Cholecalciferol 대사연구(代謝硏究)에 관(關)한 일정리(一整理) (Cholecalciferol Metabolism Metabolic Conversion and Mode of Action)

  • Cholecalciferol은 피하에서 자외선과 체온의 작용을 받아 7-dehydrocholesterol로부터 Previtamin D를 거쳐 합성이 되며 이어서 간에 빨리 축적, 25 hydroxylation을 거치게 되고 신장에서 가장 활성을 띤 형태인 $1.25-(OH)_{2}CC$로 변하게 된다. 한편 신장에서는 체내의 Ca, P이 정상으로 존재하게 되면 1-hydroxylation이 억제되는 대신 24-hydroxylation이 일어나 또 다른 active form인 $24,\;25-(OH)_{2}CC$가 되는데 24-hydroxylation의 역활이 무엇인가에 대해서는 아직 구체적으로 밝혀지지 않았다. $24,\;25-(OH)_{2}CC$$1.25-(OH)_{2}CC$는 모두 공통적으로 또 다른 active form인 $1.24,\;25-(OH)_{3}CC$를 형성할 수도 있다. 그중 $1.25-(OH)_{2}CC$는 Steroid H.으로 일컬어지기도 하는 Vit. D metabolite중에서 가장 활성을 가진 형태이며 표적기관으로 크게 소장, 뼈, 신장을 들 수 있다. 소장에서의 역활은 무엇보다도 Ca흡수에 관련되는 것으로 소장에서의 Ca흡수와 Ca BP합성에 관여한다. 골격 형성과 뼈의 mineralization에 관여하는 Vit. D metabolite를의 효과에 관해서는 아직까지 일관성있는 보고가 없다. 한편 $1.25-(OH)_{2}CC$는 side chain oxidation을 거쳐 $CO_{2}$와 미지의 물질을 생성하는데 이 mechanism이 어떤 의미를 갖는지는 분명치 않다. 그밖에 또 다른 표적기관으로서 최근 타액선의 존재가 알려졌으며 Vit. D가 배설되는 경로에 대해서는 새로운 바가 없다. Vit. D가 담즙을 통해서 우선적으로 배설되고 소량이 뇨를 통해 배설되는데 metabolite들의 배설경로는 더욱 규명되어야 할 과제이다.

뇌 정위적 방사선수술 시 Non-Coplanar Volumetric Modulated Arc Therapy의 유용성 평가 (The Evaluation of Non-Coplanar Volumetric Modulated Arc Therapy for Brain stereotactic radiosurgery)

