Effects of Aqua-acupuncture with Woolgum on the Recovery of Liver Injury in Rats (울금약침(鬱金藥針)이 백서(白鼠)의 간손상(肝損傷) 회복(恢復)에 미치는 영향(影響))

  • These studies were carried out to observe the effect of Aqua-acupuncture with Woolgum extract on the recovery from liver injury of rats. The liver injury of rats induced with 0.5ml/200g carbon tetrachloride. The Aqua-acupuncture with Woolgum extract solution inserted into corresponding locus of the Kansu(BL18.) in Rats. Experimental group was divided into 4 groups: Normal group, $CCI_4$-intoxicated group, Saline treated group after $CCI_4$-intoxicated and Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. Biochemical assays for each serum enzyme activity of GOT, GPT, LDH, total protein, triglyceride, albumin, WBC, neutrophils, lymphocyte were performed. The results were as follows : 1. GOT was decreased with statistical significance in the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. In GOT activity, the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated showed higher level significant compared with the saline aqua-acupuncture treated group after $CCI_4$-intoxicated. 2. GPT was decreased with statistical significance in the Woolgum aqua-acupuncture treated after $CCI_4$-intoxicated. In GOT activity, the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated showed higher level significant compared with the saline aqua-acupuncture treated group after $CCI_4$-intoxicated. 3. LDH was decreased with statistical significance in the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. 4. Total protein increased with statistical significance in the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. 5. Albumin was increased with statistical significance in the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. The saline aqua-acupuncture treated group after $CCI_4$-intoxicated was not recognized as compared with $CCI_4$-intoxicated group. 6. Triglyceride was increased with statistical significance in the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. 7. WBC was decreased with statistical significance in the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. In WBC count, the Woolgum aqua-acupuncture higher level significant compared with the saline aqua-acupuncture treated group after $CCI_4$-intoxicated. 8. Lymphocyte was decreased with statistical significance in the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. 9. Neutrophils was increased with statistical significance in the Woolgum aqua-acupuncture treated group after $CCI_4$-intoxicated. The saline aqua-acupuncture treated group after $CCI_4$-intoxicated was not recognized as compared with $CCI_4$-intoxicated group. From above results, it is suggested that the Woolgum aqua-acupuncture had significant effects on the injury induced by $CCI_4$.

  • This is the study of the effects of Aqua-acupuncture with Gardeniae Fructus on thc recovery of rat's liver which was damaged by 0.3ml/ea of $CCI_4$. Rats were divided into 4 groups; Normal-group(None treated group), Control-group(Not treated after $CCI_4$-intoxicated), Exp. I(Treated with Saline Aqua-acupuncture after $CCI_4$-intoxicated) and Exp. ll(Treated with Gardeniae Fructus Aqua-acupuncture after $CCI_4$-intoxicated). Biochemical assays for each serum enzyme activities of AST, ALT, Albumin, LDH, ${\gamma}$-GT, TG and Total cholesterol were performed. The results were summarized as follows: 1. AST activities in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. Albumin in serum increased in the Gardeniae Fructus Aqua- acupurkture treated group after $CCI_4$-intoxicated. 4. LDH in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after CClcintox icated, the Gardeniae Fructus Aqua acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. 5. ${\gamma}$-GT In serum significantly decreased In the Gardeniae Fructus Aqua-acupuncture trea ted group after $CCI_4$-intoxicated. In compan son with Saline-treated group after $CCI_4$-intoxicated, the Crardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated was not recognized significantly. 6. TG in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after $CCI_4$-intoxicated, the Gardeniae Fructus Aqua -acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. 7. Total cholesterol in serum decreased in the Gardenias Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after $CCI_4$-intoxicated, the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. The results from ahove show that the Gardeniae Fructus Aqua- acupuncture has highly effects on the damaged liver caused by $CCI_4$. Therefore it is expected that the Gardeniae Fructus Aqua- acupuncture could be used to cure the damaged liver.

