A Comparative Study of Hataedock versus Probiotics on Immunomodulating Effect in Intestinal Mucosa (황련감초 하태독법과 프로바이오틱스의 대장점막 내 면역조절 효과 비교연구)

  • Objectives Hataedock is a treatment that dispels toxic heat and meconium which has been accumulated to the fetus from a pregnant mother via orally administering herbal extracts to a newborn baby. This study was conducted to compare the efficacy of Hataedock, with using the extract of Coptis japonica & Glycyrrhiza uralensis, to the early administration of probiotics for immunomodulation in the intestinal mucosa. Methods NC/Nga mice were divided into three groups; Control group (no treatment), CGT group (3-week-old mice given the extract of Coptis japonica & Glycyrrhiza uralensis), and MBT group (3-week-old mice given a Bifidobacterium). After 2 weeks, the intestinal mucosa tissues of each group of mice were observed. Immunohistochemical staining for IL-4, IL-13, CD40, $Fc{\varepsilon}RI$, $p-I{\kappa}B$, EGF, and VEGF in the intestinal mucosa was performed. Results CGT group showed 65% decrease in IL-4, 67% decrease in IL-13, 58% decrease in CD40, 72% decrease in $Fc{\varepsilon}RI$, 76% decrease in $p-I{\kappa}B$, 41% increase in EGF and 100% increase in VEGF compared to the control group. MBT group also showed 50% decrease in IL-4, 63% decrease in IL-13, 33% decrease in CD40, 53% decrease in $Fc{\varepsilon}RI$, 46% decrease in $p-I{\kappa}B$, 23% increase in EGF and 151% increase in VEGF compared to the control group. Conclusions These results suggest that both Hataedock, with using the extract of Coptis japonica & Glycyrrhiza uralensis, and early administration of probiotics were effective in regulating Th2, relieving inflammation and developing intestinal mucosal tissues. Hataedock with extract of Coptis japonica & Glycyrrhiza uralensis may be more effective for immunomodulation in intestinal mucosa than probiotics.

Distribution of inorganic metals in blood of adults at urban area in Korea

  • The objective of this study was to describe the distribution of metals concentrations in blood of adults who were not occupationally exposed in Korea. The blood samples were obtained between February and August 2001 from volunteer adults in urban area of Korea. 66 male participants were 46 (20-75) years of age and 74 female were 40 (20-69) years of age. The levels of metals in blood were observed the log-normal distribution, and we calculated geometric mean (GM) and geometric standard deviation (GSD). The GM levels of metals in blood of the men were 65.88$\mu\textrm{g}$/$\ell$, 1.01$\mu\textrm{g}$/$\ell$, 0.23$\mu\textrm{g}$/$\ell$ and 0.15$\mu\textrm{g}$/$\ell$, for Pb, Cr, Ni and Cd, respectively. The GM levels of the women were 58.49$\mu\textrm{g}$/$\ell$, 1.66$\mu\textrm{g}$/$\ell$, 0.30$\mu\textrm{g}$/$\ell$ and 0.10$\mu\textrm{g}$/$\ell$, for Pb, Cr, Ni and Cd, respectively. The levels of Pb-B and Cd-B were significantly higher non-smoker than smoker, whereas those of Cr-B and Ni-B were not different by smoking habit.

Bioremediation of Heavy Metals from the Land Application of Industrial Sewage Sludge with Minari (Oenanthe stolonifer DC.) Plant