  • 목 적 : 뇌 정위적 방사선수술은 외과적 수술로 인한 합병증 발생 비율이 높은 질환을 비침습적으로 치료할 수 있다. 하지만 뇌 정위적 방사선수술은 방사선을 이용하기 때문에 분할방사선치료와 같이 방사선에 의한 부작용을 동반할 수 있다. 두경부 등의 분할방사선치료 시 방사선에 의한 부작용을 줄이기 위해 Coplanar Volumetric Modulated Arc Therapy(C-VMAT)와 Non-Coplanar Volumetric Modulated Arc Therapy(NC-VMAT)가 주변 정상조직에 미치는 영향에 대한 분석이 이루어졌지만, 뇌 정위적 방사선수술에서는 이러한 내용들이 분석되지 않았다. 본 연구에서는 뇌 정위적 방사선수술을 실시한 환자를 대상으로 C-VMAT과 NC-VMAT을 비교, 분석하여 NC-VMAT의 유용성을 평가하고자 한다. 대상 및 방법 : 뇌 정위적 방사선수술 13건을 대상으로 하였으며, C-VMAT, NC-VMAT 치료 계획을 진행하였다. 치료 계획 용적은 최소 0.78 cc에서 최대 12.26 cc의 용적을 가지고 있었으며, 처방 선량은 15~24 Gy가 처방되었다. 치료 장비는 trueBEAM Stx(Varian Medical Systems, USA)을 사용하였으며, 치료 계획에 사용된 에너지는 6 MV Flattening Filter Free(6FFF) X-선을 이용하였다. C-VMAT 치료계획은 half 2 Arc 또는 full 2 Arc를 사용하여 치료계획을 진행했으며, NC-VMAT 치료계획은 $40{\sim}190^{\circ}$의 Arc를 3~7개 사용하고, couch의 각도 또한 3~7개의 각도로 구성하여 치료계획을 진행하였다. 결 과 : 최대선량의 평균 값은 C-VMAT에서 $105.1{\pm}1.37%$로 나타났으며, NC-VMAT에서는 $105.8{\pm}1.71%$로 나타났다. C-VMAT의 처방 선량 지수(Conformity index)는 $1.08{\pm}0.08$로 나타났고, 선량 균질 지수(Homogeneity Index)는 $1.03{\pm}0.01$로 나타났다. NC-VMAT의 처방 선량 균질지수는 $1.17{\pm}0.1$, 선량 균질지수는 $1.04{\pm}0.01$로 나타났다. 뇌의 $V_2$, $V_8$, $V_{12}$, $V_{18}$, $V_{24}$는 C-VMAT에서 $176{\pm}149.36cc$, $31.50{\pm}25.03cc$, $16.53{\pm}12.63cc$, $8.60{\pm}6.87cc$, $4.03{\pm}3.43cc$로 나타났으며, NC-VMAT에서는 $135.55{\pm}115.93cc$, $24.34{\pm}17.68cc$, $14.74{\pm}10.97cc$, $8.55{\pm}6.79cc$, $4.23{\pm}3.48cc$로 나타냈다. 결 론 : 최대선량과 처방 선량 지수, 선량 균질 지수 항목에서는 C-VMAT과 NC-VMAT이 큰 차이를 보이지는 않았다. 뇌의 선량 당 용적은 $V_2{\sim}V_{18}$까지 구간에서 값의 차이는 최소 0.5 %에서 최대 48 %의 차이가 나타냈다. $V_{19}{\sim}V_{24}$까지 구간에서 값의 차이는 최소 0.4 %에서 최대 4.8 %의 차이가 나타냈다. 방사선괴사(Radionecrosis)가 발생하기 시작하는 $V_{12}$ 용적의 평균 값을 비교해본 결과 NC-VMAT 이 C-VMAT 대비 약 12.2 % 적은 용적을 포함하는 것을 알 수 있었다. 이러한 결과로 볼 때 NC-VMAT 치료 계획을 수립한다면, $V_2{\sim}V_{18}$의 용적을 줄일 수 있기 때문에 방사선괴사를 줄일 수 있을 것이라 사료된다.

TGF-β-activated Kinase-1: A Potential Prognostic Marker for Clear Cell Renal Cell Carcinoma

  • Background: TGF-${\beta}$-activated kinase-1 (TAK1) has been found to be over-expressed in a variety of solid malignancies and related to tumor growth. The aim of this study was to evaluate the expression level of TAK1 in clear cell renal cell carcinoma (ccRCC) and assess its value as a novel prognostic marker. Methods: TAK1 mRNA was assessed in 51 paired ccRCC tissues and adjacent normal tissues (ADTs) by real-time PCR. Tissue TAK1 protein was also assessed in 91 ADTs and 177 samples of ccRCC immunohistochemically for evaluation of relationships with clinical characteristics. Results: RT-PCR showed that TAK1 RNA level was significantly higher in ccRCC tissues than in the paired ADTs and immunohistochemistry confirmed higher expression of TAK1 protein in ccRCC samples compared with ADTs. TAK1 protein expression in 177 ccRCC samples was significantly correlated with T stage, N classification, metastasis, recurrence and Fuhrman grade, but not age and gender. Patients with low TAK1 levels had a better survival outcome. TAK1 expression and N stage were independent prognosis factors for the overall survival of ccRCC patients. Conclusions: Overexpression of TAK1 predicts a poor prognosis in patients with ccRCC, so that TAK1 may serve as a novel prognostic marker.

