Objectives : The purpose of this study is to investigate effects of Syzygium aromaticum (SAE) spread on the allergic contact dermatitis caused by 2,4-dinitro-chlorobezene (DNCB). Methods : Forty-two mice were divided into six groups ; normal, negative control (DNCB-treated), positive control (DNCB + 1% pimecrolimus), experimental group I, II and III. control and experimental groups were induced allergic contact dermatitis by DNCB. Experimental group I(DNCB + 0.2% SAE), II(DNCB + 1% SAE) and III(DNCB + 5% SAE) were spread SAE and positive control was spread the 1% pimecrolimus. In this study, effect of SAE on clinical aspects on the skin, histopathological change, the blood level of IgE, cytokines, histamine were investigated. In addition, effect of SAE on spleen $CD4^+/CD8^+$ T cell subset was investigated. Results : 1. In experimental group I, II and III, erythemas and edema were more reduced than negative control. 2. In experimental group I, II and III inflammatory edema and the numbers of infiltrated inflammatory cells were more reduced than negative control. 3. In experimental group I, II and III, clinical skin score was more reduced than negative control. 4. In experimental group II and III, the thickness of skin was statistically significant reduced than negative control. 5. In experimental group II and III, histamine release was statistically significant reduced than negative control in dose-dependantly. 6. In experimental group II and III, cytokines (IL-1${\beta}$, TNF-${\alpha}$, IL-4, IL-6, IL-10) were statistically significant reduced than negative control in dose-dependantly. 7. In experimental group I, II and III, the level of total IgE was statistically significant reduced than negative control in dose-dependantly. 8. In experimental group III, $CD4^+$ and $CD8^+$ T cells were statistically significant decreased similar to the positive control. Conclusions : According to above experiments, Syzygium aromaticum(SAE) was effective on allergic contact dermatitis.
To evaluate the effects of GP on contact dermatitis, we examined the composition of immune cells from drain lymph node in DNCB-induced contact dermatitis murine model NC/Nga mice. And the amount of pathologic cytokines of spleen and antioxidant activity were investigated. The results were summarized as followers; 1. GP did not show cytotoxic effect on mLFC in vitro. 2. GP did not have hepatotoxicity in vivo in the level of ALT, AST. 3. GP decreased the production of DPPH and in a dose-dependent. 4. GP significantly decreased total cell number of DLN in DNCB-induced NC/Nga mice compared to the untreated control group. 5. GP significantly decreased the number of CD3+, CD19+, CD4+, CD8+, CD3+/CD69+ and CD4+/CD45+ in DLN of DNCB-induced NC/Nga mice compared to the untreated control group. 6. GP significantly reduced the level of IL-4 and IFN-$\gamma$ in splenocytes of DNCB-induced NC/Nga mice compared to the untreated control group. Taken together above results, GP have therapeutic effects on contact dermatitis by regulating T cell activation. This study warranted further investigations of molecular mechanisms of GP on contact dermatitis.
Vaccination is the most effective way to prevent influenza virus infections. However, conventional vaccines based on hemagglutinin (HA) have to be annually updated because the HA of influenza viruses constantly mutates. In this study, we produced a 3M2e-3HA2-NP chimeric protein as a vaccine antigen candidate using an Escherichia coli expression system. The vaccination of chimeric protein (15 ㎍) conferred complete protection against A/Puerto Rico/8/1934 (H1N1; PR8) in mice. It strongly induced influenza virus-specific antibody responses, cytotoxic T lymphocyte activity, and antibody-dependent cellular cytotoxicity. To spare the dose and enhance the cross-reactivity of the chimeric, we used a complex of poly-γ-glutamic acid and alum (PGA/alum) as an adjuvant. PGA/alum-adjuvanted, low-dose chimeric protein (1 or 5 ㎍) exhibited higher cross-protective effects against influenza A viruses (PR8, CA04, and H3N2) compared with those of chimeric alone or alum-adjuvanted proteins in vaccinated mice. Moreover, the depletion of CD4+ T, CD8+ T, and NK cells reduced the survival rate and efficacy of the PGA/alum-adjuvanted chimeric protein. Collectively, the vaccination of PGA/alum-adjuvanted chimeric protein induced strong protection efficacy against homologous and heterologous influenza viruses in mice, which suggests that it may be a promising universal influenza vaccine candidate.
