PREVALENCE OF STREPTOCOCCUS MUTANS AND STREPTOCOCCUS SOBRINUS IN CHILDREN UNDER 6 YEARS OF AGE (6세 이하의 어린이에서 Streptococcus mutans와 Streptococcus sobrinus의 분포에 관한 연구)

  • Mutans streptococci have been reported to be implicated in dental caries. Of these streptococcal species, Streptococcus mutans and Streptococcus sobrinus are most commonly found in human dental caries. Prevalence of these bacterial species in dental caries is found to be varied in different races and countries. Therefore, importance of these bacteria in dental caries remains to be determined. The present study was peformed to detect S. mutans and S. sobrinus in 52 Korean children with dental caries between 2 to 6 years of age. For the study, plaque samples were collected from caries-active(CA) and caries-free(CF) teeth of each subject. DNA was extracted from the plaques and amplified by polymerase chain reaction(PCR) using primers corresponding to dex and gtf genes. The results obtained from the study were as follows: 1. Of 52 children, 37 children (71.2%) were found to harbor S. mutans and/or S. sobrinus. 2. Of the MS-infected 37 children, 3 children (5.8%) harbored S. mutans and/or S. sobrinus in the CF plaques only, 22 children (42.3%) in the CA plaques only and 12 children (23.1%) in both CF and CA plaques. 3. S mutans and/or S. sobrinus were detected in 34 CA plaques (65.4%), while 15 in CF plaques (28.8%) 4. In the case of CF plaques, 8 plaques (15.4%) were observed to harbor S. mutans only, 6 plaques (11.5%) to harbor S. sobrinus only and 1 plaque (1.9%) to harbor both species. 5. Of CA plaques, 24 plaques (46.2%) were detected to have S. mutans only, 2 plaques (3.8%) to have S. sobrinus only and 8 plaques (15.4%) to have both species. 6. In comparison with the efficiency of two different primers for PCR, it was found that the primer based on gtf gene was more effective in detecting S. mutans, while the primer corresponding to dex gene was better for S. sobrinus. Overall results suggest that MS appears to be important in dental caries of the Korean children, and S. mutans is more closely associated with than dental caries as compared to S. sobrinus.

  • Calcium Carbonate Saturation Index (LI: Langelier Index), an indicator of $CaCO_3(s)$ saturation, indicates corrosiveness of drinking water and it has been used to monitor drinking water conditions in USA, E.U, and Japan. The objective of this research was to measure LI parameters including water temperature, pH, total alkalinity, calcium ion concentration, and electric conductivity, and to evaluate possibility of using LI in domestic system. Results showed that water temperature varied from 2.0 to $26^{\circ}C$ during 15 months, indicating an average annual temperature of $23.9^{\circ}C$. Total alkalinity was from 20 to 45 mg/L. The concentration difference between total alkalinity and $HCO_3{^-}$ value was hardly observed; the concentration of total alkalinity can be replaced by that of $HCO_3{^-}$. Tap water had a medium corrosiveness since LI values were from 2.0 to 0.5. To reduce the corrosiveness and to increase LI values of drinking water, the results of this study showed that chemicals such as $Ca(OH)_2$, $CaCO_3$, NaOH, or $NaHCO_3$ should be added to water treatment plants.

  • Bacteriophage T4 endonuclease V initiates the repair of ultraviolet (UV)-induced pyrimidine dimer photoproducts in duplex DNA. The mechanism of DNA strand cleavage involves four sequential stens: linear diffusion along dsDNA, pyrimidine dimer-specific binding,l pyrimidine dimer-DNA glycosylase activity, and Af lyase activity. Although crystal structure is known for this enzyme, solution structure has not been yet known. In order to elucidate the solution structure of this enzyme NMR spectroscopy was used. As a basis for the NMR peak assignment of the protein, HSQC spectrum was obtained on the uniformly $\^$15/N-labeled T4 endonuclease V. Each amide peak of the spectrum were classified according to amino acid spin systems by interpreting the spectrum of $\^$15/N amino acid-specific labeled T4 endonuclease V. The assignment was mainly obtained from three-dimensional NMR spectra such as 3D NOESY-HMQC, 3D TOCSY-HMQC. These experiments were carried out will uniformly $\^$15/N-labeled sample. In order to assign tile resonance of backbon atom, triple-resonance theree-dimensional NMR experiments were also performed using double labeled($\^$15/N$\^$13/C) sample. 3D HNCA, HN(CO)CA, HNCO, HN(CA)HA spectra were recorded for this purpose. The results of assignments were used to interpret the interaction of this enzyme with DNA. HSQC spectrum was obtained for T4 endonuclease V with specific $\^$15/N-labeled amino acids that have been known for important residue in catalysis. By comparing the spectrum of enzyme*DNA complex with that of the enzyme, we could confirm the important role of some residues of Thr, Arg, Tyr in activity. The results of assignments were also used to predict the secondary structure by chemical shift index (CSI).

