백서(白鼠)의 족삼리(足三里)에 시술한 비파엽약침(枇杷葉藥鍼)이 천식억제 및 면역조절효과에 미치는 영향 (The Effects of Eriobotryae Folium Herbal-acupuncture at ST36 on ovalbumin-induced asthma in C57BL mouse)

  • To study the effects of EF(Eriobotryae Folium) herbal acupuncture on asthma, we injected EF-HAS into Jok-samni(ST36) of C57BL/6 mice Objective : The aim of this study was to investigate the effect of EF-HA(herbal acupuncture) at ST36 on ovalbumin-induced asthma in mice Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). Experimental groups were treated with concentrations(1%) of EF-HA at Jok-samni(ST36) for the later 8 weeks(3times/week). Result : 1. The weight and total cells of lung of the mice group treated with EF-HA decreased significantly compared with that of Control group. 2. Total Leukocytes and Eosinophils in BALF of the mice group treated with EF-HA decreased significantly compared with those of Control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with EF-HA decreased significantly compared with those of Control group. 4. The concentration of IgE, IL-4, IL-5 in BALF of the mice group treated with EF-HA decreased significantly compared with those of Control group. 5. The concentration of IL-4, IL-5, IL-13 in Serum of the mice group treated with EF-HA decreased significantly compared with those of Control group. 6. The number of $Gr-1^+/CD11b^+,\;CD3^-/CCR3^+,\;CD4^+,\;CD8^+,\;CD3e^+/CD69^+$ cells in the lungs of the mice group treated with EF-HA decreased significantly compared with those of Control group. 7. The cytokine's manifestation of mRNA of the mice group treated with EF-HA with RT-PCR decreased significantly compared with that of Control group. Conclusion : We conclude that EF-HA is effective on OVA-induced asthma of C57BL/6 mouse.

Strain Differences in the Chronic Mild Stress Animal Model of Depression and Anxiety in Mice

  • Chronic mild stress (CMS) has been reported to induce an anhedonic-like state in mice that resembles some of the symptoms of human depression. In the present study, we used a chronic mild stress animal model of depression and anxiety to examine the responses of two strains of mice that have different behavioral responsiveness. An outbred ICR and an inbred C57BL/6 strain of mice were selected because they are widely used strains in behavioral tests. The results showed that the inbred C57BL/6 and outbred ICR mice were similarly responsive to CMS treatment in sucrose intake test (SIT) and open field test (OFT). However, the two strains showed quite different responses in forced swimming test (FST) and novelty-suppressed feeding (NSF) test after 3 weeks of CMS treatment. Only C57BL/6 mice displayed the depression- and anxiety-like behavioral effects in response to CMS treatment in FST and NSF test. Our results suggest that there are differences in responsiveness to CMS according to the different types of strain of mice and behavioral tests. Therefore, these results provide useful information for the selection of appropriate behavioral methods to test depression- and anxiety-like behaviors using CMS in ICR and C57BL/6 mice.

A murine periodontitis model using coaggregation between human pathogens and a predominant mouse oral commensal bacterium

