Lewis 폐암 마우스 모델에서 Interleukin-12가 E-selectin 발현에 미치는 영향 (Effect of Interleukin-12 on the Expression of E-selectin in Mouse Model of Lewis Lung Carcinoma)

  • 연구배경: Interleukin-12 (IL-12) 는 자연살해세포와 T 림프구 활성을 증가시킬 뿐 아니라 증식을 촉진시키고 mterferon-$\gamma$와 granulocyte-macrophage colony stimulating factor 그리고 tumor necrosis factor-$\alpha$ (TNF-$\alpha$)의 생성을 유도한다고 알려져 있으며 여러 동물실험에서 악성종양의 치료에 상당한 효과가 있음이 알려져 있다. 이런 결과는 종양내 염증세포의 침윤이 증가함과 밀접한 관련이 있다고 밝혀져 있지만 이들 염증세포가 종양내로 이동하는 단계에 IL-12가 미치는 영향에 대해서는 잘 알려진 바가 없다. 본 연구에서는 IL-12가 종양의 성장 및 전이에 미치는 영향과 활성화된 내피세포에 발현되는 것으로 알려진 E-selectin의 발현에 미치는 영향을 관찰하고자 하였고 또한 E-selectin의 발현이 IL-12에 의해 생산이 증가되고 여러 유착분자의 발현을 유도하는 작용이 있는 것으로 알려진 TNF-$\alpha$와 연관이 있는지 알아보고자 하였다. 방법: C57BL/6 마우스를 대상으로 Lewis 폐암세포주를 피하주사하여 암세포를 이식한 후 각 군마다 각각 IL-12 또는 TNF-$\alpha$ 생리식염수를 복강내 투여하였다. 암세포이식 28일째 마우스를 도살하여 폐전이 결절의 수를 측정하였으며 피하종양에 대해 CD4, CD8, CD16, E-selectin에 대한 면역화학조직염색을 시행 하였다. 결과: IL-12를 투여한 군에서 대조군에 비해 암세포 이식부위의 종양 크기가 감소하였으며 폐전이 결절의 수도 감소하였다. IL-12를 투여한 군에서 대조군에 비해 CD4+ 및 CD8+ 그리고 CD16+ 세포의 종양내 침윤이 증가하였다. IL-12를 투여하지 않은 대조군에서 E-selectin은 발현되지 않았으나 IL-12 투여군에서 종양내 혈관내피세포에서의 E-selectin 발현이 증가되었다. TNF-$\alpha$ 투여시 종양내 혈관내피세포에서의 E-selectin 발현 증가가 관찰되었으며 또한 종양내 CD4+ 및 CD8+ 그리고 CD16+ 세포의 침윤이 증가되었다. 결론: 이상의 결과로 보아 IL-12는 Lewis 폐암모델에서 종양내 염증세포의 침윤을 일으켜 종양의 성장과 전이를 억제하는 작용을 나타내며 염증세포의 침윤에는 E-selectin이 관여할 것으로 생각된다. 또한 E-selectin의 발현증가에는 IL-12 에 의해 생성이 유도되는 TNF-$\alpha$가 작용할 가능성을 추정할 수 있겠다.

  • To study the effects of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Eclipta prostrata diffuse herba infusion solution(ELP-HAS), we injected ELP-HAS into Chok-samni(St36) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. We have reached the following conclusion through the effect on the number of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1+/CD3e^+$ cells in mouse PBMCs, the effect on the pulmonary colony number, and the effect on MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma The results were obtained as follows: 1. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the spleen cell proliferation in Balb/c mouse was significantly increased compared with control group. 2. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$ cells in C57Bl/6 mouse PBMCs was increased compared with control group. 3. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the pulmonary colony number of C57BL/6 mice implanted intravenously with B16-F10 melanoma was decreased significantly compared with control group. 4. In the experiment groups treated with ELP(Eclipta prostrata)Herbal-Acupuncture, MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma were increased significantly compared with control group.

