Objective : The purpose of this research is to examine the effects of Scutellariae Radix(SR) extract on immune cells in ovalbumin(OVA)-induced asthmatic mice. Methods : In vitro, C57BL/6 mice were exposed to OVA for 6 weeks. The mice lungs were taken out, chopped, grinded with collagenase and activated by rIL-3/rIL-5. The lung cells were treated with SR extract and analyzed by a flow cytometer. Results : The population of granulocytes/lymphocytes, CD3e-/CCR3+, CD69+/CD3e+, CD23+/B220+, CD4+ cells in the SR group in vitro decreased and population of CD8+ cells in the 100 and $10{\mu}g/m{\ell}$ SR group in vitro decreased, compared with that of control group. Conclusion : The results of this study support a role for SR as an effective treatment for asthma in its experimental success in significantly decreasing inflammation and asthma reactions.
We compared the quality of 'Fuji' apples (Malus ${\times}$ domestica) from trees whose leaves were not removed (no artificial defoliation; NAD) with apples from trees that underwent early defoliation (ED, treated in mid September and early October) and conventional defoliation (CD, treated in early and mid October). The experiment was conducted in three consecutive years using 15-year-old 'Fuji' apple grafted on Malus prunifolia. Fruits were harvested on November 7, 16 or 12 in 2011, 2012 and 2013, respectively. Compared to NAD treatment, ED and CD treatment reduced the fresh weight by 4.7% and 0.6%, respectively. The soluble solids content of NAD apples ($14.4^{\circ}Brix$) was slightly higher than that of CD ($14.1^{\circ}Brix$) and ED ($14.0^{\circ}Brix$) apples. Soluble sugar content, flesh firmness, water-core index, and titratable acidity were not affected by defoliation treatment regardless of treatment timing. The skin blush index of NAD apples (2.3) was inferior to that of CD (3.3) and ED (3.4)- treated apples. Furthermore, artificial defoliation treatments increased skin redness ($a^*$) and yellowness ($b^*$) and significantly improved the degree of skin blush compared to NAD fruits.

The inflammatory response during cutaneous leishmaniasis (CL) involves immune and non-immune cell cooperation to contain and eliminate Leishmania parasites. The orchestration of these responses is coordinated primarily by CD4+ T cells; however, the disease outcome depends on the Th cell predominant phenotype. Although Th1 and Th2 phenotypes are the most addressed as steers for the resolution or perpetuation of the disease, Th17 cell activities, especially IL-17 release, are recognized to be vital during CL development. Th17 cells perform vital functions during both acute and chronic phases of CL. Overall, Th17 cells induce the migration of phagocytes (neutrophils, macrophages) to the infection site and CD8+ T cells and NK cell activation. They also provoke granzyme and perforin secretion from CD8+ T cells, macrophage differentiation towards an M2 phenotype, and expansion of B and Treg cells. Likewise, immune cells from the inflammatory infiltrate have modulatory activities over Th17 cells involving their differentiation from naive CD4+ T cells and further expansion by generating a microenvironment rich in optimal cytokines such as IL-1β, TGF-β, IL-6, and IL-21. Th17 cell activities and synergies are crucial for the resistance of the infection during the early and acute stages; however, if unchecked, Th17 cells might lead to a chronic stage. This review discusses the synergies between Th17 cells and the inflammatory infiltrate and how these interactions might destine the course of CL.
In order to determine the content levels of trace metals in cow's milk, 79 market milk samples were collected from markets in major cities of Korea from July to August, 1985 and analyzed for content levels of Cu, Fe, Zn, Mn, Pb, Cd. Immunoregulative action of 70% ethyl alcohol fraction of Cervus nippon was investigated in vitro. The fraction enhanced the proliferation of thymocytes and the population of CD4$\^$+/CD8$\^$-/ single-positive cells in thymocytes. CN-E enhanced [Ca$\^$2+]$\sub$i/, the production of TNF-${\alpha}$ and IL-1 ${\beta}$, and phagocytosis in peritoneal macrophages. The fraction did not induce the proliferation of splenocytes and DNA fragmentation in thymocytes. These results indicate that 70% ethyl alcohol fraction of Cervus nippon regulates immunological action by stimulating murine thymocytes and macrophages.
In this study, a straightforward and precise nitrogen/sulphur-codoped carbon dots (N/S-CD) was produced using a microwave irradiation approach. The N/S-CD was formulated from succinic acid (SA), bis-(3-aminopropyl)-amine (BAPA), and sodium thiosulphate (STS) as sources of carbon, nitrogen, and sulphur, respectively. The synthesized N/S-CD established a valuable quantum yield (QY) of 70 % and was sensitive to aluminium ion (Al3+) with a detection limit of 0.21 µM and a linear concentration range of 0-100 µM. When detecting Al3+ in real water samples, the N/S-CD resulted in a satisfactory recovery in the range of 91.14 %-103.37 %. Thus, the proposed study is very promising for Al3+ detection in environmental water samples.
