Metastatic cancer is one of the main causes of cancer-related death since they rarely respond to available treatments. There is epidemiologic evidence that high garlic consumption decreases the incidence of cancer. Recent studies of our laboratory have revealed that a garlic-extracts is effective in suppressing metastasis. For experimental metastasis, C57BL/6 mice were injected intravenously with melanoma B16F10 cells in the tail vein, and were orally administered various concentrations (0, 50, 100 or 200 mg/kg body weight) of garlic hexane extract (GHE) for 21 days. The incidence and the area of the melanoma cell colony occupied by the poorly differentiated carcinoma were significantly lower in dose-dependent in 50, 100 and 200 mg/kg BW GHE - treated mice compared with control mice. In conclusion, the results of the present study show that GHE administration prevents lung metastasis in C57BL/6 mice.
Objective : This study was carried out to investigate effect of korean medicinal herbs-included hair tonic (MHT) and food (MF) on hair growth in an alopecia model of C57BL/6 mice. Methods : Six-weeks old mice were given laboratory rodent chow diet for 1 week. Then, we used a depilatory for hair removal on mice. The next day, mice were randomized and separated in 3 groups of 6 mice; normal group (N, vehicle epidermal application+normal diet-treated group), positive control group (C, 3% minoxidil sulfate epidermal application+normal diet-treated group), and the treatment group (T, HT epidermal application+ MF diet-treated group). The hair regrowth was determined by photograph, which was taken at 7, 14 and 21 days. And scoring indices, hair density and hair thickness, were evaluated by Folliscope at same day. In addition, the hair regrowth was analyzed insulin-like growth factor (IGF-1) in the dorsal skin of mice. Results : As a result, not only external appearance but also hair density and hair thickness on dorsal skin were increased more in C, T groups than in N group at 21 days with mixed treatment. Therefore, distinct regrowth pattern is appeared by treatment of herbal tonic and natural food for 21 days. Also, the expressions of IGF-1 on dorsal skin were higher in C, T groups than in N group. Conclusion : These results suggested that this herbal hair tonic and natural food has hair growth promoting activities and may be useful for treatment of bald or alopecia.
Objectives : The purpose of this study is to confirm the suppression effect of Asthma and Immune response improvement of Fasciculus Vascularis Luffae Herbal-acupuncture into Chok-samni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks Two experimental groups were treated with different concentrations(1%, 0.1%) of FVL-HA at Chok-samni(St36) for the later 8 weeks(3times/week). Results : 1. Lung weigh of the mice group treated with FVL-HA decreased significantly compared with that of control group. 2. Total cells of lung, total Leukocytes and Eosinophils in BALF of the mice group treated with FVL-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice group treated with FVL-HA in Photomicrographs decreased significantly compared with those of control group. 4. The concentration of IL-l3, IgE, IL-4 In serum and IL-4 in BALF of the mice group treated with FVL-HA decreased significantly compared with that of control group. 5. The number of Gr-1+/CB11b+, CD3-/CCR3+, CD4+, CD8+, CD3e+/CD69+, IgE+/B220+ cells in the lungs of the mice group treated with FVL-HA decreased significantly compared with those of control group. 6. The cytokine's manifestation of mRNA of the mice group treated with FVL-HA with RT-PCR decreased significantly compared with that of control group. Conclusion : These result suggests that Fasciculus Vascularis Luffae Herbal-acupuncture Choksamni(St36) in C57BL/6mice may be an effective part to OVA-induced asthma in C57BL/6 mice.
$\beta$-Immunan, a proteoglycan, was isolated from the mycelium of Canoderma lucidum which belongs to a medicinal mushroom. The effects of $\beta$-Immunan on cell-cell and cell-matrix interactions mediated by carbohydrate-recognition and the mechanism responsible for the inhibition of experimental metastasis of Bl6-F10 and B16/BL6 murine melanoma were studied. The results showed that $\beta$-Immunan inhibited Bl6-Fl melanoma cell's adhesion to laminin and asialofetuin-induced homotypic aggregation and reduced invasion against Bl6-F10 murine melanoma cells through matrigel in vivo assay. When $\beta$-Immunan was intraperitoneally administrated to C57B/6 mice bearing B16/BL6 murine melanoma cells, it was decreased the number of pulmonary metastatic colony by the dose dependent manner ranging from 20 to 100 mg/kg/day. The results indirectly indicate that clinical treatment with $\beta$-Immunan might be expected to exhibit anti-metastatic effect. In the pulmonary metastasis, the number of pulmonary metastatic colony of melanoma when $\beta$-Immunan was intraperitoneally administrated to C57BL/6 mice bearing B16/BL6 murine melanoma cells by intravenous injection were decreased by the dose dependent manner ranging from 20 to 100 mg/kg/day.
