  • The cycles of heavy metals, mercury, lead and cadmium, were investigated on in dynamic grassland ecosystems at a steady state in Zoysia japonica and Miscanthus sinensis in Mt. Kwanak, Korea. Estimates of decay constants of heavy metals based on experimental and methomatical model, were Hg 0.14, Pb 0.17 and Cd 0.41 of Z. japonica grassland, and Hg 0.33, Pb 0.13 and Cd 0.56 of M sinensis grassland. The durations of reaching half of initial amounts in Z. japonica and M. sinensis grassland, were Hg 4.95, Pb 4.07 and Cd 1.69 years, and Hg 2.10, Pb 5.33 and Cd 1.24 years respectively. Times needed for 99% decomposed were longer in Z. japonica than M. sinensis grassland. Decay velocity of constituents of surface soil layers were more rapidly in M. sinensis than Z. japonica grassland. Key words: Cycles of heavy metals, Mercury, Lead, Cadmium, Zoysia japonica. Effects of SPDJTK(SoPungDoJeokTangKami) and Concurrent Administration of AJ (Atopy cream, Jawoongo) Plus SPDJTK on Atopic Dermatitis-like Skin Lesions in NC/Nga Mouse Induced by BMAC (아토피양(樣) 피부염 NC/Nga 생쥐에서 소풍도적탕가미(消風導赤湯加味)와 아토피크림, 자운고(紫雲膏) 및 소풍도적탕가미(消風導赤湯加味)의 병용투여가 피부염에 미치는 영향)

  • Objectives The purpose of this study is to investigate the effect of SPDJTK(SoPungDoJeokTangKami) and concurrent administration of AJ(Atopy cream, Jawoongo)+SPDJTK on atopic dermatitis-like skin lesions by using in NC/Nga atopic dermatitis mouse induced by BMAC-induced mice. Methods Clinical skin score, hematology and Serum total IgE and IgG1 of NC/Nga atopic dermatitis mice were evaluated. Moreover, the cytokine level, total cell number, Immunohistochemical staining and Histological features of axillary lymph node(ALN), draining lymph node(DLN), peripheral blood mononuclear cells(PBMCs) and dorsal skin tissue were used in NC/Nga mice. Results Orally administrated SPDJTK with concurrent administration of SPDJTK and AJ decreased the clinical skin score, total cell number of WBC, eosinophils in blood, serum total IgE & IgG1, IL-5, IL-13, IFN-$\gamma$. Also, total cell number of ALN and dorsal skin tissue, absolute cell number of CD4+, CD8+, CD3+CD69+, CD3+CCR3+, CCR3+, CD4+CXCR5+ in ALN, absolute cell number of CD3+CCR3+, CCR3+ in DLN, granulocytes in PBMCs, activation cell number of CD3+CD69+, CCR3+, total cell number of CD3+ T cell in dorsal skin tissue were significantly decreased. Furthermore, thickness of epidermis, infiltrated inflammatory immune cell and mast cell in dermis, amount of Eotaxin2 mRNA, CCR3 mRNA in dorsal skin tissue, gene expression of IL-5, IL-13 mRNA in ALN, CD4+ Th cell in dorsal skin tissue and CCR3+ eosinophils in ALN were all significantly decreased. However, total number of DLN, absolute number of CD3e+ T cell and CD19+ B cell, absolute number of CD4+, number of Th cell in DLN and gene expression of foxp3 mRNA were significantly increased significantly. Conclusions Concurrent administration of SPDJTK and AJ on atopic dermatitis in NC/Nga atopic dermatitis mouse was very effective treatment for atopic dermatitis.

Influence of Feeding β-Cyclodextrin on Reducing the Content of Cholesterol in Pork (돼지고기의 콜레스테롤 함량 감소에 관한 β-Cyclodextrin의 급여효과)

  • The main objective of the present study was to determine the effect of dietary ${\beta}-cyclodextrin\;({\beta}CD)$ on reducing the cholesterol content of pork. Twelve swine of 50 kg body weight were randomly distributed into four groups of three heads each and fed experimental diets for 9 weeks until they reached a market weight of 110 kg. They were assigned to the four experimental diets containing 0% (control), 1.5%, 3.0% or 5.0% pure ${\beta}CD$. Daily feed intake, body weight gain and feed efficiency were not significantly different between any of the four group. The plasma total lipid, triacylglycerol and total cholesterol content of the swine in the three ${\beta}CD$-fed groups were significantly (p<0.05) decreased when compared to those in the control group, and were significantly (p<0.05) reduced by 21.80%,55.58% and 27.69%, respectively, in the swine fed on 5% ${\beta}CD$. The cholesterol content of pork belly (mg/100g) was significantly (p<0.05) decreased by 5.33 mg, 12.70 mg and 15.23 mg in the swine maintained on 1.5%, 3.0% and 5.0% ${\beta}CD$, respectively. The cholesterol content of pork belly, when expressed as the rate of reduced cholesterol, was significantly (p<0.05) decreased by 6.44%, 15.36% and 18.42% in groups of 1.5%, 3.0% and 5.0% ${\beta}CD$, respectively, when compared to that of the control group. These results suggest that dietary ${\beta}CD$ may be classified as dietary fiber which can modulate cholesterol metabolism in swine.

