heads each and fed experimental diets for 9 weeks until they reached a market weight of 110 kg. They were assigned to the four experimental diets containing 0% (control), 1.5%, 3.0% or 5.0% pure ${\beta}CD$. Daily feed intake, body weight gain and feed efficiency were not significantly different between any of the four group. The plasma total lipid, triacylglycerol and total cholesterol content of the swine in the three ${\beta}CD$-fed groups were significantly (p<0.05) decreased when compared to those in the control group, and were significantly (p<0.05) reduced by 21.80%,55.58% and 27.69%, respectively, in the swine fed on 5% ${\beta}CD$. The cholesterol content of pork belly (mg/100g) was significantly (p<0.05) decreased by 5.33 mg, 12.70 mg and 15.23 mg in the swine maintained on 1.5%, 3.0% and 5.0% ${\beta}CD$, respectively. The cholesterol content of pork belly, when expressed as the rate of reduced cholesterol, was significantly (p<0.05) decreased by 6.44%, 15.36% and 18.42% in groups of 1.5%, 3.0% and 5.0% ${\beta}CD$, respectively, when compared to that of the control group. These results suggest that dietary ${\beta}CD$ may be classified as dietary fiber which can modulate cholesterol metabolism in swine.