  • Purpose: C57BL/6 mice, which are among the most common backgrounds for genetically engineered mice, are resistant to the induction of periodontitis by oral infection with periodontal pathogens. This study aimed to develop a periodontitis model in C57BL/6 mice using coaggregation between human pathogens and the mouse oral commensal Streptococcus danieliae (Sd). Methods: The abilities of Porphyromonas gingivalis ATCC 33277 (Pg33277), P. gingivalis ATCC 49417 (Pg49417), P. gingivalis KUMC-P4 (PgP4), Fusobacterium nucleatum subsp. nucleatum ATCC 25586 (Fnn), and F. nucleatum subsp. animalis KCOM 1280 (Fna) to coaggregate with Sd were tested by a sedimentation assay. The Sd-noncoaggregating Pg33277 and 2 Sd-coaggregating strains, PgP4 and Fna, were chosen for animal experiments. Eighty C57BL/6 mice received oral gavage with Sd once and subsequently received vehicle alone (sham), Fna, Pg33277, PgP4, or Fna+PgP4 6 times at 2-day intervals. Mice were evaluated at 5 or 8 weeks after the first gavage of human strains. Results: Fnn, Fna, and PgP4 efficiently coaggregated with Sd, but Pg33277 and Pg49417 did not. Alveolar bone loss was significantly higher in the PgP4 group at both time points (weeks 5 and 8) and in all experimental groups at week 8 compared with the sham group. The PgP4 group presented greater alveolar bone loss than the other experimental groups at both time points. A higher degree of alveolar bone loss accompanied higher bacterial loads in the oral cavity, the invasion of not only PgP4 but also Sd and Fna, and the serum antibody responses to these bacteria. Conclusions: Periodontitis was successfully induced in C57BL/6 mice by oral infection with a P. gingivalis strain that persists in the oral cavity through coaggregation with a mouse oral commensal bacterium. This new model will be useful for studying the role of human oral bacteria-host interactions in periodontitis using genetically engineered mice.

Effects of Galgunhaejutang on Alcohol Consumption in C57BL/6 Mice

  • Objectives : Galgunhaejutang extract (GHT) is a Traditional Korean medical herbal mixture that has been tradtionally used to treat alcohol consumption. This study was preformed to evaluate the effects of GHT on alcohol consumption in C57BL/6 mice. Methods : Sixty three C57BL/6 mice were alcohol dependence-induced by limited access paradigm. Water, GHT 0.688 g/kg (GHT-L), or GHT 3.45 g/kg (GHT-H) were administrated for 10 days. The amounts of alcohol consumption for 2 hours, water consumption for 22 hours, food intake for 24 hours and body weight were measured. Results : There weren't significant differences in 2 hours of alcohol consumption, 22 hours of water consumption, 24 hours of food intake and body weight for ten days between vehicle group and GHT-L or GHT-H group. Conclusions : Further studies employing multi-dose and long term administration of GHT (more than 10 days) might be of benefit.

과배란처리(過排卵處理)에 대(對)한 Mouse의 배란반응(排卵反應)의 계통차(系統差) (Strains Difference of Ovulatory Response in Mice to Superovulation-treatment)

  • 본(本) 연구(硏究)는 5계통(系統)(CF#1, C57BL/6, ICR, dd, CHN)의 mouse에 있어서 과배란처리(過排卵處理)에 대한 반응차(反應差)를 조사(調査)하기 위하여 수행(遂行)되었다. 과배란처리(過排卵處理)는 5IU의 PMS를 피하(皮下)에 주사(注射)한 다음 58시간(時間) 후(後)에 5IU의 HCG를 피하주사(皮下注射)하였으며, 배란수(排卵數)의 조사(調査)는 HCG 주사(注射) 24시간후(時間後)에 도살(屠殺)하여 난관내(卵管內)의 난자수(卵子數)를 현미경하(顯微鏡下)에서 계정(計定)하였다. 평균배란수(平均排卵數)는 CF#1, C57BL/6, ICR, dd 및 CHN계통(系統)에서 각각 21.3, 17.9, 20.7, 21.0 및 20.5개였다.

옻나무 추출물이 식이에 의한 비만유도 C57BL/6 mouse에 미치는 영향 (Effects of Rhus verniciflua Stokes (RVS) Extract on Diet-induced Obesity in C57BL/6 mouse)

  • We investigated the effects of Rhus verniciflua Stokes (RVS) on high fat diet- induced obesity in C57BL/6 that received ad libitum exposure to a high fat diet for 8 weeks. Male C57BL/6 mice weighing $20.1{\pm}0.95\;g$ were divided into four groups and the body weight and food intake were weighed for 8 weeks. After the administration of RVS extract, the liver, subcutaneous and retroperitoneal adipose tissue in the mice were weighed and common blood count in whole blood and biochemical parameters in plasma were measured. It was founded that body weight increase was significantly suppressed by the oral administration of RVS extract in group with high fat diet. Weight of subcutaneous adipose tissue and concentration of triglyceride in plasma from mice which were administrated RVS extract were lower than obesity control group. These results suggest that RVS extract may effect on inhibition of diet-induced obesity.