The Effect of Aqua-acupuncture of Total Saponin on the Damaged Liver Induced by Carbon Tetrachloride in Rats (인삼 총 사포닌 약침이 사염화탄소에 의한 랫트 간손상에 미치는 영향)

  • The effect of aqua-acupuncture of total saponin on the recovery of liver function in -intoxicated rats was exemined. Rats were divided into normal, control and three experimental groups (As-I group : aqua-acupuncture of saline to Gansu locus and Aa-III group : aqua-a-cupuncture of total saponin to blank locus at the thigh). Changes of some serum biochemical parameters (ALT, AST and ALP, total protein and albumin, and albumin/globulin (A/G) ratio) were determined. Serum ALT activities were increased by $CCI_{4}$T and decreased in all aqua-acupuncture groups compared with the control group. Aa-I group showed significant decrease of ALT activities on the 2nd (P<0.05) and 3rd (P<0.05) day, respectively. Aa-II group showed significant decrease on the 3rd day (P<0.05) day. Serum AST activities were increased by $CCI_{4}$T and decreased in all apua-acupuncture groups compared with the control group. Aa-I group showed significant decrease on the 1st (P<0.05), 2nd (P<0.001) and 3rd (P<0.001) day, respectively. Aa-II group showed significant decrease on the 3rd (P<0.001) day. Aa-III group showed significant decrese on the 1st (P<0.05) and 3rd (P<0.001) day, respectrvely. Serum ALP activities were increased by $CCI_{4}$T and decreased in all aqua-a-cupuncture groups compared with the control group. Aa-I group showed significant decrease on the 3rd (P<0.01) day. Total protein, albumin and A/G ratio were decreased by $CCI_{4}$T and increased in all aqua-acupuncture groups compared with control group. Aa-I group showed significant increase only on the 3rd day in albumin (P<0.01). It was thought that the aqua-acupuncture of total saponin to Gansu locus was effective on the recovery of liver function in $CCI_{4}$T-intoxicated rats.

Effects of Godulbaegi(Ixeris Sonchifolia H.) Diets on Enzyme Activities of CCI4 Induced Hepatotoxicity in Rats (고들빼기 식이가 간 독성을 유발한 흰쥐의 효소 활성에 미치는 영향)

  • This study was carried out to investigate the effects of enzyme activities on male Sprague-Dawley rats intoxicated by CCI4 on IS(Godulbaegi) diets for 4 weeks. We divides into 5 diet groups which were normal diet(N), normal diet intoxicated by CCI4(NC) and 3 IS diets ; leaves diet(ILC), roots diet(IRC) and mixed diet of leaves and roots which were also injected by CCI4 3 times for 4 weeks. The activity of glutamic pyruvic transaminase(GPT) in serum in NC was higher than in N as we expected. The GPT activites and the values of malondial-dehyde(MDA) of IS groups were all lower than in NC, IC as lowest. The activity of superoxide dismutase(SOD) in NC was higher than in N and IS groups had values less than the values of N. Catalase showed similarity in results as above. The values of glutathione S-transferase(GST) and cytochrome P-450 in NC were lower than in N. IS groups had higher values than in NC. Godulbaegi might be important not only as one of the traditional Korean foods but also as therapeutic agent for hepatotoxicity and for shortening the recovery time in liver disease.

