외기요법(外氣療法)과 냉수처치(冷水處置)가 화상(火傷) 후 흰쥐의 혈액성분(血液成分)에 미치는 영향(影響) (Effects of Emitted-Qi therapy and Cold water hterapy on Blood Features and Serum in burned Rats)

  • In order to investigate the effects of emitted-qi therapy(EQT) and cold water therapy(CWT) on the inhibition of the blood loss, the protein loss and the electrolyte loss in burned rats. The white blood cell, hematocrit and hemoglobin values in blood, and the total protein, albumin, globulin, $Na^+,\;Cl^-,\;K^+\;and\;Ca^{2+}$ values in serum were measured. The results were obtained as follows; 1. White blood cell count in blood was significantly increased in CWT group compared with control group. 2. Hematocrit value in bolld was significantly increase in EQT and CWT group compared with control group. 3. Hemoglobin value in blood was significantly increased in CWT group compared with control group. 4. Total protein value in serum was significantly increased in CWT group compared with control group. 5. Albumin value in serum was significantly increased in EQT and CWT group compared with control group. 6. Globulin value in serum tended to be increase in EQT and CWT group compared with control group, but it was not significant. 7. A/G ratio in serum was significantly increased in EQT and CWT group compared with contort group. 8. $Na^+$, value in serum was significantly increased in EQT and CWT group compared with ontrol group. 9. $K^+$, value in serum tended to be increased in EQT group compared with control group, but it was not significant. 10. $Cl^-$ value in serum was significantly increased in EQT and CWT group compared with control group. 11. $Ca^{2+}$ value in serum tended to be increased in EQT and CWT compared with control group, but it was not significant. According to the above results, the EQT and CWT inhibited the blood loss, the protein loss and the electrolyte loss. Therefore, it is considered that the EQT and CWT can be applied to the burn therapy.

Expression and Biochemical Characterization of the Bacillus thuringiensis Cry4B ${\alpha}1$-${\alpha}5$ Pore-forming Fragment

  • Tryptic activation of the 130-kDa Bacillus thuringiensis Cry4B $\delta$-endotoxin produced protease-resistant products of ca. 47 kDa and ca. 21 kDa. The 21-kDa fragment was identified as the N-terminal five-helix bundle (${\alpha}1-{\alpha}5$,) which is a potential candidate for membrane insertion and pore formation. In this study, we constructed the recombinant clone over-expressing this putative pore-forming (PPF) fragment as inclusion bodies in Escherichia coli. The partially purified inclusions were composed of a 23-kDa protein, which cross-reacted with Cry4B antibodies, and whose N-terminus was identical to that of the 130-kDa protein. Dissimilar to protoxin inclusions, the PPF inclusions were only soluble when the carbonate buffer, pH 9.0, was supplemented with 6 M urea. After renaturation via a stepwise dialysis, the refolded PPF protein appeared to exist as an oligomer and was structurally stable upon trypsin treatment. Unlike the 130kDa protoxin, the refolded protein was able to release entrapped glucose from liposomes, and showed comparable activity to the full-length activated toxin, although it lacks larvicidal activity These results, therefore, support the notion that the PPF fragment that consists of ${\alpha}1-{\alpha}5$ of the activated Cry4B toxin is involved in membrane pore-formation.

Chemical Composition of Seed in Medicinal Soybean Collected in Korea

  • Production of medicinal soybean [Glycine max (L.) Merrill], characterized with black seed, white stripe at hilum border, yellow cotyledon and small seed, is increasing with increasing consumption. The objective of this study was to investigate the chemical composition of medicinal soybean seed and to provide basic information, for the characterization of these soybeans among genetic resources. Forty-four lines of medicinal soybeans collected from Korea and two control cultivars, 'Hwangkeumkong' (Yellow seed coat) and 'Geomjeongkong l' (Black seed coat) were planted at the Research Farm of the College of Natural Resources, Korea University, located at Namyangju City on May 25, 1996. Seeds of these lines were harvested at full maturity and analyzed for protein, oil, sugar, starch and mineral contents. Mean protein and oil content of the medicinal line seeds were 42.6 and 16.1%, respectively, and those of the control cultivars were in the middle range for protein and oil content. However, sugar and starch content of the medicinal line seeds appeared to be in the lower range of the distributions compared to the control cultivars and were 10.0 and 1.68%, respectively. Mean P, K, Ca, and Mg contents of the seeds of medicinal soybean lines were 15.9, 21.5, 3.11, and 2.81 mg/g, respectively, indicating that these lines had higher P, K, and Mg and lower Ca contents when compared to the control cultivars. Mean Na and Fe contents were 671 and 224 mg/kg, respectively, showing lower Na and similar Fe contents. The observed results provided that chemical compositions of medicinal soybean were, on average, different from those of the general soybean cultivars.

