Isolation and Identiffication of Acinetobacter koreensis sp. nov. from Jang-Baek Waterfall (장백 폭포에서 분리한 Acinetobacter koreensis sp. nov.의 보고)

  • Two isolates of genus Acinetobacter were obtained from Jang-Baek waterfall in North Korea. Morphological characteristics of the isolated 2 strains were Gram-negative, aerobic and rod shape bacteria. Physiological and biochemical characterization of the isolated 2 strains were some different aspect from those of type strains. 16S rDNA sequence analysis showed that the two isolates shared 99.9% sequence similarity. Strains JB10 and $JB15^{T}$ were shown to belong to the Gammaproteobacteria and showed the highest levels of sequence similarity to Acinetobacter tandoii $4N13^{T}$ (97.3%), Acinetobacter haemolyticus $ATCC17906^{T}$ (97.2%), Acinetobacter johnsonii $DSM6963^{T}$ (97.2%), Acinetobacter junii $DSM6964^{T}$ (96.7%), Acinetobacter schindleri $LUH5832^{T}$ (97.0%) and Acinetobacter ursingii $LUH3792^{T}$ (96.6%). The major cellular fatty acid in Acinetobacter type strains and isolated strains included $C_{18:1}\;{\omega}9c\;and\;C_{16:1}\;{\omega}7c/C_{15:0}\;iso\;2OH$. Eventhough it was ascertained that the isolated strains were closely related to genus Acinetobacter, physiological and biochemical characteristics and the result of the isolated strains 16S rDNA analysis indicate some different aspects from those of type strains of genus Acinetohacter It is considered that the isolated JB10 (=KEMC 52-093) and $JB15^{T}\;(=KEMC\;52-094^{T})$ strains be new species of genus Acinetobacter. We name it as Acinetobacter koreensis sp. nov.

Association Analysis of the Essential Hypertension Susceptibility Genes in Adolescents: Kangwha Study (청소년 고혈압 관련 유전자의 연관성 분석: Kangwha Study)

  • Objectives : In this study we examined the association between the genetic markers ACE (A-240T, C-93T, I/D, A2350G), AGT (M235T), AT1R (A1166C), CYP11B2 (T344C, V386A), REN (G2646A), ADRB2 (G46A, C79G, T47C, T1641), GNB3 (C825T) and ADD1 (G460W) and the presence of essential hypertension in adolescents. Methods : The Kangwha Study is an 18-year prospective study that is aimed at elucidating the determinants of the blood pressure level from childhood to early adulthood. For this study, we constructed a case-control dataset of size of 277 and 40 family trios data from the Kangwha Study. For this purpose, we perform a single locus-based case-control association study and a single locus-based TDT (transmission/disequilibrium test) study. Results : In the case-control study, the single locus-based association study indicated that the ADD1 (G460W) (p=0.0403), AGT (M235T) (p=0.0002), and REN (G2646A) (p=0.0101) markers were significantly associated with the risk of hypertension. These results were not confirmed on the TDT study. This study showed that genetic polymorphisms of the ADD1, AGT and REN genes might be related to the hypertension in Korean adolescents. Conclusions : This study provided useful information on genetics markers related to blood pressure. Further study will be needed to confirm the effect of the alpha adducin gene, the angiotensinogen gene and the renin gene on essential hypertension.

  • The temperature sensitive (ts) mutation on RNA1 gene of Saccharomyces cerevisiae prevents growth at restrictive temperature ($36^{\circ}C$) by accumulation of precursor tRNA, rRNA and mRNA (Hutchison et al., 1969; Shiokawa and Pogo, 1974; Hopper et al., 1978). RNA1 gene was cloned by complementation of the temperature sensitive growth defect of an rna1-1 mutant strain and identified by retransformation and concomitant loss of recombinant plasmid on non-selective condition. By deletion mapping, it was found that RNA1 gene resides within 3.5kb of BgII fragment.

A Study on the Environmental Characteristics of a Two-storey Pig House for the Control of Manure (고상식돈사의 분뇨처리에 관한 연구)

  • This research was conducted to examine proper manure treatment methods for a high-rise swine barn with results of the fact-finding survey on manure treatment methods used in high-rise swine barns and sawdust swine barns In Korea. The average temperatures on bedding of control, T1, and T2 were between $25.3{\sim}25.4^{\circ}C$ with little difference. Air flow rate of T2 was slower than those of control and T1. Dust generation in control was 54 cpm which was higher than T1 and T2. The average ammonia concentration in control was highest showing 6.0 ppm, ranged from 0.5 to 14.5 ppm, with statistical difference (p<0.05), compared to T1 (2.8 ppm) and T2 (2.6 ppm). Sawdust spreading used in control and T1 decreased ammonia concentration. T1 showed the lowest concentration of $CO_2$. Water contents of control and T2 were close to the proper water content, 65%. T1 had the highest water content and pH, and the lowest organic matter and C/N ratio. The volumes of sawdust spread per head were 0.26 and $0.27m^3$ in control and T2, respectively, which was less than T1. Operating cost such as an electricity bill for blowers was cheap in T2. Hence, T2 bedding sawdust up to 10 cm thick and periodically spreading additional sawdust from second story was recommended.

In situ Hybridization for the Detection and Localization of the Bitter Taste Receptor Tas2r108 in the Murine Submandibular Gland

  • Mammals have 3 pairs of major salivary glands i.e., the parotid, submandibular, and sublingual glands. Saliva secretion of these glands is modulated by taste perception. Salivary glands are composed mainly of acinar and ductal cells. Primary saliva is secreted by acinar cells and modified during ductal flow. Recently, of the murine 35 bitter taste receptors, Tas2r108 was expressed at highest levels in the submandibular gland by qPCR. Further, Tas2r108-transfected cells respond to a range of bitter compounds, such as denatonium, quinine, colchicine, diphenidol, caffeine and dapson. The objective of the present study was to characterize the expression of Tas2r108 mRNA in acinar and/or ductal cells of the submandibular gland using in situ hybridization (ISH). Male 42-60 days old DBA2 mice were used in the study. Messenger RNAs were extracted from the submandibular gland for generating digoxigenin (DIG) labeled-cRNA probes. These probes were transcribed in anti-sense and sense orientation using T7 RNA polymerase. Dot blot hybridization was performed using DIG labeled-cRNA probes, in order to estimate integrity and optimal diluting concentration of these probes. Subsequently, ISH was performed on murine submandibular gland to detect Tas2r108 mRNA. Dot blot hybridization data demonstrated that Tas2r108 DIG labeled-cRNA anti-sense probes specifically detected Tas2r108 cDNA. ISH results showed that the anti-sense probes labeled acinar and ductal cells in the submandibular gland, whereas no staining was visible in sense controls. Interestingly, the Tas2r108 expression levels were higher in acinar than ductal cells. These results suggested that Tas2r108 might be more associated with primary saliva secretion than with ductal modification of saliva composition.

The results of this paper are the extensions of the classical weighted Fock space $F^p_{2,{\alpha},t}$.

  • When we consider a life insurance company that sells a large number of continuous T-year term life insurance policies, it is important to find an optimal strategy which maximizes the surplus of the insurance company at time T. The purpose of this paper is to give an explicit expression for the optimal reinsurance and investment strategy which maximizes the expected exponential utility of the final value of the surplus at the end of T-th year. To do this we solve the corresponding Hamilton-Jacobi-Bellman equation.