Studies on the Physiological Activities of Caragana chamlagu Roots -Effects on the Hyperlipemia, Hyperglycemia and Liver Damage- (골담초근의 생리활성 -고지질, 고혈당 및 간손상에 미치는 영향-)

  • The studies were attempted to evaluated the therapeutic effects of various fractions(ether, methanol, butanol) of Caragana chamlagu roots on the hyperlipemia induced by feeding the diet containing 1%, cholesterol and 0.5%, cholic acid in rats, and on the hyperglycemia induced by streptozotocin in rats. Also the preventive effects of these fractions were studies on the liver damage in $CCl_4-intoxicated$ rats. The followings were obtained as the results: 1.The butanol fraction was significantly shown to down the serum lipid level in 1%, cholesterol and 0.5%, cholic acid diet-feeding rats and streptozotocin-induced hyperglycemic rats. Cholesterol level in $CCl_4-intoxicated$ rats was reduced in the case of all pre-treated groups. 2.The serum glucose level of streptozotocin-induced hyperglycemic rats was significantly decreased by the administration of various fractions of C. chamlagu roots, and the lipid-peroxidation of pancreas was significantly decreased in the case of administration of these fractions. 3.The activates of s-GOT and s-GPT were decreased by the administration of various fractions, especially in butanol fraction, of C. chamlagu roots in the $CCl_4-intoxicated$ rats. The liver lipid-peroxidation was decreased by administration of 200mg/kg of these fractions in the $CCl_4-intoxicated$ rats. In histological observation, hepatic cellular necrosis and fatty acid deposit were increased remarkably by $CCl_4-intoxication$, but the pretreatment of various fractions of C. chamlagu roots improved the pathological change of parenchymatous cell necrosis and fatty change around centrilobular area of the control.

Protective Effects of Houttuynia cordata Thunb on Carbon Tetrachloride-induced Hepatotoxicity in Rats (어성초 분획물이 사염화탄소로 유발된 흰쥐의 간손상에 대한 보호효과)

  • This study was performed to investigate the protective effect of Houttuynia cordata Thunb on hepatotoxicity in carbon tetrachloride$(CCI_4)$ intoxicated rats. The examined effects hexane, chloroform, butanol and water fractions prepared from the Houttuynia cordata Thunb methanol exlract and rats were administrated with those orally once a day for successive 6 days, fellowed by treaoent with $CCl_4$ on the sixth day. After 6 days, the activities of aminotransferase, alkalinephosphatase, ${\gamma}-glutamyl$ transpeptidase, lactate dehydrogenase and contents of triglyceride, hepatic lipid peroxide in butanol fraction pretreated rats were significantly decreased compared to the only $CCl_4$ treated rats, also depletion glutathione content induced by treatment with $CCl_4$ was prevented by butanol fraction pretreated rats. In addition, activities of hepatic superoxide dismutase, catalase, glutathione peroxidase in butanol fiaction pretreated rats were significantly decreased compared to the only $CCl_4$ treated rats, but the activity of hepatic glutathione-S-transferase was not significantly effect. These results suggest that butanol fiuction of Houttuynia cordata Thunb methanol extract have potent hepatoprotective effect against carbon tetrachloride intoxicated rats.

Changes in Liver and Serum Guanase Activity following Administration of Carbon Tetrachloride in Rats (四鹽化炭素에 依한 肝損傷時 Guanase 活性 變動)

  • This paper was intented to charily the cause of an increase of serum guanase activity in rats following injection of $CCI_4$. The cause of increasing serum guanase was focused on the change of guanase activities in both serum and liver, and additionally, these results were compared with the previously known alanine aminotransferase (ALT). Concomitantly the microscopic investigation on the histologic changes, and the determination of lipid peroxides of liver were combined in this experiment for a correlation to observe that the activity of guanase would be effected by the various degree of hepatic injury induced by $CCI_4$. The serum levels of guanase were increased about 2 fold in the fatty change stage (3-12 days), 5.2 fold representing the peak value in necrosis stage (21days), 4.5 fold in early cirrhosis stage (48 days), and 2 fold in severe cirrhosis stage (92 days). These changes of serum guanase activity showed similar patterns to those of ALT activity and lipid peroxides in liver cell. The changes of liver guanase activities showed an increase, whereas ALT activities in liver were markedly decreased. It is likely that the increase of serum guanase activity is based on the excess leaking of guanase into blood by the result of accelerated enzyme synthesis in liver cell of $CCI_4$ intoxicated rats. In addition, the possibility could not be ruled out, however, that the increase of serum guanase activity would be caused by the alteration of membrane permeability.