오마환(烏麻丸)이 백서(白鼠)의 혈액내(血液內) CD4+, CD8+ T cell 및 면역기관(免疫器官) 장기지수(臟器指數)에 미치는 영향(影響) (Effects of Omahwan(OMH) on CD4+, CD8+ T cell and Immune Organ Index in Rat)

  • OMH which is known for its properties of recruiting vitality, is composed of Polygonum multiflorum Thunb. and Sesamum indicum DC.. This formula is known to possess the properties of recruiting vitality, blackening white hair and expanding life span. 16-week-old SD-Rats were treated with OMH for 16 days. After 24 hours, the rats were treated with MTX(Mthotrexate is oral administrated for 4 days(1mg/kg/day), in order to lower immunity. Then, These rats were classified in to groups, the N-16 group(not specially tested), the Control group(MTX), the OMH-L group(2.5% OMH+MTX)&the OMH-H group(10% OMH+MTX) and 6 rats were assigned to each group. After 18 hours from MTX treatment the organ index of the rats from each group Thymus and Spleen were measured. The percentage of CD+4, CD8+ T cell were measured and compared by flow cytometer. 1) Rats from the OHM-L&OMH-H group showed higher organ indexes of the Thymus and Spleen compared to the rats from the Control Group. This proves that OMH possesses the properties to mitigate degenerations of immunity($F_{thymus}=20.162,\;F_{spleen}=5.882$, ANDVA, p<0.05). 2) The rats from the two OMH groups showed higher rates of CD4+ T cell counts compared to the control group(F=26.906, ANOVA, p<0.05). CD8+ T cell showed lower rates compared to the Control group, but showed no differences within the two OMH groups(F=1.254, ANOYA, p>0.05). CD4+/CD8+ showed higher rates in the two OMH groups compared to the control group which can be thought as a proof that OMH prevents depression of immune response(F=10.554, ANOVA, p<0.05). In this test 16 week-old rats were used, which can be considered as the middle and prime age of the human being. These rats were treated with OMH which ended up showing properties of mitigating degeneration of immune responses and maintaining T-cell rates within the blood. It was possible to study that OMH possesses the properties to increase immune responses.

말토덱스트린 또는 사이클로덱스트린을 이용한 미세캡슐화공정이 옥수수 수염 추출물의 수분흡습과 생리활성에 미치는 영향 (Effects of Microencapsulation Using Maltodextrin and/or Cyclodextrin on Water Absorption and Bioactivity of Corn Silk Extract)

  • 본 연구는 옥수수 수염 추출물을 핵물질로 사용하였고 피복물질로는 maltodextrin (MD), cyclodextrin (CD), MD와 CD를 1:1(w/w)로 혼합한(MD+CD) 세 가지 피복물질을 이용하여 미세캡슐화 하였을 때 흡습제어, DPPH radical 소거능(DRSA), hydroxyl radical 소거능(HRSA) 및 tyrosinase 저해활성(TIA)의 향상 정도를 비교 분석하였다. 대조구의 WAI 값은 0.68로 가장 높은 지수를 보였으며 처리구의 경우는 MD+CD 처리구가 0.20으로 가장 낮게 나타내었다. 이러한 현상은 피복물질에 의한 유리전이온도 상승과 같은 수분함량에서의 수분활성도 증가에 기인되기 때문이다. WSI에서는 대조구가 72.23%로 가장 낮은 지수를 나타내었으며 CD 처리구가 95.23%로 가장 높은 지수를 나타내었으며 피복물질의 수분용해도 및 미세캡슐화 과정에서의 입자크기감소가 주요원인이다. DRSA의 경우 대조구는 18.60%로 가장 낮았으며 CD 처리구에서는 89.25%이었고 HRSA는 대조구의 경우 16.89%인 반면 MD+CD 처리구는 47.64%로 나타났다. TIA에서는 대조구는 7.16%로 낮은 활성을 보인 반면 MD 처리구가 40.42%로 가장 높은 활성을 보였다. 이와 같은 생리활성도 향상은 수분용해도와 비표면적 증가에 기인되었기 때문이다. 모든 분석 치에서 대조구와 처리구간에 유의성차이를 보였다(p<0.05).

