Purification and Characterization of Anticarcinogenic Compound from Corni fructus (산수유에 함유된 항암물질의 정제 및 특성)

  • Chloroform layer from methanol extract of Corni fructus (Cornaceae) showed strong antiproliferation effect on human cancer cell lines by SRB assay. Anticarcinogenic-active compound was isolated and purified by silica gel column and thin layer chromatograpies, and identified as ursolic acid ($3{\beta}$-hydroxyrus-12-ene-28-oic acid, MW:456) by mass and IR spectrophotometries, and $^1H-and\;^{13}C-NMRs$. The compound inhibited proliferation of A549 (human lung cancer cell line) and MCF-7 (human breast cancer cell line) cells in dose-dependant manner when treated for 48 hr. Inhibition rates of both cells were over 40% and 90% compared with control cells at the $30\;{\mu}g/mL\;and\;100\;{\mu}g/mL$, respectively. Morphology of cells treated with the compound for 15 hr at $10\;{\mu}g/mL$ was distorted with shrinked cell mass, and cell number was lower than that of control cells. Cell cycle analysis showed sub-G1 phase arrest in both cell lines following 15 hr exposure to the compound; % of cell phase increased to 11.7 and 11.2% compared to the control of 4.0% and 2.1% in A549 and MCP-7 cells, respectively.

Effect of Sinapis alba L. on expression of interferon-gamma and interleukin-4 production in anti-CD3/anti-CD28-stimulated CD4(+) T cells (CD4+ T cells에서 백개자가 IFN-$\gamma$와 IL-4 생성에 미치는 영향)

  • Objective : Sinapis alba L. (SA) is a korean traditional herbal medicine that is usually used to prevent or treat inflammatory diseases, such as respiratory infection and rheumatoid arthritis. However, the effects of SA supplementation in vitro on serum antibody levels, splenocyte and peritoneal macrophage immune responses have not yet been determined. In this study, we examined the effect of SA on the production of Th1/Th2 cytokines. Methods : Splenocytes were isolated from naive C57BL/6 mice. Cells were enriched for CD4+ cell populations by first staining the cells with anti-CD4 (BD PharMingen, Calif, USA). CD4+ T cells were selected on a (CS) column, and the flow-through was collected as CD4+ T cells. Isolated cells were activated by overnight incubation on 24-well plates coated with $1{\mu}g/mL$ anti-CD3, $1{\mu}g/mL$ anti-CD28 and with SA ($100{\mu}g/mL$). Primary macrophages were collected from the peritoneal cavities of mice (8-week-old female C57BL/6). The peritoneal macrophages were washed and plated with RPMI-1640 overnight for the experiments. After 48-hours cultures, samples were centrifuged at 2000 rpm for 10 minutes, and the supernatants were stored at $-80^{\circ}C$. Mouse IL-4, IFN-$\gamma$ and TNF-$\alpha$ were quantified using ELISA kits (BioSource International, Camarillo, Calif, USA) according to the manufacturer's protocols. Results : SA at 100ug/ml decreased the generation of Th1 cytokine (IFN-$\gamma$) by 0.5-fold. However, SA has no effect on Th2 (IL-4) production. Conclusions : These results suggest that SA may play an important role in the control of T-cell-mediated autoimmunity by down-regulation of Th1 cytokine (especially IFN-$\gamma$, TNF-$\alpha$). These data may contribute to the design of new immunomodulating treatments for a group of autoimmune diseases.

Helicobacter Pylori CagA and Gastric Carcinogenesis

  • Objectives: This study aimed to demonstrate the tyrosine phosphorylation motif (TPM) and 3' region structure of the Helicobacter pylori CagA gene as well as its SHP-2 binding activity in AGS cells and relation to gastric carcinogenesis. Methods: Sixteen clinical isolate H. pylori strains from eight duodenal ulcer and eight gastric adenocarcinoma patients were studied for CagA repeat sequence EPIYA motifs, C-terminal structure, and western blot analysis of CagA protein expression, translocation, and SHP-2 binding in AGS cells. Results: Except for strain 547, all strains from the gastric adenocarcinoma patients were positive for CagA by PCR and had three EPIYA copy motifs. Western blotting showed that all strains were positive for CagA protein expression (100%), CagA protein translocation (100%), and SHP-2 binding (100%). CagA protein expression was significantly higher in the gastric adenocarcinoma patients than in the duodenal ulcer patients (P=0.0023). CagA protein translocation and SHP-2 binding in the gastric adenocarcinoma patients were higher than those in the duodenal ulcer patients, but no significant differences were found between the two groups (P=0.59, P=0.21, respectively). Conclusions: The TPMs and 3' region structures of the H. pylori CagA gene in the duodenal ulcer and gastric adenocarcinoma patients have no significant differences.

