Immuno-regulatory Effect of Lepidii Semen Herbal-acupuncture at $Joksamni(ST_{36})$ on OVA- induced Asthma in Mice (정력자약침의 천식억제(喘息抑制) 및 면역조절효과(免疫調癤效果)에 대(對)한 실험적(實驗的) 연구(硏究))

  • Objectives : The aim of this study was to investigate the asthma-suppressive and immuno-regulatory effect of Lepidii Semen Herbal-acupu ncture(LS-HA) at $Joksanmi(ST_{36})$ on ovalbumin(OVA)-induced asthma in mice. Methods : C57BL/6 mice out of all the experimental groups, except the Normal group and the LS-HA group, were sensitized and challenged with OVA. The mice in the LS-HA group and the OVA-LS-HA group were treated with LS-HA(1%) at $Joksanmi(ST_{36})$. The mice in the OVA-Saline group were injected with saline at $Joksanmi(ST_{36})$. The mice in the OVA-Needle-Prick group were treated with a single prick with an injection needle at $Joksanmi(ST_{36})$. LS-HA, saline injection and needle prick were administered for 8 weeks, three times a week. Results : The lung weight and total cells in lung of the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. Total leukocytes and eosinophils in BALF of the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. The collagen accumulation in the lung sections of the OVA-LS-HA group decreased significantly compared with that of the OVA-control group. The concentrations of IL-4, IL-5, IL-13, IgE in BALF and serum of the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. The numbers of $Gr-1^+/CD11b^+$, $CCR3^+$, $CD3e^+$, $CD19^+$, $CD3e^+/CD69^+$cells in the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. The mRNA expressions of $TNF-{\alpha}$, IL-5, IL-4 and IL-13 in lung of the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. Conclusions: These results suggest that LS-HA at $Joksamni(ST_{36})$ is considered to be effective in treating asthma and to be put to practical use in the future asthma clinic.

Effect of Qi Tonifying herbs, Ginseng, Astragali, Atractylodis, Glycyrrhizae on Mouse Cytokine Secretion (보기약물(補氣藥物)인 인삼(人參), 황기(黃?), 백출(白朮), 감초(甘草)의 물 추출액이 생쥐 면역세포의 Cytokine분비에 미치는 영향)

  • In Oriental medicine the primordial Qi and the defensive Qi are considered as important for immunity. Therefore it is anticipated that the improvement of the primordial Qi and the defensive Qi can enhance the ability of immune cells. This experiment was conducted to investigate how Ginseng Radix, Astragali Radix, Atractylodis Rhizoma Alba, Glycyrrhizae Radix, representative of Qi tonifying herbs, affect the immune system in terms of controlling and balancing immune cells. Using the MTS assay, increased proliferations were observed from herbal treated cells, among which Gins-eng showed the highest proliferation. When splenocytes were activated with anti-CD3 plus herbal extracts, levels of IFN-g and IL-4 were increased but those of IL-2 showed little change compared with the control cells. Levels of IL-2, IFN-g and IL-4 were increased in purified CD4 T cells when activated with anti-CD3/anti-CD28 but at $100\;{\mu}g/ml$ of Astragali and Atractylodis, levels of IL-2 were decreased by 11% and 42%, respectively and those of IFN-g were decreased by 55% and 12%, respectively. Under Th1/Th2 polarizing conditions, levels of IFN-g in Th1 cells treated with herbal extracts were all decreased but when it comes to IL-4, its levels were increased in Ginseng and Glycyrrhizae treated cells but decreased in Astragali and Atractylodis treated cells. Taken together, the data show that compared with other qi tonifying herbs, Ginseng and Glycyrrhizae have a tendency to favor Th2 cell differentiation in vitro.

Study on Transfer Rate of Hazardous Materials in Herbal Medicine Prescriptions (한방처방 전탕 단계별 위해물질의 이행률에 대한 연구)