위암 환자에서 수술 전 혈청 알부민수치에 따른 림프구아형의 분포양상 (The Distribution Pattern of Lymphocyte Subsets according to the Level of Serum Albumin in Preoperative Patients with Gastric Cancer)

  • 목적: 영양상태가 면역력과 상관관계가 있다는 점에 착안하여 위암 환자에서의 혈청 알부민수치와 림프구아형의 수적인 병화에 근거하여 그 상관관계를 확인하고자 본 연구를 시행하였다. 대상 및 방법: 1998년 8월부터 2004년 8월까지 위암으로 진단 받고 수술을 시행한 한자 중에서 수술 전에 말초혈액 림프구아형 검사를 시행한 환자 150명을 대상으로 하였다. 결과: 위암 환자에서 혈청 알부민수치 변화에 따른 림프구아형의 병화를 비교하였고, 각 병기에 따른 변화도 비교하였다. 3.2 mg/dl를 기준으로 환자들을 구분하였을 때 말초혈액 림프구수, CD3+ 세포, CD4+ 세포, CD8+ 세포, CD16+56 세포수는 혈청 알부민 수치가 3.2 mg/dl 이상인 군보다 3.2 mg/dl 미만인 군에서 의미 있게 감소한 것을 볼 수 있었다(P<0.05). 혈청 알부민수치가 낮은 군에서 제1기(n=59)에서는 CD16+ 세포수의 감소, CD4+/CD8+ 비율의 의미 있는 증가가 있었다(P<0.05). 제IV기(n=33)에서도 혈청 알부민수치가 낮은 군에서 CD19+세포를 제외한 나머지 모든 림프구아형의 수적인 감소가 있었고 CD4+/CD8+ 비율의 증가가 있었다(P<0.05). 결론: 혈청알부민수치가 낮은 군이 정상인 군보다 전반적인 림프구아형의 절대수가 낮다. 이에 근거하여 위암 환자에서 영양상태와 면역상태는 깊은 상관관계가 있음을 다시 확인하였다.

Oncolytic Vaccinia Virus Expressing 4-1BBL Inhibits Tumor Growth by Increasing CD8+ T Cells in B16F10 Tumor Model

  • Oncolytic viral vectors have shown good candidates for cancer treatment but have many limitations. To improve the therapeutic potential of oncolytic vaccinia virus, we developed a recombinant vaccinia virus expressing the 4-1BBL co-stimulatory molecule or CCL21. 4-1BBL and CCL21 expression was identified by FACS analysis and immunoblotting. rV-4-1BBL vaccination shows significant tumor regression compared to rV-LacZ, but rV-CCL21 shows rapid tumor growth compared to rV-LacZ in the poorly immunogenic B16 murine melanoma model. 4-1BBL expression resulted in the increase of the number of CD8+ T cells and especially the increase of effector (CD62L-CD44+) CD8+ T cells. These data suggest 4-1BBL may be the potential target for enhancement of tumor immunotherapy.

극직교 이방성 회전원판의 진동해석 및 임계속도, II : 해석결과 (Vibration Analysis and Critical Speeds of Rotating Polar Orthotropic Disks, Part II : Analysis Results)

  • This paper (Part II) provides the application results of the method presented in a companion paper (Part I) where the dynamic equation for rotating polar orthotropic disk is formulated and its solution method is considered. The natural frequencies and critical speed of polycarbonate CD are calculated to validate the present method and are shown to by very accurate. The critical speeds of typical GFRP and CFRP CD's are computed by aligning the fibers in radial and circumferential directions. The radially reinforced CFRP CD is shown to have the five times higher critical speed than that of the polycarbonate CD. The natural frequencies and critical speeds of disks with various elastic modulus ratios are obtained. The results show that the radially reinforced disk is more effective in increasing critical speed than the circumferentially reinforced disk.

