Mast cells and goblet cells have been known to protect the host against parasites. In this study, we examined the response of the mast cells and goblet cells over a period of 6 weeks in the duodenum, jejunum and ileum of C3H/HeN and C57BL/6 mice infected with Echinostoma hortense (E. hortense). In addition, we investigated whether the worm recovery rate of uninfected mice (the control group) or E. hortense-infected mice (the experimental group) was associated with the number of mast cells and goblet cells. The worm recovery rate was higher in the C3H/HeN mice than in the C57BL/6 mice. The number of goblet cells significantly increased in the experimental group of the C3H/HeN and C57BL/6 mice compared with the control group of both strains (P<0.005). Worm recovery peaked 3 weeks after the infection of the C57BL/6 mice and at 2 weeks after the infection of the C3H/HeN mice, and it was higher in the duodenum than in the jejunum and ileum. However, the infected site in the intestine had no relation with worm expulsion. In the C3H/HeN and C57BL/6 mice, the number of goblet cells in the experimental group was significantly higher than that in the control group (P<0.005). The number reached a peak 2 weeks after the infection and it even increased in duodenum, jejunum and ileum. The increased number of goblet cells was retained 6 weeks after infection. The number of goblet cells was higher in the C3H/HeN mice than in the C57BL/6 mice (P<0.01). These results indicate that goblet cells are related with the worm expulsion. Furthermore, immunohistostaining of the antral intestinal walls for lectin showed the significant increase of the number of goblet cells in the experimental group (P<0.001). The high infection rate in the duodenum was found during the early infection. An increased infection rate in the jejunum and ileum was found 3 weeks after infection and the infection rate was higher in the C3H/HeN mice than in the C57BL/6 mice. Taken together, the present study indicates that goblet cells, rather than mast cells, may play critical roles in parasite expulsion.

This experiment investigated the effect of mixed extracts obtained from Mylabris phalerata Pall. and Drynariae Rhizoma on hair growth activity of the normal and spontaneous alopecia areata model of C57BL/6N mice for 16 days. First, we examined morphological regrowth of hair in normal and spontaneous alopecia model of C57BL/6N mice. Second, we examined immunoreactive density of vascular endothelial growth factor(VEGF), c-kit and protein kinase $C-{\alpha}(PKC-{\alpha})$ in skin of normal C57BL/6N mice by immunohistochemical methods. Third, we investigated expression of $TGF-{\beta}$, prolactin and placenta lactogen after topical application of mixed extracts of Mylabris phalerata Pall. and Drynariae Rhizoma to skin by RT-PCR. The results were as follows: Hair growth effect from middle and high concentration of mixed extracts of Mylabris phalerata Pall. and Drynariae Rhizoma was observed in 80% of normal mice in whose hair had been clipped in 15th days. Hair growth effect of all concentrations of mixed extracts of Mylabris phalerata Pall. and Drynariae Rhizoma was observed in 100% of spontaneous alopecia model of C57BL/6N mice in 15th days. Immunoreactive density of VEGF, c-kit and $PKC-{\alpha}$ in skin of all concentrations of mixed extracts of Mylabris phalerata Pall. and Drynariae Rhizoma were strongly stained in epidermis, bulge, secondary hair germ cells and cutaneous trunci m. compare to control group in 10th day. In experimental III group, Immunoreactive density of VEGF, c-kit and $PKC-{\alpha}$ in skin were strongly stained in inner and outer root sheath of skin. The treatment of mixed extracts of Mylabris phalerata Pall. and Drynariae Rhizoma increased the expression of $TGF-{\beta}$, placenta lactogen and prolactin in the skin of normal C57BL/6N mice compared to control group. These experiments suggest that mixed extracts of Mylabris phalerata Pall. and Drynariae Rhizoma may stimulate the topical hair growth activity and it can be useful for treatment of alopecia areata.