  • Laboratory experiments for the removal efficiency of heavy metals in land application of sludge, the accumulation and translocation of heavy metals in x plants after transplanting, and the responses of Minari growth with different ratio of land application of sludge were conducted to determine the potential ability of bioremediation with Minari plants. The removal rate and translocation of copper. zinc. lead. and cadmium in soil and plants were compared after transplanting the Minari plants to soil treated with different ratio of sludge. The removal efficiency of heavy metals in soil incorporated with sludge was different with application ratio, but increased with growing periods of Minari plants. The removal efficiency of Cu, Zn, Pb, and Cd ranged from 67 to 74% from 51% to 63%, from 37% to 71%. and from 15% to 25% after 45 days of transplanting. respectively. The amount removed the copper value. 65.9 mg/kg, observed to be highest in soil incorporated 3% sludge after 45 days. The translocation of Cu. Zn. Pb. and Cd from shoots to roots ranged from 18 to 53%, from 17 to 32%, from 14 to 49%, and from 23 to 38% over growing periods. respectively. In plant responses it appeared to be inhibited the plant growth in the treatment compared with the control at early stage of growth. However, the fresh weights of Minari plant increased from 12.5 to 62.5% in the sludge application after 45 days relative to the control. Therefore the Minari might play a useful role in bioremediation of Cu, Zn, Pb, and Cd in the land application of sludge.

  • Anaplasma phagocytophilum major surface protein-2 (Msp2 or p44) is the immunodominant outer membrane protein of the bacterium. Recently, we disclosed that Msp2 was an A. phagocytophilum adhesin for binding to host neutrophils and HL-60 cells, probably mediated by attachment to platelet selectin glycoprotein ligand-1 (PSGL-1). In this study, we further elucidated that Msp2 bound to PSGL-1/FucT IV-transfected BJAB but not nontransfected BJAB cells. Binding of recombinant Msp2 or cell (lee bacteria to the surface of PSGL-1/FucT IV-transfected BJAB cells was significantly higher than to nontransfected BJAB cells (p<0.01 and p<0.01). Also, Msp2 monoclonal antibody and soluble recombinant Msp2 as antagonist led to concentration-dependent reductions in A. phagocytophilum adhesln (p<0.05 and p<0.01) to transfected BJAB cells. Thus, we conclude that Msp2 of. A. phagocytophilum acts as an adhesin by which the bacterium binds to PSGL-1 on host neutrophils and myeloid cells.

The Removal Rates of the Constituents of Litters in the Littoral Grassland Ecosystems in the Lake Paldangho Ⅶ.Cadmium and Lead (팔당호 연안대 초지생태계에서 낙엽 구성성분의 유실률 Ⅶ.카드뮴과 납)

  • This study was carried out to investigate the removal rates of Cd and Pb of the litters in the Phragmites communis. Miscanthus sacchariflorus, Typha angustata, Scirpus tabernaemontani grassland aquatic ecosystem in the lake Paldangho. The annual production of Cd and Pb were 0.005g /$m^2$ , 0.21g /$m^2$in P. commumis, 0.004g /$m^2$, O.08g /$m^2$ in M. sacchariflorus, 0.023g /$m^2$, 0.42g /$m^2$ in T. angustata and 0.020g /$m^2$, 0.23g /$m^2$ in S. tabernaemontani respectively. The removal rates of Cd and Pb of the litters were 0.83, 0.85 in P. communis. 0.36, 0.54 in M. sacchariflorus, 0.61, 0.51 in T. angustata and 0.76, 0.71 in S. tabernaemontani, respectively. The times required to decay 50, 95, 99 percent of the steady state level and turnover values of cadmium on the grassland floor were 0.83, 3,60, 6.00 years in P. communis. 1.90, 8.24, 13.74 years in M.sacchariflorus, 1.15, 4.96, 8.27 years in T. angustata and 0.91, 3.95, 6.58 years in S. tabernaemontani The times required to decay 50, 95, 99 percent of the steady state level and turnover values of lead on the grassland floor were 0.81, 3.51, 5.86 years in P. communis. 1.28, 5.56, 9.26 years in M. sacchariflorus, 1.37, 5.94, 9.90 years in T. angustata and 0.97, 4.21, 7.02 years in S. tabernaemontani. The Preservice Teachers등 Understanding of the International System of Units (국제단위제도에 대한 초등학교 예비 교사들의 이해)