  • Objective: To compare the contents and transfer rate of hazardous substances in crude, washing solution, crude after washing, decoction and remnant after boiling. Methods: The heavy metal contents of each step were measured by inductively coupled plasma mass spectrometer (ICP-MS) and mercury analyzer (SP-3DS). In order to analyze pesticides in each sample we used simultaneous multi-residue analysis of pesticides by GC/ECD, which was followed by GC/MSD analysis to confirm the identity of the detected pesticide in each sample. In addition, the contents of sulfur dioxide ($SO_2$) were performed by Monier-Williams distillation method. Results: 1. Contents (mg/kg) of heavy metals were not detected in decoctions of any tested herbal medicine prescriptions. 2. Transfer rates (%) of heavy metals from crude to remnant were as follows: Yijin-tang (As: 46.9, Cd: 50.0 and Pb: 100.0), Oryung-san (As: 80.0, Cd: 100.0 and Pb: 73.8), Hwangryunhaedok-tang (As: 88.9, Cd: 71.4 and Pb: 92.7), Bangpungtongseong-san (As: 100.0, Cd: 17.3 and Pb: 56.1), Oyaksungi-san (As: 47.4, Cd: 175.0 and Pb: 142.4). 3. Contents (mg/kg) of residual pesticides were not detected in any samples. 4. Transfer rate (%) of sulfur dioxide ($SO_2$) from crude to remnant in all samples were as follows: Yijin-tang (25.0), Oryung-san (166.7), Hwangryunhaedok-tang (50.0), Bangpungtongseong-san (181.8), Oyaksungi-san (50.0). Conclusion: Our results showed that the boiled herbal medicine prescriptions which we take are safe from the hazardous substances.

Expression of genes related to lipid transport in meat-type ducks divergent for low or high residual feed intake

  • Objective: This study examined the effects of divergence in residual feed intake (RFI) on expression profiles of key genes related to lipid transport in the liver and duodenal epithelium and their associations with feed efficiency traits in meat-type ducks. Methods: A total of 1,000 male ducks with similar body weight (1,042.1±87.2 g) were used in this study, and their individual RFI was calculated from 21 to 42 d of age. Finally, the 10 highest RFI (HRFI) and 10 lowest RFI (LRFI) ducks were chosen for examining the expression of key genes related to lipid transport in the liver and duodenal epithelium using quantitative polymerase chain reaction. Results: In the liver, expression levels of albumin (ALB), CD36 molecule (CD36), fatty acid hydroxylase domain containing 2 (FAXDC2), and choline kinase alpha (CHKA) were significantly higher in LRFI ducks than in HRFI ducks (p<0.01); negative correlations (p<0.05) between expression levels of ALB, CD36, FAXDC2, and CHKA and RFI were detected in the liver. Additionally, ALB expression was strongly positively correlated (p<0.05) with CD36, FAXDC2, CHKA, and apolipoprotein H (APOH) expression in the liver. In duodenal epithelium, we found that mRNA levels of ALB, CD36, FAXDC2, and APOH were significantly higher in LRFI ducks than in HRFI ducks (p<0.01); RFI was strongly negatively correlated (p<0.05) with ALB, FAXDC2, and APOH expression, while ALB expression was strongly positively correlated with APOH expression (p<0.01) in duodenal epithelium. Furthermore, expression levels of both ALB and FAXDC2 genes were significantly associated with feed conversion ratio and RFI in both liver and duodenal epithelium (p<0.05). Conclusion: Our findings therefore suggest that ALB and FAXDC2 genes might be used as potential gene markers designed to improve feed efficiency in future meat-type duck breeding programs.

Effect of Cheongyeoltang (CYT) using Bioconversion on Atopic Dermatitis (청열탕(淸熱湯)의 생물전환을 통한 항아토피피부염 효능(效能) 연구(硏究))

  • In order to improve efficacy of oriental medicines and to study the application of fermented oriental medicine in clinicals, the efficacy of CYT and CYTBH on atopic dermatitis were evaluated. The results and conclusions are as follows. CYT and CYTBH significantly improved the atopic dermatitis symptoms in NC/Nga mice by naked eye evaluation and significantly decreased clinical index in both groups. CYT and CYTBH both decreased the cell numbers of CD3+, CD11b+Gr-1+ cells in dorsal skin. Of the cells, CYT significantly decreased CD11b+Gr-1+ cells whereas CYTBH significantly decreased all immune cells. CYT and CYTBH both decreased the production rate of IL-4 and IFN-${\gamma}$ activated by CD3/CD28. In the case of CYTBH, significant decrease in all cases was observed. CYT and CYTBH decreased the production rate of IL-5, IL-13 and IL-17 in serum. Significant decrease of IL-5 in the case of CYT and IL-5 and IL-13 in the case of CYTBH were observed. CYT and CYTBH significantly decreased transcription of IL-5 mRNA and IL-13 mRNA in skin. Significant decrease in IgG1 and IgE immunoglobulins in serum were oberved in both groups. Significant decrease was only observed in the case of CYTBH. Both CYT and CYTBH significantly decreased the secretion of histamine. Both CYT and CYTBH suppressed erythema, hemorrhage, edema, excoriation, erosion of skin tissues of NC/Nga mice resulting in the decrease of thickness of epidermis. Significant decrease of infiltration of obese cells was also observed. The results above indicated that both CYT and CYTBH had significant efficacy in the treatment of atopic dermatitis through immune modulation. Animal studies showed that CYTBH had superior activity than that of CYT suggesting further and continuous studies on the changes in ingredients or absorption improvement by fermentation should follow.