Seasonal Changes of the Phytoplankton and the Periphyton Community at the Suer Stream in Kwangyang (전남 광양의 수어천 수역에 있어서 식물플랑크톤과 부착조류 군집의 계절적 변화)

  • Seasonal changes of the phytoplankton and the periphyton community were investigated from August 1998 to April 1999 at five stations at the Suer stream in Kwangyang. A total of 112 species of phytoplankton were identified. Of those, the diatoms were present at all stations but the green algae, the bluegreen algae, and the dioflagellates were present at Station 4 and Station 5 more frequently than the other stations. The phytoplankton standing crops varied from 10,100 cells/1 at Station 4 in April 1999 to 1,489,100 cells/1 at Station 4 in October 1998. The seasonal variation patterns of phytoplankton standing crops were different among stations as well as the pattern of presence. The dominant species were as follows: Achnanthes minutissima, Aulacoseira distans v. alpigena, Cocconeis placentula v. lineata, Cymbella minuta, C. silesiaca, Fragilaria arcus v. recta, Peridinium cinctum, Rhizosofenia longiseta, Synedra rumpens and filamentous algae. Of those, Achnanthes minutissima, Rhizosolenia longiseta, Synedra rumpens and filamentous algae showed the highest rate of occupation in the phytoplankton standing crops during the investigated periods. A total of 99 species of periphyton were identified. Among those, the diatoms of the periphyton community were observed frequently rather than those of the phytoplankton community. The ecological indicator values showed ${\bate}$-mesosaprobous in saprobity and was close to eutraphentic in trophic state. There were no considerable differences between the ecological indicator values by planktonic diatoms and periphytic diatoms.

  • Boron was predeposited into p (100) Si wafer at 94$0^{\circ}C$ for 60minutes to make the back surface field. High tempreature diffusion process at 1145$^{\circ}C$ for 3 hours was immediately followed without removing boron glass to obtain high surface concentration Back boron was annealed at 110$0^{\circ}C$ for 40minutes after boron glass was removed. N+ layer was formed by predepositing with POCI3 source at 90$0^{\circ}C$ for 7~15 minutes and annealed at 80$0^{\circ}C$ for 60min1es under dry Of ambient. The triple metal layers were made by evaporating Ti, Pd, Ag in that order onto front and back of diffused wafer to form the front grid and back electrode respectively. Silver was electroplated on front and back to increase the metal thickness form 1~2$\mu$m to 3~4$\mu$m and the metal electrodes are alloyed in N2 /H2 ambient at 55$0^{\circ}C$ and followed by silicon nitride antireflection film deposition process. Under artificial illumination of 100mW/$\textrm{cm}^2$ fabricated N+PP+ cells showed typically the open circuit voltage of 0.59V and short circuit current of 103 mA with fill factor of 0.80 from the whole cell area of 3.36$\textrm{cm}^2$. These numbers can be used to get the actual total area(active area) conversion efficiency of 14.4%(16.2%) which has been improved from the provious N+P cell with 11% total area efficiency by adding P+ back.

Characteristics of MCMB Anode with VGCF As a Conducting Agent for LPB (LPB용 MCMB 부극 도전재로 VGCF를 첨가한 부극의 특성 연구)

  • An investigation upon the characterization of MCMB anodes with different vapor grown carbon fiber (VGCF) content for application in lithium polymer battery(LPB) was carried out. When VGCF material was used as conducting agent with MCMB active material, the impedance and the initial coulombic efficiency of test cells were found to decrease with the increasing amount of VGCF. On the other hand, as a function of added VGCF the discharge capacity and the utilization linearly with increased. Impedance of test cell with MCMB anode containing $6wt\%$ VGCF exhibited the lowest value whereas the impedance of $8wt\%$ VUF contained anode was similar to that of MCMB anode without VGCF. Interestingly, $6wt\%$ VUF contained anode showed the best battery characteristics. Internal resistance and rate capacity of the cell were. respectively, $0.918{\Omega}\;at\;25C\;and\;93\%$ at 2C. Generally, rate capability and the cycleability of MCMB based test cells with $4\~6wt\%$ VGCF content exhibited better results than the other cells. In the case of $6wt\%$ VGCF containing anode, the discharge capacity of the cell faded slowly with an ultimate charge-discharge cycling capacity of 178mAh/g at the 100th cycle. Thereafter, the discharge capacity faded negligibly and the utilization of the cell at the 100th cycle was more than $90\%$. The effect of addition of VGCF is discussed in detail.

The Improvement of skin barrier function and anti-obesity effect of Codonopsis lanceolata by supercritical carbon dioxide extraction

  • The root of Codonopsis lanceolata has been used in traditional medicine. This study was conducted to confirm the comparative effect of ethanol solvent extraction (CLE) and supercritical carbon dioxide extraction (CLS) of C. lanceolata roots. CLS had higher antioxidant than CLE. For supercritical co-solvent modified carbon dioxide extraction (CLS), it were extracted at 250 bar 50℃ 150 min at a flow rate of ethyl alcohol 3 mL/min for 90min. In addition, CLS inhibited the adipocyte differentiation of 3T3-L1 cells. When treated with the extract at a concentration of 100 ㎍/mL, the Wnt/β-catenin pathway reporter luciferase activity of HEK 293-TOP cells increased approximately by 3-folds compared to that of the untreated control group. Also, the treatment by CLS (50 ㎍/mL) showed a significant increase of involucrin expression. These results indicate that supercritical carbon dioxide extract of C. lanceolatamay serve as a cosmeceutical agent for improving skin barrier function and the treatment of obesity.