  • This paper aims to examine how well university students, who are going to be primary school teachers, understand the International System of Units (SI), focusing on seven basic units such as 'm', '㎏', '㏖', 'A', 'K', 's', 'cd'. This study specifically investigates whether the students know the seven units and understand their uses and how they read and learn them. The subjects were 1030 students from the University of Education in Jinju, Geongsangnamdo. Data was collected through a questionnaire which was designed by this researcher and checked by an authority, and the frequency and percentage of reponses to each question were obtained and analysed. Findings show all the students knew very well that 'm' and '㎏' are included in the seven units, compared to the others which low percentage of the students considered as the elements. In terms of understanding of use, the units of the length 'm', the mass '㎏', and the time 's' are well understood, presumably, because they are often used in ordinary life and school, while the amount of substance '㏖', the electric current 'A', the thermodynamic temperature 'K', and the luminous intensity 'cd' are not as well understood. It is probably because 'A', 'cd', and 'K' are hardly used in everyday life. With respect to reading the units, the subjects read 'm', '㎏', '㏖', and's' much better than the others. As for the source of learning them, most students answered they learned them in schools, which implies school education is very important. From these results it is concluded that school education should be accompanied with use in everyday life for understanding and using the units. SI Base Units understanding was investigated by an academic year. According to this investigation, generally the right answer rate differed 4<2<3<1 by an academic year in order. For the Senior, Sophomore appeared the right answer rate more high. because, they learned before in deepening or elementary course on chemistry and physics related with SI Base Units. On the other hand, for the junior answered low rate by this reason that they couldn't remember about SI Base Units that learned before. For the Freshman appeared low rate by this reason that they were not learned or graduated from the department of liberal art in high school.

A Study of the Inverter Optimization Design for EEFL BLU (EEFL BLU 구동용 인버터 최적화 설계에 대한 연구)

  • EEFL(External Electrode Fluorescent Lamp) for LCD displays are analyzed on electrical and optical characteristics by various electrode length. The electrodes of EEFL are coated at the outside of the glass tube. Brightness and efficiency of the EEFL are affected on lamp impedance characteristic. So, the experimental models are proposed for analysis and measurements of the brightness and efficiency of the lamps according to the electrode length. The sample of LCD backlight unit is used for these experiments, EEFL arrayed BLU of 32' for commercial TV display. The parameters of the experiments were quantised for simple result reading as the length of electrodes as 15, 23 and 30 mm. The inverter was designed and manufactured in the laboratory as the Full-Bridge switching inverter. The feature of the output were measured on voltages about 1000 - 1400V at the currents of 11 - 29 mA and the brightness $15,000\;cd/m^2-40,000\;cd/m^2$. The experiments have shown that the brightness are increased by increasing of the electrode lengths which have the lamp currents increased. But at an certain conditions, the brightness and efficiency were decreased because of unmatched between the inverter output and lamp impedance. The optimum applications of the EEFL BLU of 32' in this experiments have been shown to choose the parameters for driving frequencies of 100 kHz - 150 kHz, the brightness of $18,000-19,000\;cd/m^2$ and efficiencies of 40 - 45 lm/w.

Tumour-Derived Reg3A Educates Dendritic Cells to Promote Pancreatic Cancer Progression

  • As a pancreatic inflammatory marker, regenerating islet-derived protein 3A (Reg3A) plays a key role in inflammation-associated pancreatic carcinogenesis by promoting cell proliferation, inhibiting apoptosis, and regulating cancer cell migration and invasion. This study aimed to reveal a novel immuno-regulatory mechanism by which Reg3A modulates tumour-promoting responses during pancreatic cancer (PC) progression. In an in vitro Transwell system that allowed the direct co-culture of human peripheral blood-derived dendritic cells (DCs) and Reg3A-overexpressing/ silenced human PC cells, PC cell-derived Reg3A was found to downregulate CD80, CD83 and CD86 expression on educated DCs, increase DC endocytic function, inhibit DC-induced T lymphocyte proliferation, reduce IL-12p70 production, and enhance IL-23 production by DCs. The positive effect of tumour-derived Reg3A-educated human DCs on PC progression was demonstrated in vivo by intraperitoneally transferring them into PC-implanted severe combined immunodeficiency (SCID) mice reconstituted with human T cells. A Reg3A-JAK2/STAT3 positive feedback loop was identified in DCs educated with Reg3A. In conclusion, as a tumour-derived factor, Reg3A acted to block the differentiation and maturation of the most important antigen-presenting cells, DCs, causing them to limit their potential anti-tumour responses, thus facilitating PC escape and progression.