  • In this present study, the corrosion mitigation effect of conifer cone extract (CC) was examined in the cement mortar to improve the steel rebar (SR) corrosion resistance. The corrosion inhibition properties of the SR embedded in cement mortar (CM) admixed with different percentage (0, 0.5, 1.0, 1.5, 2.0 %) of CC was studied by open circuit potential (OCP) and electrochemical impedance spectroscopy (EIS) tests. This result confirms that the CM with 0.5% of CC added has better corrosion resistance than the blank specimen (0 % of CC). Although, the percentage of CC increase above 0.5%, the CC could yield a negative impact on CM properties in terms of reducing the corrosion resistance due to the reduction of cement hydration reaction. It was highlighted that the SR embedded in CM containing 0.5% of CC had increased corrosion resistance.

간호대학생의 문화적 역량과 문화간호역량 교육요구 분석 (Level of Cultural Competence (CC) and Educational Needs for Cultural Competence in Nursing (CCN) in Undergraduate Nursing Students)

  • Purpose: This study evaluates the level of cultural competence (CC) and educational needs for cultural competence in nursing (CCN) in undergraduate nursing students. Methods: A self-report instrument was used to measure CC and educational needs for CCN. Questionnaires were completed by 330 nursing students in three nursing colleges. Descriptive statistics, a t-test, an ANOVA, Pearson correlation analyses, and multiple regression were used to analyze the data. Results: The mean CC score was 2.99 and the mean CCN educational needs score was 7.1. CC positively correlated with educational needs for CCN. The regression model explained 36% of the variance in CC. Factors affecting CC were school grades, multicultural education experience, frequency of meeting foreigners, length of encounters with foreigners, foreign-language fluency, and degree of interest in multicultural nursing. Conclusion: To strengthen the CC of students, nursing educators should develop an educational program for CCN and incorporate CCN into nursing school curricula.

고체추진제의 고압 연소속도 측정기법 (The Measurement Technique of Burning Rate in Solid Proplellant at High Pressure)

  • 본 연구에서는 내부용적이 200 cc와 700 cc인 closed bomb을 이용하여 30,000 psi까지 HTPB/AP 추진제의 연소 특성을 고찰하였다. 장전 밀도에 따라 closed bomb법에 의해 측정한 결과 연소속도는 $1,000{\sim}5,000$ psi 범위에서 strand burner법에 의해 측정한 추진제의 연소속도와 잘 일치하였으며, 측정 압력이 6,000 psi 부근에서 압력 지수가 급격히 증가하는 현상을 보였다. Closed bomb의 내부용적이 각각 200 cc와 700 cc일 때 측정된 연소속도는 내부용적의 크기에 상관없이 잘 일치하였다.

피트모스 혼합상토에 기비로 혼합된 석회질 비료가 'Red Madness' 페튜니아 플러그 묘 생장과 상토화학성에 미치는 영향 (Effect of pre-planting liming fertilization in peatmoss based substrates on plug seeding growth of 'Red Madness' petunia and changes in soil chemical properties)

  • This research was conducted to investigate the influence of application rate of liming fertilizers on changes in soil chemical properties and growth of 'Red Madness' petunia in plug production. To achieve this, dolomite (DO) with 0, 1.0, 3.5, 8.0 or 13.0 $g{\cdot}L^{-1}$ and calcium carbonate (CC) with 0, 2.0, 2.5, 3.0, 3.5, or 4.0 $g{\cdot}L^{-1}$ were incorporated into peatmoss + vermiculite (1:1, v/v) during the root substrates formulation. The treatments of 3.5 $g{\cdot}L^{-1}$ of DO and 2.5 or 3.0 $gL^{-1}$ of CC had acceptable ranges of pH and EC in soil solution such as 5.6~6.2 and 0.7~1.0 $dS{\cdot}m^{-1}$, respectively. The faster rising of pH was observed in root media containing CC rather than those of DO. This indicates that the solubility of CC is higher than DO. The soil Ca concentrations in all treatments of CC were 1.8 times as high as those of DO. The treatments of 3.5 or 8.0 $g{\cdot}L^{-1}$ of DO had the highest soil Mg concentrations, but all treatments of CC had lower soil Mg concentrations than control treatment indicating that additional application of Mg fertilizers are required. The elevated application rate of DO or CC resulted in the increase of fresh and dry weight. But plant heights were not influenced by application of liming fertilizers. The results of tissue analysis showed that application of DO or CC influenced the $PO_4{^-}P$, Ca and Mg contents, but not influenced the contents of other nutrients such as N, P, Fe, Mn, Zn and Cu.