Protective Effects of Mesangi (Capsosiphon fulvecens) on Hepatotoxicity in Carbon Tetrachloride (CCI4)-Intoxicated Rats (매생이가 사염화탄소로 유발된 흰쥐의 간손상에 미치는 영향)

  • This study investigated the hepatoprotective effects of mesangi (Capsosiphon fulvecens) in carbon tetrachloride $(CCI_4)$-induced liver injury. Thirty healthy male Sprague-Dawley rats were divided into three groups : the normal (Normal), $CCI_4$-treated $(CCI_4)$, and mesangi-treated (Exp-CF) groups. Administration of $CCI_4$ increased the levels of GOT, GPT, and LDH in serum, while the levels were significantly decreased by the addition of mesangi. Higher levels of HDL-cholesterol were found in the Normal and Exp-CF groups, which has lower levels of total and LDL-cholesterol. Administration of $CCI_4$ also increased IGFBP-1 expression in serum, but it was decreased in the Exp-CF group. This suggests that $CCI_4$ is involved in the change in IGFBP-1 expression via its hepatotoxicity. The results indicate that mesangi has a hepatoprotective effect in rats given $CCI_4$.

Effects of Butanol Extract of Aralia elata on Lipid Peroxidation (두릅나무 부탄올 추출물이 지질 과산화에 미치는 영향)

  • It is well known that lipidperoxide, formed in vivo, induced the denaturation of enzyme and destruction of cell membrane to acute injury of tissue. Aralia elata have physiological activates, the improvement of lipid metabolism, antidiabetic activity etc., which was thought to have the relationship to lipid peroxidation. The anti-lipidperoxidative effect of Aralia elata have not yet established. In this study, we examined the anti-lipidperoxidative effects of Aralia elata (Butanol fraction) on CCI$_{4}$ induced lipidperoxidation in rats, and elucidated the anti-lipidperoxidative mechanism. In rat liver homogenate intoxicated with CCI$_{4}$ (0.5 ml/100g), BuOH fraction of Aralia elata (80 mg/Kg/day) exhibited 85.41% anti-lipidperoxidative effect but in serum 69.63% inhibitory effects, respectively. In mitochondrial and microsomal fraction showed inhibition of 55.85% and 69.30%, respectively. In order to elucidate the mechanism of anti-lipidperoxidation effects of Aralia elata, enzymatic (NADPH dependent) and non-enzymatic (Ascorbic acid catalyzed) reaction, in vitro, were performed. In enzymatic reation, Aralia elata exhibited 59.43% anti-lipidperoxidation effects, but in non-enzymatic reaction exhibited 43.27% inhibition. Therefore, it is noteworthy that antioxidative power of them may mainly results from the inhibition by enzymatic reaction.

The Effect of Betaine on the $CCI_4$-Induced Hepatotoxicity in Rats (사염화탄소에 의하여 유발된 흰쥐의 간 독성에 미치는 비테인의 효과)

  • Effects of betaine, a major component of Lycii Fructus, on carbon tetrachloride intoxicated rats were evaluated. Histological studies showed that betaine had improved the typical necrosis around centrilobular area in liver tissue due to the carbon tetrachloride intoxication. Betaine, whether it was administrated simultaneously or prior to carbon tetrachloride, prevented or retarded the elevation of liver-weight/body-weight ratio due to the carbon tetrachloride intoxication. Betaine also significantly elevated the reduced activities of some enzymes, cytochrome P-450, 7-ethoxycoumarin-0-deethylase and glutathione-S-transferase, involved in xenobiotic metabolism due to carbon tetrachloride intoxication. These results demonstrate a possible hepato-protective role of betaine against fatty liver that could be easily induced by carbon tetrachloride in rat.