Combined utilization with herbal products and prescribed drugs: A result from health examinee-based national survey (일부 종합병원의 건강검진 수검자들의 한약·양약 복합투여)

  • Recently, there has been prevailing of the combined utilization (CU) with herbs and prescribed drugs in medical therapies in the world. But the information about frequency, efficacy and safety of this CU has not well known in Korea, yet. This study aimed to identify the status of CU by Koreans, and to inquire which side effects of CU represent to those people. A self-completed questionnaire survey was performed through each health examination center in twenty general hospitals and one oriental hospital. Of the initial 2,100 health examinees, 1,851 were participated in this survey, resulting in a response rate of 88.1%. The proportion of CD was 26.3%. The most commonly mentioned reason of CD was 'to promote general health and well-being' (17l persons, 35.5%). The main route of taking CD was self-purchase at drugstore or at herbs market, followed by the prescription of (oriental)physicians. 33.0% (151 persons) of those who took the CD rated it as effective. 93 respondents (19.8%) were experienced several adverse effects including nausea, fatigue, and dizziness. The growing simultaneous use of herbal products and pharmaceutical drugs by Korean consumers may be continuously increased the risk of herb-drug interactions. The medical professionals should be provided with comprehensive and up-to-date information about potential benefits and risks of herbs and prescribed drugs. In the future studies it may be recommended to deal common cold, health promotion, indigestion, headache, and hypertension for the effect and safety of the CD by well-organized prospective study design.

Immunological Characteristics of Hyperprogressive Disease in Patients with Non-small Cell Lung Cancer Treated with Anti-PD-1/PD-L1 Abs

  • Hyperprogressive disease (HPD) is a distinct pattern of progression characterized by acceleration of tumor growth after treatment with anti-PD-1/PD-L1 Abs. However, the immunological characteristics have not been fully elucidated in patients with HPD. We prospectively recruited patients with metastatic non-small cell lung cancer treated with anti-PD-1/PD-L1 Abs between April 2015 and April 2018, and collected peripheral blood before treatment and 7-days post-treatment. HPD was defined as ≥2-fold increase in both tumor growth kinetics and tumor growth rate between pre-treatment and post-treatment. Peripheral blood mononuclear cells were analyzed by multi-color flow cytometry to phenotype the immune cells. Of 115 patients, 19 (16.5%) developed HPD, 52 experienced durable clinical benefit (DCB; partial response or stable disease ≥6 months), and 44 experienced non-hyperprogressive progression (NHPD). Patients with HPD had significantly lower progression-free survival (p<0.001) and overall survival (p<0.001). When peripheral blood immune cells were examined, the pre-treatment frequency of CD39+ cells among CD8+ T cells was significantly higher in patients with HPD compared to those with NHPD, although it showed borderline significance to predict HPD. Other parameters regarding regulatory T cells or myeloid derived suppressor cells did not significantly differ among patient groups. Our findings suggest high pre-treatment frequency of CD39+CD8+ T cells might be a characteristic of HPD. Further investigations in a larger cohort are needed to confirm our results and better delineate the immune landscape of HPD.

Vermicomposting of Sludge from Milk Processing Industry (MPS) (지렁이를 이용한 우유가공 폐수처리장 슬러지의 효율적 퇴비화)

  • This study was conducted to determine the optimal ratio of sludges from milk processing industry (MPS), paper-mill industry (PMS) and night-soil treatment plant (NSS) for vermicomposting. Five different ratios, 0 : 80 : 20 (MPS-0), 25 : 60 : 15 (MPS-25), 50 : 40 : 10 (MPS-50), 75 : 20 : 5 (MPS-75), and 100 : 0 : 0 (MPS-100 : control) MPS : PMS : NSS by wet weight were tested in a small plot experiment. The experiment for each mixing ratio was performed for 2 weeks with the three replications. MPS-100 (100 : 0 : 0) only had the highest decomposition rate with 19.9%, followed by MPS-25, MPS-50, MPS-75 and MPS-0 with 19.5, 19.1, 17.6 and 16.7%, respectively. Except for MPS-100, Vermicomposting resulted in increase in ash, T-P, $NO_2{^-}-N$, $NO_3{^-}-N$, Mg, K, As, Cd and Cu, whereas moisture, VS (Volatile Substance), TKN (Total Kjeldahl Nitrogen), $NH_4{^+}-N$, Ca, Hg and Pb were lower in the final cast than the initial feed mixture. Meanwhile Zn showed very slight difference and Cr and Ni did not show any tendency between the feed mixture and the final cast. In the case of MPS-100, where the decomposition rate was the highest, all the heavy metals in the final cast except for Hg were increased. All the vermicomposts produced from five different mixing ratios of the vermicomposting sludges met the Korea Standard as by-product compost.

