Neuronal Phenotypes and Gene Expression Profiles of the Human Adipose Tissue-Derived Stromal Cells in the Neuronal Induction (신경 분화 유도한 인체 지방조직 유래 간질세포의 신경 표현형과 유전자 발현)

  • Purpose: Human adipose tissue-derived stromal cells(hADSCs) can be expanded in vitro and induced to differentiate into multiple mesenchymal cell types. In this study we have examined various neuronal phenotypes and gene expression profiles of the hADSCs in the neuronal induction. Methods: The hADSCs were isolated from human adipose tissue and they were characterized by the flow cytometry analysis using CD13, CD29, CD34, CD45, CD49d, CD90, CD105 and HLA-DR cell surface markers. We differentiated the hADSCs into the neuronal lineage by using chemical induction medium and observed the cells with contrast microscopy. The immunocytochemistry and western blotting were performed using the NSE, NeuN, Trk-A, Vimentin, N-CAM, S-100 and ${\beta}$-Tubulin III antibodies. Results: The hADSCs were positive for CD13($90.3{\pm}4%$), CD29($98.9{\pm}0.7%$), CD49d($13.6{\pm}6%$), CD90 ($99.4{\pm}0.1%$), CD105($96%{\pm}2.8%$) but negative for CD34, CD45 and HLA-DR. The untreated cultures of hADSCs predominately consisted of spindle shaped cells and a few large, flat cells. Three hours after the addition of induction medium, the hADSCs had changed morphology and adopted neuronal-like phenotypes. The result of immunocytochemistry and western blotting showed that NSE, NeuN, Trk-A, Vimentin, N-CAM, S-100 and ${\beta}$-Tubulin III were expressed. However, NSE, NeuN, Vimentin were weakly expressed in the control. Conclusion: Theses results indicate that hADSCs have the capabillity of differentiating into neuronal lineage in a specialized culture medium. hADSCs may be useful in the treatment of a wide variety of neurological disorders.

Flow cytometric immunophenotyping of canine adipose-derived mesenchymal stem cells (ADMSCs) and feline ADMSCs using anti-human antibodies

  • Various trials have been conducted to develop therapies for serious untreatable diseases. Among these, those using stem cells have shown great promise, and adipose-derived mesenchymal stem cells (ADMSCs) are easier to obtain than other types of stem cells. Prior to clinical trials, characterization of ADMSCs with monoclonal antibodies should be performed. However, it is difficult to use species-specific antibodies for veterinarians. This study was conducted to confirm the panel of human antibodies applicable for use in immunophenotypic characterization of canine adipose-derived stem cells and feline ADMSCs extracted from subcutaneous adipose tissue collected during ovariohysterectomy. For flow cytometric immunophenotyping, the third passages of canine ADMSC and feline ADMSC and human CD31, CD34, CD42, CD44, CD62 and CD133 antibodies were used. Of these, CD133 reacted with canine cells (3.74%) and feline cells (1.34%). CD133 is known as a marker related with more primitive stem cell phenotype than other CD series. Because this human CD133 was not a species-specific antibody, accurate percentages of immunoreactivity were not confirmed. Nevertheless, the results of this study confirmed human CD133 as a meaningful marker in canine and feline ADMSCs.

  • Luminescent characteristics of FLs were studied according to the structure of electrode for LCD backlight applications. The luminance and luminous efficiency of the FLs fabricated under same conditions were measured and evaluated as functions of magnitude of applied voltage and widths of external electrode. The luminance and luminous efficiency of CCFL at 12 V were 27600 cd/$m^2$ and 35.3 lm/w, respectively The luminance of EEFLS increased as the widths of external electrode increased, and the luminous efficiency of EEFLS showed to increase to 20 mm of electrode width) and to decrease at wider than 20 m of electrode widths. The luminance and luminous efficiency of EEFL with 20 mm of electrode widths were 21600 cd/$m^2$ and 26500 cd/$m^2$, 35.6 lm/w and 34.8 lm/w at 12 V, 14 V, respectively.

The Effects of a CD-ROM Educational Program for Sexual Violence Prevention on Sexual Knowledge of Middle School Students (성희롱.성폭력 예방교육 CD-ROM이 중학생의 성폭력예방 지식에 미치는 효과)

  • Purpose: This study analyzed the effects of a CD- ROM educational program for the prevention of sexual violence in relation to the sexual knowledge of middle school students. Method: A one group, pre-test-post-test design was used. The subjects of this study were 171 middle school students in Kangwon Province, Korea. The questionnaire included 34 items relating to overall sexual knowledge, and included questions on sexual violence. The collected data were analyzed using the mean, standard deviation and paired- t tests. Result: There were significant differences in the sexual knowledge by demographic characteristics between the pre- and post- tests. Of the 34 items, 20 showed significant differences in the sexual knowledge between the pre- and post- tests. Conclus ion: The overall level of sexual knowledge of the students increased after taking the CD- ROM educational program. Further testing will be required to determine the effects of the CD- ROM using a control group and random sampling.

