  • 고탁수 장기화 문제를 해결하기 위한 방법으로는 고탁수층을 선택취수하여 우선 배제함으로써 하류하천의 고탁수 발생일수를 최소화하는 방법이 있는데, 이와 같은 선택적 취수기법은 저수지 운영에 있어서 고탁수층을 우선 배제한 후 홍수기 이후에 저수지내로 유입되는 청수를 담수하여 호소 내 탁도 저감 효과를 얻을 수 있다. 본 연구에서는 선택취수 시 성층수괴의 동수역학적 변화를 분석하기 위하여 국내에서 처음으로 Two-tank 기법을 이용하여 성층구조를 구현해 내었는데, 소금물의 밀도변화를 이용하여 수심 1m의 성층수조를 만들었고, 밀도경사가 상이한 성층구조에서 취수조건을 변화시키면서 비교란 유속계인 PIV 시스템을 이용해 유속의 흐름을 Vector와 Contour로 분석하였다. 선택취수에 대한 흐름의 동수역학적 분석결과 취수유량보다는 밀도성층경사 변화에 더 민감한 반응을 보였다. 취수유량을 줄이거나 밀도성층경사를 급하게 했을 때에는 선택취수 영역(withdrawal zone)의 수직방향 폭은 줄어드는 반면, X축으로의 영향범위는 증가함을 나타냈다. 취수유량을 증가시키거나 성층밀도경사를 완만히 했을 땐 선택취수 영역(withdrawal zone)의 수직방향 폭은 증가하였고 X축으로의 영향범위는 축소됨을 나타내었다. 이 결과는 Richardson 수로도 판단되어지는데, Richardson 수가 증가하면 유속에 비해 상대적으로 밀도성층경사가 크다는 것인데 이럴 경우 선택취수 영역(withdrawal zone)의 수직방향 폭 최대가 되고 선택취수 영역(withdrawal zone)의 수직방향 폭은 최소가 된다. 선택취수 영역(withdrawal zone)의 수직방향 폭이 최소가 되면 취수구 직경 D의 1.8배의 값을 가지고 Richradson 수가 최소가 되더라도 취수구 직경 D의 3.3배를 벗어나지 않는다는 결과를 도출할 수 있었다.링 목적으로 사용될 수 있다. 본 연구에서 개발한 영상수위계는 한강홍수통제소 관할의 전류, 청담대교 등 4개소 낙동강 홍수통제소 2개소, 지자체 등에 적용되었으며, 적용 결과 비교적 안정적이면서 정확하게 수위를 측정하는 것으로 나타났다. 한편 기존 CCD 카메라 이외에 CCTV를 이용한 영상수위계를 개발하여 영상의 화질 개선뿐 아니라 하천화상 감시 기능을 강화하였다.소류의 섭취율은 높았다. 집단간의 상관도를 보면 교육별로 김치, 장아찌, 콩이 각각 p>0.5 수준에서 유의한 차가 없었고, 나머지는 유의한 차가 있었다. 연령별로는 멸치가 유의한 차가 없었고(p>0.5), 수입별로는 콩이 유의한 차가 없었다(p>0.5). 4. 영양지식(營養知識) 검토 가정생활(家庭生活)에 필요(必要)한 일반적(一般的)인 영양지식(營養知識)은 대체적으로 낮은 편이었다. 어린이 영양, 편식의 해로움, 비만증의 해로움, 임신부 그리고 수유부 영양에 대하여는 일반적으로 알고 있다고 하였으며, 그다음으로 이유기 영양, 어린이 발육에 필요한 식품, 식품과 영양소와의 관계, 우유의 성분, 노인영양에 대하여 잘 알고 있는 비율이 낮았으며, 인체의 영양소, 식단작성여부, 간식의 이론, 식품감별법에 대하여는 가장 낮은 비율을 나타냈다. 각 영양지식은 교육정도가 높을수록 영양지식이 높았고, 교육별 집단간의 유의한 차가 나타났다.   • 급속한 도시화 및 지구온난화로 인한 집중호우로 홍수피해가 해마다 증가하고 있다. 홍수피해를 최소화하기 위하여 4대강 중심의 홍수예경보시스템이 구축되는 등 다양한 제도적 장치가 마련되고 있으나 중소하천이 분포되어 있는 도시유역에서의 홍수예측기능은 부족한 실정이다. 본 연구에서는 중소 도시하천에 적용 가능한 실시간 도시홍수예측서비스 시스템(Realtime Urban Flood Forecasting Service, U-FFS)을 개발하였다. 경기도 성남에 위치한 탄천을 대상유역으로 선정하고 실시간 강우 및 수위관측소를 설치하여 수문데이타를 수집하였으며 이를 바탕으로 수위예측모형을 구축하였다. 모형구축에는 이미 국내외 학계에서 그 정확도가 입증된 바 있는 Data-driven 모델의 일종인 ANFIS(Adaptive Neuro-Fuzzy Inference System)를 이용하였다. 개발된 수위예측모형은 지정된 시간에 자동으로 작동 가능한 실행파일로 프로그래밍되어 최종적으로 홍수예측 웹서비스와 연동된다.   • In this thesis, a new measurement system is implemented for depth data extraction based on the camera calibration of the known pattern. The relation between 3D world coordinate and 2D image coordinate is analyzed. A new camera calibration algorithm is established from the analysis and then, the internal variables and external variables of the CCD camera are obtained. Suppose that the measurement plane is horizontal plane, from the 2D plane equation and coordinate transformation equation the approximation values corresponding minimum values using Newton-Rabbson method is obtained and they are stored into the look-up table for real time processing . A slit laser light is projected onto the object, and a 2D image obtained on the x-z plane in the measurement system. A 3D shape image can be obtained as the 2D (x-z)images are continuously acquired, during the object is moving to the y direction. The 3D shape images are displayed on computer monitor by use of OpenGL software. In a measuremental result, we found that the resolution of pixels have $\pm$ 1% of error in depth data. It seems that the error components are due to the vibration of mechanic and optical system. We expect that the measurement system need some of mechanic stability and precision optical system in order to improve the system.