육미지황탕가미방(六味地黃湯加味方)이 C57BL/6JHam-ob/ob mice의 비만(肥滿) 및 제2형 당뇨병(糖尿病)에 미치는 영향(影響) (Antiobese and Antidiabetic Effects of Yookmijihwang-tang-gamibang, a Traditional Polyherbal Formula on the Obese and Type II Diabetic C57BL/6JHam-ob/ob Mice)

  • Objectives : The object of this study was to observe the pharmacological effects of Yookmijihwang-tang-gamibang (Liuweidihuang-tang-jiaweifang, YGB) consisted of 17 types of herbs has been traditionally used in Korean medicine for treating various disorders in clinics, aqueous extracts on the genetically obese and type II diabetic C57BL/6JHam-ob/ob(ob/ob) mice. Methods :Three different dosages of YGB were orally administered, once a day for 28 days to ob/ob mice with ob/ob control and C57BL/6JJms normoglycemic intact mice. Four weeks after treatments of YGB: the changes on the body weight, food consumption, blood glucose levels, leptin and adiponectin contents were observed for monitoring the antiobese and antidiabetic effects of YGB. The effects were compared to those of CLA(conjugated linoleic acid) which improve type II diabetes and inhibit related obesity. Results : After end of 28 days of continuous treatments, ob/ob control showed increases of adipocyte hypertrophy, vasodilated atrophic glomerulus which were detected with marked hyperplasia of pancreatic islets, insulin and glucagon producing cells. These obese and related type II diabetes induced in ob/ob mice were markedly and significantly inhibited by 28 days of continuous treatment of three dosages of YGB. The YGB 50mg/kg showed similar favorable effects on the diabetes and related diabetic complications as compared with CLA 750mg/kg in ob/ob mice of the present study. Conclusions : The results obtained in this study suggest that over 25mg/kg of YGB extracts favorably retarded the obese and type II diabetes in genetically obese and type II diabetic ob/ob mice.

인삼백합탕(人蔘百合湯)이 B16세포에 대한 세포독성능 및 C57BL/6계 생쥐의 폐전이암의 억제에 미치는 영향 (Cytotoxicity and Antitumor Effects of Insambaekhaptang on C57BL/6 Mice Melanoma-induced Lung Metastasis)

  • 연구배경 종양은 그 발생원인과 성장기전이 자세히 밝혀져 있지 않는 질병으로 최근 사망원인의 제1원인으로 급격히 발생빈도가 증가하고 있다. 종양환자의 수는 점점 증가하는 추세이며, 그에 따른 사망자의 수도 늘고 있다. 종양을 치료하는 방법으로 수술요법 면역요법 방사선요법 화학약물요법 골수이식 호르몬요법등이 사용되어 왔으며, 최근에는 전통적인 한약재를 이용한 종양파괴 및 종양의 성장을 억제하는 약재의 연구가 활발히 시도되고 있다. 본 연구에서는 '노수토홍(勞嗽吐紅)'에 사용되는 인삼백합탕(人蔘百合湯)을 사용하여 종양파괴 및 종양의 성장억제능을 연구하고자 하였다. 연구방법 인삼백합탕을 전이암 실험에 다용(多用)되는 B16세포를 대상으로 세포독성을 MTT검사법에 의하여 실험한 후 $IC_{50}$을 측정하였다. 그 후 동물실험으로 C57BL/6에 B16세포를 주입시킨 후 고형암의 중량과 폐에 전이된 흑생종의 집락의 수를 측정하였고, 생존기간의 연장정도를 측정하였다. 연구결과 인삼백합탕 구성약물의 시험관내 세포독성을 측정하기 위하여 MTT검사법으로 측정한 결과 농도에 비례하여 생존율은 감소하였고, $IC_{50}$을 산출한 결과 백출, 홍화, 계피, 인삼등이 낮은 수치를 나타내었다. 인삼백합탕엑스의 시험관내 세포독성은 농도에 비례하여 생존율이 감소하였고, $IC_{50}$$0.0002437{\mu}g/ml$을 나타내었다. B16세포를C57BL/6의 복강에 주입시켜 고형종양의 무게를 측정한 결과 대조군에 비하여 유의성있는 감소를 보였다(p<0.05). B16세포를 C57BL/6의 꼬리정맥에 주입하여 폐전이암을 유발시킨 후 폐의 표면에 생긴 집락의 수를 측정한 결과 대조군에 비하여 유의성있는 감소를 보였다(p<0.001). B16세포를 C57BL/6의 꼬리정맥에 주입하여 폐전이암을 유발시킨 후 생존일수를 측정한 결과 평균치가 대조군은 23일, 투여군은 26일을 나타내어 113%의 증가를 보였다.