Nutritional evaluation of new alternative types of dog foods including raw and cooked homemade-style diets

  • Background: New alternative types of pet foods such as raw and cooked homemadestyle diets containing human food ingredients have been introduced due to a trend of pet humanization and diversification of consumer needs. Objectives: To evaluate nutritional adequacy of new alternative types of dog foods containing human food ingredients as maintenance diets for dogs. Methods: Eleven homemade-style foods for adult dogs were purchased from online channel in Korea and analyzed to evaluate nutritional adequacy for adult dogs. Nutrients analyzed included crude protein, amino acids, crude fat, fatty acids, and minerals. Results: Crude protein and amino acids in all products satisfied Association of American Feed Control Officials (AAFCO) requirements. Crude fat in one of 11 products did not meet AAFCO requirements. The most deficient minerals were selenium (10 of 11, 90.9%), copper (five of 11, 45.5%), zinc (five of 11, 45.5%), potassium (three of 11, 27.3%), calcium (three of 11, 27.3%), iron (two of 11, 18.2%), and magnesium (one of 11, 9.1%). Six products were not in the range of the recommended Ca:P ratio in AAFCO dog food maintenance nutrient profiles. Conclusions: This study performed nutritional evaluation of raw and cooked homemadestyle foods as maintenance diets for adult dogs. Some nutritional inadequacies were observed including some minerals, Ca:P ratio, and omega-6:omega-3 fatty acid ratio, although three products (26.2%) satisfied the AAFCO standard except selenium. Overall, the data suggest a need for accurate nutritional adequacy statement for consumers based on proper methods to validate the formula.

영유아 보육시설 어린이들의 영양, 건강상태 (Nutrition and Health Status of Day-Care Center Children)

  • Although it is rapidly increasing the number of day-care centers in Korea, the quality of food and nutrition services is not improved sufficiently. The purpose of this study was to investigate the nutrition and health status of children in day-care center. Menus for lunch of 253 centers were collected and the content of nutrients was analyzed. Nutrient intakes from lunch and snacks of 90 children were investigated using weighing method and those from breakfast and dinner by 24-hour recall method surrogated from their mothers. Anthropometric indices of height, weight, skinfold thickness, the ratio of fat were measured. The lunch menus for children contained $437.0{\pm}138.9\;Kcal$, $17.6{\pm}6.7gr$ protein, $153.9{\pm}87.7mg\;Ca$, $3.63{\pm}1.6mg\;Fe$, $164.5{\pm}158.2RE\;Vit\;A$. and $4.46{\pm}3.04mg\;Vit\;B_1$. Energy, Ca and Fe supplied were below one third of RDAs. Mean daily calorie intake of children aged 3 years was $1303.0{\pm}474.0Kcal/day$, $1322.3{\pm}442.4Kcal$ in aged four, $1307.0{\pm}545.9Kcal$ in aged five and $1497.1{\pm}93.5kcal$ in six year of age. Intake of iron, vitamin A and vitamin $B_1$ were below RDAs.

우렁이의 생체부위별 일반성분, 아미노산, 무기질 및 지방산 분석 (Proximate Composition, Amino acid, Fatty acid and Inorganic Matter of Apple Snail)

  • 우렁이 생체에 대하여 일반성분을 분석한 결과 수분함량은 74.8%, 조단백질 12.7% 및 조지방 0.6%의 함량을 보여 고단백 저지방의 식품재료로써 그 이용 가능성을 찾을 수 있었다. 또한 우렁이를 껍질, 내장 및 근육 3부분으로 분류하여 무기성분을 분석한 결과 우렁이에 함유되어 있는 주요 무기성분으로는 P, Ca, Mg 및 Na이었으며, 그 중 껍질 및 내장, 근육모두에서는 Ca이 가장 많이 함유되어 있어 중요한 Ca 급원으로 이용 가능하다. 우렁이 생체의 아미노산을 분석한 결과 arginine이 31.7% 로 가장 높게 나타났으며, 다음으로 alanine 21.2%, 와 glutamic acid 7.1% 순이었다. 아미노산의 경우 식용할 수 있는 근육에서 조개의 감칠맛과 풍미성분인 alanine 과 glutamic acid 조성이 많으므로 가공식품제조에 바람직하였으며, 또한 arginine 역시 매우 풍부 해 건강식품으로도 이용 가능성을 보여 주었다. 지방산의 경우 포화지방산이 생체, 내장에서 각각 5.4,2.8%로 나타났다. 이러한 결과는 우렁이의 이용성 제고 및 가공 제품화에 중요한 기초 자료로 활용될 수 있을 것으로 생각된다.