The purpose of this study was to establish an in vitro culture method of tumor-specific T cells, and determine the efficacy of the cultured tumor-specific cytotoxic T-lymphocytes (CTL) as an agent of anti-tumor immunotherapy against a murine lymphoma, TIMI.4. Tumor-specific T-lymphocytes derived from C57BL/6 mice (thy-1.2) immune to TIMI.4 were activated by in vitro stimulation with the irradiated TIMI.4 cells, and expanded by restimulation with TIMI.4 in the presence of the concanavalin A-stimulated rat spleen culture supernatant, and splenic antigen-presenting cells. In vitro restimulation enhanced markedly the proportion of $CD8^+$, a predominant surface marker of CTL and the cytotoxic activity in the cultured immune T cell population. The resulting TIMI.4-specific T cells were adoptively transferred into nude mice. The tumor cells residing in the host after 7 days of adoptive transfer to B6.PL (thy-1.1) mice were quantified by use of an antibody directed to the thy-1.2 allele. The TIMI.4 cells in the recipient nude mice were decreased in a dose-dependent manner. Anti-tumor activity of the TIMI.4-specific T cells was also demonstrated by a survival test, where the tumor-bearing nu/nu mice which received the activated T-cells survived about 30% longer than the control mice which received the tumor cells alone. These suggest that adoptive transfer of TIMI.4-specific T cells could be a candidate for effective therapy of the murine lymphoma.
Atopic dermitiis(AD) is a chronic inflammatory skin disease, which requires safe and effective medicinal therapy. Over production of Th2 cytokines and chemokines as well as IgE, which are mediated by highly activated immune cells, have been considered as pathologic factors in this disease. We found that Gamipaidok-san(GPDS), which is a traditional herbal medicine clinically prescribing for atopic dermitis patients in the hospital, has suppressive effects on the development of DNC8 induced dermatitis in Nc/Nga atopic model. Oral administration of GPDS at the concentration of 250 mg/Kg for 12 weeks significantly suppressed the clinical severity of the dermatitis including pruities, edema, eczematous and dryness. Histological examination revealed that thickness of dermis and epidermis were considerably reduced, and the number of infiltrated inflammatory immune cells including mast cells, CCR3+, and CD4+ T cells were decreased in the affected skin and ear, and consistantly, the number of CD3+/CCR3+ cells in Iymph nodes were decreased. The levels of Th2 cytokines produced by activated splenocyte from atopic mice were also down-regulated by GPDS. Furthermore, the serum levels of IgE were considerably reduced, which accompanied by a decrease in the number of B220+IgE+ B cells in the Iymph nodes. Taken together, these results suggested that oral administration of GPDS, a traditional herbal medicine, has suppressive effects on atopic dermitis of Nc/Nga mouse by the modulation of the immune system, therefore GPDS has potential as a natural therapeutic for treatment of atopic dermatitis.
Cortex Mori radicis has been used as a components of antidiabetics, antiasthma and diuresis in Oriental Medicine. This experiment examined the effect of an extracts, obtained from the acetone(CM-A) and aqueous(CM-W) extracts of Cortex Mori radicis, on hair growing activity of the C57BL/6N mice after topical application to skin. We investigated the number of hair follicle and mast cells, and changes of subpopulation of splenocytes and thymocytes in skin for 16 day. The results were as follows : The Hair growing effect in experimental groups was more increased in 85%(CM-A) and 90%(CM-W) than control group(10%) in hair depilated area. The number of hair follicle in experimental groups(CM-A and CM-W) was more increased than control group. Splenic B/T lymphocytes of CM-A group were decreased compare to control group. CD4/CD8 positive TH cells in splenic T lymphocytes of CM-W group were increased compare to control group. These results suggest that CM-A and CM-W may be used in treatment of alopecia areata.