Mylabris is the dried body of the chinese blister beetle. The species used in medicine are Mylabris phalerata and M. cichorii. In recent studies, it has been found that Mylabris possesses antitumor properties, increases the number of leukocytes, and has irritant effects on the urinary organs. The crude extracts of Mylabris have been noted for their highly irritant action and other traditional uses of Mylabris include treatment of poor local blood circulation. The active constituent of Mylabris is cantharidin. The chemical is notable for its vesicant properties, but with severe side effects such as nephrotoxicity. This experiment examined the effect of extracts and fractions, obtained from Mylabris phalerata Pall. on hair growth activity of the C57BL/6N mice after topical application to skin. First, we examined the effect of an extracts, obtained from the alcohol extracts of dried Mylabris phalerata Pall. on hair growth activity of the C57BL/6N mice after topical application to skin. Second, we examined on hair growth activity of the cantharidin fraction of Mylabris phalerata Pall. compared to the control and 1% minoxidil groups. Third, we investigated the number of hair follicle and mast cells after topical application of extracts of Mylabris phalerata Pall. to skin for 16 days. The results were as follows: Hair growth effect from the extracts of Mylabris phalerata Pall.(0.312%) was observed in 80% of mice whose hair had been removed in 13 days. Hair growth effect from the extract of Mylabris phalerata Pall.(0.312 and 0.625%) and 1% minoxidil group was observed in 100% of mice whose hair had been clipped in 20 days. Hair growth effect from the cantharidin fraction(0.5%) and water fraction(0.5%) of Mylabris phalerata Pall. was observed in 100% of mice whose hair had been clipped in 24 days. The hair growth effect on the cantharidin fraction(0.125%) was observed to be strong compared with the minoxidil(3%) group, commercial hair growth agents, in mice whose hair had been clipped in 19 days. In the spontaneous alopecia mice model, the hair growth effect from the cantharidin fraction (0.125%) was observed to be strong as compared with the states before the 13 days experiment. These experiments suggest that extracts and fractions of Mylabris phalerata Pall. may stimulate the topical hair growth activity in low doses.

Obesity induced by high-fat diet (HFD) is verified as a strong risk factor and negative prognostic factor for prostate cancer in several genetically engineered mice although it was not examined in the normal mice. To investigate whether HFD-induced obesity can affect the development and progression of cancer in the prostate of normal mice, alterations in the weight and histological structure of the prostate as well as the expression of cancer-related proteins were analyzed in obese C57BL/6N mice fed with 60% HFD for 16 weeks. First, HFD-induced obesity, including an increase in organ weight, body weight, fat accumulation, and serum lipid profile, was successfully induced in C57BL/6N mice after HFD treatment. The total weight of the prostate significantly increased HFD-induced obesity in the model mice compared with the control group. Among the four lobes of the prostate, the weight of the ventral prostate (VP) and anterior prostate (AP) were higher in HFD-induced obesity model mice than in the control group, although the weights of the lateral prostate (DLP) and seminal vesicle (SV) were constantly maintained. In addition, the incidences of hyperplasia and non-hodgkin's lymphoma (NHL) in the histological structure were remarkably increased in HFD-induced obesity model mice, while the epithelial thickness was higher in the same group. A significant increase in the phosphorylation levels of key proteins in the AKT (protein kinase B) signaling pathway was detected in HFD-induced obesity model mice. Therefore, these results suggest that HFD-induced obesity can promote hyperplasia and NHL in the prostates of C57BL/6N mice through the activation of the AKT signaling pathway.

This study investigated the effects of cell cultured Acanthopanax senticosus extract (ASE) supplementation and swimming exercise on lipid profiles and carnitine concentrations in C57BL/6J mice fed high fat diets. Male C57BL/6J mice (n=50), aged 4 weeks, were divided into 5 groups based on exercise and/or ASE supplementation (0.5 g/kg of body weight): normal diet (N-C), high fat diet (H-C), high fat diet non-supplement & exercise (H-NSE), high fat diet supplement & no exercise (H-SNE), high fat diet supplement & exercis (H-SE). Liver nonesterified carnitine (NEC) was significantly higher in the H-SNE group than in the H-C group, and liver total carnitine (TCNE) levels were significantly higher in the H-SNE group than in the H-NSE and H-SE groups. Liver and muscle carnitine palmitoyltransferase-I (CPT-I) mRNA levels tended to be higher with ASE supplementation and/or exercise. These results suggest that supplementation with ASE and/or exercise might have a role in improving lipid oxidation.