Effect of Ultrathin Al2O3 Layer on TiO2 Surface in CdS/CdSe Co-Sensitized Quantum Dot Solar Cells

  • In order to enhance the photovoltaic property of the CdS/CdSe co-sensitized quantum dot sensitized solar cells (QDSSCs), the surface of nanoporous $TiO_2$ photoanode was modified by ultrathin $Al_2O_3$ layer before the deposition of quantum dots (QDs). The $Al_2O_3$ layer, dip-coated by 0.10 M Al precursor solution, exhibited the optimized performance in blocking the back-reaction of the photo-injected electrons from $TiO_2$ conduction band (CB) to polysulfide electrolyte. Transient photocurrent spectra revealed that the electron lifetime (${\tau}_e$) increased significantly by introducing the ultrathin $Al_2O_3$ layer on $TiO_2$ surface, whereas the electron diffusion coefficient ($D_e$) was not varied. As a result, the $V_{oc}$ increased from 0.487 to 0.545 V, without appreciable change in short circuit current ($J_{sc}$), thus inducing the enhancement of photovoltaic conversion efficiency (${\eta}$) from 3.01% to 3.38%.

Forced Expression of HoxB4 Enhances Hematopoietic Differentiation by Human Embryonic Stem Cells

  • HoxB4 has been shown to enhance hematopoietic engraftment by hematopoietic stem cells (HSC) from differentiating mouse embryonic stem cell (mESC) cultures. Here we examined the effect of ectopic expression of HoxB4 in differentiated human embryonic stem cells (hESCs). Stable HoxB4-expressing hESCs were established by lentiviral transduction, and the forced expression of HoxB4 did not affect stem cell features. HoxB4-expressing hESC-derived CD34+ cells generated higher numbers of erythroid and blast-like colonies than controls. The number of CD34+ cells increased but CD45+ and KDR+ cell numbers were not significantly affected. When the hESC derived CD34+ cells were transplanted into $NOD/SCID{\beta}2m-/-$ mice, the ectopic expression of HoxB4 did not alter their repopulating capacity. Our findings show that overexpression of HoxB4 in differentiating hESCs increases hematopoietic colony formation and hematopoietic cell formation in vitro, but does not affect in vivo repopulation in adult mice hosts.

The Effect of $Cd^{2+}$ on Stomatal Apertures of Epidermal Strips in Commelina communis L. ($Cd^{2+}$이 닭의장풀의 기공개폐에 미치는 영향)

  • The effect of $Cd^{2+}$ on stomatal apertures of epidermal strips and intact leaves in Commelina communis L. was investigated. Cadmium stimulated stomatal opening. The stomata, treated with 100 $\mu$M $Cd^{2+}$ opened to a degree of about 8.38 fm, but the stomata, treated with no cadmium opened to 3.74 ${\mu}{\textrm}{m}$. In order to show that there was no mechanical or osmotic impediment preventing the stomata in the epidermal strips, salicylic acid (SA) and abscisic acid (ABA) were used. The treatment of 100 $\mu$M SA and 1 $\mu$M ABA inhibited 14% and 28% of stomatal opening, respectively. Other heavy metals such as $Al^{3+} and Ag^{2+}$ were also used to investigate the effect of the stomatal apertures. The treatment of $Al^{3+} and Ag^{2+}$ also stimulated 19% and 41% of stomatal opening. To understand how cadmium open stomata, the effect of cadmium on the K+ influx into the epidermal strips was investigated. $Cd^{2+}$, SA, ABA inhibited 98%, 28%, 34% of K+ uptake respectively. 3-weeks old Commelina was transferred to and grown in Hoagland solution (0,5 mM $Cd^{2+}$, 10 mM $Cd^{2+}$) for 4 days and stomatal conductance were measured. The treatment of 5 mM $Cd^{2+}$ and 10 mM $Cd^{2+}$ showed about 51% and 70% inhibition of stomatal conductance, respectively Therefore, it could be concluded that stomata in epidermal strips and intact leaves behaved differently and cadmium- stimulated stomatal opening was due to the result of cadmium uptake into the epidermal strips instead of K+. Correlation of Contrast-Enhanced Ultrasonographic Features with Microvessel Density in Papillary Thyroid Carcinomas