We here document discovery of a new and simple model of tumor seeding involving the mouse peritoneum. Irradiated tumor cells administered by i.p. injection provided effective vaccination against peritoneal carcinomatosis and distal metastasis with colorectal carcinomas. In flow cytometric analysis, CD4+ and CD8+ T lymphocytes, macrophages and myeloid-derived suppressor cells (MDSCs), which are easy to obtain in the peritoneal cavity, were revealed to have significant differences between immunized and non-immunized mice and these contributed to antitumor responses. We also observed that both serum and peritoneal lavage fluid harvested from immunized mice showed the presence of CT26-specific autoantibodies. In addition, increase in level of TGF-${\beta}1$ and IL-10 in serum but a decrease of TGF-${\beta}1$ in peritoneum was found. Taken together, these findings may provide a new vaccine strategy for the prevention of peritoneal and even systemic metastasis of carcinomas through induction of an autoimmune response in the peritoneum.
Rejection is one of the life-threatening complications after organ transplantation. An eight-month-old, intact male, mixed breed dog was presented with acute rejection after renal allograft. The heterotopic renal transplantation with bilateral nephrectomy was performed in the dog. The triple drug protocol for immunosuppression was applied for prevention of the acute rejection. Postoperative care was done according to the transplantation protocol of VMTH, Kangwon National University. The dog was euthanized when the serum creatinine concentration exceeded 5 mg/dL followed by tile signs of illness. The transplanted kidney was enlarged. The renal cortex lesions were characterized by necrosis of the renal tubules and the glomeruli. Interstitial lesions were characterized by hemorrhage and severe infiltration of lymphoid cells. Intrarenal arteries showed necrosis of the walls and infiltration of perivascular lymphoid cells. In immunohistochemical (IHC) findings, infiltration of the CD4 and the CD8 positive T lymphocytes was examined. In this case, acute rejection was shown by humoral and cellular immunity on the basis of histopathologic and IHC evaluation.

Background: We aimed to investigate mucosal immunity related to forkhead box P3 ($FOXP3^+$) regulatory T (Treg) cells, T helper 17 (Th17) cells and cytokines in pediatric inflammatory bowel disease (IBD). Methods: Mucosal tissues from terminal ileum and colon and serum samples were collected from twelve children with IBD and seven control children. Immunohistochemical staining was done using anti-human FOXP3 and anti-$ROR{\gamma}t$ antibodies. Serum levels of cytokines were analyzed using a multiplex assay covering interleukin $(IL)-1{\beta}$, IL-4, IL-6, IL-10, IL-17A/F, IL-21, IL-22, IL-23, IL-25, IL-31, IL-33, interferon $(IFN)-{\gamma}$, soluble CD40L, and tumor necrosis factor-${\alpha}$. Results: $FOXP3^+$ Treg cells in the lamina propria (LP) of terminal ileum of patients with Crohn's disease were significantly (P < 0.05) higher than those in the healthy controls. $ROR{\gamma}t^+$ T cells of terminal ileum tended to be higher in Crohn's disease than those in the control. In the multiplex assay, serum concentrations (pg/mL) of IL-4 ($9.6{\pm}1.5$ vs. $12.7{\pm}3.0$), IL-21 ($14.9{\pm}1.5$ vs. $26.4{\pm}9.1$), IL-33 ($14.3{\pm}0.9$ vs. $19.1{\pm}5.3$), and $IFN-{\gamma}$ ($15.2{\pm}5.9$ vs. $50.2{\pm}42.4$) were significantly lower in Crohn's disease than those in the control group. However, serum concentration of IL-6 ($119.1{\pm}79.6$ vs. $52.9{\pm}39.1$) was higher in Crohn's disease than that in the control. Serum concentrations of IL-17A ($64.2{\pm}17.2$ vs. $28.3{\pm}10.0$) and IL-22 ($37.5{\pm}8.8$ vs. $27.2{\pm}3.7$) were significantly higher in ulcerative colitis than those in Crohn's disease. Conclusion: Mucosal immunity analysis showed increased $FOXP3^+$ T reg cells in the LP with Crohn's disease while Th17 cell polarizing and signature cytokines were decreased in the serum samples of Crohn's disease but increased in ulcerative colitis.