Properties of Zeolite A Synthesized by Natural Bentonite (천연 벤토나이트로부터 합성된 제올라이트 A의 특성)

  • Synthetic zeolite was prepared by using of natural bentonite from Kampo area and the application of detergent builder was investigated. The optimum synthetic condition was SiO$_2$/Al$_2$O$_3$= 2, Na$_2$O/A1$_2$O$_3$=1, H$_2$O/A1$_2$O$_3$=30 at 90$^{\circ}C$ for 3hr and it was found by XRD analysis that the zeolite synthesized under this condition was type A. When the zeolite A synthesized under the optimum condition was contacted with 40$^{\circ}$Dh CaCl$_2$solution at 30$^{\circ}C$ for 15min, the cation exchange capacity was 264.9mg CaO/g-zeolite. And the whiteness of the sample was 89% and the mean particle size was 9.95$\mu\textrm{m}$.

Roles of Neutrophil/Lymphocyte and Platelet/Lymphocyte Ratios in the Early Diagnosis of Malignant Ovarian Masses

  • Background: The present study aimed to investigate the utility and importance of the various parameters of complete blood count panel for benign-malignant differentiation of adnexal masses. Materials and Methods: This retrospective study involved 316 patients with documented benign and 253 patients with malignant adnexal masses who underwent primary surgical treatment at a tertiary referral center. Prior to the study, all benign and malignant cases were compared within their own groups and then the benign and malignant cases were compared to each other. For all cases, cut-off, sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV) were calculated for the neutrophil lymphocyte ratio (NLR), platelet lymphocyte ratio (PLR), neutrophil, lymphocyte, platelet and CA-125 parameters, and the results were compared in regards to the groups. Results: NLR, PLR, neutrophil, CA-125, and platelet values were higher in the malignant compared to the benign cases (p<0.01). The lymphocyte value was lower in the malignant cases (p<0.01). No significant differences were found for basophils and eosinophils (p > 0.05). For CA-125, the sensitivity, specificity, PPV and NPV for all cases were 78%, 62%, 62% and 78%, respectively. For NLR, they were 65.6%, 72.1%, 65.3%, and 72.3%, and for PLR, 48%, 81%, 67%, and 66%. Additionally, the sensitivity and specificity were 55% and 77% for CA-125, 66% and 58% for NLR, and 61% and 58% for PLR in early malignant cases. Conclusions: NLR and PLR appear to be useful methods that can be applied together with CA-125 due to the relatively high sensitivity values for the malign-benign differentiation of ovarian masses. Although the specificity of these parameters is lower than CA-125, especially in cases with early malignant ovarian pathology, their sensitivity being higher is promising for the early diagnosis of ovarian cancer. It can be used to detect ovarian malignancies in the early stages, and it will increase the treatment options and improve survival rates.

Recovery of Calcium Phosphate from Sewage Sludge Ash (하수슬러지 소각재로부터 Ca-P 형태의 인 회수)

  • In this study, optimum extraction conditions for phosphorus recovery from sewage sludge ash(SSA) were investigated. For this purpose, an experiment was conducted to determine optimal recovery conditions for Ca-P type phosphorus by using calcium component in the recycled aggregate residue. The phosphorus content of sewage sludge ash was confirmed to be 5.0 %. When $H_2SO_4$ was used as an extract, concentration of 1 N $H_2SO_4$, L/S ratio of 10, and extraction time of 30 min were found to be the optimal extraction conditions. Phosphorus was extracted by using optimal extraction conditions, and then the heavy metals eluted with phosphorus were removed using 1~20 g of cation exchange resin. In 20 g of cation exchange resin, Fe 71.3%, Cu 82.4%, Zn 79.9%, and Cr 15% were removed. After that, the mixing ratio of the calcium extract obtained from the recycled aggregate residue (RAR) was changed to 1:1, 1:5, 1:10. The pH of the SSA to RAR mixture was adjusted to 2, 4, 8 and 12 by the addition of 5 N NaOH to the mixture of 1:5, and the phosphorus was recovered as Ca-P type precipitate. The optimum pH was 8. When recycled aggregate residues were used, the weight of calcium phosphate increased, but the amount of wastewater generated also increased. Therefore, it was concluded that the use of recycled aggregate residue was not economically feasible.