To investigate the hair growth activity of fractions and extract of Arisaematis Rhizoma in the hair removed skin of normal and spontaneous alopecia areata model in C57B/6N mice. These experiments were performed with the macroscopic, microscopic, immunohistochemical(VEGF, c-kit, PKC-${\alpha}$, TGF and FGF) and RT-PCR(TGF-${\beta}$, IGF, prolactin and placenta lactogen) methods. The results were as follows: Macroscopic observation after topical application of vehicle, 50% EtOH as control and extract of Arisaematis Rhizoma to the hair removed skin of C57BL/6N mice on the 9th, 11th and 15th day. Extensive hair growth activity was observed in treated group with extract of Arisaematis Rhizoma on the 9th, 11th and 15th day. In Arisaematis Rhizoma extracts treated group, hair follicles of middle stage of anagen was observed and it were grown down to subcutaneous tissue of skin in all the normal mice on 15th day. But in control group, most of hair follicles of telogen phase was observed in skin. The treatment of extract of Arisaematis Rhizoma increased expression of IGF(145%) and placenta lactogen(108%) in the skin of normal C57BL/6N mice on the 11th day compared to control group(100%). But expression of TGF-${\beta}$(90%) and prolactin(91%) decreased in the skin of normal C57B/6N mice on the 11th day compared to control group(100%). After application of fractions(chloroform, ethyl acetate and water fractions) of Arisaematis Rhizoma extract for 9th day, hair growth effect was observed in whole skin area in 50% of normal mice. But in control group, hair growth effect was not observed in whole skin area of normal mice. Immunoreactive density of VEGF, c-kit, PKC-${\alpha$ and FGF in skin of fractions of Arisaematis Rhizoma extracts was strongly stained in epidermis, bulge, secondary hair germ cells, cutaneous trunci m., subcutaneous tissue, root sheath compare to control group on the 9th day. In spontaneous alopecia areata model, The hair growth activity of Arisaematis Rhizoma extrat treated group(75%) was observed to be strong compared to control group(O%) on 7th day. These experiments suggest that fractions and extracts of Arisaematis Rhizoma may stimulate the topical hair growth activity. Thus it can be useful for treatment of alopecia areata.
Objective : This study was carried out to investigate effect of korean medicinal herbs-included hair tonic (MHT) and food (MF) on hair growth in an alopecia model of C57BL/6 mice. Methods : Six-weeks old mice were given laboratory rodent chow diet for 1 week. Then, we used a depilatory for hair removal on mice. The next day, mice were randomized and separated in 3 groups of 6 mice; normal group (N, vehicle epidermal application+normal diet-treated group), positive control group (C, 3% minoxidil sulfate epidermal application+normal diet-treated group), and the treatment group (T, HT epidermal application+ MF diet-treated group). The hair regrowth was determined by photograph, which was taken at 7, 14 and 21 days. And scoring indices, hair density and hair thickness, were evaluated by Folliscope at same day. In addition, the hair regrowth was analyzed insulin-like growth factor (IGF-1) in the dorsal skin of mice. Results : As a result, not only external appearance but also hair density and hair thickness on dorsal skin were increased more in C, T groups than in N group at 21 days with mixed treatment. Therefore, distinct regrowth pattern is appeared by treatment of herbal tonic and natural food for 21 days. Also, the expressions of IGF-1 on dorsal skin were higher in C, T groups than in N group. Conclusion : These results suggested that this herbal hair tonic and natural food has hair growth promoting activities and may be useful for treatment of bald or alopecia.