개인정보 보호를 위한 의료영상 복사발급 지침에 대한 고찰 (Study for Guideline of CD copy Issue to Secure Personal Information)

  • Images of medical treatment on PACS environment are digitized and they make saving and transmission of patient's information easy with CD format causing increase of patients transfer rate between hospital and data transmission as well. Figuring out the configuration of confining status of copied images of medical treatment with CD format and present the appropriate way of identification for personal information. 20 general hospital over 500 beds which are utilizing PACS currently. Questionnaire investigation focusing on PACS operation center and it's management people. In accordance with the law of securing personal information of public organizations, it says "Anyone who wants to see his or her information via representative he or she needs to make a letter of attorney and turn in this to the president of it's possessing facility" in the article 16. Based on the investigation of general hospital for CD copy issue status near metropolitan area, 8 out of 20, were issuing CD copy through personal identification process(the person oneself and representative) and 10 general hospital were doing by getting signature of receiptor only without identification of the person oneself and 2 general hospital confirmation process is not at all. By the law of securing personal information of public organization, article 16, confirmation process is supposed to mandate lawfully with a letter of attorney when there is any request of reading and/or copies of one's information. But in reality, there were no consistent rules on reading, the process and coverage of copy issue, rejection coverage of copy issue of CD copy in each general hospital PACS operation center. Therefore there is a big need for consolidated format which is applicable by law when the representative requests to issue of CD copy that is storing medical treatment images. By issue through this consolidated format, securing medical information of individual and systemic operation and management will be valid and effective.

  • CdTe crystals were grown by the vertical Bridgman method. P-type DcTe crystals were grown with Cd:Te= 1:1.001 wt. % ratio, while n-type CdTe crystals were 1:1 Also, CdTe:In crystals were investigated, Lattic constants were 6.489${\AA}$ for p-type 6.480${\AA}$for n=type and 6.483${\AA}$ for CdTe:In EPD was 10$\^$-3/-10$\^$4/cm$\^$-2/ for n-, p-type CdTd, 10$\^$4/-10$\^$5/cm$\^$-2 for Cd:Te:In using by E-Ag solution for (111) plane The carrier concentration, the resistivity and the Hall carrier mobility measured by the van der Pauw method were p=5.78${\times}$10$\^$15/cm$\^$-3/, $\rho$=20.2$\Omega$cm, ${\mu}$$\sub$p/=75.6cm$\^$-2/ V$\^$-1/ sec$\^$-1/ for p-typem n=2.98${\times}$10$\^$16/cm$\^$-3/, $\rho$=0.214$\Omega$cm, ${\mu}$$\sub$p/=978.9cm$\^$-2/ V$\^$-1/ sec$\^$-1/ for n-type and n=7.45${\times}$10$\^$16/cm$\^$-3/, $\rho$=1.54 ${\times}$10$\^$3/$\Omega$cm, ${\mu}$$\sub$p/=658.4 cm$\^$-2/ V$\^$-1/ sec$\^$-1/ for CdTe:In crystals, Transmittance of p-type CdTe was 61% that of n-type was 65%, Cd:Te:In showed 60% IR transmittance.

Tumor-derived CD4+CD25+ Tregs Inhibit the Maturation and Antigen-Presenting Function of Dendritic Cells

  • CD4+CD25+regulatory T cells (Tregs) play a key role in regulation of immnue response and maintenance of self-tolerance. Studies have found Tregs could suppress tumor-specific T cell-mediated immune response and promote cancer progression. Depletion of Tregs can enhance antitumor immunity. Dendritic cells (DCs) are professional antigen-presenting cells and capable of activating antigen-specific immune responses, which make them ideal candidate for cancer immunotherapy. Now various DC vaccines are considered as effective treatment for cancers. The aim of this study was to evaluate variation of Tregs in BALB/C mice with hepatocellular carcinoma and investigate the interaction between tumor-derived Tregs, effector T cells (Teff) and splenic DCs. We found the percentages of Tregs/CD4+ in the peripheral blood of tumor-bearing mice were higher than in normal mice. Tumor-derived Tregs diminished the up-regulation of costimulatory molecule expression on splenic DCs, even in the presence of Teff cells and simultaneously inhibited IL-12 and $TNF-{\alpha}$ secretion by DCs.
