1H, 15N, and 13C Resonance Assignments of the Anti-CRISPR AcrIIA4 from Listeria monocytogenes Prophages

  • The CRISPR-Cas system is the adaptive immune system in bacteria and archaea against invading phages or foreign plasmids. In the type II CRISPR-Cas system, an endonuclease Cas9 cleaves DNA targets of phages as directed by guide RNA comprising crRNA and tracrRNA. To avoid targeting and destruction by Cas9, phages employ anti-CRISPR (Acr) proteins that act against host bacterial immunity by inactivating the CRISPR-Cas system. Here we report the backbone $^1H$, $^{15}N$, and $^{13}C$ resonance assignments of AcrIIA4 that inhibits endonuclease activity of type II-A Listeria monocytogenes Cas9 and also Streptococcus pyogenesis Cas9 using triple resonance nuclear magnetic resonance spectroscopy. The secondary structures of AcrIIA4 predicted by the backbone chemical shifts show an ${\alpha}{\beta}{\beta}{\beta}{\alpha}{\alpha}$ fold, which is used to determine the solution structure.

Reliability and Validity of a Korean Version of the Cultural Awareness Scale (K-CAS)

  • Few instruments are available to measure nurses' perceptions of cultural competence in South Korea. Furthermore, the equivalence of factors between original and translated instruments has not been evaluated. The specific aims of this study were to identify the validity and reliability of a Korean version of the Cultural Awareness Scale (K-CAS) and to evaluate the equivalence of factors between the original CAS and the K-CAS. A total of 515 nursing students completed the 26-item K-CAS, 28-item Caffrey Cultural Competence in Healthcare Scale (CCCHS), and eight-item Openness to Diversity and Challenge Scale (ODCS). The K-CAS exhibited good reliability (${\alpha}=.83$) and construct validity by correlating with the CCCHS (r=.344, p<.001) and ODCS (r=.394, p<.001). Confirmatory factor analysis results of the K-CAS confirmed the same factor structure as the original CAS. The K-CAS could be a useful tool to assess the concept of cultural competence among nursing students and nurses.

A Novel Oxidation Model with Photolysis for Degradation of Trichlorobenzenes (TCBs)

  • First- and second-order kinetic oxidation rates of trichlorobenzenes (TCBs) were obtained and compared by a chemical activation system (CAS) which mimics mixed functional oxidase activity. The system consists of EDTA, ferrous sulfate, ascorbic acid, and $H_2O_2$ in potassium phosphdte buffer (monobasic at pH 7.4). The rate of transformation in CAS was enhanced in the presence and absence of catalase in the sequence 1, 2, 3-TCB < 1, 2, 4-TCB < 1, 3, 5-TCB. In general, the rates of degradation were greater in the test media with catalase. The effect of photolysis on the degradation of the TCBs with the CAS were examined. Sensitized photolysis with nitrite, Fenton's reagent, TiO$_2$ and triethylamine (TEA) studied in concert with the CAS demonstrated significant enhancement of the degradation rate of TCBs. Disappearance rates of TCBs in CAS with prior photolysis or prior photosensitization were at least 10-fold higher than the sum of the rate for each single experiment. This study proves that the combination of the CAS and photolysis can be used as a suitable technique for enhancing degradation of TCBs in aqueous systems.

Photosensitization Effect and Structure-Activity Relationship on Mutagenic Potential of Alkylating Agents by 4-(Nitrobenzyl)Pyridine (4-NBP) test (4-(Nitrobenzyl)Pyridine에 의한 알킬화합물들의 잠재적 변이원성에 대한 구조활성 및 광화학효과의 연구)

  • The NBP assay was conducted to determine the photomutagenic or photocarcinogenic potential of alkylating agents. Using a 4-NBP in vitro technique, whereby photochemical treatment on CAS (Chemical Activation System) was performed to investigate the enhancement effect, 20 compounds were shown to undergo alkylating mechanisms with 4-NBP. Chemically meaningful results were obtained with different sets of 20 compounds for the alkylating activities due to the UV irradiation, demonstrating that all of the testing compounds showed increasing photoalkylating effects either in the presence or absence of CAS in comparison with previously reported data, except furoic acid and fumaric acid that showed decreasing effect in the presence of a CAS. Caffeine did not show a meaningful result either. However, these findings demonstrate the effects of potential photoalkylating activity in chemical activation system (CAS) and suggest a potential risk-ranking system for the in vivo assays.