Heavy metals in the mangrove sediments of Chuuk and Kosrae, Federated States of Micronesia were analyzed to examine the pollution levels of heavy metals using enrichment factor (EF) and pollution load index (PLI). The mean concentrations of Cr, Ni, Cu, Zn, As, Cd and Pb in surface mangrove sediments were 642, 125, 46.9, 149, 15.6, 0.14 and 8.55 ${\mu}g$, respectively. Kosrae mangrove sediments showed the highest concentrations of Cr and Ni while Chuuk contains more of other metals such as Cu, Zn, As, Cd and Pb. Compared to those from other mangrove regions of the world, Cr, Ni and As levels in mangrove sediments from Micronesia were at higher levels whereas Cu, Zn, Cd and Pb were at lower to median levels. In core sediment of Chuuk, metal concentrations in the upper part were higher than those in the lower part. Based on the EF and PLI values, As is evaluated as the heaviest contaminant in the surface sediment from Micronesia whilst other metals (Cr, Ni, Cu, Zn, Cd and Pb) are present at slightly lesser levels.
Background: The leukocyte common antigen (CD45) is a transmembrane-type protein tyrosine phosphatase that has five isoforms. Methods: We generated seven murine mAbs against human CD45 by injecting cells from different origins, such as human thymocytes, PBMCs, and leukemic cell lines. By using various immunological methods including flow cytometry, immunohistochemistry, and immunoprecipitation, we evaluated the reactivity of those mAbs to CD45 of thymus as well as tonsil lysates. Furthermore, we transiently transfected COS-7 cells with each of gene constructs that express five human CD45 isoforms respectively, and examined the specificities of the mAbs against the transfected isoforms. Results: In case of thymocytes, lymphocytes, and monocytes, all the seven mAbs demonstrated positive reactivities whereas none was reactive to erythrocytes and platelets. The majority of immune cells in formalin-fixed paraffin-embedded thymus and tonsil tissues displayed strong membranous immunoreactivity, and the main antigen was detected near 220 kDa in all cases. Among the mAbs, four mAbs (AP4, DN11, SHL-1, and P6) recognized a region commonly present in all the five isoforms. One mAb, YG27, recognized four isoforms (ABC, AB, BC, and O). Two mAbs, P1 and P14, recognized the isoforms that contain exon A encoded regions (ABC and AB). Conclusion: In this study, we confirmed that AP4, DN11, SHL-1, YG27 and P6, are mAbs reactive with the CD45 antigen whereas P1 and P14 are reactive with the CD45RA antigen.

JL1, a specific epitope on CD43, is a potential biomarker for the diagnosis of acute leukemia. Although qualitative assays for detecting leukemia-specific CD43 exist, there is a need to develop quantitative assays for the same. Here, we developed two novel monoclonal antibodies (mAbs), 2C8 and 8E10, recognizing different epitopes on CD43. These clones are capable of pairing with YG5, another mAb against JL1 epitope, because they were selectively obtained using sandwich ELISA. Antigens recognized by 2C8 and 8E10 were confirmed as CD43 by western blotting using the CD43-hFC recombinant protein. When expression on various leukemic cell lines was investigated, 2C8 and 8E10 displayed a disparity in the distribution of the epitope. Enzyme assays revealed that these mAbs recognized a sialic acid-dependent epitope on CD43. Using normal thymus and lymph node paraffin-embedded tissues, we confirmed a difference in the epitopes recognized by the two mAbs that was predicted based on the maturity of the cells in the tissue. In summary, we developed and characterized two mAbs, 2C8 and 8E10, which can be used with YG5 in a sandwich ELISA for detecting leukemia-specific CD43.
This study was conducted in order to get the scientific background for soil pollution criteria. The 92 soil samples derived from various geological units were taken and analyzed to survey natural heavy metal background levels using aqua regia digestion method and 0.1N HCl extraction method. From these results, the average natural contents of metals were 0.287 mg/kg for Cd, 15.26 mg/kg for Cu, 18.43 mg/kg for Pb, 25.36 mg/kg for Cr, 54.27 mg/kg for Zn, 17.68 mg/kg for Ni, 6.83 mg/kg for As by the aqua regia method, and 0.040 mg/kg for Cd, 0.48 mg/kg for Cu, 3.06 mg/kg for Pb, 0.09 mg/kg for Cr, 1.54 mg/kg for Zn, 0.27 mg/kg for Ni, 0.089 mg/kg for As by the 0.1N HCl extraction method. Ratios of soluble contents and total contents were Cd 0.14, Cu 0.03, Pb 0.17, Cr 0.004, Zn 0.03, Ni 0.02, As 0.013 and the correlation coefficients of soluble contents and total contents were 0.24(As), 0.88(Cd), 0.43(Cr), 0.65(Cu), 0.70(Pb), 0.61(Ni), 0.24(Zn). The correlation factor decreased in the order of Cd > Pb > Cu > Ni > Cr > Zn $\approx$ As.