Result Analysis according to Population Distribution Trends when Reagents are Changed (시약 변경 시 모집단 분포 추이에 따른 결과 분석)

  • Purpose In this study, the monthly population distribution was calculated for statistical verification of 10 tests (CA19-9, CA15-3, testosterone, PTH, calcitonin, AFP, CEA, CA72-4, PSA, estradiol) with changed reagents, and the trend of change By comparing and analyzing the results, we want to verify the accuracy of the results and improve the reliability of the test. Materials and Methods From June 2021 to February 2023, for the 10 items in which the reagent was changed, the monthly percentage and standard deviation index before and after the reagent change were compared, respectively. At this time, the reference value was arbitrarily set for the comparison group in consideration of the reference value of the test item, and the groups were subdivided and compared, and the standard deviation index allowed range was -2.0 or more and 2.0 or less. Results For CA19-9, CA15-3, AFP, CEA, and calcitonin 5 test items, the change in monthly ratio in all test sections before and after the reagent change was kept constant. On the other hand, for CA72-4, PSA, testosterone, PTH, and estradiol items, the standard deviation index exceeded the acceptable standard after changing the reagent. CA72-4 test items showed an increasing trend in standard deviation index in the test section exceeding the reference value. In the case of the PSA test item, the maintenance ratio of 0.04 ng/mL was significantly decreased after changing the reagent. The testosterone test item had a standard deviation index of -2.5 in the test section exceeding 10.1 ng/mL after changing the reagent, and the standard deviation index of the PTH test item was out of the acceptable range in all test sections. It was confirmed that the estradiol test item showed an overall increase in the result value. Conclusion Through this study, the continuity and accuracy of the test results could be verified. It is considered that the stability of the test can be secured by analyzing the factors affecting the test result and solving the cause for the test item whose standard deviation index is out of the acceptable standard.

Effect of Nanosilica on the Mechanical Properties and AC Electrical Breakdown Strength of Epoxy/Microsilica/Nanosilica Composite

  • Epoxy/microsilica (65 phr)/nanosilica (0~5 phr) composites (EMNC) were prepared in order to develop a high-voltage insulation material, where phr means parts per hundred relative to the epoxy oligomer. Tensile and flexural tests of the composites were carried out, and the AC electrical breakdown strength was measured, after which all the data were estimated by Weibull statistical analysis. As the nanosilica content increased, the tensile strength increased, and the highest value was 117.7 MPa in the EMNC system with 3 phr nanosilica, which was ca. 10% higher than that of the system without nanosilica. The value then decreased after 3 phr. The flexural strength and AC electrical breakdown strength showed the same tendencies as the tensile strength. The highest value of the flexural strength was 184.6 MPa in the EMNC system with 3 phr of nanosilica, which was ca. 15% higher than that of the system without nanosilica. The strongest value of the AC electrical breakdown strength was 79.0 kV/0.5 mm in the EMNC system with 3 phr of nanosilica, which was ca. 34% higher than that of the system without nanosilica.

Removal of sulfate ion from semiconductor wastewater by ettringite precipitation

  • This study seeks towards an optimal way to control sulfate ions in semiconductor wastewater effluent with potential eco-toxicity. We developed a system based on ettringite (Ca6Al2(SO4)3(OH)12·26H2O). The basic idea is that the pH of the water is raised to approximately 12 with Ca(OH)2. After, aluminium salt is added, leading to the precipitation of ettringite. Lab-scale batch and continuous experiment results with real semiconductor wastewater demonstrated that 1.5 and 1 of stoichiometric quantities for Ca2+ and A3+ with pH above 12.7 could be considered as the optimal operation condition with 15% of sludge recycle to the influent. A mixed AlCl3 + Fe reagent was selected as the beneficial Al3+ source in ettringite process, which resulted in 80% of sludge volume reduction and improved sludge dewaterability. The results of continuous experiment showed that with precipitation as ettringite, sulfate concentration can be stably reduced to less than 50 mg/L in effluent from the influent 2,050 ± 175 mg/L on average (1,705 ~ 2,633 mg/L).

Antibiotic and Heavy Metal Resistance of Coliform Bacteria Isolated from Mineral Water (약수에서 分離한 大腸菌群의 일부 중금속 및 抗生劑耐性에 관한 연구)

  • The purposes of this study were to find out the heavy metal and antibiotic resistant coliform bacteria from mineral water and the resistant factors. For the experiment, mineral water samples were taken from A area and B area during the period from march to July, 1988. The results of the experiment were as follows 1. From mineral water, eleven resistant coliforms and one susceptible coliform were isolated. 2. All resistant isolates harbored diverse plasmids of ranged ca. 14-54kb. 3. Susceptible coliform harbored a only plasmid of ca. 2.8 kb. 4. All resistant isolates harbored common size of plasmid of ca. 14kb. 5. As a result of the transformation and agarose gel electrophoresis experiments, resistant factor was R-plasmid. In conclusion, It is suggested that heavy metal contamination of mineral water is the selective pressure for the plasmid encoding the tolerance. Heavy metal resistance, in some case, is present with antibiotic resistance. Therefore, heavy metal contamination of mineral water induces antibiotic resistant bacteria.