  • Background: The purpose of this study was to investigate the correlation of contrast-enhanced ultrasonographic (CEUS) features with microvessel density (MVD) in papillary thyroid carcinomas (PTCs). Materials and Methods: Contrast-enhanced ultrasonography (CEUS) was performed in 62 patients (17 men and 45 women) with PTC. Tomtec software was applied to analyze the time intensity curve of CEUS. Immunohistochemistry was performed to evaluate the level of MVD in papillary thyroid carcinoma. Then the relationship between quantitative feature and the level of MVD was analyzed using SPSS 16.0 software. Results: The mean peak intensity of PTC tissues was lower than that of peripheral thyroid parenchyma ($61.9{\pm}11.8%$ vs 100%, p<0.05). The MVDs of CD34 and CD31 antibodies staining were $38.0{\pm}6.1$ and $37.9{\pm}5.1$ respectively in 62 PTC samples. A significantly positive correlation was observed between peak intensity and MVD in PTC tissues ($P_{CD34}$<0.01, $r_{CD34}$=0.838, $P_{CD31}$<0.01, $r_{CD31}$=0.837). Conclusions: The peak intensity in CEUS could reflect the MVD in PTC tissues. Therefore, quantification of CEUS seems to be helpful for assessment of MVD in PTC tissues.

A Study of Peripheral Blood Stem Cell Collection and Bone Marrow Engraftment after Peripheral Blood Stem Cell Transplantation (말초혈액 조혈모세포 채혈 및 이식 후 생착에 관한 연구)

  • Peripheral bood stem cell collection (PBSCC), including peripheral blood stem cell transplantation (PBSCT), has been utilized worldwide as a very beneficial treatment method instead of allogenic Bone Marrow Transplantation (BMT) because it has many advantages such as rapid bone marrow engraftment and hematopoietic recovery, easy and safe accessibility and lower risk of rejection compared with allogenic BMT. In order to identify most the observable parameter in PBSCC, we analyzed various hematological parameters before and after PBSCC, and evaluated the correlation between the time of bone marrow engraftment and the number of CD34+ cells. Thirteen patients, who underwent 54 PBSCCs from January, 2003 to August, 2004 at Chungnam National University Hospital due to various systemic neoplasms, were analyzed in aspects of various hematological parameters including CD34+ cells using by Flow Cytometry (FCM). PBSCC harvests are described below: Mononuclear cells (MNC) $2.3{\pm}1.4{\times}10^8/kg$ and CD34+ cells $0.63{\pm}0.35{\times}10^6/kg$ on average, respectively. There was a statistical significance in Hb and Hct before and after PBSCC, but not in WBC and platelet counts. The period to reach the hematological bone marrow engraftment was 13.4(10~21) days and 19.5(11~38) days according to the criteria of absolute neutrophile counts (ANC) ${\geq}500/uL$ and platelet counts ${\geq}50,000/{\mu}L$ in peripheral blood, respectively. There was a significant correlation between the numbers of CD34+ cell and ANC (p<0.05), and a borderline significance between MNC and ANC (p=0.051). We found that a group of patients, who were infused with CD34+ cells more than $3.5{\times}10^6/kg$, reached more rapidly the period of bone marrow engraftment in platelet counts (p=0.040). This present study suggested that Hb and Hct were the most useful parameters and should be closely monitored before and after PBSCC, that a PBSCT with the dosage of more than $3.5{\times}10^6/kg$ of CD34+ cells was needed to perform successful bone marrow engraftment, and additionally that platelet counts could be more useful in indicating bone marrow engraftment than ANC.

Study on the Inclusion Behavior of Sulfobutylether-β-Cyclodextrin with Perphenazine by Flow Injection Chemiluminescence

  • The inclusion behavior of sulfobutylether-${\beta}$-cyclodextrin (SBE-${\beta}$-CD) with perphenazine (PPH) was first studied by flow injection (FI)-chemiluminescence (CL) analysis with proposed $lg[(I_0-I_s)/I_s]=lgK_{P-CD}+nlg[C_{PPH}]$ model and molecular docking. Results showed that a 1:1 complex of SBE-${\beta}$-CD/PPH could online form, with the formation constant $K_{P-CD}$ of $2.57{\times}10^7Lmol^{-1}$ at 298 K. The thermodynamic parameters showed that the inclusion behavior of SBE-${\beta}$-CD/PPH was a spontaneous process by hydrophobic interaction. The molecular docking results revealed PPH entered into the larger cavity of SBE-${\beta}$-CD with two hydrogen bonds. Based on the linear relationship of the decrement of luminol/SBE-${\beta}$-CD/PPH CL intensity against the logarithm of PPH concentration ranging from 0.03 to 30.0 ng $mL^{-1}$, the present FI-CL analysis using luminol/SBE-${\beta}$-CD/PPH system was successfully applied to PPH determination in biological fluids and tablets with recoveries from 94.5 to 105.6% and RSDs less than 2.6% (n = 5).