An Analysis of the Cognitive Processes of 5-Year-Old Children : A Focus on a Performance of Cognitive Assessment System Based on Gender, Monthly Age, and Tendencies towards Hyperactivity (만 5세 유아의 인지과정 특성 분석 : 성별, 월령, 과잉행동성향에 따른 CAS 수행 결과를 중심으로)

  • This study investigated the cognitive process of 5-year-old children, with a particular focus on gender, monthly age, and their tendencies towards hyperactivity through the performance of the Cognitive Assessment System (CAS; Das & Naglieri, 1997). The children with tendencies towards hyperactivity were identified based on Conners Teachers' Rating Scale (CTRS). The subjects were 75 five-year-old children in Seoul and surrounding metropolitan areas. Data were analyzed by means of descriptive statistics, an independent sample t-test, Pearson's correlation coefficient, one-way ANOVA, and by K-mean cluster analysis. Our results were as follows : (1) The CAS and CTRS' sub-factors were correlated negatively, except the positive correlation between planning factor and hyperactivity factor. (2) Girls exhibited significantly higher CAS scores in planning & sequential processing than boys. (3) The upper monthly age group (68-71 months) showed significantly higher score in terms of planning than the lower monthly age group (60-63 months). (4) The CAS scores of the children with tendencies towards hyperactivity was lower than that of normal children. (5) The CAS profile of 5-year-old children was divided into 4 groups with distinctive characteristics by means of K-mean cluster analysis.

  • In this paper, we study how to use CAS to solve certain minmax problems. More specifically, we reduce some to the problems about the packing of certain convex sets in the plane, and use Recognize algorithm to obtain good hypotheses for results. This method can be widely used to solve same types of mathematical problems.

  • This letter presents a hybrid conditional access system (CAS) for advanced terrestrial digital multimedia broadcasting (AT-DMB). The proposed architecture is characterized by its use of a unified CAS channel and various communication networks for CAS message transmissions. We implement a prototype CAS based on the hybrid architecture, which improves the CAS message transmission efficiency greatly compared to the existing T-DMB CAS standard and supports various AT-DMB interlayer services more easily and efficiently.

Ginseng total saponin modulates podocyte p130Cas in diabetic condition

  • Proteinuric conditions demonstrate structural and compositional changes of the foot processes and slit diaphragms between podocytes. p130Cas in podocytes serves as an adapter protein anchoring glomerular basement membrane to actin filaments of podocyte cytoskeleton. To investigate the effect of ginseng total saponin (GTS) on the pathologic changes of podocyte p130Cas induced by diabetic conditions, we cultured mouse podocytes under: 1) normal glucose (5 mM, control); 2) high glucose (HG, 30 mM); 3) advanced glycosylation endproducts (AGE)-added; or 4) HG plus AGE-added conditions and treated with GTS. In confocal imaging, p130Cas colocalized with zonula occludens-1 and synaptopodin connecting to F-actin. However, diabetic conditions relocalized p130Cas molecules at perinuclear cytoplasmic area and reduced the intensity of p130Cas. In Western blotting, diabetic conditions, especially HG plus AGE-added condition, decreased cellular p130Cas protein levels at 24 and 48 h. GTS improved such quantitative and qualitative changes. These findings imply that HG and AGE have an influence on the redistribution and amount of p130Cas of podocytes, which can be reversed by GTS.

Structural and dynamic views of the CRISPR-Cas system at the single-molecule level

  • The CRISPR-Cas system has emerged as a fascinating and important genome editing tool. It is now widely used in biology, biotechnology, and biomedical research in both academic and industrial settings. To improve the specificity and efficiency of Cas nucleases and to extend the applications of these systems for other areas of research, an understanding of their precise working mechanisms is crucial. In this review, we summarize current studies on the molecular structures and dynamic functions of type I and type II Cas nucleases, with a focus on target DNA searching and cleavage processes as revealed by single-molecule observations.

Efficient Generation of Human IgG1 Light Kappa Constant Region Knock-in Mouse by CRISPR/Cas9 System

  • Mice with specific modified genes are useful means of studying development and disease. The CRISPR/Cas9 system is a very powerful and effective tool for generating genetically modified mice in a simple and fast manner. To generate human IgG light kappa constant knock-in mice, we tested by microinjection of a mixture of Cas9 protein, single-guide RNA and target homologous recombinant donor DNA into zygotes. We found that the injection of 10 ng/μL of Cas9 protein and crRNA/tracrRNA, rather than single guide RNA, induced the production of knock-in mice more effectively. Thus, our study provides valuable information that will help to improve the production of knock-in mice and contribute the successful generation of humanized Ab-producing mice in Korea.