Performance of Column Type Bioreactor Packed with Immobilized Cyclodextrin Glucanotransferase for Cyclodextrin Production

  • Performance of column type bioreactor packed with immobilized cyclodextrin glucanotransferase (CGTase) on chitosan and Amberite IRA 900 was evaluated for cyclodextrin(CD) production. For CGTase immobilized on chitosan, the maximum CD conversion yield of 42% was achieved at the range of 88-168 units of immobilizied CGTase per gram of chitosan, retention time of 0.3 hr, and from 5.0% (w/v) of partially cyclized soluble starch. On the other hand, for CGTase immobilized on Amberite IRA 900, the maximum conversion yield of 40% was obtained at the range of 3.6-11.0 units of immobilized CGTase per gram of carrier and retention time of 1.2 hr from 5.0% of substrate. Above CD conversion yields are almost identical level with that can be obtained with soluble CGTase of 47%. The productivities of bioreactor packed with immobilized CGTase were 17.0g of CD/lㆍhr for amberite IRA 900 and 15.5g of CD/lㆍhr for chitosan. The partially cyclized starch with soluble CGTase were more suitable as substrate to achieve better CD conversion yield, and 5% (w/v) of partially cyclized soluble starch containing 10% (w/w) of CD was found to be most suitable to obtain maximum CD conversion yield.

Inhibitory Effects of Changchuldoin-tanggamibang on Collagen Induced Arthritis in DBA/1J Mouse (창출도인탕가미방(蒼朮桃仁湯加味方)이 DBA/1J 생쥐의 collagen 유발 관절염 억제에 미치는 영향)

  • Objectives: This study was carried out to find the effects of Changchuldoin-tanggamibang (hereinafter referred to CDIT) on the inhibition of arthritis induced by collagen on DBA/1J mouse. Methods: The experimental mice were divided into four groups: normal group (Nr), control group (CIA-CT), methotrexate group (CIA-MTX), and Changchuldoin-tanggamibang group (CIA-CDIT). Cytotoxicity, hepatotoxicity, arthritis index, value of immunocytes in draining lymph node and paw joint, and rheumatoid factor (IgG, IgM) in serum were measured in vivo. Results: 1. Cytotoxicity against hFCs was not shown in any concentration. 2. Hepatotoxicity was low in the CDIT-treated group compared with the MTX group. 3. The arthritis index decreased significantly. 4. In total cell counts of DLN and paw joint, the cells in DLN increased significantly while there was a significant decrease in paw joint. 5. In lymph nodes, CD19+, CD3+, CD4+, CD8+, CD3+/CD8+, CD3+/CD69+, CD4+/CD25+, CD3+/CD49b+, and CD4+/CD44+ cells increased significantly, while B220+/CD23+, and CD11c+/MHCII+ cells decreased significantly. 6. In joints, CD3+, CD4+, CD4+/CD25+, and CD11b+/Gr-1+ cells decreased significantly. 7. The level of IgG decreased and the level of IgM significantly decreased compared with the control. 8. Anti-collagen II in serum decreased compared with the control. 9. Around the joint of the CDIT group, infiltration of inflammation, synovial hyperplasia, invasion of cytokine, of cartilage, deposition of collagen and synovial injury decreased compared with the control in histopathologic observation (HE, MT staining). Conclusions: Comparison of the results for this study showed that CDIT had immunomodulatory effects. We expect that CDIT could be used as a effective drug for not only rheumatoid arthritis but also another auto-immune diseases. Therefore, we have to survey continuously, looking for effective substances and mechanisms in the future.

A Study on the Effect of Herbal-acupuncture with Phellodendri Cortex Solution on Collagen-induced Arthritis in DBA/1J Mice (황백약침이 콜라겐 유도 관절염 생쥐에 미치는 영향)

