Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of HYT on the recovery of dermatitic symptoms by how HYT influenced the immune related factors. The results are as below: 1. Compared to the control, HYT treated group showed recovery of atopic dermatitis by the naked eye observation, and significant reduction of dermatits index was observed after 14 weeks. 2. HYT treated group showed significant decrease of the ratio of CCR3+, B220+/IgE+, and Gr-1+/CD11b+ immune cells in dorsal skin by 40.5%, 34.2%, and 48.1%, respectively. 3. HYT treated group showed increase in the ratio of CD19+ immune cells within PBMC by 10.8%, as well as decrease in CD3+, CD3+/CD69+, NKT+ ratios by 5.3%, 35.2%, and 44.9%, respectively. 4. HYT treated group showed increase in the expression of IFN-$\gamma$ in serum by 589.3%, whereas the expression of IL-4, IL-5, IL-6, IL-13, TNF-$\alpha$, MCP-1 and RANTES were decreased by 31.4%, 82.1%, 97.1%, 39.5%, 83.7%, 26.1%, 48.6%, respectively. A 47.2% decrease in IgE expression was also observed. The results above strongly supported the improvement of atopic dermatitis by HYT treatment through immune modulation. Further studies on the synergistic effect of each ingredients of HYT and therapeutic effects according to the dosage of each ingredient should be followed for clinical applications. This work was (partly) supported by the RIC program of MKE(Ministry of Knowledge Economy) in Daejeon University.
Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of Hyunggaeyunkyotanggamibang(HYGB) on the recovery of dermatitic symptoms through its influence on the immune related factors. The results are as below: 1. HYGB treated group showed improvement of atopic dermatitis with naked eye observation, and significant decrease of dermatitis index was observed after 14 weeks. 2. HYGB treated group showed significant decrease of the ratio of CCR3+, B220+/IgE+, and CD11b+/Gr-1+ immune cells in dorsal skin by 41.7%, 21.5%, and 23.8%, respectively. 3. HYGB treated group showed an increase of CD19+ immune cells by 10.3% in PBMC, whereas CD3+, CD3+/CD69+, NKT+ immune cells were decreased by 4.3%, 42.9%, and 21.7%, respectively. 4. HYGB treated group showed an increase in the expression of IFN-$\gamma$ in the serum by 514.3%. However, the expressions of IL-4, IL-5, IL-6, IL-13, TNF-$\alpha$, MCP-1 and RANTES were decreased by 21.2%, 69.8%, 90.5%, 28.7%, 72.2%, 26.1%, and 19.9%, respectively. Also, the expression of IgE was decreased by 44.3%. 5. HYGB treated group showed a decrease of the expression of IL-4 and IL-5 by 43% and 44.3%. The results above indicated that HYGB clinically used for atopic dermatitis treatment has objective validity, and therefore can be provided as the basic data for EBM(Evidence based medicine) construction for anti-allergic and anti-inflammatory studies.
This laboratory has recently reported the solubility and in vivo absorption enhancement of diclofenac sodium by ${\beta}-cyclodextrin$ complexation. The acute gastroduodenal mucosa injury provoked by administration of 34 mg/kg and 68 mg/kg of a diclofenac sodium (DS) and equivalent dose of new formulation [diclofenac sodium-beta-cyclodextrin complexation$(DS-{\beta}-CD)$] was evaluated and compared. Microscopic examinations, performed after 18-hrs treatment, demonstrated that $DS-{\beta}-CD$ was less gastrolesive than DS. The drop in gastrophy after a single dose of the assigned drug was considerably greater for DS than for $DS-{\beta}-CD$, which registered similar values to control. Since gastrophy is an expression of the anatomy-functional integrity of the gastric barrier, the results indicate that $DS-{\beta}-CD$ exerts less direct acute damage on the gastric mucosa. Therefore, when administered short-term, $DS-{\beta}-CD$ appears to be less gastrolesive than the standard DS formulation.