전분이용성 효모 Sporobolomyces holsaticus의 균체성분 분석에 대하여 (Analysis of Cellular Components of Starch-Utilizing Yeast Sporobolomyces holsaticus)

  • 미생물을 이용한 식량자원개발의 일환으로 분리배양한 전분이용성 효모, Sporobolomyces holsaticus FRI Y-5의 균체성분에 대하여 검토하였다. 효모균체의 일반조 성분으로서 총 탄수화물이 33.08%, 조단백질이 45.63%, 조지방이 20.01%, 회분이 3.04%였으며, 그 밖에 색소수출물이 4.46%였다. 완전세포(Whole cell)의 가용성단백질함량은 cold and hot NaOH extraction 시 40.89%였으며, 이때 조단백질과 가용성단백질 함량으로부터 핵산함량이 7.6%로 추정되었다. 균체단백질의 아미노산 분석결과, 표준단백질에 비하여 필수아미노산 중 threonine, valine, isoleucine이 약간 적었으며 그때 제한아미노산인 valine의 단백가는 89.4였다. 균체지방의 지방산 조성은 $C_{16:0}$이 6.5%였고, $C_{18:0}$이 2.4%, $C_{18:1}$이 81.9%, $C_{18:2}$이 3.2%, $C_{18:3}$이 6.0%였으며, 이 때 $C_{18:1}$이 특이하게 많았다. 또한 이 효모균체는 미량원소로서 Fe나 Ca에 비하여 Mg과 K을 많이 함유하고 있었으며, 각각 365.33mg/100g, 282.75mg/100g이었고, 비타민 중 $Vit.\;B_2$는 3.7mg/100g이나 niacin은 검출되지 않았다. 색소수출물의 흡광도 실험결과, carotenoid계 색소가 확인되었으며 자외선부분에서 태양광에 파괴되지 않는 물질을 발견하였다.

Ribgrass Mosaic Tobamovirus Occurred on Chinese Cabbage in Korea

  • A tobamovirus, Ribgrass mosaic virus (RMV), was identified newly from chinese cabbage (Brassica campestris L. pekinensis) in Korea. Virus disease incidence of RMV on chinese cabbage was 37.9% in alpine area on August in 1993. RMV induced the symptoms of necrotic ring spots, necrotic streak on midrib and malformation. RMV, Ca1 and Ca3 isolate, could infect 35 species out of 45 plants including Chenopodium amaranticolor. Physical properties of RMV Ca1 isolate were very stable as 10.8 over for dilution end point, $95^{\circ}C$ for temperature inactivation point and 18 weeks for longevity in vitro. RMV had the soil transmission rate of 75.0% for the chinese cabbages, 'Chunhawang' and 'Seoul' cultivars. The purified virions of RMV had the typical ultraviolet absorption spectrum of maximum at 260 nm and minimum at 247 nm. RMV of Ca1 isolate was related serologically with antisera of Tobacco mosaic virus (TMV)-Cym, TMV-O and Pepper mottle virus, but not related with antiserum of Odontoglossum ring spot virus. coat protein gene of RMV-Ca1, sized 473 nucleotides, encoded 158 amino acid residues. Nucleotide identity of RMV-Ca1 CP gene was 96.4% with RMV-Shanghai (GenBank accession No. of AF185272) from China and 96.0% with RMV-Impatiens (GenBank accession No. of AM040974) from Germany. Identity of amino acids between RMV-Ca1 and the two RMV isolates was 96.8%. Specific three primers were selected for rapid and easy genetic detection of RMV using Virion Captured (VC)/RT-PCR method.