  • We obtained precise linear transverse displacements from the results of signal processing on moire fringes measured by the superposition of a fixed self-imaging elongated circular(EC) grating and a moved EC grating. The linear displacement less than one pitch of the moving EC grating was calculated from the position of a bright moire fringe generated on the linear grating part of the EC gratings. The moire signals were high-frequency-filtered, smoothed, curve-fitted, and first-differentiated moire signals in sequence. Also the linear displacement created by movement by the unit of integer times of a pitch was readily measured by the count of the even number of zero-crossing points corresponding to bright moire fringes obtained by the above same processing without curve fitting. Then we can measure linear transverse displacements with the accuracy of better than 3 %, which are more accurate values than those by the traditional visual method within the displacement of 80 ${\mu}{\textrm}{m}$.

Optimization of the re-extraction process of ethanol from catechin components from Korean green tea extract (한국산 녹차추출물 catechin 성분의 ethanol 재추출 공정 최적화)

  • The purpose of this study was to determine the optimum re-extraction conditions of ethanol from catechin in Korean green tea (Camellia sinensis L.) using response surface methodology (RSM). The experiments were carried out according to a five level and two variable central composite design (CCD). The two independent variables were solvent ratio to sample content (1, 4, 7, 10, 13 mL/g) and extraction temperature (-20, -10, 0, 10, $20^{\circ}C$) on the dependent variables including yield, epigallocatechin (EGC), epicathchin (EC), epigallocatechingallate (EGCG), epicatechingallate (ECG), total catechin and caffeine. ANOVA results showed that Coefficients of determination ($R^2$) of estimated models for dependent variables were ranged from 0.9054~0.9778, while $R^2$ of caffeine were estimated 0.8770. The optimum ranges for the maximized extraction including yield, EGC, EC, EGCG, ECG, caffeine and total catechin were 4.5~7.5 mL/g in ratio of ethanol to sample and $-8{\sim}8^{\circ}C$ in extraction temperature. The actual values of yield, EGC, EC, EGCG, ECG, caffeine and total catechin, respectively, at the optimized conditions were 35.02%, 13.31%, 3.978%, 19.11%, 4.29%, 5.30% and 40.68%.