In order to elucidate the relationships between cadmium, copper, mercury, zinc levels and metallothionein in the liver and kidney cortex of Korean, the levels of Cd, Zn, Hg, Cu and metallothionein (MT) were determined in the kidney cortex and liver of 50 subjects deceased in the period of January-November, 2001 in the area of Seoul and Gyeonggi Province of Korea. The mean age of the population studied was 36.3+/-12.3 years. The tissues were digested with microwave digestion system and the elements were determined by inductively coupled plasma atomic emission spectrometry. MT was determined by the Cd-hemoglobin affinity assay. The determined levels (mean+/-SD) were: 33.9+/-18.9 micrograms Cd/g wet weight; 47.5+/-12.6 micrograms Zn/g wet weight; 2.5+/-0.57 microgram Cu/g wet weight; 0.26+/-0.31 micrograms Hg/g wet weight, 4.0+/-3.1 mg MT/g wet weight in renal cortex and 2.5+/-1.9 micrograms Cd/g wet weight; 46.9+/-15.0 micrograms Zn/g wet weight; 6.2+/-2.5 micrograms Cu/g wet weight; 0.10+/-0.15 micrograms Hg/g wet weight, 0.92+/-0.57 mg MT/g wet weight in the liver. Positive relationships between Cd and MT, sum of four divalent metal and MT in the kidney cortex were observed. No other correlation was found between Cu and MT, Hg and MT, Zn and MT in either organs.
Road runoff water includes various heavy metals (zinc, Zn; lead, Pb; copper, Cu; chrome, Cr; cadmium, Cd; etc.) and pathogens (E-coli and coliform). Since these pollutants are significantly harmful to human beings and have negative impact on water streams, numerous studies have been conducted to determine the characterization of these non-point pollutants from road runoff water. However, since these non-point pollutant concentrations vary depending on road traffic, road construction, and road maintenance, measurement of pollutant loadings in different site is necessary to estimate the effect of road runoff water on drinking water source. The objective of this study was to examine the quality of road runoff water from a city bridge in Seoul, Korea. This study was conducted for two years to assess annual discharge pollution loads. In this study, five key heavy metals (Zn, Pb, Cu, Cr, and Cd) and two pathogens (E-coli and coliform) were measured at 18 different events. The pollutant load mass transported was always higher than the corresponding runoff volume for Zn, Cu, and Cd, while Pb and Cr showed similar values between the load mass transported and the corresponding runoff volume. The event mean concentrations were Zn (0.908 mg/L), Pb (0.092 mg/L), Cu (0.141 mg/L), Cr (0.023 mg/L), and Cd (0.006 mg/L). Like Zn, Cu, and Cd, E-coli and coliform values (relatively high in Summer and Fall) are higher at the beginning of each event and decrease afterwards.
In order to investigate the efficacy of HYGBH on atopic dermatitis, various immune related factors were studied. The results and conclusions are as follows. Atopic dermatitis symptoms were improved in HYGBH treated group and significant decrease in dermatitis index were observed in 12 and 14 weeks. HYGBH treated group showed significant decrease in CD4+, CD3+/CD69+ immune cell ratio in PBMC by 18% and 40.6% respectively. HYGBH treated group showed significant decrease in CD3+, CD11b+/Gr-1+ immune cell ratio in dorsal skin by 44.6% and 53.1% respectively. HYGBH treated group showed significant decrease in IL-4, IFN-${\gamma}$ in spleen by 29.5%, 7.7% respectively. HYGBH treated group showed decrease in the expression of IL-5, IL-13, IL-17 and histamine by 21%, 9.6%, 14%, and 32.2% respectively. Also the group showed decrease in the expression of IgE by 6.8% respectively. HYGBH treated group showed significant decrease in the transcription of IL-5 and IL-3 mRNA in skin by 35.5% and 23.2% respectively. The results above indicated that treatment of HYGBH improved atopic dermatitis symptoms by anti-oxidant activity as well as immune modulation activity as a clinical evidence. Also, to increase the application of fermented oriental medicine, different fermentation conditions using various microbial strains should be accumulated as the clinical evidence in the future.
The objectives of this study were to investigate the efficiencies of the stabilizers such as mono-potassium phosphate (MKP), phosphate fertilizer and red mud in treating the mine tailings contaminated with heavy metals and to characterize the changes in fractionations of the heavy metals during the stabilization. The TCLP results showed that the stabilization efficiencies of Cd, Pb and Zn increased with the increase in the stabilizer dosage and the reaction times. MKP showed the highest efficiencies for the heavy metals stabilization among the stabilizers tested. When the mine tailings were amended with MKP, the TCLP concentrations of Cd, Pb and Zn were reduced by 79~97%, 61~84%, and 89~99%, respectively. When the composite stabilizers, MKP/phosphate fertilizer or MKP/red mud, were used, the stabilization efficiencies were lower than when MKP was used as a single stabilizer. The sequential extraction results showed that carbonates fraction of Cd and Zn increased generally. Especially, when red mud was used, carbonates fraction of Cd and Zn increased 5 and 18 times, respectively. In the case of Pb, the treatment with MKP increased residual fraction by 10 times. The results showed that MKP was the most effective in stabilizing the heavy metals (Cd, Pb and Zn) to improve the efficacy of the composite binders.