연구배경: 유육종증은 전신을 침범하는 만성 육아종성 질환으로 단핵구성 폐포염이 선행되어 육아종형성을 유도하며, 여러 종류의 cytokine들이 육아종성 염증과 그에 따른 섬유화의명태생리가전에 관여한다고 알려져 있다. 특히 최근에는 IL-6와 IL-8이 유육종증에서 광범위하게 염증반응을 진행시키는 것으로 보고되고 있고, 반면에 IL-10은 단핵구/대식세포와 T림프구의 기능을 억제하여 IL-1, IL-6, TNF-$\alpha$, IFN-$\gamma$ 등의 분비를 감소시키므로써 육아종성 염증반응을 억제한다고 보고되고 있다. 본 연구에서는 유육종증환자들의 폐포세척액내에서 IL-6, IL-8, IL-10의 농도를 측정하고 유육종증의 활성화를 나타내는 임상적 지표들과 비교해 봄으로써 이러한 lnterleukin들이 유육종증의 병태생리기전에 관여하는지 알아보고자 하였다. 대상 및 방법: 폐조직검사를 통해 조직학적으로 확진된 유육종증환자 14명과 건강한 정상대조군 6명을 대상으로 기관지폐포세척술을 시행하였다. 유육종증환자들은 중요 장기(심장, 중추신경, 후방 포도막등)의 침범여부, 고칼숨혈증, 심각한 뼈기능감소 및 질환의 진행여부에 따라 활동성(n=7)과 비활동성(n=7)으로 분류하였고 회수한 폐포세척액을 동일한 조건에서 10배로 농축한 후 ELISA방법을 이용하여 IL-6, IL-8, IL-10을 측정하여 이 Interleukin들의 상호관계와 유육종증의 활성도를 비교하였다. 결 과: 기관지폐포세척액내 IL-6농도는 특히 활동성 유육종증환자군에서 비활동성 유육종증환자군 및 정상대조군보다 유의하게 높았다(각각 p<0.05). IL-8 농도는 활동성 유육종증환자군에서 비활동성 유육종증환자군 및 정상대조군보다 높은 경향을 보였으나 통계적 유의성은 없었고 IL-10농도는 특히 활동성 유육종증환자군에서 정상대조군 및 비활동성 유육종증환자군보다 낮은 경향을 보였으나 통계적 유의성은 없었다. 활동성 유육종증환자군의 기관지폐포세척액내 IL-6농도는 기관지폐포세척액내 IL-8 농도 및 CD4/CD8 비(比)와 서로 의미 있는 상관관계를 보였다(p<0.05). 결 론: 활동성 유육종증환자군의 기관지폐포세척액내에서 IL-6농도가 비활동성 유육종증환자군과 정상대조군에서 보다 유의하게 높았고 이러한 IL-6는 기관지폐포세척액내 CD4/CD8 비(比) 및 IL-8농도와도 상관관계를 보여, 주로 IL-6이 유육종증의 진행과정과 활성화에 종요한 역할을 하고 있는 것으로 생각된다. 또한 세포성 면역반응을 억제할 수 있는 cytokine으로 생각되는 IL-10의 농도가 유육종증환자의 폐내에서 증가되지 않은 것도 이 질환의 발병이나 진행에 기여할 것으로 추정되며 앞으로 유육종증의 치료에 IL-6의 단일클론성 항체나 IL-10이 이용될 수도 있을 것으로 생각된다.