EFFECT OF ANTICURVATURE FILING METHOD ON PREPARATION OF THE CURVED ROOT CANAL USING PROFILE (PROFILE을 이용한 근관형성 시 ANTICURVATURE FILING방법의 영향)

  • This study investigated the effect of anticurvature filing method on preparation of the curved root canal using ProFile. Thirty six resin blocks were divided equally into three groups by instrumentation motions: anticurvature filing motion. circumferential filing motion and straight up-and-down motion. Each resin block was sectioned at 8mm level from the apex and at the greatest curvature of the canal and reassembled in metal mold by a modified Bramante technique. All groups were instrumented with the ProFile system. At each levels. image of sectioned surface were taken using CCD camera under a stereomicroscope at $\times40$ magnification and stored. Distances of transportation at the inner and outer area of curvature and the centering ratio were determined and compared by statistical analysis. along with the assessment of the increase of root canal cross-sectional area. The results were as follows; 1. In all groups. there was no statistical difference in the mean increase of root canal cross-sectional area. the centering ratio. and the mean distances of transportation at the inner area of curvature at each level. 2. At 8mm level from the apex. the mean distances of transportation at the outer area of curvature decreases in following order anticurvature filing motion. circumferential filing motion. straight up-and­down motion but. no significant difference at the greatest curvature of the canal among three groups. Effect of anticurvature filing motion using ProFile does not seem to be different from other instrumentation motions at the inner area of curvature in curved root canal.

Mass Screening of Lovastatin High-yielding Mutants through Statistical Optimization of Sporulation Medium and Application of Miniaturized Fungal Cell Cultures (Lovastatin 고생산성 변이주의 신속 선별을 위해 통계적 방법을 적용한 Sporulation 배지 개발 및 Miniature 배양 방법 개발)

  • For large and rapid screening of high-yielding mutants of lovastatin produced by filamentous fungal cells of Aspergillus terreus, one of the most important stage is to test as large amounts of mutated strains as possible. For this purpose, we intended to develop a miniaturized cultivation method using $7m{\ell}$ culture tube instead of traditional $250m{\ell}$ flask (working volume $50m{\ell}$). For obtaining large amounts of conidiospores to be used as inoculums for miniaturized cultures, 4 components i.e., glucose, sucrose, yeast extract and $KH_2PO_4$ were intensively investigated, which had been observed to show positive effect on enhancement of spore production through Plackett-Burman design experimet. When optimum concentrations of these components that were determined through application of response surface method (RSM) based on central composite design (CCD) were used, maximum spore numbers amounting to $1.9\times10^{10}$ spores/plate were obtained, resulting in approximately 190 fold increase as compared to the commonly used PDA sporulation medium. Using the miniaturized cultures, intensive strain development programs were carried out for screening of lovastatin high-yielding as well as highly reproducible mutants. It was observed that, for maximum production of lovastatin, the producers should be activated through 'PaB' adaptation process during the early solid culture stage. In addition, they should be proliferated in condensed filamentous forms in miniaturized growth cultures, so that optimum amounts of highly active cells could be transferred to the production culture-tube as reproducible inoculums. Under these highly controlled fermentation conditions, compact-pelleted morphology of optimum size (less than 1 mm in diameter) was successfully induced in the miniaturized production cultures, which proved essential for maximal utilization of the producers' physiology leading to significantly enhanced production of lovastatin. As a result of continuous screening in the miniaturized cultures, lovastatin production levels of the 81% of the daughter cells derived from the high-yielding producers turned out to be in the range of 80%$\sim$120% of the lovastatin production level of the parallel flask cultures. These results demonstrate that the miniaturized cultivation method developed in this study is efficient high throughput system for large and rapid screening of highly stable and productive strains.