Objectives : Bombyx batryticatus L. is the dried larval form of the silkworm (Bombyx mori L.) infected by Beauveria bassiania (Bals.) Vuill. It is used as a food and medicinal resource to treat asthma, headaches, epilepsy, and convulsions in traditional Korean and Chinese medicines. However, the research of the toxicity about B. batryticatus is not enough yet. Here, we investigate the effects of potential subacute toxicity following the repeated oral administration of B. batryticatus water extract to C57BL/6 mice, at various doses of 0, 50, 150, and 450 mg/kg/day during a two-week period. Methods : The following parameters were examined during the study period: body weight, gross findings, clinical signs, organ weight, hematology, serum biochemistry, histopathology, and mortality. At the end of the treatment period, all the mice were euthanized. Results : No changes were observed in the body weights, gross findings, clinical signs, organ weights, and mortality after two weeks of administration of the B. batryticatus extract. In addition, compared with the normal control group, no noticeable treatment-related changes were observed in the hematological, serum biochemical, and histopathological parameters in the treated group following treatment with doses of up to 450 mg/kg/day. Conclusion : Based on these findings, we conclude that the treatment of mice with the water extract of B. batryticatus did not cause considerable C57BL/6 toxicity, and therefore, it could be considered safe for further pharmacological studies.

Total leukocytes and eosinophils in BALF of the mice group treated with FF-HA decreased remarkably compared with those of control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with FF-HA decreased significantly compared with those of Control group. 4. The concentrations of IgE, IL-4, IL-5 in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with FF-HA decreased significantly compared with those of control group 5. The number of Gr-l+/CDllb+, CDll+ cells, CCR3+ cells, CD4+ cells, and CD3e+/CD69+ in the lungs of the mice group treated with FF-HA decreased significantly compared with those of control group. 6. The mRNA expression of TNF-a, IL-4, IL-5, IL-13 in the mice group treated with FF-HA with RT-PCR decreased remarkably compared with those of control group. Conclusion : These result suggests that Farfarae Flos Herbal-acupuncture at Joksamni(ST36) in C57BL/6mice may be an effective part to OVA-induced asthma in C57BL/6 mice.
This study was conducted to evaluate the hair-growth promotion effect in C57BL/6 mice of a new substance mixed with Allium cepa (red)-bioconversion extract and Angelica gigas Nakai. The ethanol extract of Allium cepa (red) was bioconverted through the use of the Bacillus subtilis KJ-3 (BS3) strain, which was named Red-BCQ. The quercetin content of Red-BCQ increased by about 7.4-fold after bioconversion. Angelica gigas Nakai extract (Agnex) contains a large amount of coumarins such as decursin (D) and decursinol angelate (DA). A 1 mg portion of Agnex contained 0.4146 mg of D and 0.3659 mg of DA. Minoxidil has been known to promote hair growth. In this study, the hair-growth promotion effects of Red-BCQ, Agnex, and a mixture of both Red-BCQ and Agnex were compared with 5% minoxidil. Twenty-five mice were divided into five experimental groups including saline (CON), 5% minoxidil (PCON), Red-BCQ (RA), Agnex (AG), and a Red-BCQ-Agnex mixture (RAG)-treated group. Samples were administered orally once a day at a fixed time for 4 weeks. Hair growth was monitored by photograph at 7, 14, 21, and 28 days. We also observed 5α-reductase, alkaline phosphatase (ALP), γ-glutamyl transpeptidase (γ-GT), insulin-like growth factor (IGF)-1, transforming growth factor (TGF)-β1, antioxidant enzyme, and the hair follicles of the skin tissue. In all the results, the RAG-administered group showed greater antioxidative and hair-growth promotion effects than the other groups. These data suggest that RAG has potent stimulating activity on hair growth in C57BL/6 mice.