C57BL6 생쥐에서 NDM 헤어토닉의 모발성장 촉진 효과 (Effect of NDM Hair Tonic on Hair Growth Promotion in C57BL6 Mice)

  • NDM hair tonic is composed of several plant extracts which are known to be used in oriental medicine. This study was carried out to investigate the effect of NDM hair tonic on hair regrowth in an alopecia model of C57BL6 mice. Hair of six-weeks old mice were trimmed by electric clippers and electric shavers, so as not to damage the skin. The next day, mice without visible scraches were selected, randomized and separated in groups of 6 mice. There were four experimental groups including saline (negative control), 50% ethanol (vehicle control), 3% minoxidil (MXD), and NDM tonic. The test compounds were topically treated with 0.15 $m\ell$ per mouse per day for 2 weeks. The hair regrowth was determined photographically and histologically and the quantity of endocrine factors, IGF-1 and TGF-$\beta$, in the skin of mice was measured by PCR. No clinical signs were found in all animals. The topical treatment of NDM tonic for 2 weeks to dorsal skin accelerated hair regrowth faster than either the controls or MXD treatment. The NDM tonic treatment also promoted hair follicle development and elongation as compared with the controls or MXD treatment. Both NDM tonic and MXD treatment significantly increased the expression of IGF-1 and TGF-$\beta$ in the skin of C57BL6 mice as compared with the controls (p<0.05). These results suggest that NDM tonic has a hair growth activity and be useful for the treatment of baldness or alopecia.

Effects of Gyeongshingangjeehwan 18 on Pancreatic Fibroinflammation in High-Fat Diet-Fed Obese C57BL/6J Mice

  • The polyherbal drug Gyeongshingangjeehwan 18 (GGEx18) from Rheum palmatum L. (Polygonaceae), Laminaria japonica Aresch (Laminariaceae), and Ephedra sinica Stapf (Ephedraceae) has traditionally been used as an antiobesity drug in Korean local clinics. This study investigates the effects of GGEx18 on pancreatic fibroinflammation in high-fat diet (HFD)-fed obese C57BL/6J mice and the molecular mechanism involved in this process. After HFD-fed obese C57BL/6J mice were treated with GGEx18 (125, 250, and 500 mg/kg) for 12 weeks, variables and determinants of obesity, pancreatic inflammation, and fibrosis were measured using histology, immunohistochemistry, and real-time polymerase chain reaction. Administration of GGEx18 at 500 mg/kg/day to obese mice decreased body weight gain, mesenteric adipose tissue mass, and adipocyte size. GGEx18 treatment not only reduced mast cells and CD68-immunoreactive cells, but also decreased collagen levels and ${\alpha}$-smooth muscle actin-positive cells in the pancreas of HFD-fed mice. Concomitantly, GGEx18 decreased the expression of genes for inflammation (i.e., CD68 and tumor necrosis factor ${\alpha}$) and fibrosis (i.e., collagen ${\alpha}1$ and transforming growth factor ${\beta}$) in the pancreas of obese mice. These results suggest that GGEx18 may inhibit visceral obesity and related pancreatic fibroinflammation in HFD-fed obese mice.