  • Objectives & Methods : The purpose of this study is to observe the effects of Phellodendri Cortex Herbal-acupuncture solution (PC-HAS) at Joksamni (ST36) on collagen II induced arthritis in DBA-1J mice. The author performed several experimental items to analyze arthritis evaluation, change of weight, spleen size and adhesion rate, change of cytokine level, IgG, IgM and anti-collagen II, chang of immunocyte count, histological change of CIA mouse joint. Results : 1. In the PC-HA group, arthritis index, the incidence of arthritis and joint edema were significantly decreased. 2. In the PC-HA group, the change of spleen size, spleen adhesion rate and the knee joint were significantly decreased. 3. The levels of $IL-1{\beta}$, IL-6 and INF- in serum of the CIA mouse were significantly decreased by PC-HA. 4. The levels of IgG, IgM and anti-collagen II in serum of the CIA mouse were significantly decreased by PC-HA. 5. In the CIA mouse spleen cell culture, the levels of IFN- , IFN- / IL-4, IL-10 were significantly decreased by PC-HA, but the level of IL-4 was significantly increased by PC-HA. 6. In the PC-HA group, the ratios of $CD3e^+$ to $CD45R^+$ cell, $CD4^+$ to $CD8^+$ cell and $CD4^+/CD25^+$ cell were similarly maintained as normal group in the CIA mouse spleen cell. 7. In the PC-HA group, $CD4^+CD25^+$ and $CD45R^+/CD69^+$ cell were significantly decreased in the lymph nodes. 8. In the PC-HA group, $CD3^+/CD69^+$ and $CD11b^+/Gr-1^+$ cell were significantly decreased in knee joint. 9. In histology, the cartilage destruction and synovial cell proliferation in the PC-HA group were similar with that of the normal group and the collagen fiber expressions in the PC-HA group were similar with that of the normal group. Conclusions : Form the result above, the results suggest that the PC-HA at ST36 has significant effect on collagen-induced arthritis, and can be put to practical use in the future rheumatoid arthritis clinic.

Effects of Astragali Radix Herbal-acupuncture at $ST_{36}$ (Jok-samni) on Collagen-induced Arthritis in Mice (황기약침이 Collagen으로 유발된 생쥐의 Arthritis에 미치는 영향)

  • Objectives : The aim of the present study was to observe the effect of Astragali Radix Herbal-Acupuncture Solution(AR-HAS) at $ST_{36}$(jok-samni, $Z\acute{u}s\bar{a}n$ Li) on collagen- II -induced arthritis(CIA) in mice. Methods : DBA/1J mice were immunized with bovine type II collagen(CII) on days 0 and 21 to induce arthritis. The mice were divided into 5 groups : normal group(no CIA), control group(CIA+no treatment), needle prick group(CIA+single prick with an injection needle), saline group(CIA+saline injection) and ARHA group(CIA+ R-HA treatment). The needle prick, saline injection, and AR-HA groups were injected on the right $ST_{36}$(jok-samni) of mice for 9 weeks, 3 times a week, beginning 4 weeks after the booster immunization. Results : 1. The incidence of arthritis, AI(arthritis index), and joint edema decreased in the AR-HA group. 2. Weight gain, hypertrophy of the spleen, adhesion of the tissues, and transformation of the joint were restrained in the AR-HA group. 3. The concentrations of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, $IFN-{\gamma}$ in CIA mouse serum and $IFN-{\gamma}$, IL-10 in the CIA mouse spleen cell culture decreased significantly at $ST_{36}$ in the AR-HA group. 4. Total cell counts increased significantly, and the ratio of $CD3e^+$ to $CD45R^+$, $CD4^+$ to $CD8^+$, and $CD4^+$ to $CD25^+$ cells decreased significantly at $ST_{36}$ in the CIA mouse spleen cell culture of the AR-HA group. 5. Total cell counts decreased significantly, and $CD4^+/CD25^+$ and $CD45R^+/CD69^+$ decreased significantly at $ST_{36}$ in the CIA mouse lymph nodes of the AR-HA group. 6. $CD3e^+/CD69^+$ and $CD11b^+/Gr-1^+$ cells decreased significantly at $ST_{36}$ in the CIA mouse joints of the AR-HA group. 7. The histological examination showed that cartilage destruction and synoviocyte proliferation decreased in the CIA mouse joints of the AR-HA group, and collagen fiber was expressed similar to that seen in the normal group. Conclusions : Our experiments show that at $ST_{36}$, an anti-inflammatory mechanism of AR-HA controls synovial cell proliferation and protects against cartilage destruction in rheumatoid arthritis.