Objectives This study was carried out to investigate the effect of Ganoderma lucidum extract (GLE) and microneedle therapy system (MTS) on hair growth in an alopecia model of C57BL/6N mice. Methods Five-week old mice were depilated and separated in 4 groups ; CON (50% EtOH), MXD (5% Minoxidil), MTS and GLE + MTS. The treatments were applied twice a week for 3 weeks. The hair growth was determined photographically, the hair density, thickness and length were identified by Folliscope and the weights of body and organs were measured. In dorsal skin tissue, the expression of hair growth-related gene and protein was analyzed by RT - PCR or Western blot. In addition, the hair follicles in the dermis were observed by H&E staining. Results The promotion of hair growth was observed in GLE + MTS and MTS compared to CON. The hair density, thickness and length were also improved in GLE + MTS and MTS compared to CON. The mRNA expression of TGF-${\alpha}$, TGF-${\beta}1$, IGF-1, PRL and PL and the protein expression of VEGF and IGF-1 were increased in GLE + MTS and MTS compared to CON. The hair follicles and hair root growth were improved in GLE + MTS and MTS compared to CON. In the above results, GLE + MTS were more effective than MTS. Conclusions These results suggest that GLE and MTS has a hair growth activity and can be useful for the treatment of alopecia.

Drynariae Rhizoma has been used for promotes mending of the sinews and bone, tonifies the kidney for such symptoms as weak low back and knees, and stimulates the growth of hair as a tinctute for alopecia in oriental medicine. This experiment examined the effect of an acetone extracts of Drynariae Rhizomas(GSB-1), its EtoAc fraction(GB-2) and n-buOH fraction(GSB-3), on hair growth activity of the C57BL/6L mice after topical application to skin. First, We examined on hair growth activity of extracts of Drynariae Rhizomas compare to control and 1 % minoxidil groups. Second, We investigated on the number of hair follicle and mast cells after topical application of extracts of the Drynariae Rhizomas to skin for 16 day. Third, We investigated immunoreactive density of vascular endothelial growth factor(VEGF), protein kinase C-${\alpha}$(PKC-${\alpha}$) and stem(mast) cell factor(SCF) in skin of C57BL/6N mice by immunohistochemical methods. The results were as follows : Hair growth effect of acetone extracts of Drynariae Rhizomas, its EtoAc fraction and n-BuOH fraction was observed in 98 %, 96 % and 60 % in hair removed skin area in 16 day respectively, Immunoreactive density of VEGF in skin of GSB-1 group was weakly stained compare to control group in 10 day, But GSB-2 and GSB-3 groups were mildy stained in bulge and root sheath of skin. Immunolocalization of SCF antigens was observed weakly stained density in epidermis, bulge, stem cells and dermal papilla of control gruop. but in experimental group, immunoreactivity of SCF antigens was observed mildly stained density in bulge, epidermis and root sheath of GSB-1 gruop, heavily stained density in epidermis, bulge and root sheath of GSB-2 and GSB-3 groups to the hair removal skin of C57BL/6N mice on day 10. These experiment suggest that acetone extracts of Drynariae Rhizomas and its EtoAc fraction may be used for topical treatment of alopecia areata.
Chitosan derivative, of a sulfated N-acetyl chitosan was synthesized, and the inhibitory effects of this compound on the experimental and spontaneous lung metastallc B16/BL6 melanoma bearing mice were investigated. Position of substitution with sulfate in water-soluble sulfated derivatives of chitosan were analysed by 13C-nmr. The structure of N-acetyl chitosan 3,6 0-disulfate were confirmed. The tumor growth inhibition of B16/BL6 melanoma cells has been shown at the highest level of 77.6% when sulfated N-acetyl chitosan were administered at the dose of 100mg/kg. In the lung metastasls, the sulfated N-atetyl chitosan was administered to C57BL/6B mice bearing B16/BL6 melanoma cells by I.V. injection and the number of metastasis foci of melanoma were decreased by the dose dependent manner ranging from 20 to 100mg/kg. In the spontaneous metastasis, I.V. administrations of sulfated N-acetyl chitosan after tumor inoculation resulted in marked reduction of metastatic colonies. A sulfated N-acetyl chitosan was able to partially inhibit the tumor cell adhesion by migration to laminin. These results suggested that chitosan derivative, a sulfated N-acetyl chitoasn was able to inhibit to the experimental and spontaneous metastasis models as well as cell adhesion ability.