Face recognition using a sparse population coding model for receptive field formation of the simple cells in the primary visual cortex (주 시각피질에서의 단순세포 수용영역 형성에 대한 성긴 집단부호 모델을 이용한 얼굴이식)

  • In this paper, we present a method that can recognize face images by use of a sparse population code that is a learning model about a receptive fields of the simple cells in the primary visual cortex. Twenty front-view facial images form twenty persons were used for the training process, and 200 varied facial images, 20 per person, were used for test. The correct recognition rate was 100% for only the front-view test facial images, which include the images either with spectacles or of various expressions, while it was 90% in average for the total input images that include rotated faces. We analyzed the effect of nonlinear functon that determine the sparseness, and compared recognition rate using the sparese population code with that using eigenvectors (eigenfaces), which is compact code that makes contrast with the sparse population code.

Effects of C18 Fatty Acids on Intracellular $Ca^{2+}$ Mobilization and Histamine Release in RBL-2H3 Cells

  • To investigate the underlying mechanisms of C18 fatty acids (stearic acid, oleic acid, linoleic acid and ${\alpha}$-linolenic acid) on mast cells, we measured the effect of C18 fatty acids on intracellular $Ca^{2+}$ mobilization and histamine release in RBL-2H3 mast cells. Stearic acid rapidly increased initial peak of intracellular $Ca^{2+}$ mobilization, whereas linoleic acid and ${\alpha}$-linolenic acid gradually increased this mobilization. In the absence of extracellular $Ca^{2+}$, stearic acid ($100{\mu}M$) did not cause any increase of intracellular $Ca^{2+}$ mobilization. Both linoleic acid and ${\alpha}$-linolenic acid increased intracellular $Ca^{2+}$ mobilization, but the increase was smaller than that in the presence of extracellular $Ca^{2+}$. These results suggest that C18 fatty acid-induced intracellular $Ca^{2+}$ mobilization is mainly dependent on extracellular $Ca^{2+}$ influx. Verapamil dose-dependently inhibited stearic acid-induced intracellular $Ca^{2+}$ mobilization, but did not affect both linoleic acid- and ${\alpha}$-linolenic acid-induced intracellular $Ca^{2+}$ mobilization. These data suggest that the underlying mechanism of stearic acid, linoleic acid and ${\alpha}$-linolenic acid on intracellular $Ca^{2+}$ mobilization may differ. Linoleic acid and ${\alpha}$-linolenic acid significantly increased histamine release. Linoleic acid (C18:2: ${\omega}$-6)-induced intracellular $Ca^{2+}$ mobilization and histamine release were more prominent than ${\alpha}$-linolenic acid (C18:3: ${\omega}$-3). These data support the view that the intake of more ${\alpha}$-linolenic acid than linoleic acid is useful in preventing inflammation.

Inhibitory Effects of Verapamil and TMB-8 on Tonic Contraction Are Accompanied by Inhibition of Phospholipase C Activity in Intact Gastric Smooth Muscle Cells

  • Gastric smooth muscle of guinea pigs was used to investigate whether the inhibitory effect of calcium antagonists on tonic contraction was accompanied by inhibition of phospholipase C activity. Tonic contraction and $[^{3}H]$ inositol phosphate (IP) formation in response to acetylcholine were measured after pretreatment with verapamil, nifedipine, 8-(N,N-diethylamino)octyl 3,4,5-trimethoxy-benzoate (TMB-8) or EGTA. Verapamil $(10\;{\mu}M)$, TMB-8 $(10\;{\mu}M)$ or EGTA (2 mM) significantly inhibited acetylcholine $(1\;{\mu}M)$-stimulated tonic contraction but nifedipine (100 nM) did not. Acetylcholine dose-dependently increased the formation of $[^{3}H]IP$. This effect was not observed in the presence of 2 mM EGTA. Both verapamil and TMB-8 significantly inhibited $[^{3}H]IP$ formation induced by $10\;{\mu}M$ acetylcholine, whereas nifedipine did not. In a subsequent study, we measured phospholipase C activity in gastric muscle cell homogenate and in permeabilized cells to determine whether calcium antagonists could inhibit the activity directly. The calcium antagonists did not change the phospholipase C activity of the cell homogenate or the permeabilized cells. But EGTA decreased phospholipase C activity by 50%. These results suggest that the inhibitory effects of verapamil and TMB-8 on acetylcholine-stimulated tonic contraction may be accompanied by inhibition of phospholipase C activity.
