Five secondary metabolites, including a new isopimarane derivative xylaroisopimaranin A (1), were isolated from the endophytic fungus Xylaralyce sp. (HM-1), and their structures were elucidated by 1D, 2D NMR, MS and CD spectra. Their bioactivities were performed to antibacterial, Hep G2 cells cytotoxicity and brine shrimp inhibition. The biological evaluation results showed that the xylaroisopimaranin A (1), xylabisboein B (2), griseofulvin (3), 5-methylmellein (4) and mellein-5-carboxlic acid (5) displayed no significant Hep G2 cells cytotoxicity and antibacterial acitivity, but they inhibited the brine shrimp with $IC_{50}$ from 0.5 to $25{\mu}mol/mL$.
The Korea Atomic Energy Research Institute (KAERI) has designed a Hybrid-Low Power Research Reactor (H-LPRR) which can be used for critical assembly and conventional research reactor as well. It is an open tank-in-pool type research reactor (Thermal Power: 50 kWth) of which the most important applications are Neutron Activation Analysis (NAA), Radioisotope (RI) production, education and training. There are eight irradiation holes on the edge of the reactor core: IR (6 holes for RI production) and NA (2 holes for NAA) holes. In order to quantify the elemental concentration in target samples through the Instrumental Neutron Activation Analysis (INAA), it is necessary to measure neutron spectrum parameters such as thermal neutron flux, the deviation from the ideal 1/E epithermal neutron flux distribution (α), and the thermal-to-epithermal neutron flux ratio (f) for the irradiation holes. In this study, the MCNP6.1 code and FORTRAN 90 language are applied to determine the parameters for the two irradiation holes (NA-SW and NA-NW) in H-LPRR, and in particular its α and f parameters are compared to values of other research reactors. The results confirmed that the neutron irradiation holes in H-LPRR are designed to be sufficiently applied to neutron activation analysis, and its performance is comparable to that of foreign research reactors including the TRIGA MARK II.

Phoswich 검출기를 제작하기 위하여 무기 섬광체인 CsI(Tl), $CdWO_4(CWO),\;Bi_4Ge_3O_{12}(BGO)$와 $Gd_2SiO_5:Ce(GSO)$의 특성을 연구하였다. CsI(Tl), CWO, BGO 및 GSO 섬광체의 radioluminescence 중심파장은 550 nm, 475 nm, 490 nm 및 440 nm이였고, neutral filter를 사용하여 측정한 CsI(Tl), CWO, BGO 및 GSO 섬광체의 절대광량은 각각 54890 phonon/MeV, 17762 phonon/MeV, 8322 phonon/MeV 및 8932 phonon/MeV이였으며, single photon method로 측정한 형광감쇠시간은 각각 $1.3{\mu}s,\;8.17{\mu}s$, 213 ns 및 37 ns이였다. 플라스틱 섬광체와 CsI(Tl) 섬광체를 사용하여 phoswich 검출기를 제작하였고 PSD(pulse shape discriminator) 방법으로 ${\beta}$ 입자와 ${\gamma}$ 선을 구별하며 각각의 방사선에 대한 파고 스펙트럼을 측정하였다.
Tachykinin peptide의 구조와 생리활성간의 상관관계를 조사하기 위해 고상법으로 합성한 어류 유래의 neuropeptide $\gamma$(Mammalian-, Bowfin- 그리고 Shark-neuropeptide $\gamma$)를 사용하였다. 이들의 이차 구조를 알아보기 위해 circular dichroism (CD)을 이용하였다. Carp intestine에 서 bowfin-neuropeptide $\gamma$>shark- neuropeptide $\gamma$>mammalian-neuropeptide $\gamma$순으로 활성 이 나타났다. 그러나, guinea-pig 회장과 rat 십이지장에 대해서 mammalian-NP$\gamma$의 활성이 어류 유래의 neuropeptide $\gamma$들보다 높게 나타났다. 이러한 결과들은 neuropeptide $\gamma$가 종-특이적인 활성을 나타냄을 제시한다.