  • Objectives : The purpose of this study is to observe the effects of Carthami Flos herbal-acupuncture (CF-HA) at Joksamni($ST_{36}$) on arthritis in mice induced by Collagen II. Methods : The author performed several experimental items, including arthritis evaluation, change in weight, spleen size and stenosis rate, change in cytokine level, IgG, IgM and anti-collagen II, change of immunocyte count and histological change of the CIA mouse joint. Conclusions are as follows: Results : 1. In the CF-HA, the arthritis index and rate and the incidence of arthritis were decreased as the experiment proceeded. 2. In the CF-HA, spleen swell and stenosis, joint edema and change were decreased. 3. In the CF-HA, the level of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ and $IFN-{\gamma}$ in blood serum were significantly decreased. 4. In the CF-HA, the level of IgG, IgM and anti-collagen II were decreased. 5. In the CF- HA, $IFN-{\gamma}$, $IFN-{\gamma}/IL-4$, IL-10 of the culture fluid was decreased. 6. In the CF-HA, the cell rate of $CD3e^+$ and $CD45R^+$, $CD4^+$ and $CD8^+$, $CD4^+/CD25^+$ in spleen was similar to the cell rate of the normal group. 7. In the CF-HA, the cell rate of $CD4^+/CD25^+$, $CD45R^+/CD69^+$ in a lymph node was decreased as in the normal group. 8. In the CF-HA, the cell rate of $CD3^+/CD69^+$, $CD11b^+/Gr-1^+$ in joints was decreased as in the normal group. 9. In the CF-HA, the cartilage destruction and the inflammation cell growth in the H&E stain were decreased. The collagen fiber in the M&T stain were less destructed, therefore the result was similar to the normal group. Conclusions : These results suggest that CH-HA at $ST_{36}$ has an effect in controlling immune reaction and suppressing inflammation in rheumatoid arthritis therefore, the continuous flow of the following study is expected.

Inhibitory Effects of Gwanjul9-bang on Collagen Induced Arthritis in DBA/1J Mouse (관절9호방이 생쥐의 Collagen II 유발 관절염에 미치는 영향)

  • This study was carried out to know the effects of Gwanjul9-bang (hereinafter reffered to GJ9) on the inhibition of arthritis induced by collagen on DBA/1J mouse. For this purpose, GJ9 was orally administered to mouse with arthritis induced by collagen II. Cytotoxicity, hepatotoxicity, arthritis index, value of immunocyte in draining lymph node and paw joint, rheumatoid factor (IgG, IgM) in serum were measured in vivo. The cytotoxicity against hFCs was not measured in any concentration. The hepatotoxicity was low in GJ9 treated group compared with MTX group. The arthritis index was decreased significantly. In total cell counts of DLN and paw joint, the cells in DLN increased significantly while there was significantly decrease in paw joint. In lymph nodes, $CD19^+$, $CD3^+$, $CD4^+$, $CD3^+CD69^+$, $CD8^+$, $CD4^+CD25^+$, $CD3^+CD49b^+$, $CD4^+CD44^+$, $CD3^+CD8^+$ cells increased significantly, $B220^+CD23^+$cells decreased significantly. In joints, $CD3^+$, $CD4^+$, $CD4^+CD25^+$, $CD11b^+Gr-1^+$ cells decreased significantly. The levels of IgG and IgM was significantly decreased compared with control. Anti-collagen II in serum was significantly decreased compared with control. The degree of arthritis induced damage of joint of GJ9 group is slight compared with control group in histopathologic observation (Hematoxylin & Eosin, Masson's Trichrome). Comparison of the results for this study showed that GJ9 had immunomodulatory effects. So we expect that GJ9 should be used as a effective drugs for not only rheumatoid arthritis but also another auto-immune disease. Therefore we have to survey continuously in looking for the effective substance and mechanism in the future.

The Experimental Study on the Suppression Effect of Asthma and Immune Response Improvement of Platycodi Radix Herbal-acupuncture (길경약침(桔梗藥鍼)의 천식억제(喘息抑制) 및 면역조절효과(免疫調節效果)에 대(對)한 실험적(實驗的) 연구(硏究))

  • Objectives : The aim of this study was to investigate the effect of Asthma-depression and Immunoregulation with PR-HAS(Herbal-acupuncture with Platycodi Radix infusion solution) injection at Joksamni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). The experimental group(OVA-PR-HA) wase treated with concentrations(1%) of PR-HAS at Joksamni(St36) for the later 8 weeks(3times/week). The second experimental group(OVA-Needle prick) was treated with Needle-Prick at Joksamni(St36) for the later 8 weeks(3times/week). Results : 1. The weight and total cells in the mice lung treated with PR-HA decreased significantly compared with that of control group. 2. Total leukocytes and eosinophils in BALF of the mice group treated with PR-HA decreased remarkably compared with those of control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with PR-HA decreased significantly compared with those of control group. 4. The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. 5. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The mRNA expression of ${\beta}-actin,\;TNF-{\alpha}$, IL-4, IL-5,IL-13 in the mice group treated with PR-HA with RT-PCR decreased significantly compared with those of control group. Conclusion : The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. The mRNA expression of ${\beta}-actin,\;TNF-{\alpha}$, IL-4, IL-5, IL-13 in the mice group treated with PR-HA with RT-PCR decreased significantly compared with those of control group. These result suggests that Platycodi Radix Herbal-acupuncture at Joksamni(St36) in C57BL/6mice may be an effictive part to OVA-induced asthma in C57BL/6 mice.