보리와 완두의 혼파재배에서 혼파비율과 예취시기에 따른 사료가치의 변화 (Changes in Feed Value of Barley and Pea by Different Seeding Rates and Cutting Dates in Mixed Sowing Cultivation)

  • 본 연구는 우리나라 중서부지역인 충청지역에서 사료작물의 새로운 작부체계로 보리와 완두를 혼파재배할 때 기초자료로 이용하고자 혼파비율과 예취시기에 따른 사료가치변화를 구명하기 위하여 수행하였고 결과를 요약하면 다음과 같다. 건물수량은 보리의 파종비율이 높을수록 증가하였으며, 보리와 완두 혼파비율이 85 : 15% 파종구에서 5월 16일에 예취할 때 가장 많았다. 조단백질, 가급태단백질 및 가소화 단백질은 4월 25일 예취시에는 혼파비율간 유의차가 없으나, 5월 2일 예취 이후부터는 완두의 혼파비율이 높을수록 증가하는 경향이었다. 보리의 혼파비율이 높을수록, 예취시기가 늦을수록 ADF와 NDF는 증가하는 경향이었으며, RFV는 오히려 감소하는 경향이었다. TDN함량은 완두의 혼파비율이 높을수록 증가하였으나, 예취시기별로는 5월 9일 예취시까지는 예취시기가 늦을수록 감소하였고 그 이후는 예취시기가 늦을수록 증가하는 경향이었다. P와 Mg함량은 혼파비율간 유의차가 없었으며, 완두의 혼파비율이 높을수록, Ca은 증가하였고, K은 감소하였다. 예취시기별로는 P, Ca 및 K은 예취시기가 늦을수록 감소하였으나. Mg은 5월 9일 예취시기까지는 별 변동이 없었고, 그 이후 예취시 부터는 예취시기가 늦을수록 증가하였다. 에너지함량에서 ENE, NEM 및 NEG는 혼파비율과 예취시기 간에 유의성이 있어 완두의 혼파비율이 높을수록, 예취시기가 빠를수록 증가하였다. 건물중에 각 성분의 함량을 적한 수량에서 보면 단백질, P, Ca, K 및 Mg 등의 무기물 수량은 보리와 완두의 혼파비율이 75 : 25% 파종구에서, ENE, NEL, NEM 및 NEG 등의 에너지 수량과 TDN수량은 보리와 완두의 혼파비율이 85 : 15% 파종구에서 가장 높았다. 예취시기별로는 TDN 수량, 모든 무기물 및 에너지수량은 5월 16일에 예취하는 것이 가장 높았다.

사춘기 혈압에 영향을 미치는 식이 인자 및 혈액과 뇨중 무기 이온 농도 (Dietary Factors and Serum and Urinary Electrolytes Affecting Blood Pressure in Adolescents)

  • Correlations of adolescents' blood pressure with dietary factors and blood or urinary electrolytes were investigated in this study. Through the screening for blood pressures of 960 middle school students aged 13-16 years, 30 students with the mean value of SBP and DBP above 96th percentile(high blood pressure group) and the other 30 with the mean blood pressure between 48th and 52th percentiles(normal blood pressure group) were selected as the subjects. SBP/DBP of the high and talc normal blood pressure groups were $141.8{\pm}9.0$ / $83.6{\pm}5.1$ mmHg and $116.4{\pm}3.5$ / $69.8{\pm}3.7 mmHg$, respectively. The average values of age, weight, height, BMI, degree of relative crude physical activity, and family income were not different between two groups. Among nutrients for which intakes were determined by 24 hour-recall and expressed as percent RDA, intakes of total and animal calcium were lower in the high blood pressure group compared to the normal, and were negatively correlated with both SBP and DBP even when the effects of related general and other dietary factors were excluded. Whereas energy, total, animal and vegetable protein, total and animal lipid, and riboflavin were higher in intake in the high blood pressure group than in fille normal, and they were in positive correlations with SBP and/or DBP. Vitamin A and ascorbic acid intakes were also negatively correlated with DBP, though ascorbic acid intake was not different between two groups. High blood pressure group preferred higher concentration of thin rice gruel than normal group and talc preferred salt concentrations was ill positive correlation with SBP. Serum and urinary levels of Ca, Mg, Ca/Mg ratio, Na, K and Na/K ratio did not show any differences between two groups. However serum Ca/Mg retio was in negative and se겨m Mg was ill positive correlations with SBP, and urinary Na excretion was negatively correlated with SBP. Calcium intake showed negative correlation with serum Ha and Na/K ratio. Our data indicated that dietary intake of Ca, energy, protein, lipid, vitamin A and C as well as salt preference were associated with blood pressure in adolescents. It might be assumed that Ca lowered blood pressure by increasing serum Ca/Mg ratio and decreasing serum Ca/K ratio though urinary excretion of Na.