In Vitro Effect of Interleukin-11 (IL-11) on Megakaryopoiesis from Umbilical Cord Blood Cells (생체 외 제대혈 배양에서 거대핵세포 조혈에 대한 Interleukin-11 (IL-11)의 효과)

  • Background: The megakaryopoiesis and platelet production is regulated by several hematopoietc factors such as thrombopoietin (TPO), interleukin-11 (IL-11) and interleukin- 3 (IL-3). IL-11 is a potent stimulator of megakaryopoiesis in vivo, and acts primarily as a megakaryocyte maturation factor in vitro and it can act synergistically with IL-3 and TPO. We performed this study to investigate the effects of recombinant human IL-11 (rhIL-11) with other hematopoietic factors on megakaryocyte colony formation in vitro. Methods: CD34+ cells were separated from umbilical cord blood and megakaryocyte colonies using MegaCult Assay Kit were cultured with rhIL-11, recombinant human IL-3 (rhIL-3), and recombinant human TPO (rhTPO) for 7 and 14 days. The number and percentage of CD34+ and CD41a+ cells were determined by flowcytometry. Results: The number of CD41a+ cells were $0.54{\pm}0.05{\times}10^4$ (rhIL-11 100 ng/ml), $5.32{\pm}0.23{\times}10^4$ (rhIL-3 100 ng/ml), and $8.76{\pm}0.15{\times}10^4$ (rhTPO 50 ng/ml) of total expanded cells during the culture of the purified CD34+ cells in liquid phase for 7 days. The number of CD41a+ cells were increased to $7.47{\pm}0.69{\times}10^4$ (rhIL-3+ rhIL-11), $11.92{\pm}0.19{\times}10^4$ (rhTPO+rhIL-11) of total expanded cells, respectively, during the culture of the purified CD34+ cells in liquid phase for 7 days in the presence of rhIL-11 (100 ng/ml). When the purified CD34+ cells were cultured in semisolid mediaincluding various concentration of rhIL-11, the megakaryocyte colonies were not formed. When the purified CD34+ cells were cultured with rhIL-11 and rhTPO or with rhIL-11 and rhIL-3, the number of megakaryocyte colonies were increased compared with rhTPO or rhIL-3 alone. Conclusion: These results indicate that IL-11 exerts a potent proliferative activity to colony forming unit-megakaryocyte from human umbilical cord blood, and it acts with other hematopoietic factors synergistically.

Distribution Properties of Heavy Metals in Goseong Cu Mine Area, Kyungsangnam-do, Korea and Their Pollution Criteria: Applicability of Frequency Analysis and Probability Plot (경남 고성 구리광산 지역의 중금속 분산특성과 오염기준: 빈도분석과 확률도의 적용성)

  • The frequency analysis and the probability plot were applied to heavy metal contents of soils collected from the Goseong Cu mine area as a statistic method for the determination of the threshold value which was able to partition a population comprising largely dispersed heavy metal contents into the background and the anomalous populations. Almost all the heavy metal contents of soil showed a positively skewed distributions and their cumulative percentage frequencies plotted as a curved lines on logarithmic probability plot which represent a mixture of two or more overlapping populations. Total Cu, Pb and Cd data and extractable Cu and Pb data could be partitioned into background and anomalous populations by using the inflection in each curve. The others showed a normally distributed population or an largely overlapped populations. The threshold values obtained from replotted frequency distributions with the partitioned populations were Cu 400 mg/kg, Pb 450 mg/kg and Cd 3.5 mg/kg in total contents and Cu 40 mg/kg and Pb 12 mg/kg in extractable contents, respectively. The thresholds for total contents are much higher than the tolerable level of soil pollution proposed by Kloke(Cu 100 mg/kg, Pb 100 mg/kg, Cd 3 mg/kg), but those for extractable contents are not exceeded the worrying level of soil pollution proposed by Ministry of Environment(Cu 50 mg/kg, Pb 100 mg/kg). When the threshold values were used as the criteria of soil pollution in the study area, $9{\sim}19%$ of investigated soil population was in polluted level. The spatial distributions of heavy metal contents greater than threshold values showed that polluted soils with heavy metals are restricted within the mountain soils in the vicinity of abandoned mines.