DFA (Difructose anhydride)는 특유의 구조적인 안정성 때문에 당뇨병 환자를 위한 당원으로써 적합하다는 연구가 보고 되어 있다. DFA에는 4가지 type이 있는데 inulin에 의한 DFA I DFA III DFAV가 있고 levan에 의한 DFA IV가 있는 것으로 알려져 있다. 특히 DFA IV는 당뇨병 환자를 위한 당원 뿐 만 아니라 rat을 이용한 연구에서 칼슘의 흡수를 도와 준다는 보고가 있었다. 이러한 DFAIV를 생성하는 데 쓰이는 Microbacterium sp. AL-210에서 유래한 LFTase (Levan fructotransferase)의 wild-type과 mutants (D63A, D195N, N85S)의 구조적 특성을 밝히기 위해 정제하였다. LFTase의 wild-type과 mutants들을 대량 발현시킨 후 흡착 크로마토그래피, 이온교환 크로마토그래피 그리고 젤 여과 크로마토그래피를 이용하여 고순도로 분리 정제하였으며 이를 SDS-PAGE를 통하여 확인하였다. 분리 정제된 단백질을 JNET 이차 구조 예측 프로그램, solubility 측정, CD (원 편광 이색성 분광편광계), fluorescence spectroscopy (형광분석법), DSC (시차주사열량계)를 이용하여 분석하였다. 또한 다중 정렬과 2차 구조 예측 프로그램을 이용하여 wild-type의 2차 구조를 분석하였다. Solubility 측정에서 가장 적합한 온도는 $55^{\circ}C$, 최상의 pH는 7.5로 나타났다. CD 분석에서 wild-type과 비교한 결과 다른 mutant에 비해 N85S의 $\alpha$-helix가 많이 감소한 것과 $\beta$ strand와 random coil이 증가한 것을 확인하였다. 또한 DSC 분석을 통해 wild-type이 다른 mutants에 비해 안정적인 구조를 지닌 것을 확인하였다. 형광분석에서 N85S가 wild-type과 가장 유사하게 나타났으며 D63A와 D195N은 wild-type에 비해 높은 강도를 나타내었다. 또한 wild-type의 sequence를 Exo-inulinase from Aspegillus awamori, a plant fructan 1-exohydrolase from Cichorium intybus 그리고 invertase from Thermotogo maritime (Tm)의 sequence와 다중 정렬한 결과 Exo-inulinase와 높은 identity를 보였다.
The nsp14 protein is an exoribonuclease that is encoded by severe acute respiratory syndrome coronavirus (SARS-CoV). We have cloned and expressed the nsp14 protein in Escherichia coli, and characterized the nature and the role(s) of the metal ions in the reaction chemistry. The purified recombinant nsp14 protein digested a 5'-labeled RNA molecule, but failed to digest the RNA substrate that is modified with fluorescein group at the 3'-hydroxyl group, suggesting a 3'-to-5' exoribonuclease activity. The exoribonuclease activity requires $Mg^{2+}$ as a cofactor. Isothermal titration calorimetry (ITC) analysis indicated a two-metal binding mode for divalent cations by nsp14. Endogenous tryptophan fluorescence and circular dichroism (CD) spectra measurements showed that there was a structural change of nsp14 when binding with metal ions. We propose that the conformational change induced by metal ions may be a prerequisite for catalytic activity by correctly positioning the side chains of the residues located in the active site of the enzyme.
Since heavy metal pollution is a significant global environmental problem and very dangerous to human health, the improved methods for detecting heavy metals are required recently. Colorimetric chemosensors are now considered as one of the most effective analytical method used in the environment monitoring. New direct dyes having the function of colorimetric chemosensors were synthesized. When metal ions such as $Al^{3+}$, $Ca^{2+}$, $Cd^{2+}$, $Cr^{3+}$, $Cu^{2+}$, $Fe^{2+}$, $Fe^{3+}$, $Hg^{2+}$, $Li^+$, $Mg^{2+}$, $Na^+$, $Ni^{2+}$, $Pb^{2+}$ and $Zn^{2+}$ were added each solution of new direct dyes, the color of solution was changed and can be easily detected with naked eyes without expensive experimental equipment such as atomic absorption spectrometer (AAS) or inductively coupled plasma?mass spectrometer (ICP-MS). The new benzidine analogues were diazotized and reacted with couplers such as H-acid, J-acid, Chromotropic acid, Nevill-winther acid and gamma acid to synthesize new direct dyes. The structures of the new direct dyes were confirmed by high resolution mass spectrometer (FAB ionization) and evaluated with UV-Vis spectroscopy. The UV-VIS spectroscopy was measured for the dye solutions by adding various concentrations of metal ions. It was observed that the absorbance in UV-Vis spectra was changed as the heavy metal ions were added.
Seasonal fluctuations of physico-chemical and biological aspects of the environment were studied in Vladivostok harbour (Golden Horn Bay, the East Sea/Sea of Japan). The benthic community structure was described with a focus on size-spectra (bacteria, meio- and macrofauna) related with the chemical environment and chemical fluxes in sediment and to reveal their possible ecological role in the process of bioremediation of the environment. Samples from two sites with different concentrations of heavy metals (Fe, Zn, Cu, Pb, Mn, Cr, Ni Cd, Co) and petroleum hydrocarbon were assessed by a number of methods. These included plate counts of culturable bacteria, observation through a scanning electron (SEM) and transmission electron microscope (TEM). These approaches were complemented with microscopic assessments of the diversity of the benthic community. The specific communities had a limited number of species, tolerant to abnormally high levels of toxic compounds. The dominant species were presented by several sho.1-lived small polychaetes (Capitella capitata) and nematodes (Oncholaimium ramosum). The highest population density was recorded in microbenthos, in various diatoms, various physiological groups of bacteria which participate in biomineralization: marine heterotrophic bacteria, which oxidized oil, black oil in addition to groups resistant to heavy metals. They have the entire set of mechanisms for neutralizing the negative effect of those compounds, forming the detrital food web and biogeochemical circulation of material in sediments, which results in the biological self-recycling of sea basins.