Evidence for a Common Molecular Basis for Sequence Recognition of N3-Guanine and N3-Adenine DNA Adducts Involving the Covalent Bonding Reaction of (+)-CC-1065

  • The antitumor antibiotic (+)-CC-1065 can alkylate N3 of guanine in certain sequences. A previous high-field $^1H$ NMR study on the$(+)-CC-1065d[GCGCAATTG*CGC]_2$ adduct ($^*$ indicates the drug alkylation site) showed that drag modification on N3 of guanine results in protonation of the cross-strand cytosine [Park, H-J.; Hurley, L. H. J. Am. Chem. Soc.1997, 119,629]. In this contribution we describe a further analysis of the NMR data sets together with restrained molecular dynamics. This study provides not only a solution structure of the (+)-CC-1065(N3- guanine) DNA duplex adduct but also new insight into the molecular basis for the sequence- specific interaction between (+)-CC-1065 and N3-guanine in the DNA duplex. On the basis of NOESY data, we propose that the narrow minor groove at the 7T8T step and conformational kinks at the junctions of 16C17A and 18A19T are both related to DNA bending in the drugDNA adduct. Analysis of the one-dimensional $^1H$ NMR (in $H_2O$) data and rMD trajectories strongly suggests that hydrogen bonding linkages between the 8-OH group of the (+)-CC-1065 A-sub-unit and the 9G10C phosphate via a water molecule are present. All the phenomena observed here in the (+)-CC-1065(N3-guanine) adduct at 5'$-AATTG^*$are reminiscent of those obtained from the studies on the (+)-CC-1065(N3-adenine) adduct at $5'-AGTTA^*$, suggesting that (+)-CC-1065 takes advantage of the conformational flexibility of the 5'-TPu step to entrap the bent structure required for the covalent bonding reaction. This study reveals a common molecular basis for (+)-CC-1065 alkylation at both $5'-TTG^*$ and $5'-TTA^*$, which involves a trapping out of sequence-dependent DNA conformational flexibility as well as sequence-dependent general acid and general base catalysis by duplex DNA.

  • Functional and nutritional components of 7 cultivars of Chinese cabbage (CC; Brassica rapa subspecies campestris) from China were analyzed to compare the effects of soil pH (6.9 vs. 7.6). The CC grew on the soil of pH6.9 (CC-6.9) contained significantly increased amounts (2.3~4-fold) of pectin, crude protein, vitamin C and vitamin E compared to the control. The contents of ash and minerals (Ca, Fe, Na, Zn, K and Mn) were also significantly increased in CC-6.9. However, CC-6.9 contained 16-75% lower contents of reducing sugars, cellulose and crude fat than CC-7.6. CC-7.6 contained more glucosinolates than CC-6.9; gluconasturtiin (20.68 vs. 0.84 $nmol{\cdot}g^{-1}$ wet wt) and gluconapin (202.55 vs. 0.15 $nmol{\cdot}g^{-1}$ wet wt). In conclusion, CC-6.9 had an improved texture (high pectin and low cellulose) and nutritional value (high in protein, Ca, Fe, Zn, vitamin C, and E), whereas CC-7.6 had better taste (high in reducing sugars) and anticancer functionality (high in glucosinolates).

  • Objectives : The present study was designed to investigate corelation between mix proportion of Scutellaria baicalensis (SB) and Coptis chinensis (CC) on lipopolysaccharide (LPS)-induced TYPE-1 interferon production. Methods : I examined TYPE-1 interferon, interferon regulating factor (IRF)-1,7 and interleukin(IL)-10 production on LPS-induced RAW264.7 cells to evaluate inhibitory effect of mix proportion of SB and CC using real time PCR. Results : Mixture of SB and CC regulated TYPE-1 interferon and IRF-1,7 mRNA expression with SB dose dependent manner, while maintained IL-10 mRNA expression on LPS-induced RAW264.7 cells. Conclusion : In mixture of SB and CC, SB plays a key role in reducing TYPE-1 interferon through inactivation IRF-1,7. Furthermore mixture of SB and CC maintained IL-10 mRNA level. Collectively, this results suggest that SB confer beneficial effects in autoimmune diseases clinically.