Effect of black chokeberry on skeletal muscle damage and neuronal cell death

  • [Purpose] Numerous epidemiological studies have shown that it is possible to prescribe exercise for neurodegenerative disease, such as Alzheimer's disease and Parkinson's disease. However, despite the availability of diverse scientific knowledge, the effects of exercise in this regard are still unclear. Therefore, this study attempted to investigate a substance, such as black chokeberry (Aronia melanocapa L.) that could improve the ability of the treatment and enhance the benefits of exercising in neurodegenerative diseases. [Methods] The cell viability was tested with 2,3-bis[2-methyloxy-4-nitro-5-sulfophenyl]-2H-tetrazolim-5-carboxanilide and the cells were stained with ethidium homodimer-1 solution. The mRNA expression levels were evaluated by microarray. The active compounds of black chokeberry ethanolic extract (BCE) were analyzed by gas chromatography. The chemical shift analysis in the brain was performed using magnetic resonance spectroscopy. [Results] BCE treatment decreased hydrogen peroxide-induced L6 cell death and beta amyloid induced primary neuronal cell death. Furthermore, BCE treatment significantly reduced the mRNA levels of the inflammatory factors, such as IL-1α, Cxcl13, IL36rn, Itgb2, Epha2, Slamf8, Itgb6, Kdm6b, Acvr1, Cd6, Adora3, Cd27, Gata3, Tnfrsf25, Cd40lg, Clec10a, and Slc11a1, in the primary neuronal cells. Next, we identified 16 active compounds from BCE, including D-mannitol. In vivo, BCE (administered orally at a dosage of 50 mg/kg) significantly regulated chemical shift in the brain. [Conclusion] Our findings suggest that BCE can serve as a candidate for neurodegenerative disease therapy owing to its cyto-protective and anti-inflammatory effects. Therefore, BCE treatment is expected to prevent damage to the muscles and neurons of the athletes who continue high intensity exercise. In future studies, it would be necessary to elucidate the effects of combined BCE intake and exercise.

Effect of enzymolytic soybean meal supplementation on performance, nitrogen excretion, serum biochemical parameters and intestinal morphology in broilers fed low-protein diets

  • Objective: The objective of this study was to investigate the effect of supplementation with enzymolytic soybean meal (ESBM) on broilers fed low crude protein (CP) diets. Methods: A total of 360 one-day-old broilers were randomly assigned to six treatments with 6 replicates per treatment and 10 chicks per replicate for a period of 42 days. Chicks were fed a basal standard high-CP diet as a positive control (PC), a low-CP diet (reducing 10 g/kg CP from the PC) as a negative control (NC), or an NC + 0.5%, 1.0%, 1.5%, or 2.0% ESBM diet. Results: Compared to chicks fed the PC, chicks fed the NC had a decreased body weight gain (BWG, p<0.05) from 1 to 42 days, but supplementation with 2.0% ESBM restored BWG (p<0.05) and even linearly improved the feed conversion rate (FCR, p<0.05). Digestibility of CP and ether extract was increased (p<0.05) in chicks fed a 1.0% ESBM diet compared to the PC. With increasing levels of ESBM, nitrogen (N) excretion decreased (p<0.05). The addition of ESBM to the diet did not affect (p>0.05) serum concentrations of total protein, albumin and total cholesterol but led to a descending trend in triglycerides and an ascending trend in calcium and urea N at 42 days (p<0.10). There were no differences (p>0.05) in villus height (VH), crypt depth (CD), and VH/CD (V/C) of the duodenum and jejunum between the PC and NC at both 21 days and 42 days, while increasing dietary ESBM levels linearly (p<0.05) decreased CD and increased V/C of the duodenum and jejunum at both 21 days and 42 days. Conclusion: The findings indicated that ESBM could be used in broiler low-CP diets to improve production performance, decrease N excretion, and enhance intestinal health.