Objectives : Increasing evidence supports the biological and pharmacological activities of essential oils on the central nervous system such as pain, anxiety, attention, arousal, relaxation, sedation and learning and memory. The purpose of present work is to investigate the protective effect and molecular mechanism of cypress essential oil (CEO) against scopolamine (SCO)-induced cognitive impairments in C57BL/6 mice. Methods : A series of behavior tests such as Morris water maze, passive avoidance, and fear conditioning tests were conducted to monitor learning and memory functions. Immunoblotting and RT-PCR were also performed in the hippocampal tissue to determine the underlying mechanism of CEO. Results : SCO induced cognitive impairments as assessed by decreased step-through latency in passive avoidance test, relatively low freezing time in fear conditioning test, and increased time spent to find the hidden platform in Morris water maze test. Conversely, CEO inhalation significantly reversed the SCO-induced cognitive impairments in C57BL/6 mice comparable to control levels. To elucidate the molecular mechanisms of memory enhancing effect of CEO we have examined the expression of brain-derived neurotrophic factor (BDNF) in the hippocampus. CEO effectively elevated the protein as well as mRNA expression of BDNF via activation of cAMP response element binding protein (CREB). Conclusions : Our findings suggest that CEO inhalation effectively restored the SCO-impaired cognitive functions in C56BL/6 mice. This learning and memory enhancing effect of CEO was partly mediated by up-regulation of BDNF via activation of CREB.

Hwanggumgung (HGG) is a hair-care product which is composed of several plant extracts used in oriental medicine. This study was carried out to investigate effect of HGG on hair regrowth in a shaving model of C57BL6 mice. Five-week-old mice were acclimated for 1 week under 23$\pm$3$^{\circ}C$, 50$\pm10\%$ relative humidity and 12 h of a light/dark cycle before beginning experiment. There were four experimental groups including distilled water (D.W., control), 10$\%$ ethanol (EtOH, vehicle control), a positive control of 3$\%$ minoxidil (MXD), and HGG for female and male mice, respectively; Six-weeks old mice were trimmed by electric clippers so as not to damage the skin. The next day; mice without visible scraches were selected, randomized and separated in groups of 11 mice. The test compounds were topically treated with 0.15ml per mouse per day for 21 days. The hair regrowth was photographically and histologically determined during the experimental period of 21 days. Enzyme activities of $\gamma$-glutamyl transpeptidase and alkaline phosphatase were also determined using a rate assay method. There were no clinical signs in all experimental groups. The topical application of 3$\%$ MXD and HGG in female mice promoted hair regrowth earlier and faster than the control groups. In male mice, the topical application of 3$\%$ MXD and HGG also accelerated hair growth compared with the controls. Ten percent ethanol also promoted hair growth faster than D.W group. The histology of hair growth in experimental groups was strongly associated with the hair regrowth. 3$\%$ MXD and HGG promoted elongation of hair follicles compared with the controls in both female and male mice. Activities of alkaline phosphatase and $\gamma$-glutamyl transpeptidase, enzymes related to hair growth, significantly increased after treatments of 3$\%$ MXD and HGG for 2 weeks in both female and male mice (p < 0.05). These results suggest that HGG has hair growth promoting activities and it can be for treatment for alopecia.
Objective : Recently, hair has become one of the important aspects of beauty. So, there are many studies about prevention and treatment of alopecia. Finasteride and minoxidil have been known to drug for alopecia treatment. However, these agents have side effects such as irritation, pruritus, and erythema when they were used for a long time. So, we assessed hair growth effect on herbal mixture (RAA) include in Rehmanniae Radix Preparata, Achyranthis Radix, and Acanthopanacis Cortex. Methods : 6 weeks aged C57BL/6 mice were divided three treatment groups. : CON group (vehicle solution), MNXD group (positive control, 3% minoxidil), RAA group (15% RAA solution). And we applied 200 ㎕ of three groups to shaved dorsal skin every day. Hair growth effects of treatment were determined through phototrichogram by folliscope and hair follicle morphometry by H&E staining. And we assessed hair growth-related gene (VEGF, IGF-1, TGF-β1) expressions by western blot and 5-α reductase II analysis of dermal (skin) or internal organ (prostate gland). Results : Hair density and hair follicle size in MNXD or RAA group was enhanced compared to those of CON group, respectively. Also, the protein expression levels in dermal of VEGF, IGF-1 increased but TGF-β1 decreased in RAA and MNXD group, compared to CON group, respectively. 5-α reductase II levels of tissues in MNXD or RAA group significantly decreased compared to those of CON group, respectively. Conclusion : These results suggest that RAA has the promoting effect on hair growth.