  • The purpose of this study was to determine the optimum Platycodon grandiflorum root concentrate (PGRC, $65^{\circ}Brix$), fermented P. grandiflorum root extract by Lactobacillus plantarum (FPGRE, $2^{\circ}Brix$), and cactus Chounnyouncho extract (Cactus-E, $2^{\circ}Brix$) for preparation of PGRC stick product with FPGRE using response surface methodology (RSM). The experimental conditions were designed according to a central composite design with 20 experimental points, including three replicates for three independent variables such as amount of PGRC (8~12 g), FPGRE (0~20 g), and Cactus-E (0~20 g). The experimental data for the sensory evaluation and functional properties based on antioxidant activity and antimicrobial activity were fitted with the quadratic model, and accuracy of equations was analyzed by ANOVA. For the responses, sensory and functional properties showed significant correlation with contents of three independent variables. The results indicate that addition of PGRC contributed to increased bitterness and acridity based on the sensory test and antimicrobial activity, addition of FPGRE contributed to increased antioxidant activity and antimicrobial activity, and addition of Cactus-E contributed to increased fluidity based on the sensory test, antioxidant activity, and antimicrobial activity. Based on the results of RSM, the optimum formulation of PGRC stick product was calculated as PGRC 8.456 g, FPGRE 20.00 g, and Cactus-Ex 20.00 g with minimal bitterness and acridity, as well as optimized fluidity, antioxidant activity, and antimicrobial activity.

Assembly and Testing of a Visible and Near-infrared Spectrometer with a Shack-Hartmann Wavefront Sensor (샤크-하트만 센서를 이용한 가시광 및 근적외선 분광기 조립 및 평가)

  • We report the assembly procedure and performance evaluation of a visible and near-infrared spectrometer in the wavelength region of 400-900 nm, which is later to be combined with fore-optics (a telescope) to form a f/2.5 imaging spectrometer with a field of view of ${\pm}7.68^{\circ}$. The detector at the final image plane is a $640{\times}480$ charge-coupled device with a $24{\mu}m$ pixel size. The spectrometer is in an Offner relay configuration consisting of two concentric, spherical mirrors, the secondary of which is replaced by a convex grating mirror. A double-pass test method with an interferometer is often applied in the assembly process of precision optics, but was excluded from our study due to a large residual wavefront error (WFE) in optical design of 210 nm ($0.35{\lambda}$ at 600 nm) root-mean-square (RMS). This results in a single-path test method with a Shack-Hartmann sensor. The final assembly was tested to have a RMS WFE increase of less than 90 nm over the entire field of view, a keystone of 0.08 pixels, a smile of 1.13 pixels and a spectral resolution of 4.32 nm. During the procedure, we confirmed the validity of using a Shack-Hartmann wavefront sensor to monitor alignment in the assembly of an Offner-like spectrometer.

Statistical Optimization of Production Medium for Enhanced Production of Itaconic Acid Biosynthesized by Fungal Cells of Aspergillus terreus (Aspergillus terreus에 의해 생합성되는 이타콘산의 생산성 증가를 위한 통계적 생산배지 최적화)

  • Statistical optimization of the production medium was carried out in order to find an optimal medium composition in itaconic acid fermentation process. Itaconic acid utilized in the manufacture of various synthetic resins is a dicarboxylic acid biosynthesized by fungal cells of Aspergillus terreus in a branch of the TCA cycle via decarboxylation of cis-aconitate. Through OFAT (one factor at a time) experiments, six components (glucose, fructose, sucrose, soluble starch, soybean meal and cottonseed flour) were found to have significant effects on itaconic production among various carbon- and nitrogen-sources. Hence, using these six factors, interactive effects were investigated via fractional factorial design, showing that the initial concentrations of sucrose and cottonseed flour should be high for enhanced production of itaconic acid. Furthermore, through full factorial design (FFD) experiments, negative effects of $KH_2PO_4$ and $MgSO_4$ on itaconic acid biosynthesis were demonstrated, when excess amounts of the each component were initially added. Based on the FFD analysis, further statistical experiments were conducted along the steepest ascent path, followed by response surface method (RSM) in order to obtain optimal concentrations of the constituent nutrients. As a result, optimized concentrations of sucrose and cottonseed flour were found to be 90.4g/L and 53.8g/L respectively, with the corresponding production level of itaconic acid to be 4.36 g/L (about 7 fold higher productivity as compared to the previous production medium). From these experimental results, it was assumed that optimum ratio of the constituent carbon (sucrose) and nitrogen (cottonseed flour) sources was one of the most important factors for the enhanced production of itaconic acid.
